| 1 | Blossom of CRISPR technologies and applications in disease treatment显示文摘Since 2013,the CRISPR-based bacterial antiviral defense systems have revolutionized the genome editing field.In addition to genome editing,CRISPR has been developed as a variety of tools for gene expression regulations,live cell chromatin imaging,base editing,epigenome editing,and nucleic acid detection.Moreover,in the context of further boosting the usability and feasibility of CRISPR systems,novel CRISPR systems and engineered CRISPR protein mutants have been explored and studied actively.With the flourish of CRISPR technologies,they have been applied in disease treatment recently,as in gene therapy,cell therapy,immunotherapy,and antimicrobial therapy.Here we present the developments of CRISPR technologies and describe the applications of these CRISPR-based technologies in disease treatment. | Huayi Liu Lian Wang Yunzi Luo | 2018 | Synthetic and Systems Biotechnology2018,3,4: | 2 |

| 9 | A method for Absolute Protein Expression Quantity Measurement Employing Insulator RiboJ显示文摘Measuring the absolute protein expression quantity for a specific promoter is necessary in the fields of both molecular biology and synthetic biology.The strength of a promoter is traditionally characterized by measuring the fluorescent intensity of the fluorescent protein downstream of the promoter.Until now,measurement of the absolute protein expression quantity for a promoter,however,has been unsuccessful in synthetic biology.The fact that the protein coding sequence influences the expression level for different proteins,and the inconvenience of measuring the absolute protein expression level,present a challenge to absolute quantitative measurement.Here,we introduce a new method that combines the insulator RiboJ with the standard fluorescence curve in order to measure the absolute protein expression quantity quickly;this method has been validated by modeling verification.Using this method,we successfully measured nine constitutive promoters in the Anderson promoter family.Our method provides data with higher accuracy for pathway design and is a straightforward way to standardize the strength of different promoters. | Hongbin Yu Zheng Wang Hanyue Xu Jiusi Guo Qingge Ma Xiangxu Mu Yunzi Luo | 2018 | Engineering2018,4,6: | 1 |

| 11 | Hypoxia induces mitochondrial protein lactylation to limit oxidative phosphorylation显示文摘Oxidative phosphorylation(OXPHOS)consumes oxygen to produce ATP.However,the mechanism that balances OXPHOS activity and intracellular oxygen availability remains elusive.Here,we report that mitochondrial protein lactylation is induced by intracellular hypoxia to constrain OXPHOS.We show that mitochondrial alanyl-tRNA synthetase(AARS2)is a protein lysine lactyltransferase,whose proteasomal degradation is enhanced by proline 377 hydroxylation catalyzed by the oxygen-sensing hydroxylase PHD2.Hypoxia induces AARS2 accumulation to lactylate PDHA1 lysine 336 in the pyruvate dehydrogenase complex and carnitine palmitoyltransferase 2(CPT2)lysine 457/8,inactivating both enzymes and inhibiting OXPHOS by limiting acetyl-CoA influx from pyruvate and fatty acid oxidation,respectively.PDHA1 and CPT2 lactylation can be reversed by SIRT3 to activate OXPHOS.In mouse muscle cells,lactylation is induced by lactate oxidation-induced intracellular hypoxia during exercise to constrain high-intensity endurance running exhaustion time,which can be increased or decreased by decreasing or increasing lactylation levels,respectively.Our results reveal that mitochondrial protein lactylation integrates intracellular hypoxia and lactate signals to regulate OXPHOS. | Yunzi Mao Jiaojiao Zhang Qian Zhou Xiadi He Zhifang Zheng Yun Wei Kaiqiang Zhou Yan Lin Haowen Yu Haihui Zhang Yineng Zhou Pengcheng Lin Baixing Wu Yiyuan Yuan Jianyuan Zhao Wei Xu Shimin Zhao | 2024 | Cell Research2024,34,1: | 0 |
| 12 | Peptide WCPFSRSF ameliorates excitotoxicity and elevates synaptic plasticity in glutamate-damaged SH-SY5Y cells by modulating the PI3K/mTOR/EIF4E and BDNF/CREB/TrkB pathways显示文摘The purpose of this study was to illustrate the beneficial effects and underlying mechanisms of peptide Trp-Cys-Pro-Phe-Ser-Arg-Ser-Phe (WCPFSRSF) against excitotoxicity by transcriptome analysis combined with western blot in glutamate-treated neuronal cells.Our results demonstrated that WCPFSRSF restored the cell membrane damage induced by glutamate and promoted cell survival in SH-SY5Y cells.The RNA-sequencing analysis illustrated 376 genes (98 upregulated and 278 downregulated) were differentially expressed between the glutamate- and WCPFSRSF-treated cells.Furthermore,RNA-sequencing screening and further western blot results showed that WCPFSRSF regulated cAMP-related signaling and synapse-related function,which further promoted the translation and protein synthesis,followed by cell survival,neurotrophic pathway and long-term memory.The potential mechanisms of WCPFSRSF were related to the Akt/mTOR/EIF4E signaling pathway and CREB/TrkB/BDNF pathway.Taken together,our study demonstrated that WCPFSRSF could be used as a regulator for synaptic plasticity and neurotrophic-related pathway,which provided evidence that WCPFSRSF is a promising therapeutic strategy for neuroprotection and memory enhancement. | Qi Zhang Lin Zheng Guowan Su Donghui Luo Mingtao Huang Yunzi Feng Mouming Zhao | 2022 | Food Bioscience2022,47,3: | 0 |
