1Discrimination of mitochondrial DNA 10400 locus by SNP-operated on/off Switch显示文摘Objective:To apply reformed AS-PCR, which combined phosphorothioate-modified primers with exo+ polymerase, in single nucle- otide polymorphism discrimination of mitochondrial DNA 10400 locus. Methods: We used the mtDNA 10400 locus to design unmodi- fied and 3′phosphorothioate-modified allele-specific primers for PCR, which was performed using polymerases with and without 3′exonuclease activities. The effects of these primers on primer-extension were evaluated by agarose gel electrophoresis. Results: The unmodified primers were extended by both exo- and exo+ polymerase irrespective of whether the primers were matched or mismatched with the templates. However, the 3′phosphorothioate-modified primers with a terminal mismatch triggered an' off- switch'of exo+ polymerase when compared to exo-polymerase. Conclusion: The'on/off'switch constituted by the combination of 3′phosphorothioate-modified primers with exo+ polymerase is a cost-effective, high-throughput and reliable method for SNP typing, which will be of enormous application in association studies by single nucleotide polymorphism screening.Mei Hong Enben Su Ziqing Chen Xiaobing Ju Qi Chen Rong Zhou 2008Journal of Nanjing Medical University2008,22,6:1
2Unconventional Petroleum Sedimentology:A Key to Understanding Unconventional Hydrocarbon Accumulation显示文摘The commercial exploitation of unconventional petroleum resources(e.g.,shale oil/gas and tight oil/gas)has drastically changed the global energy structure within the past two decades.Sweet-spot intervals(areas),the most prolific unconventional hydrocarbon resources,generally consist of extraordinarily high organic matter(EHOM)deposits or closely associated sandstones/carbonate rocks.The formation of sweet-spot intervals(areas)is fundamentally controlled by their depositional and subsequent diagenetic settings,which result from the coupled sedimentation of global or regional geological events,such as tectonic activity,sea level(lake level)fluctuations,climate change,bottom water anoxia,volcanic activity,biotic mass extinction or radiation,and gravity flows during a certain geological period.Black shales with EHOM content and their associated high-quality reservoir rocks deposited by the coupling of major geological events provide not only a prerequisite for massive hydrocarbon generation but also abundant hydrocarbon storage space.The Ordovician-Silurian Wufeng-Longmaxi shale of the Sichuan Basin,Devonian Marcellus shale of the Appalachian Basin,Devonian-Carboniferous Bakken Formation of the Williston Basin,and Triassic Yanchang Formation of the Ordos Basin are four typical unconventional hydrocarbon systems selected as case studies herein.In each case,the formation of sweet-spot intervals for unconventional hydrocarbon resources was controlled by the coupled sedimentation of different global or regional geological events,collectively resulting in a favorable environment for the production,preservation,and accumulation of organic matter,as well as for the generation,migration,accumulation,and exploitation of hydrocarbons.Unconventional petroleum sedimentology,which focuses on coupled sedimentation during dramatic environmental changes driven by major geological events,is key to improve the understanding of the formation and distribution of sweet-spot intervals(areas)in unconventional petroleum systems.Caineng Zou Zhen Qiu Jiaqiang Zhang Zhiyang Li Hengye Wei Bei Liu Jianhua Zhao Tian Yang Shifa Zhu Huifei Tao Fengyuan Zhang Yuman Wang Qin Zhang Wen Liu Hanlin Liu Ziqing Feng Dan Liu Jinliang Gao Rong Liu Yifan Li 2022Engineering2022,,11:1
3Dithienocarbazole- and benzothiadiazole-based donor-acceptor conjugated polymers for bulk heterojunction polymer solar cells显示文摘Donor-acceptor(D-A)-conjugated polymers P(BT-C1)and P(BT-C2),with dithieno[2,3-b;7,6-b]carbazole(C1)or dithieno[3,2-b;6,7-b]carbazole(C2)as D-unit and benzothiadiazole(BT)as A-unit,were synthesized.The optical bandgaps of the polymers are similar(1.84 and 1.88 e V,respectively).The structures of donor units noticeably influence the energy levels and backbone curvature of the polymers.P(BT-C1)shows a large backbone curvature;its highest occupied molecular orbital(HOMO)energy level is 5.18 e V,whereas P(BT-C2)displays a pseudo-straight backbone and has a HOMO energy level of 5.37 e V.The hole mobilities of the polymers without thermal annealing are 1.9×10 3 and 2.7×10 3 cm2 V 1 s 1 for P(BT-C1)and P(BT-C2),respectively,as measured by organic thin-film transistors(OTFTs).Polymer solar cells using P(BT-C1)and P(BT-C2)as the donor and phenyl-C71-butyric acid methyl ester(PC71BM)as the acceptor were fabricated.Power conversion efficiencies(PCEs)of 4.9%and 5.0%were achieved for P(BT-C1)and P(BT-C2),respectively.The devices based on P(BT-C2)exhibited a higher Voc due to the deeper HOMO level of the polymer,which led to a slightly higher PCE.Ziqing Rong Yunfeng Deng Zhiyuan Xie Yanhou Geng Fosong Wang 2015Science China Chemistry2015,58,2:0
