2A transgene-free method for rapid and efficient generation of precisely edited pigs without monoclonal selection显示文摘Gene-edited pigs for agricultural and biomedical applications are typically generated using somatic cell nuclear transfer(SCNT).However, SCNT requires the use of monoclonal cells as donors, and the time-consuming and laborious monoclonal selection process limits the production of large populations of gene-edited animals. Here, we developed a rapid and efficient method named RE-DSRNP(reporter RNA enriched dual-sg RNA/CRISPR-Cas9 ribonucleoproteins) for generating gene-edited donor cells. RE-DSRNP takes advantage of the precise and efficient editing features of dual-sg RNA and the high editing efficiency, low off-target effects, transgene-free nature, and low cytotoxic characteristics of reporter RNA enriched RNPs(CRISPR-Cas9ribonucleoproteins), thus eliminating the need for the selection of monoclonal cells and thereby greatly reducing the generation time of donor cells from 3–4 weeks to 1 week, while also reducing the extent of apoptosis and chromosomal aneuploidy of donor cells. We applied RE-DSRNP to produce cloned pigs bearing a deletion edit of the wild-type p53-induced phosphatase 1(WIP1)gene: among 32 weaned cloned pigs, 31(97%) carried WIP1 edits, and 15(47%) were homozygous for the designed fragment deletion, and no off-target event was detected. The WIP1 knockout(KO) pigs exhibited male reproductive disorders, illustrating the utility of RE-DSRNP for rapidly generating precisely edited animals for functional genomics and disease research. 7Screening of phenolic compounds in australian grown grapes and their potential antioxidant activities显示文摘Grapes(Vitis Vinifera)are one of the world's most utilised and prized fruit.Grapes are a great source of antioxidants due to the abundance of phenolic compounds.Grape pulp is the main part of the grape being consumed.The pulp of five Australian grown grapes–Dawn Seedless(DS),Calmeria(CG),Golden Globe(GG),Menindee Seedless(MS),and Thompson Seedless(TS)will be qualitatively and quantitatively analysed for its phenolic composition and antioxidant capacity.The general trend followed:GG>TS>MS>DS>CG,and the phenolic content of the pulp also presented a significant positive correlation with the antioxidant capacity.GG revealing the highest levels of TPC(4.31±0.11 mg GAE/g)and TFC(0.48±0.09 mg QE/g)while TS were highest in TTC(0.94±0.12 mg CE/g).Similarly,GG exhibited higher antioxidant capacity in DPPH(5.84±0.17 mg AAE/g),FRAP(4.91±0.14 mg AAE/g),and TAC(6.78±0.12 mg AAE/g),while TS revealed highest radical scavenging ability for ABTS(4.56±0.19 mg AAE/g).The LC-ESI-QTOF-MS/MS tentatively identified 78 phenolic compounds consisting of flavonoids(36),phenolic acids(31),lignans(3),stilbenes(1),and other polyphenols(7)in five grapes samples.Ten most abundant fruit-based targeted polyphenols were quantified.GG had a higher phenolic concentration in HPLC-PDA quantification,particularly for phenolic acids(gallic acid and chlorogenic acid)and flavonoids(kaempferol).Our study provides relatively adequate information on grape pulp's phenolic content and antioxidant capacity,providing confidence and research prospects to promote grape pulp use in different food,pharmaceutical and supplement industries,and even influencing grape cultivation.Gia Toan Vo Ziyao Liu Osbert Chou Biming Zhong Colin J.Barrow Frank R.Dunshea Hafiz A.R.Suleria 2022Food Bioscience2022,47,3:2

9Laser mass spectrometry for high sensitive and multi-component analysis of exhaust gas from vehicles显示文摘Laser mass spectrometry is a newly developed method for pollutant detection. It combines resonance-enhanced multiphoton ionization (REMPI) and time-of-flight mass spectro- metry (TOF-MS). It may detect pollutants with high sensitivity, high selectivity and in a multi-component way. In this note, laser mass spectrometry was used to detect the pollutants in exhaust gases from vehicles. With one-color REMPI (at 266 nm), several aromatic hydrocarbons, including benzene, toluene, xylene, C3-benzene, etc., were detected. These substances were selectively ionized by (1+1) REMPI and their mass resolution was detected by a TOF mass spectrometer. 10Absence of hexavalent chromium in marine carbonates:implications for chromium isotopes as paleoenvironment proxy显示文摘The oxygenation of Earth’s atmosphere is widely regarded to have played an important role in early-life evolution.Chromium(Cr)isotopes recorded in sedimentary rocks have been used to constrain the atmospheric oxygen level(AOL)over geological times based on the fact that a positive Cr isotopic signature is linked to the presence of Cr(Ⅵ)as a result of oxidative continental weathering.However,there is no direct evidence of the presence of Cr(Ⅵ)in sedimentary rocks yet.Carbonates are most widely distributed over geological times and were thought to have incorporated Cr(Ⅵ)directly from seawater.Here,we present results of Cr valence states in carbonates which show Cr(Ⅲ)is the dominant species in all samples spanning a wide range of geological times.These findings indicate that Cr(Ⅵ)in seawater was reduced either before or after carbonate precipitation,which might have caused Cr isotopic fractionation between seawater and carbonates,or marine carbonates preferentially uptake Cr(Ⅲ)from seawater.As Cr(Ⅲ)can come from non-redox Cr cycling,which also can cause isotopic fractionation,we suggest that positively fractionated Cr isotopic values do not necessarily correspond to the rise in AOL.Ziyao Fang Liping Qin Wei Liu Tao Yao Xiaoyan Chen Shiqiang Wei 2021National Science Review2021,8,3:2
11Steady increase in water clarity in Jiaozhou Bay in the Yellow Sea from 2000 to 2018:Observations from MODIS显示文摘The Moderate Resolution Imaging Spectroradiometer(MODIS)surface reflectance data were used to analyze the temporal and spatial distribution characteristics of water clarity(Z_(sd))in the Jiaozhou Bay,Qingdao,China,in the Yellow Sea from 2000 to 2018.Z_(sd)retrieval models were regionally optimized using in-situ data with coincident MODIS images,and then were used to retrieve the Z_(sd) products in Jiaozhou Bay from 2000-2018.The analysis of the Z_(sd) results suggests that the spatial distribution of relative Z_(sd) spatial characteristics in Jiaozhou Bay was stable,being higher Z_(sd) in the southeast and a lower Z_(sd) in the northwest.The annual mean Z_(sd) in Jiaozhou Bay showed a significant upward trend,with an annual increase of approximately 0.02 m.Water depth and wind speed were important factors affecting the spatial distribution and annual variation of Z_(sd) in Jiaozhou Bay,respectively.Ziyao YIN Junsheng LI Jue HUANG Shenglei WANG Fangfang ZHANG Bing ZHANG 2021Journal of Oceanology and Limnology2021,39,3:2
12An overview of multi-antenna technologies for space-ground integrated networks显示文摘Multi-antenna technologies have already achieved a series of great successes in the development of information networks. For future space-ground integrated networks(SGINs), the traditional various kinds of separated information networks will converge to a whole fully connected information network to provide more flexible and reliable services on a world scale. Regarding their great successes in existing systems, multiantenna technologies will be of critical importance for the realization of SGINs and multi-antenna technologies are definitely one of the most important enabling technologies for future converged SGINs. In this article, a comprehensive overview on multi-antenna technologies is given. We first investigate multi-antenna technologies from a theoretical viewpoint. It is shown that we can understand multi-antenna technologies in a general and unified point of view. This fact has two-fold meanings. First, the research on multi-antennas can help us understand the relationships between different technologies e.g., OFDMA, CDMA, etc. On the other hand,multi-antenna technologies are easy to integrate into various information systems. Following that, we discuss in depth the potentials and challenges of the multi-antenna technologies on different platforms and in different applications case by case. 13Formation of a new intermediate phase and its evolution toward θ’ during aging of pre-deformed Al-Cu alloys显示文摘The frequent occurrence of hitherto unknown phase Pre-θ′-2 and unusual 1.5 cθ′thick θ′precipitate was observed by atomic-resolution scanning transmission electron microscopy in the well-studied Al-Cu alloys. This phenomenon is associated with heterogeneous precipitate nucleation and growth on preexisting dislocations introduced by slight deformation prior to aging. In this study, the precise structure details of Pre-θ′-2 was determined by atomic scale imaging, image simulation based on image forming theories and first principle calculations. Pre-θ′-2 has a well-defined ordered structure sandwiched between two 2 aAl(~1.5 cθ′) spaced Cu layers on {200}Alplanes. 16Growth behavior of CVD diamond films with enhanced electron field emission properties over a wide range of experimental parameters显示文摘In this study, diamond films were synthesized on silicon substrates by microwave plasma enhanced chemical vapor deposition(CVD) over a wide range of experimental parameters. The effects of the microwave power,CH_4/H_2 ratio and gas pressure on the morphology, growth rate, composition, and quality of diamond films were investigated by means of scanning electron microscope(SEM), X-ray diffraction(XRD), Raman spectroscopy and X-ray photoelectron spectroscopy(XPS). A rise of microwave power can lead to an increasing pyrolysis of hydrogen and methane, so that the microcrystalline diamond film could be synthesized at low CH_4/H_2 levels. Gas pressure has similar effect in changing the morphology of diamond films, and high gas pressure also results in dramatically increased grain size. However,diamond film is deteriorated at high CH_4/H_2 ratio due to the abundant graphite content including in the films. Under an extreme condition of high microwave power of 10 kW and high CH_4 concentration, a hybrid film composed of diamond/graphite was successfully formed in the absence of N_2 or Ar,which is different from other reports. This composite structure has an excellent measured sheet resistance of 10-100 Ω/Sqr. which allows it to be utilized as field electron emitter. 17Crystalline InGaZnO quaternary nanowires with superlattice structure for high-performance thin-film transistors显示文摘Amorphous indium-gallium-zinc oxide (a-IGZO) materials have been widely explored for various thin-film transistor (TFT) applications;however, their device performance is still restricted by the intrinsic material issues especially due to their non-crystalline nature. In this study, highly crystalline superlattice-structured IGZO nanowires (NWs) with different Ga concentration are successfully fabricated by enhanced ambient-pressure chemical vapor deposition (CVD). The unique superlattice structure together with the optimal Ga concentration (i.e., 31 at.%) are found to effectively modulate the carrier concentration as well as efficiently suppress the oxygen vacancy formation for the superior NW device performance. In specific, the In1.8Ga1.8Zn2.4O7 NW field-effect transistor exhibit impressive device characteristics with the average electron mobility of ~ 110 cm^2-V^-1·s^-1 and on/off current ratio of ~ 10^6. Importantly, these NWs can also be integrated into NW parallel arrays for the construction of high-performance TFT devices, in which their performance is comparable to many state-of-the-art IGZO TFTs. 18A Comparative Investigation of Single Crystal and Polycrystalline Ni-Rich NCMs as Cathodes for Lithium-Ion Batteries显示文摘Nickel-rich LiNi_(1-x-y)Co_(x)Mn_(y)O_(2)(NCM,1-x-y≥0.6)is known as a promising cathode material for lithium-ion batteries since its superiority of high voltage and large capacity.However,polycrystalline Ni-rich NCMs suffer from poor cycle stability,limiting its further application.Herein,single crystal and polycrystalline LiNi_(0.84)Co_(0.07)Mn_(0.09)O_(2)cathode materials are compared to figure out the relation of the morphology and the electrochemical storage performance.According to the Li^(+)diffusion coefficient,the lower capacity of single crystal samples is mainly ascribed to the limited Li+diffusion in the large bulk.In situ XRD illustrates that the polycrystalline and single crystal NCMs show a virtually identical manner and magnitude in lattice contraction and expansion during cycling.Also,the electrochemically active surface area(ECSA)measurement is employed in lithium-ion battery study for the first time,and these two cathodes show huge discrepancy in the ECSA after the initial cycle.These results suggest that the single crystal sample exhibits reduced cracking,surface side reaction,and Ni/Li mixing but suffers the lower Li^(+)diffusion kinetics.This work offers a view of how the morphology of Ni-rich NCM effects the electrochemical performance,which is instructive for developing a promising strategy to achieve good rate performance and excellent cycling stability.Xianming Deng Rui Zhang Kai Zhou Ziyao Gao Wei He Lihan Zhang Cuiping Han Feiyu Kang Baohua Li 2023Energy & Environmental Materials2023,6,3:1
19Electrostatic Interaction Tailored Anion-Rich Solvation Sheath Stabilizing High-Voltage Lithium Metal Batteries显示文摘Through tailoring interfacial chemistry,electrolyte engineering is a facile yet effective strategy for highperformance lithium(Li)metal batteries,where the solvation structure is critical for interfacial chemistry.Herein,the effect of electrostatic interaction on regulating an anion-rich solvation is firstly proposed.The moderate electrostatic interaction between anion and solvent promotes anion to enter the solvation sheath,inducing stable solid electrolyte interphase with fast Li+transport kinetics on the anode.This asdesigned electrolyte exhibits excellent compatibility with Li metal anode(a Li deposition/stripping Coulombic efficiency of 99.3%)and high-voltage LiCoO_(2) cathode.Consequently,the 50μm-thin Li||high-loading LiCoO_(2) cells achieve significantly improved cycling performance under stringent conditions of high voltage over 4.5 V,lean electrolyte,and wide temperature range(-20 to 60℃).This work inspires a groundbreaking strategy to manipulate the solvation structure through regulating the interactions of solvent and anion for highperformance Li metal batteries.Junru Wu Ziyao Gao Yao Wang Xu Yang Qi Liu Dong Zhou Xianshu Wang Feiyu Kang Baohua Li 2022Nano-Micro Letters2022,14,9:1
