1Get effective polyclonal antisera in one month显示文摘According to the traditional immunization procedure, after the first injection of the sample A (emulsion of aimed antigen and Freund's complete adjuvant) to immunize rabbit, successive injections of the sample B (emulsion of aimed antigen and Freund's incomplete adjuvant) were followed every 2-4 weeks. In general,high titer of the corresponding polyclonal antisera will be observed after 4-5 injections of sample B in 3-4months. This report presents a simply modified procedure that was able to stimulate the antisera formation in one month and achieve enough avidity to satisfy either Western blot or immunohistochemistry analysis.It just applied an additional injection of the sample A to the rabbit at the 3rd day after the primary immunization injection. You could gain the high titer of the antisera right after the first sample B injection in one month. 4Effects of Friction Heat on the Tribological Properties of the Woven Self-lubricating Liner显示文摘In the dry-sliding process of the woven self-lubricating liner which is used in the self-lubricating spherical plain bearing, the friction heat plays an important role in the tribological performances of the liner. It has important value to study on the relationship between tribological performances of the liner and the friction heat. Unfortunately, up to now, published work on this relationship is quite scarce. Therefore, the effect of friction heat on the tribological performances of the liner was investigated in the present work. The tribological behaviors of the liner were evaluated by using the high temperature end surface wear tester. Scanning electron microscopy (SEM) was utilized to examine the morphologies of worn surfaces of the liner and study the failure modes. Differential scanning calorimetry (DSC) measurement and X-ray diffraction (XRD) analysis were performed to study the behaviors of the wear debris. The temperature rise on the worn surface was calculated according to classical models. SEM observation shows that the dominating wear mechanism for the liner is mainly affected by friction shear force, contact pressure and friction heat. Higher fusion heat for the wear debris than that for the pure polytetrafluroethylene (PTFE) indicates that the PTFE is the main portion of the wear debris, and, the PTFE in the wear debris shows a higher crystallisation degree owing to the effects of friction shear force and the friction heat. Combining the calculated temperature rise results with the wear rate of the liner, it can be concluded that the effects of temperature rise on the tribological performances of the liner become more obvious when the temperature rise exceeds the glass transition temperature (Tg) of the PTFE. The wear resistance of the liner deteriorates dramatically when the temperature rise approaches to the melting point (Ton) of the PTFE. The tribological performances of the liner can be improved when the temperature rise exceeds Tg but is far lower than Ton. 5Characteristics of the microbial community in rhizosphere of Camptotheca acuminata cultured with exotic invasive plant Eupatorium adenophorum显示文摘The traditional culture-dependent plate counting and culture-independent small-subunit-ribosomal RNA gene-targeted molecular techniques, Single-Strand Conformation Polymorphism (SSCP) and ter-minal Restriction Fragment Length Polymorphism (tRFLP) combined with 16S rDNA clone library were adopted to investigate the impacts of secretion from Camptotheca acuminata (abbreviated to Ca) roots on the quantities and structure of eukaryotic microbes and bacteria in the rhizosphere, and the possi-bility that Ca controls exotic invasive plant Eupatorium adenophorum (Ea). The counting results indi-cated that the number of bacteria increased in turn in rhizospheres of Ea, Ca-Ea mixed culture and Ca, while that of eukaryotic microbes decreased. PCR-SSCP profiles showed eukaryotic microbial bands (corresponding to biodiversity) in rhizosphere of Ea were more complex than those of Ca and CE. Meristolohmannia sp., Termitomyces sp. and Rhodophyllus sp. were the dominant populations in the rhizosphere of Ca. Bacterial terminal restriction fragments (TRFs) profiles showed no difference among three kinds of rhizospheres, and the sequences of the 16S rDNA clone library from Ca rhizospheres were distributed in 10 known phyla, in which phylum Proteobacteria were the absolute dominant group and accounted for 24.71% of the cloned sequences (δ-Proteobacteria accounted for up to 17.65%), and phyla Acidobacteria and Bacteroidetes accounted for 16.47% and 10.59% of the cloned sequences, respectively. In addition, high performance liquid chromatography detected a trace amount of camp-tothecin and hydroxycamptothecin in the rhizospheric soil of Ca and CE, but examined neither camp-tothecin nor hydroxycamptothecin in rhizospheric soil of Ea. Therefore, invasion and diffusion of Ea evidently depended on distinguishing the eukaryotic community structure, but not on that of the bac-terial pattern. Ca was able to alter the eukaryotic community structure of invasive Ea by secreting camptothecin and hydroxycamptothecin into rhizospheres, and may benefit the control of overspread of Ea. 6Direct observation of ultrafast plasmonic hot electron transfer in the strong coupling regime显示文摘Achieving strong coupling between plasmonic oscillators can significantly modulate their intrinsic optical properties.Here,we report the direct observation of ultrafast plasmonic hot electron transfer from an Au grating array to an MoS_(2) monolayer in the strong coupling regime between localized surface plasmons(LSPs)and surface plasmon polaritons(SPPs).By means of femtosecond pump-probe spectroscopy,the measured hot electron transfer time is approximately 40 fs with a maximum external quantum yield of 1.65%.Our results suggest that strong coupling between LSPs and SPPs has synergetic effects on the generation of plasmonic hot carriers,where SPPs with a unique nonradiative feature can act as an‘energy recycle bin’to reuse the radiative energy of LSPs and contribute to hot carrier generation.Coherent energy exchange between plasmonic modes in the strong coupling regime can further enhance the vertical electric field and promote the transfer of hot electrons between the Au grating and the MoS_(2) monolayer.Our proposed plasmonic strong coupling configuration overcomes the challenge associated with utilizing hot carriers and is instructive in terms of improving the performance of plasmonic opto-electronic devices.Hangyong Shan Ying Yu Xingli Wang Yang Luo Shuai Zu Bowen Du Tianyang Han Bowen Li Yu Li Jiarui Wu Feng Lin Kebin Shi Beng Kang Tay Zheng Liu Xing Zhu Zheyu Fang 2019Light(Science & Applications)2019,8,1:7
7Simulation and Field Testing of Multiple Vehicles Collision Avoidance Algorithms显示文摘A global planning algorithm for intelligent vehicles is designed based on the A* algorithm, which provides intelligent vehicles with a global path towards their destinations. A distributed real-time multiple vehicle collision avoidance(MVCA)algorithm is proposed by extending the reciprocal n-body collision avoidance method. MVCA enables the intelligent vehicles to choose their destinations and control inputs independently,without needing to negotiate with each other or with the coordinator. Compared to the centralized trajectory-planning algorithm, MVCA reduces computation costs and greatly improves the robustness of the system. Because the destination of each intelligent vehicle can be regarded as private, which can be protected by MVCA, at the same time MVCA can provide a real-time trajectory planning for intelligent vehicles. Therefore,MVCA can better improve the safety of intelligent vehicles. The simulation was conducted in MATLAB, including crossroads scene simulation and circular exchange position simulation. The results show that MVCA behaves safely and reliably. The effects of latency and packet loss on MVCA are also statistically investigated through theoretically formulating broadcasting process based on one-dimensional Markov chain. The results uncover that the tolerant delay should not exceed the half of deciding cycle of trajectory planning, and shortening the sending interval could alleviate the negative effects caused by the packet loss to an extent. The cases of short delay(< 100100 ms) and low packet loss(< 5%) can bring little influence to those trajectory planning algorithms that only depend on V2 V to sense the context, but the unpredictable collision may occur if the delay and packet loss are further worsened. 8Different Dosages of Intravitreal Triamcinolone Acetonide Injections for Macular Edema Secondary to Central Retinal Vein Occlusion显示文摘Purpose:.To study the effect of intravitreal injections of triamcinolone acetonide (TA) for the treatment of macular edema secondary to central retinal vein occlusion.(CRVO).in a sample of Chinese patients from Shaanxi province. Methods:.The 50 eyes from 50 patients were separated into three TA treatment groups:.17 patients were given 4 mg/0.1 ml,.19 patients were given 8 mg/0.2 ml,.and 14 patients were given 16 mg/0.4 ml. Patients were followed up for 12 months. Foveal thickness, intraocular pressure (IOP), and best-corrected visual acuity (BCVA) were measured. Results:.Macular edema responded well both anatomically and functionally to the TA injections. After the initial intravitreal injection,.macular edema recurred at 2-4 months in the low-dose group.(4 mg),.at 3-5 months in the medium-dose group (8 mg), and at 6-9 months in the high-dose group (16 mg)..No significant difference in BCVA or in foveal thickness were observed between the first intravitreal injection and the re-injection. There was no increase in IOP after re-injection of 16 mg TA,.if the patient did not have an elevated IOP after the initial intravitreal injection of 4/8 mg TA. 9Sliding Wear of the Hybrid Kevlar/PTFE Fabric Reinforced Phenolic Composite Filled with Nano-titania显示文摘The Kevlar/polytetrafluroethylene(Kevlar/PTFE) fabric composite can be used as a self-lubricating liner of the self-lubricating bearing.Many types of nano-particles can improve the tribological performance of the polymer-based composite.Unfortunately,up to now,published work on the effect of nano-particles on the tribological performance of the fabric composite which can be used as a self-lubricating liner is quite scarce.Therefore,for the purpose of exploring a way to significantly improve the tribological performance of the fabric composite,the tribological performance of the Kevlar/PTFE fabric composite filled with nano-titania is evaluated by using the block-on-ring wear tester.The scanning electron microscopy is utilized to observe the morphologies of worn surfaces of the fabric composites and the counterparts.The tensile properties of the composites are evaluated on the universal material testing machine.The test results show that the addition of nano-titania at a proper mass fraction of the matrix resin improves the wear resistance and the tensile strength,decreases the friction coefficient,and makes the wear volume of the composite reach a relative steady state more quickly;plastic deformation and microcutting are important for the wear of the fabric composite;a lubricating layer is formed on the worn surface of the composite during sliding,and the lubricating layer is critical for the tribological performance of the composite;the formation and properties of the lubricating layer are influenced by the nano-titania particles.The proposed study on the effect of nano-titania on the tribological performance of the Kevlar/PTFE fabric composite,especially on the evolution of the worn surface of the composite,provides the basis for further understanding of the influence mechanism of the nano-particles on the tribological performance of the composite and explores a method of improving the tribological performance of the composite.YANG Yulin HUANG Shijun ZU Dalei ZHANG Ruijun QI Xiaowen 2011Chinese Journal of Mechanical Engineering2011,24,1:4
10Epigallocatechin gallate content change of the fresh tea leaf homogenates extracted by different methods in extraction and preservation显示文摘The fresh leaves of China green tea, Camellia sinensis, were collected from Fuyang, Zhejiang Province, China, in April. The tea polyphenols was extracted by four different methods (homogenized with distilled water at room temperature, homogenized with 0.3% citric acid (w/v) at room temperature, 5- min boiling and homogenized with distilled water at room temperature, homogenized with 85°C distilled water), and after preserving at room temperature, the change of the Epigallocatechin gallate (EGCG) contents of the extracts was investigated. Results indicated that the EGCG content of homogenate extracted with 85°C distilled water was the highest before the extract was preserved, followed by that of the extract homogenized with 0.3% citric acid at room temperature. During preservation, EGCG content changed obviously. The EGCG contents of homogenates extracted with distilled water at room temperature and 85°C distilled water declined quickly and separately reduced to 21.52% and 54.6% of their initial contents after preservation for 12 h. The EGCG contents extracted by 0.3% citric acid (w/v) solvent at room temperature and 5- min boiling/homogenized with distilled water at room temperature declined relatively slowly ,and separately reduced to 76.9% and 85.16% of their initial contents after preservation for 12 h. 11Has the stilling of the surface wind speed ended in China?显示文摘Since the 1960 s,the global land surface wind speed(SWS)has significantly weakened,a phenomenon known as global terrestrial stilling.The latest research found that the stilling reversed around 2010,and since then the global SWS has been strengthening.However,there is still a lack of systematic quantitative analysis in China.We analyzed the transition and regional differences in the long-term trends of the SWS in China based on observational SWS data from 1971 to 2019.The results showed that annual mean SWS in China underwent a reversal from a continuously weakening trend to a significantly strengthening trend around 2014 and implying that stilling may have ended in 2014.The reversal had obvious regional and seasonal variations.In Northeast China,Western Xinjiang as well as on the Tibetan Plateau,the years with both annual and seasonal mean SWS changing from weakening to strengthening were around 2013/2014,1993/1994,and 2000.However,in the west of North China,SWS showed an obviously strengthening trend only in autumn and winter after 2007;while only autumn mean SWS showed a strengthening trend after 2012 in South China.It should be noted that stilling is ongoing in the eastern and southern coastal areas,North China and Eastern Xinjiang.Qing YANG Mingxing LI Ziqing ZU Zhuguo MA 2021Science China Earth Sciences2021,64,7:4

15Ubiquitin-specific protease 22 enhances intestinal cell proliferation and tissue regeneration after intestinal ischemia reperfusion injury显示文摘BACKGROUND Intestinal ischemia reperfusion(I/R) injury is a serious but common pathophysiological process of many diseases, resulting in a high mortality rate in clinical practice. Ubiquitin-specific protease 22(USP22) acts as regulator of cell cycle progression, proliferation, and tumor invasion. Depleted USP22 expression has been reported to contribute to arrested cell cycle and disrupted generation of differentiated cell types in crypts and villi. However, the role of USP22 in intestinal damage recovery has not been investigated. Therefore, elucidation of the underlying mechanism of USP22 in intestinal I/R injury may help to improve the tissue repair and patient prognosis in clinical practice.AIM To investigate the role of USP22 in intestinal cell proliferation and regeneration after intestinal I/R injury.METHODS An animal model of intestinal I/R injury was generated in male Sprague-Dawley rats by occlusion of the superior mesenteric artery followed by reperfusion.Chiu's scoring system was used to grade the damage to the intestinal mucosa. An in vitro model was developed by incubating rat intestinal epithelial IEC-6 cells in hypoxia/reoxygenation conditions in order to simulate I/R in vivo. siRNA and overexpression plasmid were used to regulate the expression of USP22. USP22,Cyclin D1, and proliferating cell nuclear antigen(PCNA) expression levels were measured by Western blot analysis and immunohistochemistry staining. Cell survival(viability) and cell cycle were evaluated using the Cell Counting Kit-8and flow cytometry, respectively.RESULTS USP22 expression was positively correlated with the expression levels of PCNA and Cyclin D1 both in vivo and in vitro, which confirmed that USP22 was involved in cell proliferation and intestinal regeneration after intestinal I/R injury. Decreased levels of Cyclin D1 and cell cycle arrest were observed in the USP22 knockdown group(P < 0.05), while opposite results were observed in the USP22 overexpression group(P < 0.05). In addition, increased expression of USP22 was related to improved intestinal pathology or IEC-6 cell viability after I/R or hypoxia/reoxygenation. 19Simultaneous quantitative determination of five alkaloids in Catharanthus roseus by HPLC-ESI-MS/MS显示文摘AIM: To establish a method to simultaneously determine the main five alkaloids of Catharanthus roseus for trace samples, a high-performance liquid chromatography–electrospray ionization-tandem mass spectrometry(HPLC-ESI-MS/MS) analysis method was developed. METHOD: The five Catharanthus alkaloids, vinblastine, vincristine, vinleurosine, vindoline, and catharanthine were chromatographically separated on a C18 HPLC column. The mobile phase was methanol-15 nmol?L–1 ammonium acetate containing 0.02% formic acid(65 : 35, V/V). The quantification of these alkaloids was based on the Multiple Reaction Monitoring(MRM) mode. RESULTS: This method was validated, and the results achieved the aims of the study. The intra- and inter-day precision and accuracy of the five alkaloids were within 1.2%-11.5%(RSD%) and-10.9%-10.5%(RE%). The recovery rates of the five alkaloids of samples were from 79.9% to 91.5%. The five analytes were stable at room temperature for 2 h, at 4 °C for 12 h, and at-20 °C for two weeks. The developed method was applied successfully to determine the content of the five alkaloids in three plant parts of three batches of C. roseus with a minute amount collected from three regions of China.
