1Phase II clinical trial using camrelizumab combined with apatinib and chemotherapy as the first-line treatment of advanced esophageal squamous cell carcinoma显示文摘Background:Effective therapeutic options are limited for patients with advanced esophageal squamous cell carcinoma(ESCC).The incorporation of an immune checkpoint inhibitor and a molecular anti-angiogenic agent into the commonly adopted chemotherapy may produce synergistic effects.Therefore,we aimed to investigate the efficacy and safety of camrelizumab plus apatinib combined with chemotherapy as the first-line treatment of advanced ESCC.Methods:In this single-arm prospective phase II trial,patients with unresectable locally advanced or recurrent/metastatic ESCC received camrelizumab 200 mg,liposomal paclitaxel 150 mg/m2,and nedaplatin 50 mg/m2 on day 1,and apatinib 250 mg on days 1-14.The treatments were repeated every 14 days for up to 9 cycles,followed by maintenance therapy with camrelizumab and apatinib.The primary endpoint was objective response rate(ORR)according to the Response Evaluation Criteria in Solid Tumors(version 1.1).Secondary endpoints included disease control rate(DCR),progression-free survival(PFS),overall survival(OS),and safety.Results:We enrolled 30 patients between August 7,2018 and February 23,2019.The median follow-up was 24.98 months(95%confidence interval[CI]:23.05-26.16 months).The centrally assessed ORR was 80.0%(95%CI:61.4%-92.3%),with a median duration of response of 9.77 months(range:1.54 to 24.82+months).The DCR reached 96.7%(95%CI:82.8%-99.9%).The median PFS was 6.85 months(95%CI:4.46-14.20 months),and the median OS was 19.43 months(95%CI:9.93 months–not reached).The most common grade 3-4 treatmentrelated adverse events(AEs)were leukopenia(83.3%),neutropenia(60.0%),and increased aspartate aminotransferase level(26.7%).Treatment-related serious AEs included febrile neutropenia,leukopenia,and anorexia in one patient(3.3%),and single cases of increased blood bilirubin level(3.3%)and toxic epidermal necrolysis(3.3%).No treatment-related deaths occurred.Conclusions:Camrelizumab plus apatinib combined with liposomal paclitaxel and nedaplatin as first-line treatment demonstrated feasible anti-tumor activity and manageable safety in patients with advanced ESCC.Randomized trials to evaluate this new combination strategy are warranted.Trial registration:This trial was registered on July 27,2018,at ClinicalTrials.gov(identifier:NCT03603756).Bo Zhang Ling Qi Xi Wang Jianping Xu Yun Liu Lan Mu Xingyuan Wang Lidan Bai Jing Huang 2020Cancer Communications2020,40,12:38
2Overexpression of p27^(KIP1)induced cell cycle arrest in G_1 phase and subsequent apoptosis in HCC-9204 cell line显示文摘AIM We have previously reported that inducibleover-expression of Bak may prolong cell cycle inG1 phase and lead to apoptosis in HCC-9204 cells.This study is to investigate whether p27KIP1playsan important role in this process.METHODS In order to elucidate the exactfunction of p27KIP1in this process,a zinc induciblep27KIP1stable transfectant and transient p27KIP1-GFP fusion transfectant were constructed.Theeffects of inducible,p27KIP1on cell growth,cellcycle arrest and apoptosis were examined in themock,control pMD vector,and pMD-KIP1transfected HCC-9204 cells.RESULTS This p27KIP1-GFP transfectant maytransiently express the fusion gene.The cellgrowth was reduced by 35% at 48 h of p27KIP1induction with zinc treatment as determined bytrypan blue exclusion assay.These differencesremained the same after 72 h of p27KIP1expression,p27KIP1caused cell cycle arrest after24 h of induction,with 40% increase in G1population.Prolonged p27KIP1expression in thiscell line induced apoptotic cell death reflected byTUNEL assay.Fourty-eight h and 72 h of p27KIP1expression showed a characteristic DNA ladder onagarose gel electrophoresis. 3PROPULSIVE PERFORMANCE AND VORTEX SHEDDING OF A FOIL IN FLAPPING MOTION显示文摘The propulsive performance and vortex shedding of oscillating foil, which mimics biological locomotion, were numerically investigated. The objectives of this study were to deal with unsteady force, in particular thrust force, exerted on the foil in pitching and plunging motions, and to explore the relation of the propulsive performance with vortex structures near the foil and vortex shedding in the near wake. The two dimensional incompressible Navier Stokes equations in the vorticity and stream function formulation were solved by fourth order essentially compact finite difference schemes for the space derivatives and a fourth order Runge Kutta scheme for the time advancement. To reveal the mechanism of the propulsive performance, the unsteady force and the shedding of the trailing and leading edge vortices of the foil were analyzed. 4Protective effects of Lingguizhugan decoction on amyloid-beta peptide (25-35)-induced cell injury Anti-inflammatory effects显示文摘In the present study, a human neuroblastoma cell line (SH-SY5Y) and BV-2 microglia were treated with amyloid-β peptide (25-35) , as a model of Alzheimer's disease, to evaluate the protective effects of 10-3-10-8 g/mL Lingguizhugan decoction and to examine the underlying anti-inflammatory mechanism. Lingguizhugan decoction significantly enhanced the viability of SH-SY5Y cells with amyloid-β peptide-induced injury, and lowered levels of interleukin-1β, interleukin-6, tumor necrosis factor-α and nitric oxide in the culture supernatant of activated BV-2 microglia. The effects of 10-3 g/mL Lingguizhugan decoction were more significant. 5Distribution of nitric oxide synthase in stomach myenteric plexus of rats显示文摘AIM: To study the distribution of nitric oxide synthase (NOS) in rat stomach myenteric plexus.METHODS: The distribution of NOS in gastric wall was studied in quantity and location by the NADPH-diaphorase (NDP) histochemical staining method and whole mount preparation technique.RESULTS: NOS was distributed in whole stomach wall, most of them were located in myenteric plexus, and distributed in submucosal plexus. The shape of NOS positive neurons was basically similar, most of them being round and oval in shape. But their density, size and staining intensity varied greatly in the different parts of stomach. The density was 62 -± 38 cells/mm2(antrum), 43 ± 32 cells/mm2(body), and 32 ± 28 cells/mm2 (fundus), respectively. The size and staining intensity of NOS positive neurons in the fundus were basically the same, the neurons being large and dark stained, while they were obviously different in antrum. In the body of the stomach, the NOS positive neurons were in an intermediate state from fundus to antrum. 6Senescence-like changes induced by expression of p21^(Waf1/Cip1) in NIH3T3 cell line显示文摘P21Waf1/Cip1 is a potent cyclin-dependent kinase inhibitor. As a downstream mediator of p53, p21Waf1/cip1involves in cell cycle arrest, differentiation and apoptosis. Previous studies in human cells provided evidencefor a link between p21Waf1/cip1 and cellular senescence. While in murine cells, the role of p21Waf1/Cip1is indefinite. We explored this issue using NIH3T3 cells with inducible p21Waf1/cip1 expression. Induc-tion of p21Waf1/Cip1 triggered G1 growth arrest, and NIH3T3-p21 cells exhibited morphologic features,such as enlarged and flattened cellular shape, specific to the senescence phenotype. We also showed thatp21Waf1/Cip1-transduced NIH3T3 cells expressedβ-galactosidase activity at pH 6.0, which is known to bea marker of senescence. 7Inconsistent seasonal variation of antibiotics between surface water and groundwater in the Jianghan Plain:Risks and linkage to land uses显示文摘Antibiotics are widely used in humans and animals,but their transformation from surface water to groundwater and the impact of land uses on them remain unclear.In this study,14 antibioticswere systematically surveyed in a complex agricultural area in Central China.Results indicated that the selected antibiotic concentrations in surface waters were higher in winter(average:32.7 ng/L)than in summer(average:17.9 ng/L),while the seasonal variation in groundwaters showed an opposite trend(2.2 ng/L in dry winter vs.8.0 ng/L in summer).Macrolides were the predominant antibiotics in this area,with a detected frequency of over 90%.A significant correlation between surface water and groundwater antibiotics was only observed in winter(R^(2)=0.58).This study further confirmed the impact of land uses on these contaminants,with optimal buffer radii of 2500 m in winter and 500 m in summer.Risk assessment indicated that clarithromycin posed high risks in this area.Overall,this study identified the spatiotemporal variability of antibiotics in a typical agricultural area in Central China and revealed the impact of land uses on antibiotic pollution in aquatic environments.Xi Liu Zhi Wang Lu Zhang Weiying Fan Chao Yang Enhua Li Yun Du Xuelei Wang 2021Journal of Environmental Sciences2021,33,11:9
8Study of Static Adsorption Capacity of ACF for Xenon at 201 K显示文摘The static adsorption performances of a series of active carbon fiber(ACF)for xenon at 201 K were measured with a model ASAP2010M specific surface area and aperture distribution instrument by changing the working gas of instrument from N 2 to Xenon. 9The 1470 nm diode laser with an intralesional fiber device:a proposed solution for the treatment of inflamed and infected keloids显示文摘Background:Keloids are the result of abnormal wound healing and often are subject to infections and recurrent inflammation.We present a study conducted with a 1470 nm diode laser using an intralesional optical fiber device for the treatment of inflamed keloid scars.We evaluate its efficacy as a novel alternative method to decrease keloid infection and inflammation.Methods:The patients who underwent 1470 nm laser treatment from February 2016 to February 2018 at the plastic and reconstructive surgery department of the Shanghai Ninth People’s Hospital Affiliated to Shanghai Jiao Tong University with keloid accompanying serious local infection and fester were included.Patients took curative effect evaluation before and 1 year after the treatment.The test items included infection frequency in each year;pain,by visual analogue scale(VAS);itch,using VAS;quality of life(QOL),using QOL scale;and blood supply,using PeriCam PSI.Results:A total of 19 patients(mean age 35.21 years,range 11–66)with history of inflamed keloids with episodes of infection or abscess were enrolled.Patients underwent to a 1470 nm laser therapy for average of 1.16 times.After treatment,infection frequency and blood supply in keloids were reduced(p<0.001).Pain,itching,and QOL were improved(p<0.001).Conclusion:The present study shows that 1470 nm fiber laser treatment could improve inflamed keloids fairly well by decreasing inflammation,and a relative stabilization of collagen composition.Therefore,it is an effective minimally invasive scar therapy,but further studies are essential to confirm the present results.Ke Li Fabio Nicoli Wen Jing Xi Zheng Zhang Chunxiao Cui Ahmed Al-Mousawi Alberto Balzani Yun Tong Yixin Zhang 2019Burns & Trauma2019,7,1:8

12The effects of claudin 14 during early Wallerian degeneration after sciatic nerve injury显示文摘Claudin 14 has been shown to promote nerve repair and regeneration in the early stages of Wallerian degeneration(0–4 days) in rats with sciatic nerve injury, but the mechanism underlying this process remains poorly understood. This study reported the effects of claudin 14 on nerve degeneration and regeneration during early Wallerian degeneration. Claudin 14 expression was up-regulated in sciatic nerve 4 days after Wallerian degeneration. The altered expression of claudin 14 in Schwann cells resulted in expression changes of cytokines in vitro. Expression of claudin 14 affected c-Jun, but not Akt and ERK1/2 pathways. Further studies revealed that enhanced expression of claudin 14 could promote Schwann cell proliferation and migration. Silencing of claudin 14 expression resulted in Schwann cell apoptosis and reduction in Schwann cell proliferation. 13Metabolic Disorders Increase the Risk to Incident Cardiovascular Disease in Middle-aged and Elderly Chinese显示文摘Objective The association of metabolic syndrome (MetS) with cardiovascular diseases (CVD) has not been adequately explored in middle-aged and elderly Chinese.This study aimed to investigate MetS' prevalence and its impact on the CVD incidence in this specific population group.Methods A data set of a community-based prospective cohort study was analyzed.A total of 2300 subjects aged 40-94 years were followed up for the CVD events.MetS defined according to the JCDCG criteria was assessed at baseline,and the middle-aged and elderly groups were classified by the WHO definition.Results As compared with the middle-aged group,the prevalence of MetS increased by 0.6 times (34.6% vs.21.3%) and the incidence density of CVD increased by 4.9 times in the elderly group (52.3/1000 person-year vs.8.9/1000 person-year).Furthermore,the multivariate Cox regression revealed that the risk to CVD incidence was independently related to increased waist circumference in the middle-aged group (HR=2.23,P<0.01) and to elevated blood glucose in the elderly group (HR=1.39,P<0.01).Conclusion MetS was highly prevalent in middle-aged and elderly Chinese.MetS significantly increased the risk to CVD incidence in the elderly.All individuals with metabolic disorders should receive active clinical care to reduce the incidence of CVD.ZHANG Ming Liang HOU Xu Hong ZHU Yun Xia LU Jun Xi PENG Liang Pu GU Hui Lin JIA Wei Ping 2012Biomedical and Environmental Sciences2012,25,1:7
14Berbamine inhibits proliferation and induces apoptosis of KU812 cells by increasing Smad3 activity显示文摘Objective: The cytotoxic effect of berbamine on chronic myeloid leukemia (CML) cell line KU812 was evaluated,and the mechanisms of its action were explored.Methods: The effect of berbamine on the KU812 cell growth was determined by methyl thiazolyl tetrazolium (MTT) assay.Flow cytometry was used to profile cell cycle alteration upon berbamine treatment.Reverse transcription polymerase chain reaction (RT-PCR) was carried out to determine the transcripts of transforming growth factor-β (TGF-β) receptors (TβRs),Smad3,c-Myc,cyclin D1,p21Cip1(p21),and p27Kip1(p27).Changes in the protein levels of total Smad3,phosphorylated Smad3,the downstream targets of Smad3,and specific apoptosis-related factors were evaluated by Western blotting.Results: Berbamine inhibited KU812 cell proliferation in a doseand time-dependent manner,and the half maximal inhibitory concentration (IC50) values for treatments of 24,48,and 72 h were 5.83,3.43,and 0.75 μg/ml,respectively.Berbamine induced G1 arrest as well as apoptosis in KU812 cells.Transcriptions of Smad3 and p21 were up-regulated,while those of TβRI,TβRII,c-Myc,cyclin D1 and p27 were not changed significantly.The protein levels of both total Smad3 and phosphorylated Smad3 were both up-regulated after berbamine treatment,together with decreased c-Myc and cyclin D1 and increased p21.Meanwhile,the levels of the anti-apoptotic proteins,such as Bcl-2 and Bcl-xL,were decreased,whereas pro-apoptotic Bax was increased.Conclusions: Berbamine suppresses KU812 cell proliferation through induction of cell cycle arrest in G1 and apoptosis.It activates Smad3 without additional stimulation of TGF-β,and alters the levels of the Smad3 downstream targets,including c-Myc,cyclin D1 and p21.Our findings suggest that berbamine is a promising drug in the treatment of advanced stage patients with CML.Yun LIANG Xi QIU Rong-zhen XU Xiao-ying ZHAO 2011Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2011,12,7:7
15Endocrine Disruption Activity of 30-day Dietary Exposure to Decabromodiphenyl Ethane in Balb/C Mouse显示文摘Objective This study aimed to evaluate the hepatotoxicity,metabolic disturbance activity and endocrine disrupting activity of mice treated by Decabromodiphenyl ethane(DBDPE).Methods In this study,Balb/C mice were treated orally by gavage with various doses of DBDPE.After 30 days of treatment,mice were sacrificed; blood,livers and thyroid glands were obtained,and hepatic microsomes were isolated.Biochemical parameters including 8 clinical chemistry parameters,blood glucose and hormone levels including insulin and thyroid hormone were assayed.The effects of DBDPE on hepatic cytochrome P450(CYP) levels and activities and uridinediphosphate-glucuronosyltransferase(UDPGT) activities were investigated.Liver and thyroid glands were observed.Results There were no obvious signs of toxicity and no significant treatment effect on body weight,or liver-to-body weight ratios between treatment groups.The levels of ALT and AST of higher dose treatment groups were markedly increased.Blood glucose levels of treatment groups were higher than those of control group.There was also an induction in TSH,T3,and f T3.UDPGT,PROD,and EROD activities were found to have been increased significantly in the high dose group.Histopathologic liver changes were characterized by hepatocyte hypertrophy and cytoplasmic vacuolization.Our findings suggest that DBDPE can cause a certain degree of mouse liver damage and insufficiency.Conclusion DBDPE has the activity of endocrine disruptors in Bal/C mice,which may induce drug-metabolizing enzymes including CYPs and UDPGT,and interfere with thyroid hormone levels mediated by Ah R and CAR signaling pathways.Endocrine disrupting activity of DBDPE could also affect the glucose metabolism homeostasis.SUN Ru Bao SHANG Shuai ZHANG Wei LIN Ben Cheng WANG Qiang SHI Yun XI Zhu Ge 2018Biomedical and Environmental Sciences2018,31,1:7

17Two Cases of Multi-antibiotic Resistant Cronobacter spp. Infections of Infants in China*显示文摘Infections by Cronobacter spp. are hazardous to infants since they can lead to neonatal meningitis, bacteremia, and necrotizing enterocolitis. Cronobacter spp. are frequently resistant to β-lactam derivatives, macrolides, and aminoglycosides. In addition, multi-resistant strains have also been detected. In China, the isolation rate of Cronobacter spp. from commercial powdered infant formula(PIF) or follow-up formula(FUF) is relatively high. Nevertheless, clinical cases of Cronobacter infection have been ignored to date. Here we describe two cases of Cronobacter infection detected at the Wuhan Women and Children Medical Care Center Hospital(Wuhan City, China). We provide the genomic analysis of the isolates and the antibiotic-resistance profiles of the two strains. The Cronobacter strains identified in this study were not susceptible to third-generation cephalosporins, aminoglycoside, and/or trimethoprim-sulfamethoxazole. Whole genome sequencing revealed various genes known to encode antibiotic resistance. Future studies are needed to determine whether the genes predicted in this study are functional. 19Novel constant-cladding tapered-core ytterbium-doped fiber for high-power fiber laser oscillator显示文摘Power scaling based on traditional ytterbium-doped fibers(YDFs)is limited by optical nonlinear effects and transverse mode instability(TMI)in high-power fiber lasers.Here,we propose a novel long tapered fiber with a constant cladding and tapered core(CCTC)along its axis direction.The tapered-core region of the fiber is designed to enhance the stimulated Raman scattering(SRS)threshold and suppress higher-order mode resonance in the laser cavity.The CCTC YDF was fabricated successfully with a modified chemical vapor deposition(MCVD)method combined with solution doping technology,which has a cladding diameter of 400µm and a varying core with a diameter of~24μm at both ends and~31μm in the middle.To test the performance of the CCTC fiber during high-power operation,an all-fiber laser oscillator based on a CCTC YDF was investigated experimentally.As a result,a maximum output power of 3.42 kW was achieved with an optical-to-optical efficiency of 55.2%,although the TMI effect was observed at an output power of~3.12 kW.The measured beam quality(M^(2)factor)was~1.7,and no sign of the Raman component was observed in the spectrum.We believe that CCTC YDF has great potential to simultaneously mitigate the SRS and TMI effects,and further power scaling is promising by optimizing the structure of the YDF.Yun Ye Xianfeng Lin Xiaoming Xi Chen Shi Baolai Yang Hanwei Zhang Xiaolin Wang Jinyan Li Xiaojun Xu 2021High Power Laser Science and Engineering2021,9,2:5
20Cells in G_2/M phase increased in human nasopharyngeal carcinoma cell line by EBV-LMP1 through activation of NF-кB and AP-1显示文摘Although previous studies showed that the principal oncoprotein encoded by Epstein-Barr virus, latent membrane protein 1 (LMP1), could induce the nasopharyngeal carcinoma cells in G2/M phase increased, littleis known about the target molecules and mechanisms. The present study demonstrated that LMP1 couldinduce the accumulation of p53 protein and upregulate its transactivity in a dose dependent manner, whichresulted in the decrease of the kinase activity of cdc2/cyclin B complex and inducing arrest at G2/M phasethrough the activation of NF-kB and AP-1 signaling pathways, and the effect of NF-kB was more obviousthan that of AP-1.
