| 1 | From Golden Rice to aSTARice:Bioengineering Astaxanthin Biosynthesis in Rice Endosperm显示文摘Carotenoids are important phytonutrients with antioxidant properties,and are widely used in foods and feedstuffs as Supplements.Astaxanthin,a red-colored ketocarotenoid,has strong antioxidant activity and thus can benefit human health.However,astaxanthin is not produced in most higher plants.Here we report the bioengineering of astaxanthin biosynthesis in rice endosperm by introducing four synthetic genes,sZmPSY1,sPaCrtl,sCrBKT,and sHpBHY,which encode the enzymes phytoene synthase,phytoene desaturase,β-carotene ketolase,and β-carotene hydroxylase,respectively.Transgneic overexpression of two (sZmPSY1 and sPaCrtl),three (sZmPSY1,sPaCrtl and sCrBKT),and all these four genes driven by rice endosperm-specific promoters established the Carotenoid/ketocarotenoid/astaxanthin biosynthetic pathways in the endosperm and thus resulted in various types of germplasm,from the yellow-grained β-caro- tene-enriched Golden Rice to orange-red-grained Canthaxanthin Rice and Astaxanthin Rice,respectively. | 3 | Biogenic production of DMSP and its degradation to DMS—their roles in the global sulfur cycle显示文摘Dimethyl sulfide(DMS) is the most abundant form of volatile sulfur in Earth’s oceans, and is mainly produced by the enzymatic clevage of dimethylsulfoniopropionate(DMSP). DMS and DMSP play important roles in driving the global sulfur cycle and may affect climate. DMSP is proposed to serve as an osmolyte, a grazing deterrent, a signaling molecule, an antioxidant, a cryoprotectant and/or as a sink for excess sulfur. It was long believed that only marine eukaryotes such as phytoplankton produce DMSP. However, we recently discovered that marine heterotrophic bacteria can also produce DMSP, making them a potentially important source of DMSP. At present, one prokaryotic and two eukaryotic DMSP synthesis enzymes have been identified.Marine heterotrophic bacteria are likely the major degraders of DMSP, using two known pathways: demethylation and cleavage.Many phytoplankton and some fungi can also cleave DMSP. So far seven different prokaryotic and one eukaryotic DMSP lyases have been identified. Hundreds of genes are involved in building such a natural masterpiece. However, the plant cell wall is the least understood cellular structure in plants. Due to great progress in plant functional genomics,manyachievementshavebeenmadein uncovering cell wall biosynthesis, assembly, and architecture, as well as cell wall regulation and signaling. Such information has significantly advanced our understanding of the roles of the cell wall in many biological and physiological processes and has enhanced our utilization of cell wall materials. The use of cutting-edge technologies such as single-molecule imaging,nuclear magnetic resonance spectroscopy, and atomic force microscopy has provided much insight into the plant cell wall as an intricate nanoscale network, opening up unprecedented possibilities for cell wall research. | 6 | Editing of Rice Isoamylase Gene ISA1 Provides Insights into Its Function in Starch Formation显示文摘Isoamylase 1(ISA1) is an isoamylase-type debranching enzyme which plays a predominant role in amylopectin synthesis. In this study, the clustered regularly interspaced short palindromic repeats(CRISPR)/CRISPR-associated endonuclease 9(CRISPR/Cas9) system was used to edit ISA1 gene in rice via Agrobacterium-mediated transformation. We identified 36 genetic edited lines from 55 T_0 transgenic events, and classified the mutation forms into 7 types. Of those, two homozygous mutants, cr-isa1-1(type 1, with an adenine insertion) and cr-isa1-2(type 3, with a cytosine deletion) were selected for further analysis. Seed sizes of both cr-isa1-1 and cr-isa1-2 were affected, and the two mutants also displayed a shrunken endosperm with significantly lower grain weight. Electron microscopy analysis showed that abnormal starch granules and amyloplasts were found in cr-isa1-1 and cr-isa1-2 endosperm cells. The contents of total starch, amylose and amylopectin in the endosperm of the cr-isa1 mutants were significantly reduced, whereas sugar content and starch gel consistency were observably increased compared to the wild-type. The gelatinization temperature and starch chain length distributions of the cr-isa1 mutants were also altered. Moreover, transcript levels of most starch synthesis-related genes were significantly lower in cr-isa1 mutants. | 7 | Brassinosteroids function in spikelet differentiation and degeneration in rice显示文摘Brassinosteroids(BRs) play crucial roles in many aspects of plant development. However, their function in spikelet differentiation and degeneration in rice(Oryza sativa L.) remains unclear. Here, we investigated the roles of these phytohormones in spikelet development in fieldgrown rice subjected to five different nitrogen(N)fertilization treatments during panicle differentiation. BR levels and expression of genes involved in BR biosynthesis and signal transduction were measured in spikelets. Pollen fertility and the number of differentiated spikelets were closely associated with 24-epicastasterone(24-epiCS) and 28-homobrassinolide(28-homoBL) levels in spikelets.Enhanced BR biosynthesis and signal transduction, in response to N treatment, enhanced spikelet differentiation, reduced spikelet degeneration, and increased grain yield. Increases in proton-pumping ATPase activity, ATPconcentration, energy charge, and antioxidant system(AOS) levels were consistent with 24-epiCS and28-homoBL concentrations. Exogenous application of 24-epiCS or 28-homoBL on young panicles induced a marked increase in endogenous 24-epiCS or 28-homoBL levels, energy charge, AOS levels, spikelet differentiation, and panicle weight. The opposite effects were observed following treatment with a BR biosynthesis inhibitor. | 13 | Effect of exogenous GA3 on flowering quality, endogenous hormones, and hormone-and flowering-associated gene expression in forcing-cultured tree peony(Paeonia suffruticosa)显示文摘Gibberellins(GAs)promote flowering in the forcing-cultured tree peony(Paeonia suffruticosa),however,the mechanism of regulating flowering is not fully understood.In this study,exogenous GA3 was applied to five-year-old Luoyang Hong plants to explore responses in terms of endogenous hormones,flowering quality,and the hormone-and flowering-associated gene expression.Exogenous GA3 application significantly promoted flower bud development and new branch growth,as well as improved flowering quality.Exogenous GA3 application also stimulated the synthesis of endogenous GA3 and indole-3-acetic acid(IAA)but reduced abscisic acid(ABA)levels.To further elucidate the regulatory mechanism,eight genes for GA biosynthesis and signaling,including PsCPS,PsKS,PsGA3ox,PsGA2ox,PsGID1b,PsGID1c,PsDELLA,and PsGID2 were cloned for the first time,and sequence analysis was also performed.The results suggested that all the cloned genes have conserved structure as each homologous gene reported in the other species.Phylogenetic trees constructed by the each cloned gene showed that the phylogenetic evolutionary relationship of P.suffruticosa was closely related to Vitis vinifera.The expression patterns of the above genes,and genes for ABA and IAA biosynthetic and signaling,and the flowering time were also investigated.Most of the above genes showed higher expression in the control buds than those in the GA3 treated buds at six developmental stages,whereas the expression levels of PsSOC1 and PsSPL9 were up-regulated by GA3 treatment.The results also showed that the GA-biosynthetic and signaling pathways are conserved in tree peony,and the PsCPS,PsGA3ox,PsGA2ox,PsGID1,PsDELLA,and PsGID2 genes are necessary for feedback regulation of GAs.Furthermore,hormone changes promoted PsSOC1 and PsSPL9 expression,and repressed PsSVP expression,which contributed to the improvement flowering quality in tree peony of forcing culture. | GUAN Yan-ren XUE Jing-qi XUE Yu-qian YANG Ruo-wen WANG Shun-li ZHANG Xiu-xin | 2019 | Journal of Integrative Agriculture2019,18,6: | 6 |

Existing therapeutic approaches have encountered many problems, such as lack of autogenous allogeneic bone and immunological rejection to allogeneic implant. Synthetic hydroxyapatite (HA) provided solutions for bone repair, since the HA is the main inorganic component of animals' bone. Although HA has good biocompatibility, but only the limited osteogenic capability, which is of significance for modern bone repair materials. Si is an essential trace element in bone tissue, and it has been demonstrated to be able to promote bone formation. Therefore, silicate-doped hydroxyapatite (Si-HA) may serve as a promising material for bone repair, and promote bone regeneration in the repair. | 15 | Fig fruit ripening is regulated by the interaction between ethylene and abscisic acid显示文摘Fleshy fruit ripening is typically regulated by ethylene in climacteric fruits and abscisic acid(ABA)in non-climacteric fruits.Common fig(Ficus carica)shows a dual-ripening mechanism,which is not fully understood.Here,we detected separate peaks of ethylene and ABA in fig fruits at the onset-and onripening stages,in conjunction with a sharp rise in glucose and fructose contents.In a newly-designed split-fruit system,exogenous ethylene failed to rescue fluridone-inhibited fruit ripening,whereas exogenous ABA rescued 2-amino-ethoxy-vinyl glycine(AVG)-inhibited fruit ripening.Transcriptome analysis revealed changes in the expression of genes key to both ABA and ethylene biosynthesis and perception during fig fruit ripening.At the degreening stage,downregulation of FcACO2 or FcPYL8 retarded ripening,but downregulation of FcETR1/2 did not;unexpectedly,downregulation of FcAAO3 promoted ripening,but it inhibited ripening only before the de-greening stage.Furthermore,we detected an increase in ethylene emissions in the FcAAO3-RNAi ripening fruit and a decrease in ABA levels in the FcACO2-RNAi unripening fruit.Importantly,FcPYL8 can bind to ABA,suggesting that it functions as an ABA receptor.Our findings support the hypothesis that ethylene regulates the fig fruit ripening in an ABA-dependent manner.We propose a model for the role of the ABA-ethylene interaction in climacteric/non-climacteric processes. | Han Qiao Han Zhang Zhun Wang Yuanyue Shen | 2021 | Journal of Integrative Plant Biology2021,63,3: | 5 |

| 17 | Purification and characterization of a novel cholesterol-lowering protein from the seeds of Senna obtusifolia显示文摘'Juemingzi', a source of traditional Chinese herbal medicine, has been demonstrated to play a role in decreasing serum cholesterol concentration. In this study, a novel protein, which has shown an in-hibitory effect on cholesterol biosynthesis, was isolated from Senna obtusifolia L. seed by gel filtration and ion exchange chromatography. The novel protein’s molecular mass was 19.7 kD and its pI was 4.80. Both SDS-PAGE and isoelectric-focusing (IEF) revealed a single Coomassie brilliant blue stained band, indicating that the novel protein was a single peptide. The N-terminal amino acid sequence of the pro-tein was IPYISASFPLNIEFLPSE, which had no similarity with any other protein sequences in the NCBI protein database. | 18 | Biosynthesis of silver nanoparticles by using mangrove plant extract and their potential mosquito larvicidal property显示文摘Objective:To identify the larvicidal activities of silver nano particles synthesised with Rhizophora mucronata(R.mucronata) leaf extract against the larvae of Aedes aegypti(Ae.aegypti) and Culex quinquefasciatus(Cx.quinquefasciatus).Methods:In vitro larvicidal activities such as LC50 and LC90 were assessed for the Ae.aegypti and Cx.quinquefasciatus larval species.Further, characterisation such as UV,XRD,FTIR and AFM analysis were carried out for the synthesised silver nano particles.Results:The LC50 value of the synthesised silver nano particle was identified as 0.585 and 0.891 mgg/L for Ae.aegypti and Cx.quinquefasciatus larvae respectively. | 20 | A general aerosol-assisted biosynthesis of functional bulk nanocomposites显示文摘Although a variety of nanoparticles with better-than-bulk material performances can be synthesized, it remains a challenge to scale the extraordinary properties of individual nanoscale units to the macroscopic level for bulk nanostructured materials. Here, we report a general and scalable biosynthesis strategy that involves simultaneous growth of cellulose nanofibrils through microbial fermentation and co-deposition of various kinds of nanoscale building blocks(NBBs) through aerosol feeding on solid culture substrates. We employ this biosynthesis strategy to assemble a wide range of NBBs into cellulose nanofibril-based bulk nanocomposites. In particular, the biosynthesized carbon nanotubes/bacterial cellulose nanocomposites that consist of integrated 3D cellulose nanofibril networks simultaneously achieve an extremely high mechanical strength and electrical conductivity, and thus exhibit outstanding performance as high-strength lightweight electromagnetic interference shielding materials. The biosynthesis approach represents a general and efficient strategy for large-scale production of functional bulk nanocomposites with enhanced performances for practical applications.
