2Improvement in affinity and thermostability of a fully human antibody against interleukin-17A by yeast-display technology and CDR grafting显示文摘Monoclonal antibodies(m Abs) are widely used in many fields due to their high specificity and ability to recognize a broad range of antigens. IL-17 A can induce a rapid inflammatory response both alone and synergistically with other proinflammatory cytokines. Accumulating evidence suggests that therapeutic intervention of IL-17 A signaling offers an attractive treatment option for autoimmune diseases and cancer. Here, we present a combinatorial approach for optimizing the affinity and thermostability of a novel anti-h IL-17 A antibody. From a large na?ve phage-displayed library, we isolated the anti-IL-17 A m Ab 7 H9 that can neutralize the effects of recombinant human IL-17 A. However, the modest neutralization potency and poor thermostability limit its therapeutic applications. In vitro affinity optimization was then used to generate 8 D3 by using yeast-displayed random mutagenesis libraries.This resulted in four key amino acid changes and provided an approximately 15-fold potency increase in a cell-based neutralization assay. Complementarity-determining regions(CDRs) of 8 D3 were further grafted onto the stable framework of the hu Fv 4 D5 to improve thermostability. The resulting hybrid antibody9 NT/S has superior stabilization and affinities beyond its original antibody. Human fibrosarcoma cellbased assays and in vivo analyses in mice indicated that the anti-IL-17 A antibody 9 NT/S efficiently inhibited the secretion of IL-17 A-induced proinflammatory cytokines. 11Programmable complex pumping field induced color-on-demand random lasing in fiber-integrated microbelts for speckle free imaging显示文摘Miniaturized lasers with on-demand color emission have immense potential in the development of integrated imaging sources. Random lasers(RLs) with cavity-free structures and low spatial coherence are promising candidates for multicolor imaging sources. However, the wide tuning range and high tuning accuracy of continuous and reversible random lasing color with a defined emissive direction remain challenging. Here, random lasing with on-demand color emission and defined emissive direction is realized in fiber-integrated microbelts through programmable pumping. Individual RL microbelts as RL units doped with blue(B)-, green(G)-, and red(R)-emissive dyes and titanium dioxide nanoparticles are assembled on an optical fiber, resulting in high-performance B/G/R lasing. The optical fiber functions as a waveguide that guides the multicolor RL to ensure a defined directional emission. By manipulating the combination of the B/G/R RL units on the waveguide, distinctive lasing colors covering the entire visible spectrum are obtained, including white-colored random lasing. To realize dynamically controllable lasing for on-demand color emission, a pumping strategy with programmable excitation is proposed by shaping the pump beams into different beam arrays for precisely pumping the RL unit combination. 12The Synthesis of Some 17-(2'-Oxazolyl)-androsta-5,16-diene Deriva-tives as 17α-Hydroxylase/C17,20 -Lyase Inhibitors显示文摘Several 4'- and 5'-substituted 17-(2'-oxazolyl)-androsta-5,16-diene derivatives were designed and synthesized as inhibitors of 17α-hydroxylase/C17,20 -Lyase (P45017α) for the treatment of prostatic cancer, the results of the preliminary pharmacological screening showed that compound 6c, i.e. 17-(2'-oxazoly)-androsta-5,16-diene-3-ol was a strong inhibitor, comparable with that of the reference compound VN-85. The introduction ofmethyl or phenyl group at the 4' or 5' position of oxazole ring decreased the activity. The in vitro activities of 3- acetate 5a-c and 8 were lower than their 3-ol counterparts 6a-c and 9 as expected. 13Pan-cancer analysis of the prognostic and immunological role of nucleophosmin/nucleoplasmin 3(NPM3)and its potential significance in lung adenocarcinoma显示文摘Background Nucleophosmin/nucleoplasmin 3(NPM3),a member of the NPM protein family,is widely expressed in various human tissues.Although previous studies identified elevated NPM3 expression in several cancers,a systematic pan-cancer analysis remains lacking.In this study,we conducted a comprehensive analysis of NPM3 to determine its role in tumorigenesis and tumor development.Methods Using data from The Cancer Genome Atlas(TCGA)and various bioinformatics analysis tools,we conducted a pan-cancer analysis of NPM3.Additionally,we collected gene expression and clinical data from 890 patients with lung adenocarcinoma(LUAD)from TCGA and the Gene Expression Omnibus database.We performed Cox regression analyses to explore the independent prognostic value of NPM3 expression in LUAD and plotted a nomogram to predict patient survival.We also used real-time quantitative polymerase chain reaction(RT-qPCR)to examine the expression levels of NPM3 in seven pairs of LUAD and paraneoplastic tissue samples.Results NPM3 expression was significantly increased in 20 types of cancer and was associated with poor prognosis in five types(P<0.05).NPM3 expression was negatively correlated with DNA methylation and positively correlated with copy number variation.NPM3 was also significantly associated with immune cell infiltration in various cancers.Cox regression analyses revealed that NPM3 expression could serve as an independent prognostic marker of LUAD.Moreover,our nomogram demonstrated good predictive ability for the prognosis of patients with LUAD.Finally,the high expression of NPM3 in LUAD was verified using RT-qPCR.Conclusion NPM3 is a promising biomarker for predicting pan-cancer prognosis and immunotherapeutic efficacy.Qianhui Wei Jing Zhou Xinyue Wang Zhaona Li Xiuqiong Chen Kaidi Chen Richeng Jiang 2023Cancer Pathogenesis and Therapy2023,1,4:0
