1Laser performance of the SG-Ⅲ laser facility显示文摘SG-Ⅲ laser facility is now the largest laser driver for inertial confinement fusion research in China. 3Identification and characterization of rice blast resistance gene Pid4 by a combination of transcriptomic profiling and genome analysis显示文摘Map-based cloning of plant disease resistance (R) genes is time-consuming. Here, we reported the isolation of blast R gene Pid4 using comparative transcriptomic profiling and genome-wide sequence analysis. Pid4 encodes a coiled-coil nucleotide-binding site leucine-rich repeat(CC-NBS-LRR) protein and is constitutively expressed at diverse developmental stages in the rice variety Digu. The Pid4 protein is localized in both the nucleus and cytoplasm. Introduction of Pid4 into susceptible rice cultivars confers race-specific resistance to leaf and neck blast. Amino acid sequence comparison and blast resistance spectrum tests showed that Pid4 is a novel R gene, different from the previously reported R genes located in the same gene cluster. A Pid4 Indel marker was developed to facilitate the identification of Pid4 in different rice varieties. 5A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 7Microstructural evolution of Au-Sn solder prepared by laminate rolling during annealing process显示文摘The microstructural evolution and interfacial reaction of the Au/Sn/Au/Sn/Au/Sn/Au couples were investigated during annealing at 453,523,and 543 K for up to 240 h.The Au/Sn combination formed a rapid diffusion system.Even in rolled Au-Sn solder,three phases,such as AuSn,AuSn2,and AuSn4,were formed.After initial annealing at 453 K,the diffusion layers of AuSn,AuSn2,and AuSn4,which were formed after rolling,expanded gradually and then fully transformed into ζ phase(containing Sn from 10% to 18.5%,mole fraction) and δ(AuSn) phase.As a whole,the microstructure of the couple was stable during annealing at 453 K.The solid-state interfacial reaction was much faster at 523 K than at 453 K.After annealing at 523 K for 6 h,the AuSn,AuSn2,and AuSn4 were fully transformed into the ζ phase and δ phase(AuSn) .In spite of the prolonged annealing time for up to 240 h,no significant change of the interfacial microstructure occurred,and the microstructure of the couple was stable during annealing at 523 K.When annealing at 543 K,however,the interfacial of Au/Sn was transformed into solid-liquid state,and the whole couple formed a eutectic structure rapidly,causing the solder to be brittle.The study results clearly demonstrate that the service temperature of the Au-Sn solder should be lower than 543 K.WEI Xiaofeng WANG Richu FENG Yan ZHU Xuewei PENG Chaoqun 2011Rare Metals2011,30,6:5
8Differential expression analysis and regulatory network reconstruction for genes associated with muscle growth and adipose deposition in obese and lean pigs显示文摘During the growth and development of skeletal muscle cells and adipose cells, the regulatory mechanism of micro-effect polygenes determines porcine meat quality, carcass characteristics and other relative quantitative traits. Obese and lean type pig breeds show obvi- ous differences in muscle growth and adipose deposition; however, the molecular mechanism underlying this phenotypic variation remains unknown. We used pathway-focused oligo microarray studies to examine the expression changes of 140 genes associated with muscle growth and adipose deposition in longissimus dorsi muscle at six growth stages (birth, 1, 2, 3, 4 and 5 months) of Landrace (a leaner, Western breed) and Taihu pigs (a fatty, indigenous, Chinese breed). Variance analysis (ANOVA) revealed that differences in the expression of 18 genes in Landrace pigs and three genes in Taihu pigs were very significant (FDR-adjusted permutation, P < 0.01) and di?erences for 22 genes in Landrace pigs and seven genes in Taihu pigs were significant (FDR-adjusted permutation, P < 0.05) among six growth stages. Clustering analysis revealed a high level of signi?cance (FDR-adjusted, P < 0.01) for four gene expression patterns, in which genes that strongly up-regulated were mainly associated with the positive regulation of myofiber formation and fatty acid biogenesis and genes that strongly down-regulated were mainly associated with the inhibition of cell proliferation and positive regulation of fatty acid b-oxidation. Based on a dynamic Bayesian network (DBN) model, gene regulatory networks (GRNs) were reconstructed from time-series data for each pig breed. These two GRNs initially revealed the distinct differences in physiological and biochemical aspects of muscle growth and adipose deposition between the two pig breeds; from these results, some potential key genes could be identified. Quantitative real-time RT-PCR (QRT-PCR) was used to verify the microarray data for five modulated genes, and a good correlation between the two measures of expression was observed for both two pig breeds at different growth stages (r = 0.876 ± 0.095). 9Fine mapping of the rice bacterial blight resistance gene Xa-4 and its co-segregation marker显示文摘An F2 population developed from the Xa-4 near isogenic lines, IR24 and IRBB4, was used for fine mapping of the rice bacterial blight resistance gene, Xa-4. Some restriction fragment length polymorphism (RFLP) markers on the high-density map constructed by Harushima et al. and the amplified DMA fragments homologous to the conserved domains of plant disease resistance (R) genes were used to construct the genetic linkage map around the gene Xa-4 by scoring susceptible individuals in the population. Xa-4 was mapped between the RFLP marker G181 and the polymerase chain reaction (PCR) marker M55. The R gene homologous fragment marker RS13 was found co-segregating with Xa-4 by analyzing all the plants in the population. 10Alterations in cecal microbiota of Jinhua piglets fostered by a Yorkshire sow显示文摘Gut microbiota play important roles in host health and diseases.Several factors,in particular antibiotics,affect the gut microbiota of pigs.Cross-fostering has been applied as a regular practice to equalize litter size,reduce pre-weaning mortality and increase body weight.However,the effect of cross-fostering on cecal microbiota is unclear.In this study,we fostered three Jinhua pigs to a Yorkshire sow.The fostered Jinhua piglets grew significantly faster than their biological siblings.To explore whether the cecal microbiota of piglets will alter during fostering,we characterized the cecal microbiota of allpiglets by examining the V3 hypervariable region of 16S rDNA.We observed altered cecal microbiota in these piglets using the Illumina HiSeq 2000 platform,and this was accompanied with an increase in growth rate after fostering.The relative abundance of Bacteroidetes in the fostered Jinhua piglets was decreased compared with their biological siblings,although still higher in comparison with their new littermates.Beta-diversity analysis also showed that the cecal microbiota of the adopted Jinhua piglets differed from their biological siblings with a shift toward their step-siblings.Our data show that cecal microbiota of piglets were altered after cross-fostering while the growth rate increased.Lingjin Xian Ying Li Zhi Jiang Jideng Ma Long Jin Lei Chen Chaowei Zhou Jie Zhang Yingkai Liu Li Zhu An-an Jiang Guoqing Tang Jian Xiao Jinyong Wang Bo Zeng Mingzhou Li Xuewei Li 2014Chinese Science Bulletin2014,59,32:4
11Early assessment of the safety and immunogenicity of a third dose(booster)of COVID-19 immunization in Chinese adults显示文摘Inducing durable and effective immunity against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)via vaccination is essential to combat the current pandemic of coronavirus disease 2019(COVID-19).It has been noticed that the strength of anti-COVID-19 vaccination-induced immunity fades over time,which calls for an additional vaccination regime,as known as booster immunization,to restore immunity among previously vaccinated populations.Here we report a pilot open-label trial of a third dose of BBIBP-CorV,an inactivated SARS-CoV-2 vaccine(Vero cell),on 136 participants aged between 18 to 63 years.Safety and immunogenicity in terms of neutralizing antibody titers and cytokine/chemokine responses were analyzed as the main endpoint until day 28.While systemic reactogenicity was either absent or mild,SARS-CoV-2-specific neutralizing antibody titers rapidly arose in all participants within 4 weeks,surpassing the peak antibody titers elicited by the initial two-dose immunization regime.Broad increases of cellular immunity-associated cytokines and chemokines were also detected in the majority of participants after the third vaccination.Furthermore,in an exploratory study,a newly developed recombinant protein vaccine,NVSI-06-08(CHO Cells),was found to be safe and even more effective than BBIBP-CorV in eliciting humoral immune responses in BBIBP-CorV-primed individuals.Together,these results indicate that a third immunization schedule with either homologous or heterologous vaccine showed favorable safety profiles and restored potent SARS-CoV-2-specific immunity,providing support for further trials of booster vaccination in larger populations.Yuntao Zhang Yunkai Yang Niu Qiao Xuewei Wang Ling Ding Xiujuan Zhu Yu Liang Zibo Han Feng Liu Xinxin Zhang Xiaoming Yang 2022Frontiers of Medicine2022,16,1:3

14Self-healing anti-icing coatings prepared from PDMS polyurea显示文摘With the help of double hydrogen bonds between urea groups, a self-healing anti-icing coating was prepared from polydimethylsiloxane(PDMS)-based polyurea(PDMSPU) that was synthesized from bis(3-aminopropyl)-terminated PDMS and2,4-toluene diisocyanate. Furthermore, by incorporating a lubricant of inert silicone oil and the cross-linking agent of 1,3,5-tris-(4-aminophenoxy)benzene(TAPOB), the anti-icing coatings exhibited elastic, slippery and durable properties. The wettability,ice shear strength, abrasion-resistance, mechanical and self-healing properties of the prepared anti-icing coatings were systematically investigated. Results showed that the ice shear strength of the PDMSPU anti-icing coating with 150 wt% silicone oil was 7.9±2.9 kPa, significantly lower than that of pristine PDMSPU(74.7±7.2 kPa). The additional introduction of 2 wt%TAPOB could effectively counteract the adverse effect of the lubricant so that the tensile strength(~1 MPa) of the coating could be comparable to pristine PDMSPU(855±96 kPa). The PDMSPU coating containing 150 wt% silicone oil and 2 wt% TAPOB could maintain the ice shear strength below 50 kPa against 10 icing/deicing cycles and at 12.0±3.3 kPa after 5000 abrasion cycles, and exhibit self-healing properties after treating at 60°C for 10 or 15 h.
