5Directed Transport of Interacting Particle Systems: Recent Progress显示文摘Recent developments in studies of directed transport processes in interacting particle systems are retrospected. Due to the interactions among elements, the directed transport process exhibits complicated and novel cooperative dynamics. We considered various possibilities in achieving ratchet motion by breaking different symmetries of many-body systems. 10A Stable Upstream Stem-loop Structure Enhances Selection of the First 5'-ORF-AUG as a Main Start Codon for Translation Initiation of Human ACAT1 mRNA显示文摘Human ACAT1 cDNA K1 was first cloned and functionally expressed in 1993. There are two adjacent in-frame AUG codons, AUG1397–1399 and AUG1415–1417, at 5’-terminus of the open reading frame (ORF, nt 1397–3049) of human ACAT1 mRNA corresponding to cDNA K1. In current work, these two adjacent in- frame AUGs at 5’-terminus of the predicted ORF (5’-ORF-AUGs) as start codons for translation initiation of human ACAT1 mRNA were characterized in detail. Codon mutations indicated that both of these two adja- cent 5’-ORF-AUGs can be selected as start codons but the first 5’-ORF-AUG1397–1399 is a main start codon consistent with that of the predicted ORF of human ACAT1 mRNA. Further deletion and mutation analyses demonstrated that a stable upstream stem-loop structure enhanced the selection of the first 5’-ORF-AUG1397– as a main start codon, in addition to upstream nucleotide A in the –3 position, which is a key site of Kozak 1399 sequence. In addition, result of ACAT1 enzymatic activity assay showed no obvious difference between these two ACAT1 proteins respectively initiated from the two adjacent 5’-ORF-AUGs. 11Extraction of Posterior Polar Intraocular Foreign Bodies with Sodium Hyaluronate Backsiphon Technique显示文摘Purpose: To explore the new technique on the removal of posterior polar intraocular foreign bodies (IOFBs).Methods: Dint of viscoelasticity and cohesiveness of ophthalmic viscoelastics (sodium hyaluronate) was used to wrap, dissociate and suspend IOFBs. Then IOFBs were approached and backsiphoned by using self-made backsiphon syringe needles which were selected according to the size and shape of IOFBs. The IOFBs are removed with the inflow of sodium hyaluronate naturally surging into the needles.Results: Posterior polar IOFBs in 13 cases (13 eyes) were all removed successfully at one time. There were no accidental injuries or complications. The postoperative vision in 11 cases (11 eyes) was improved, unchanged in 1 case (1 eye) and decreased in 1 case ( 1 eye).Conclusions: The technique using viscoelastics to dissociate, suspend and backsiphon IOFBs during the removal of the posterior polar IOFBs can evidently decrease operative difficulty, injury and complications.JiuminYuan ZhengyanWang 2005Eye Science2005,21,1:0

13The Role of Osteoclasts in Psoriatic Arthritis显示文摘Psoriatic arthritis (PsA) is a chronic immune-mediated inflammatory disease related to psoriasis involving bone and cartilage. It is a heterogeneous disorder with a variety of clinical manifestations, which can include peripheral arthritis, axial spondylitis, enthesitis, skin and nail disease, dactylitis, uveitis, osteitis, inflammatory bowel disease. The distinctive feature of PsA is enthesitis. The characteristic bone erosion at the bone-pannus junction in PsA is mediated by osteoclasts, which are multinucleated giant cells derived from hematopoietic stem cells. Although the pathological mechanism of osteoclasts in PsA is mainly related to the destruction of the diseased joint, the exact pathogenesis of PsA is complex and the factors involved in initiation and termination of osteoclast need to be further explored. Much attention has been paid to the importance of osteoclast in psoriasis arthritis for decades. Based on the role of osteoclasts in PsA, our review discusses the formation and characteristics of multinucleated osteoclasts in PsA, summarizes current developments in osteoclast-related pathways in PsA including classical receptor activator of nuclear factor-κB-receptor activator of nuclear factor-κB ligand-osteoprotegerin pathway and immunomodulatory factors, as well as their advances and corresponding treatment. 14Selective Oxidation of Isobutane over NH_4CsFePVMoAs Heteropolycompound Catalysts显示文摘A series of NH4Cs1.5Fe0.08HxPVMo11AsaOy heteropolycompound catalysts for the selective oxidation of isobutane, having Keggin structure, were synthesized by using co-precipitation method. The catalysts were characterized by FT-IR, H2-TPR, TG-DTA, SEM and XRD. Effects of the As content,reaction time, reaction temperature and molar ratio of isobutane to oxygen in feedstock on the activity and selectivity of the catalyst were investigated. The activation energy of the catalysts was measured by kinetics researches. Results showed the introduction of Cs+ into the catalysts shortened the stable period of them and enhanced their catalytic activity for the selective oxidation of isobutane. The highest conversion of isobutane and the total selectivity to liquid products were 18.6% and 81.2%, respectively,which were obtained at 380 ℃ with a space velocity of 975 h-1 over the NH4Cs1.5Fe0.08HxPVMo11As0.3Oy heteropolycompound catalyst. 16Enhancement of human ACAT1 gene expression to promote the macrophage-derived foam cell formation by dexamethasone显示文摘In macrophages, the accumulation of cholesteryl esters synthesized by the activated acyl-coenzyme A:cholesterol acyltransferase-1 (ACAT1) results in the foam cell formation, a hallmark of early atherosclerotic lesions. In this study,with the treatment of a glucocorticoid hormone dexamethasone (Dex), lipid staining results clearly showed the large accumulation of lipid droplets containing cholesteryl esters in THP- 1-derived macrophages exposed to lower concentration of the oxidized low-density lipoprotein (ox-LDL). More notably, when treated together with specific anti-ACAT inhibitors, the abundant cholesteryl ester accumulation was markedly diminished in THP-l-derived macrophages, confirming that ACAT is the key enzyme responsible for intracellular cholesteryl ester synthesis. RT-PCR and Western blot results indicated that Dex caused up-regulation of human ACAT1 expression at both the mRNA and protein levels in THP-1 and THP- 1-derived macrophages. The luciferase activity assay demonstrated that Dex could enhance the activity of human ACAT1 gene P1 promoter, a major factor leading to the ACAT1 activation, in a cell-specific manner.Further experimental evidences showed that a glucocorticoid response element (GRE) located within human ACAT1gene P1 promoter to response to the elevation of human ACAT1 gene expression by Dex could be functionally bound with glucocorticoid receptor (GR) proteins.
