1CTLA4 Silencing with siRNA Promotes Deviation of Th1/Th2 in Chronic Hepatitis B Patients显示文摘To determine whether RNA interference (RNAi) could block cytotoxic T-lymphocyte antigen 4 (CTLA4) in human lymphocytes in vitro and promote IFN-γ and IL-2 secretions, three small interfering RNAs (siRNAs) were selected based on target specificity sequences of human CTLA4 and transfected into human lymphocytes of chronic HBV patients. As a result, the expression of human CTLA4 mRNA was efficiently suppressed by all the three siRNAs. Compared with negative control (siRNA-co), siRNA-1 inhibited the expression of CTLA4 most efficiently and was used in the further study. The expressions of IFN-γ and IL-2 were upregulated and the level of IL-4 was almost unchanged in lymphocytes transfected with siRNA-1 compared with the blank control. These results indicated that siRNA-1 led to IFN-γ and IL-2 secretions, which is a main response of Th1/Th2. In a conclusion, RNAi significantly suppressed the expression of human CTLA4 mRNA in human lymphocytes in vitro, and could induce Th1/Th2 response. 9SEL1L preserves CD8^(+) T-cell survival and homeostasis by fine-tuning PERK signaling and the IL-15 receptor-mediated mTORC1 axis显示文摘SEL1L-mediated endoplasmic reticulum-associated degradation(ERAD)plays critical roles in controlling protein homeostasis by degrading misfolded or terminal unfolded proteins.However,it remains unclear how SEL1L regulates peripheral T-cell survival and homeostasis.Herein,we found that SEL1L deficiency led to a greatly reduced frequency and number of mature T cells,which was further validated by adoptive transfer experiments or bone marrow chimera experiments,accompanied by the induction of multiple forms of cell death.Furthermore,SEL1L deficiency selectively disrupted naïve CD8+T-cell homeostasis,as indicated by the severe loss of the naïve T-cell subset but an increase in the memory T-cell subset.We also found that SEL1L deficiency fueled mTORC1/c-MYC activation and induced a metabolic shift,which was largely attributable to enhanced expression of the IL-15 receptorαandβchains.Mechanistically,single-cell transcriptomic profiling and biochemical analyses further revealed that Sel1l−/−CD8+T cells harbored excessive ER stress,particularly aberrant activation of the PERK-ATF4-CHOP-Bim pathway,which was alleviated by supplementing IL-7 or IL-15.Importantly,PERK inhibition greatly resolved the survival defects of Sel1l−/−CD8+T cells.In addition,IRE1αdeficiency decreased mTORC1 signaling in Sel1l−/−naïve CD8+T cells by downregulating the IL-15 receptorαchain.Altogether,these observations suggest that the ERAD adaptor molecule SEL1L acts as an important checkpoint for preserving the survival and homeostasis of peripheral T cells by regulating the PERK signaling cascade and IL-15 receptor-mediated mTORC1 axis.Yafeng Gao Wenhui Li Zhenghao Wang Cangang Zhang Yaping He Xiaowei Liu Kexin Tang Weiguo Zhang Qiaoming Long Yong Liu Jinping Zhang Baojun Zhang Lianjun Zhang 2023Cellular & Molecular Immunology2023,20,10:0
10USP9x promotes CD8^(+)T-cell dysfunction in association with autophagy inhibition in septic liver injury显示文摘Sepsis is a life-threatening condition manifested by concurrent inflammation and immunosuppression.Ubiquitin-specific peptidase 9,X-linked(USP9x),is a USP domain-containing deubiquitinase which is required in T-cell development.In the present study,we investigate whether USP9x plays a role in hepatic CD8^(+)T-cell dysfunction in septic mice.We find that CD8^(+)T cells are decreased in the blood of septic patients with liver injury compared with those without liver injury,the CD4/CD8 ratio is increased,and the levels of cytolytic factors,granzyme B and perforin are downregulated.The number of hepatic CD8^(+)T cells and USP9x expression are both increased 24 h after cecal ligation and puncture-induced sepsis in a mouse model,a pattern similar to liver injury.The mechanism involves promotion of CD8^(+)T-cell dysfunction by USP9x associated with suppression of cell cytolytic activity via autophagy inhibition,which is reversed by the USP9x inhibitor WP1130.In the in vivo studies,autophagy is significantly increased in hepatic CD8^(+)T cells of septic mice with conditional knockout of mammalian target of rapamycin.This study shows that USP9x has the potential to be used as a therapeutic target in septic liver injury.Lulu Sheng Juntao Chen Yiqing Tong Yi Zhang Qiming Feng Zhenghao Tang 2022Acta Biochimica et Biophysica Sinica2022,54,12:0
11Urotensin Ⅱ increases endothelin production by vascular smooth muscle cells in rats显示文摘The aim of this study was to observe the effects of urotensin Ⅱ (UII) on production of endothelin (ET) in rat aortic vascular smooth muscle cells (VSMC). Cultured VSMCs incubated with various concentrations of UII were used to measure the YSMC 3H-TdR incorporation, the amount of ET mRNA and ET production in VSMCs. In this work we found that UII (10-10-10-8 mol/L) promoted VSMC 3H-TdR incorporation (47%-83%, P<0.01) and increased the amount of ET mRNA by 17.1% (P < 0.05) to 112.8% (P < 0.01), respectively, in a concentration dependent manner compared with control. After 4 and 8 h incubation, 10-10-10-8 mol/L of UII elevated the ET synthesis and release in a concentration dependent manner. After 4 h incubation, the content of ET in medium was 4.9, 5.36 and 7.12 pg/mL (P < 0.01). After 8 h incubation, the ET content released from VSMCs was 12.6, 12.07 and 17.17 pg/mL (P < 0.01). 12Mold-free self-assembled scalable microlens arrays with ultrasmooth surface and record-high resolution显示文摘Microlens arrays(MLAs)based on the selective wetting have opened new avenues for developing compact and miniaturized imaging and display techniques with ultrahigh resolution beyond the traditional bulky and volumetric optics.However,the selective wetting lenses explored so far have been constrained by the lack of precisely defined pattern for highly controllable wettability contrast,thus limiting the available droplet curvature and numerical aperture,which is a major challenge towards the practical high-performance MLAs.Here we report a mold-free and self-assembly approach of mass-production of scalable MLAs,which can also have ultrasmooth surface,ultrahigh resolution,and the large tuning range of the curvatures.The selective surface modification based on tunable oxygen plasma can facilitate the precise pattern with adjusted chemical contrast,thus creating large-scale microdroplets array with controlled curvature.The numerical aperture of the MLAs can be up to 0.26 and precisely tuned by adjusting the modification intensity or the droplet dose.The fabricated MLAs have high-quality surface with subnanometer roughness and allow for record-high resolution imaging up to equivalently 10,328 ppi,as we demonstrated.This study shows a cost-effective roadmap for mass-production of high-performance MLAs,which may find applications in the rapid proliferating integral imaging industry and high-resolution display.Zhihao Liu Guangwei Hu Huapeng Ye Miaoyang Wei Zhenghao Guo Kexu Chen Chen Liu Biao Tang Guofu Zhou 2023Light(Science & Applications)2023,12,7:0
13THE ADSORPTION BEHAVIORS OF TOLUENE ON HYPERCROSSLINKED RESINS显示文摘Investigation of the adsorption characteristics of toluene on two hypercrosslinked resins. The static and the kinetic adsorption were studied. The equilibrium adsorption date was fitted to freundlich adsorption isotherm models to evaluate the model parameters. The enthalpy, free energy, entropy are indicative of an exothermic, a spontaneous and disorder decreasing process.
