Expert consensus on the use of human serum albumin in critically ill patients显示文摘Introduction Human serum albumin(HSA)is a non-glycosylated,negatively charged,single-chain polypeptide composed of 585 amino acid residues with a relative molecular mass of 66.438 kD.It is synthesized by the liver at a rate of approximately 200 mg·kg^(-1)·day^(-1),with a half-life of 21 days, and subjected to catabolism in the muscles, liver, and kidneys at a rate of 4%per day.[1]Albumin,accounting for 60%of the total plasma protein, has various physiological functions,[2]such as maintaining 70%to 80%of effective plasma colloid osmotic pressure, coordinating vascular endothelial integrity,anti-oxidant and anti-inflammatory activities, maintaining the acidbase balance, and participating in the transport, distribution, and metabolism of a variety of endogenous and exogenous substances.Yue-Tian Yu Jiao Liu Bo Hu Rui-Lan Wang Xiang-Hong Yang Xiu-Ling Shang Gang Wang Chang-Song Wang Bai-Ling Li Ye Gong Sheng Zhang Xin Li Lu Wang Min Shao Mei Meng Feng Zhu You Shang Qiang-Hong Xu Zhi-Xiong Wu De-Chang Chen 2021Chinese Medical Journal2021,,14:22
Noncoding RNAs in gastric cancer: Research progress and prospects显示文摘Noncoding RNAs(nc RNAs) have attracted much attention in cancer research field. They are involved in cellular development, proliferation, differentiation and apoptosis. The dysregulation of nc RNAs has been reported in tumor initiation, progression, invasion and metastasis in various cancers, including gastric cancer(GC). In the past few years, an accumulating body of evidence has deepened our understanding of nc RNAs, and several emerging nc RNAs have been identified, such as PIWI-interacting RNAs(pi RNAs) and circular RNAs(circ RNAs). The competing endogenous RNA(ce RNA) networks include m RNAs, micro RNAs, long nc RNAs(lnc RNAs) and circ RNAs, which play critical roles in the tumorigenesis of GC. This review summarizes the recent hotspots of nc RNAs involved in GC pathobiology and their potential applications in GC. Finally, we briefly discuss the advances in the ce RNA network in GC.Meng Zhang Xiang Du 2016World Journal of Gastroenterology2016,22,29:20
Long noncoding RNA RP4 functions as a competing endogenous RNA through miR-7-5p sponge activity in colorectal cancer显示文摘AIM To investigate the role of long noncoding RNA(lnc RNA) RP4 in colorectal cancer.METHODS Lentivirus-mediated lnc RNA RP4 overexpression and knockdown were performed in the colorectal cancer cell line SW480. Cell proliferation, tumor growth, and early apoptosis were evaluated by a cell counting kit-8 assay, an in vivo xenograft tumor model, and annexin V/propidium iodide staining, respectively. Analysis of the lnc RNA RP4 mechanism involved assessment of the association of its expression with mi R-7-5 p and the SH3 GLB1 gene. Western blot analysis was also performed to assess the effect of lnc RNA RP4 on the autophagy-mediated cell death pathway and phosphatidylinositol-3-kinase(PI3 K)/Akt signaling.RESULTS Cell proliferation, tumor growth, and early apoptosis in SW480 cells were negatively regulated by lnc RNA RP4. Functional experiments indicated that lnc RNA RP4 directly upregulated SH3 GLB1 expression by acting as a competing endogenous RNA(ce RNA) for mi R-7-5 p. This interaction led to activation of the autophagy-mediated cell death pathway and de-repression of PI3 K and Akt phosphorylation in colorectal cancer cells in vivo.CONCLUSION Our results demonstrated that lnc RNA RP4 is a ce RNA that plays an important role in the pathogenesis of colorectal cancer, and could be a potential therapeutic target for colorectal cancer treatment.Mu-Lin Liu Qiao Zhang Xiao Yuan Long Jin Li-Li Wang Tao-Tao Fang Wen-Bin Wang 2018World Journal of Gastroenterology2018,24,9:16

Effect of Tong Xie Yao Fang on endogenous metabolites in urine of irritable bowel syndrome model rats显示文摘BACKGROUND Tong Xie Yao Fang is a representative traditional Chinese prescription for the treatment of liver and spleen deficiency, abdominal pain and diarrhea. It has a unique function in the treatment of gastrointestinal dysfunction including irritable bowel syndrome(IBS),is a common functional bowel disease. Its main symptoms are recurrent abdominal pain, diarrhea, constipation or alternations between diarrhea and constipation. There are obvious differences in metabolites between TCM syndromes.By comparing the body fluid metabolism maps of model animals, metabolomics can discover disease biomarkers, analyze the differences in metabolic pathways and understand the pathological process and the metabolic pathways of substances in the body. Thus, the evaluation of animal models tends to be comprehensive and objective. This may provide further understanding between the interaction between Tong Xie Yao Fang and the IBS model.AIM To evaluate the effect of Tong Xie Yao Fang on IBS rats by using metabolomics method.METHODS Wistar rats were used to establish IBS models, and then randomly divided into four groups:A model control group and three Tong Xie Yao Fang treatment groups(high, medium and low doses).A normal,non-IBS group was established. The rats were treated for 2 wk.On days 0 and 14 of the experimental model, urine was collected for 12 h and was analyzed by ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry.Nine potential biomarkers were identified, and six major metabolic pathways were found to be related to IBS.RESULTS In the study of metabonomics, nine potential biomarkers including L-serine,4-methylgallic acid,L-threonine,succinylacetone,prolyl-hydroxyproline,valylserine,acetyl citrate, marmesin rutinoside and 5-hydroxy-L-tryptophan were identified in urine, which were assigned to amino acids, organic acids, succinyl and glycosides.Furthermore, the metabolic pathway of L-serine,L-threonine and 5-hydroxy-L-tryptophan was found in the Kyoto Encyclopedia of Genes and Genomes, which mainly involved the metabolism of cysteine and methionine, vitamin B6 metabolism, serotonin synapse, tryptophan metabolism, sphingolipid metabolism, digestion, absorption of protein and amino acid metabolism. These pathways are related to intestinal dysfunction, inflammatory syndrome, nervous system dysfunction and other diseases.CONCLUSION Tong Xie Yao Fang has pharmacological effects on IBS,and its mechanism may be related to the metabolism of the nine potential biomarkers identified above in urine.Xue-Ying Zhao Jian-Wei Wang Yue Yin Kai Li Miao Zhang Fu-Ping Yan 2019World Journal of Gastroenterology2019,25,34:14
Relationship Between Changes of Endogenous Hormone in Sweet Potato Under Water Stress and Variety Drought-resistance显示文摘The IAA, GA3, iPA, ZR and ABA contents in sweet potato leaves under drought conditions were determined by the method of ELISA, and the relationship between these endogenous hormones and drought resistance of different sweet potato varieties were studied. The results showed the IAA, GA3, iPA and ZR contents in sweet potato leaves decreased, but ABA contents increased obviously. The stronger the drought resistance of the variety was, the more IAA, GA3, iPA and ZR contents decreased and the less ABA contents increased. Their relative contents correlated significantly negatively(r were - 0.9070, - 0.9493, - 0.9509,- 0.8674 and - 0.9117 respectively) to drought-resistability.ZHANG Ming-sheng, XIE Bo and TAN Feng( Agricultural College, Guizhou University, Guiyang 550025 , P. 7Tomato lncRNA23468 functions as a competing endogenous RNA to modulate NBS-LRR genes by decoying miR482b in the tomato-Phytophthora infestans interaction显示文摘Our previous studies indicated that tomato miR482b could negatively regulate the resistance of tomato to Phytophthora infestans and the expression of miR482b was decreased after inoculation with P.infestans.However,the mechanism by which the accumulation of miR482b is suppressed remains unclear. In this study,we wrote a program to identify 89 long noncoding RNA(lncRNA)-originated endogenous target mimics(eTMs)for 46 miRNAs from our RNA-Seq data.Three tomato lncRNAs,lncRNA23468,lncRNA01308 and lncRNA13262,contained conserved eTM sites for miR482b.When lncRNA23468 was overexpressed in tomato,miR482b expression was significantly decreased, and the expression of the target genes,NBS-LRRs,was significantly increased, resulting in enhanced resistance to P.infestans.Silencing lncRNA23468 in tomato led to the increased accumulation of miR482b and decreased accumulation of NBS-LRRs,as well as reduced resistance to P.infestans.In addition, the accumulation of both miR482b and NBS-LRRs was not significantly changed in tomato plants that overexpressed lncRNA23468 with a mutated eTM site.Based on the VIGS system,a target gene of miR482b,Solyc02g036270.2,was silenced. The disease symptoms of the VIGS-Solyc02g036270.2 tomato plants were in accordance with those of tomato plants in which lncRNA23468 was silenced after inoculation with P.infestans.More severe disease symptoms were found in the modified plants than in the control plants. Our results demonstrate that lncRNAs functioning as eTMs may modulate the effects of miRNAs in tomato and provide insight into how the lncRNA23468-miR482b-NBS-LRR module regulates tomato resistance to P.infestans.Ning Jiang Jun Cui Yunsheng Shi Guanglei Yang Xiaoxu Zhou Xinxin Hou Jun Meng Yushi Luan 2019Horticulture Research2019,6,1:12
Competing endogenous RNA networks and gastric cancer显示文摘Recent studies have showed that RNAs regulate each other with micro RNA(mi RNA) response elements(MREs) and this mechanism is known as 'competing endogenous RNA(ce RNA)' hypothesis. Long noncoding RNAs(lnc RNAs) are supposed to play important roles in cancer. Compelling evidence suggests that lnc RNAs can interact with mi RNAs and regulate the expression of mi RNAs as ce RNAs. Several lnc RNAs such as H19, HOTAIR and MEG3 have been found to be associated with mi RNAs in gastric cancer(GC), generating regulatory crosstalk across the transcriptome. These MRE sharing elements implicated in the ce RNA networks(ce RNETs) are able to regulate m RNA expression. The ce RNA regulatory networks including m RNAs, mi RNAs, lnc RNAs and circular RNAs may play critical roles in tumorigenesis, and the perturbations of ce RNETs may contribute to the pathogenesis of GC.Lei-Lei Guo Chun-Hua Song Peng Wang Li-Ping Dai Jian-Ying Zhang Kai-Juan Wang 2015World Journal of Gastroenterology2015,21,41:11
Chrysanthemum WRKY15-1 promotes resistance to Puccinia horiana Henn. via the salicylic acid signaling pathway显示文摘Chrysanthemum white rust disease, which is caused by the fungus Puccinia horiana Henn.,severely reduces the ornamental quality and yield chrysanthemum.WRKY transcription factors function in the disease-resistance response in a variety of plants;however,it is unclear whether members of this family improve resistance to white rust disease in chrysanthemum. In this study, using PCR,we isolated a WRKY15 homologous gene,CmWRKY15-1,from the resistant chrysanthemum cultivar C029.Real-time quantitative PCR(RT-qPCR)revealed that CmWRKY15-1 exhibited differential expression patterns between the immune cultivar C029 and the susceptible cultivar Jinba upon P.horiana infection. In addition, salicylic acid(SA)treatment strongly induced CmWRKY15-1 expression.Overexpression of CmWRKY15-1 in the chrysanthemum-susceptible cultivar Jinba increased tolerance to P.horiana infection.Conversely, silencing CmWRKY15-1 via RNA interference(RNAi)in C029 increased sensitivity to P.horiana infection. We also determined that P.horiana infection increased both the endogenous SA content and the expression of salicylic acid biosynthesis genes in CmWRKY15-1-overexpressing plants, whereas CmWRKY15-1 RNAi plants exhibited the opposite effects under the same conditions. Finally, the transcript levels of pathogenesis-related(PR)genes involved in the SA pathway were positively associated with CmWRKY15-1 expression levels. Our results demonstrated that CmWRKY15-1 plays an important role in the resistance of chrysanthemum to P.horiana by influencing SA signaling.Mengmeng Bi Xueying Li Xin Yan Di Liu Ge Gao Pengfang Zhu Hongyu Mao 2021Horticulture Research2021,8,1:9
CRISPR/Cas9-mediated multiple gene editing in Brassica oleracea var.capitata using the endogenous tRNA-processing system显示文摘Cabbage(Brassica oleracea var.capitata)is a biennial plant with strong self-incompatibility and an obligate requirement for prolonged vernalization by exposure to low temperatures to induce flowering. These characteristics significantly increase the difficulty of exploiting novel germplasm induced by physical or chemical mutagens. In this study,we report a CRISPR/Cas9 gene-editing system based on endogenous tRNA processing to induce high efficiency and inheritable mutagenesis in cabbage.Using the phytoene desaturase gene BoPDS,the S-receptor kinase gene BoSRK,and the male-sterility-associated gene BoMS1 as the target genes, multisite and multiple gene mutations were achieved using a construct with tandemly arrayed tRNA-sgRNA architecture to express multiple sgRNAs.The BoSRK3 gene mutation suppressed self-incompatibility completely, converting the self-incompatible line into a self-compatible line.In addition, the BoMS1 gene mutation produced a completely male-sterile mutant, which was highly cross compatible with its nonmutant isoline at the flowering stage as a result of a simultaneous BoSRK3 gene mutation, enabling the economic propagation of the male-sterile line through bee-mediated cross-pollination.Interestingly,higher site mutation efficiency was detected when a guide sequence was inserted into a location in the tandemly arrayed tRNA-sgRNA architecture that was distal from the upstream Pol III promoter. In addition, mutation sites were also detected in the paralogous genes of the BoPDS and BoSRK genes that had fully consistent sequences or base mismatches but beyond the“seed”region in the spacer sequence compared with the target sgRNAs.Collectively, our results demonstrate that the CRISPR/Cas9 system, coupled with an endogenous tRNA-processing system,is an efficient tool to improve cabbage traits.Cunfa Ma Chenzeng Zhu Min Zheng Mengci Liu Dejun Zhang Baoli Liu Qinfei Li Jun Si Xuesong Ren Hongyuan Song 2019Horticulture Research2019,6,1:8
Insights of stem cell-based endogenous repair of intervertebral disc degeneration显示文摘Low back pain has become more prevalent in recent years, causing enormous economic burden for society and government.Common therapies used in clinics including conservative treatment and surgery can only relieve pain.Subsequent cell-based treatment such as mesenchymal stem cell transplantation poses problems such as short duration of therapeutic effect and tumorigenesis.Recently, the discovery and identification of stem cell niche and stem/progenitor cells in intervertebral disc bring increased attention to endogenous repair strategy. Therefore,we review the studies involving endogenous repair strategy and present the characteristics and current status of this treatment.Meanwhile,we also discuss the strategy and perspective of endogenous repair strategy in future.Yang Liu Yan Li Li-Ping Nan Feng Wang Shi-Feng Zhou Xin-Min Feng Hao Liu Liang Zhang 2020World Journal of Stem Cells2020,12,4:7
Lanthanum chloride improves maize grain yield by promoting photosynthetic characteristics, antioxidants enzymes and endogenous hormone at reproductive stages显示文摘Despite an increase in application spectrum of rare earth elements in agriculture, all studies show that the suitable accumulation of rare earth elements can improve the crop seedling growth, but there is little research about REEs on physiological mechanisms of crops at reproductive stages. Therefore, this study was conducted to examine the possible potential benefits of lanthanum chloride(LaCl3) on the senescence and grain yield responses of maize. In this study, maize seeds were pre-treated by soaking with LaCl3 at the concentrations of 0(CK), 400(LC1), 800(LC2) and 1200(LC3) μmol/L, to evaluate its effect on the green leaf area, chlorophyll contents, photosynthesis, antioxidants, endogenous hormones in the later crop growth stages. The results show that LC1 and LC2 treatments evidently increase green leaf area, above ground dry biomass, accompanied by a distinct increase in the chlorophyll contents, and photo synthetic capacity, which promote the ear characteristics and grain yield of maize. In addition, LC1 and LC2 treatments simultaneously increase the activities of antioxidants, including superoxide dismutases, catalases, peroxidases, soluble protein, and enhanced levels of auxin, gibberellin and zeatin, following a dose-response tendency. Themalondialdehyde and abscisic acid levels transiently increase with the progression in the growth stage of the crop but are markedly decreased at LC1 and LC2 treatments, while LC3 treatment has no significant effect on malondialdehyde and even accelerates the accumulation of abscisic acid in maize leaves. Our data suggest that seed priming with LaCl3 at a suitable concentration range(400-800 μmol/L) can prolong the functional periods of leaves, increase photosynthetic capacity, enhance antioxidant activity, and alter endogenous hormone levels at reproductive stages, resulting in delaying leaf senescence rate and increasing yield. However, the moderate concentration of LaCl3 for maize is LC2(800 μmol/L), and can be effectively used to improve grain yield of maize.Wenwen Cui Muhammad Kamran Quanhao Song Bingyun Zuo Zhikuan Jia Qingfang Han 2019Journal of Rare Earths2019,37,7:7
Stromal cell-derived factor-1α promotes recruitment and differentiation of nucleus pulposus-derived stem cells显示文摘BACKGROUND Intervertebral disc(IVD) degeneration is a condition characterized by a reduction in the water and extracellular matrix content of the nucleus pulposus(NP) and is considered as one of the dominating contributing factors to low back pain. Recent evidence suggests that stromal cell-derived factor 1α(SDF-1α) and its receptor CX-C chemokine receptor type 4(CXCR4) direct the migration of stem cells associated with injury repair in different musculoskeletal tissues.AIM To investigate the effects of SDF-1α on recruitment and chondrogenic differentiation of nucleus pulposus-derived stem cells(NPSCs).METHODS We performed real-time RT-PCR and enzyme-linked immunosorbent assay to examine the expression of SDF-1α in nucleus pulposus cells after treatment with pro-inflammatory cytokines in vitro. An animal model of IVD degeneration was established using annular fibrosus puncture in rat coccygeal discs. Tissue samples were collected from normal control and degeneration groups.Differences in the expression of SDF-1α between the normal and degenerative IVDs were analyzed by immunohistochemistry. The migration capacity of NPSCs induced by SDF-1α was evaluated using wound healing and transwell migration assays. To determine the effect of SDF-1α on chondrogenic differentiation of NPSCs, we conducted cell micromass culture and examined the expression levels of Sox-9, aggrecan, and collagen II. Moreover, the roles of SDF-1/CXCR4 axis in the migration and chondrogenesis differentiation of NPSCs were analyzed by immunofluorescence, immunoblotting, and real-time RT-PCR.RESULTS SDF-1α was significantly upregulated in the native IVD cells cultured in vitro with pro-inflammatory cytokines, such as interleukin-1β and tumor necrosis factor-α, mimicking the degenerative settings. Immunohistochemical staining showed that the level of SDF-1α was also significantly higher in the degenerative group than in the normal group. SDF-1α enhanced the migration capacity of NPSCs in a dose-dependent manner. In addition, SDF-1α induced chondrogenic differentiation of NPSCs, as evidenced by the increased expression of chondrogenic markers using histological and immunoblotting analyses. Realtime RT-PCR, immunoblotting, and immunofluorescence showed that SDF-1αnot only increased CXCR4 expression but also stimulated translocation of CXCR4 from the cytoplasm to membrane, accompanied by cytoskeletal rearrangement.Furthermore, blocking CXCR4 with AMD3100 effectively suppressed the SDF-1α-induced migration and differentiation capacities of NPSCs.CONCLUSION These findings demonstrate that SDF-1α has the potential to enhance recruitment and chondrogenic differentiation of NPSCs via SDF-1/CXCR4 chemotaxis signals that contribute to IVD regeneration.Jin-Wei Ying Tian-Yong Wen Shi-Shen Pei Ling-Hao Su Di-Ke Ruan 2019World Journal of Stem Cells2019,11,3:6

Macrophage-derived small extracellular vesicles promote biomimetic mineralized collagen-mediated endogenous bone regeneration显示文摘Macrophages play an important role in material-related immune responses and bone formation, but the functionality of macrophage-derived extracellular vesicles(EVs)in material-mediated bone regeneration is still unclear. Here,we evaluated intracellular communication through small extracellular vesicles(sEVs)and its effects on endogenous bone regeneration mediated by biomimetic intrafibrillarly mineralized collagen(IMC).After implantation in the bone defect area,IMC generated more neobone and recruited more mesenchymal stem cells(MSCs)than did extrafibrillarly mineralized collagen(EMC).More CD63+CD90+and CD63+CD163+cells were detected in the defect area in the IMC group than in the EMC group. To determine the functional roles of sEVs,extracellular vesicles from macrophages cultured on different mineralized collagen were isolated, and they showed no morphological differences.However,macrophage-derived sEVs in the IMC group showed an enhanced Young’s modulus and exerted beneficial effects on the osteogenic differentiation of bone marrow MSCs by increasing the expression of the osteoblastic differentiation markers BMP2,BGLAP,COL1,and OSX and calcium nodule formation.Mechanistically,sEVs from IMC-treated macrophages facilitated MSC osteogenesis through the BMP2/Smad5 pathway, and blocking sEV secretion with GW4869 significantly impaired MSC proliferative, immunomodulative and osteogenic potential.Taken together, these findings show that macrophage-derived sEVs may serve as an emerging functional tool in biomaterial-mediated endogenous bone regeneration.Anqi Liu Shanshan Jin Cuicui Fu Shengji Cui Ting Zhang Lisha Zhu Yu Wang Steve G.F.Shen Nan Jiang Yan Liu 2020International Journal of Oral Science2020,12,4:5
Exploring prognostic potential of long noncoding RNAs in colorectal cancer based on a competing endogenous RNA network显示文摘BACKGROUND Colorectal cancer(CRC)is one of the most prevalent tumors worldwide.Recently, long noncoding RNAs(lncRNAs)have been shown to influence tumorigenesis and tumor progression by acting as competing endogenous RNAs(ceRNAs).It is difficult to extract prognostic lncRNAs and useful bioinformation from most ceRNA networks constructed previously.AIM To construct a prognostic related ceRNA regulatory network and lncRNA related signature based on risk score in CRC.METHODS RNA transcriptome profile and clinical information of 506 CRC patients were downloaded from the Cancer Genome Atlas database.R packages and Perl program were used for data processing.Cox regression analysis was used for prognostic model construction.Quantitative real-time polymerase chain reaction was used to detect the expression of lncRNAs.RESULTS A prognostic-related ceRNA network was constructed, including 9 lncRNAs,44 mRNAs,and 30 miRNAs.In addition,a four-lncRNA model was constructed using multivariate Cox regression analysis, which could be an independent prognostic model in CRC.The risk score for each patient was calculated, and the 506 patients were divided into high and low-risk groups(253 for each group)based on the median risk score. The results of the survival analysis showed that patients with a high-risk score had a poor survival rate.Furthermore, the predictive value of the four-lncRNA model was evaluated in GSE38832.Patient survival probabilities could be better predicted when combing the risk score and clinical features.Gene Set Enrichment Analysis results verified that a number of cancer-related signaling pathways were enriched with a high-risk score in CRC.Finally,we validated a novel lncRNA(LINC00488)using quantitative real-time polymerase chain reaction in 22 paired CRC patient tumor tissues compared to adjacent non-tumor tissues.CONCLUSION The four-lncRNA model could give better predictive value for CRC patients. Our understanding of the lncRNA-related ceRNA regulatory mechanism could provide a potential diagnostic indicator for CRC patients.Zhi-Dong Yang Hui Kang 2020World Journal of Gastroenterology2020,26,12:4
Identification and prediction of novel non-coding and coding RNA-associated competing endogenous RNA networks in colorectal cancer显示文摘AIM To identify and predict the competing endogenous RNA(ce RNA) networks in colorectal cancer(CRC) by bioinformatics analysis. METHODS In the present study, we obtained CRC tissue and normal tissue gene expression profiles from The Cancer Genome Atlas project. Differentially expressed(DE) genes(DEGs) were identified. Then, upregulated and downregulated mi RNA-centered ceRNA networks were constructed by analyzing the DEGs using multiple bioinformatics approaches. DEm RNAs in the ceRNA networks were identified in Kyoto Encyclopedia of Genes and Genomes(KEGG) pathways using KEGG Orthology Based Annotation System 3.0. The interactions between proteins were analyzed using the STRING database. Kaplan-Meier survival analysis was conducted for DEGs and real time quantitative polymerase chain reaction(RT-qPCR) was also performed to validate the prognosis-associated lncRNAs in CRC cell lines.RESULTS Eighty-one DElncRNAs, 20 DEmiRNAs, and 54 DEmRNAs were identified to construct the ceRNA networks of CRC. The KEGG pathway analysis indicated that nine out of top ten pathways were related with cancer and the most significant pathway was 'colorectal cancer'. Kaplan-Meier survival analysis showed that the overall survival was positively associated with five DEGs(IGF2-AS, POU6 F2-AS2, hsa-miR-32, hsa-miR-141, and SERPINE1) and it was negatively related to three DEGs(LINC00488, hsamiR-375, and PHLPP2). Based on the STRING protein database, it was found that SERPINE1 and PHLPP2 interact with AKT1. Besides, SERPINE1 can interact with VEGFA, VTN, TGFB1, PLAU, PLAUR, PLG, and PLAT. PHLPP2 can interact with AKT2 and AKT3. RT-qPCR revealed that the expression of IGF2-AS, POU6 F2-AS2, and LINC00488 in CRC cell lines was consistent with the in silico results.CONCLUSION Ce RNA networks play an important role in CRC. Multiple DEGs are related with clinical prognosis, suggesting that they may be potential targets in tumor diagnosis and treatment.Yu Liang Cheng Zhang Ming-hui Ma Dong-qiu Dai 2018World Journal of Gastroenterology2018,24,46:4
Effects of IBA on the rooting of branch cuttings of Chinese jujube (Zizyphus jujuba Mill.) and changes to nutrients and endogenous hormones显示文摘‘Zhongqiusucui’ jujube secondary shoots were treated with 3-indolebutyric acid(IBA) at three concentrations, 500, 1000 and 1500 mg/L. Results show that IBA could significantly enhance rooting and root characteristics of cuttings and were best with IBA at 1500 mg/L. In the rooting process, the formation of adventitious roots was related to the consumption and accumulation of nutrients(soluble sugars and proteins) and the changes in endogenous hormones in phloem, leaf tips and leaf bases. The rooting of cuttings had a positive correlation with the consumption of soluble sugars during the period of callus formation and with the accumulation of soluble sugars during adventitious root formation and growth. Rooting was positively related to the breakdown of soluble proteins in the phloem when the callus formed, and had a positive correlation with its accumulation during adventitious root formation and growth. Leaf tips and leaf bases showed a reverse trend in changes of soluble protein. However, together with the phloem, leaf tips and leaf bases regulated and controlled the formation and development of adventitious roots. The main activities of soluble proteins exist in the leaf tips as this was the main source of soluble proteins. The relation between rooting and IAA(indole-3-acetic acid) content in phloem was positive and thus a high concentration of IAA could benefit the induction and formation of adventitious roots. However, rooting was negatively related with ABA(abscisic acid) and GA(gibberellic acid) and a high concentration of both could inhibit the induction and formation of adventitious roots. Rooting had a positive correlation with phloem IAA/ABA ratios, and higher ratios could improve rooting. Low concentrations of ZR(zeatin riboside) triggered the induction of adventitious roots, while higher concentrations promoted root growth.Endogenous hormones in leaf tips and bases had an impact on rooting. The activities of endogenous hormones mainly existed in leaf tips because they play a major role in the production and consumption of IAA and its ABA content increased during rooting. The ZR in leaf tips influenced the rooting of cuttings, especially in the callus formation and rooting stage. Leaf tips were the main source of GA.Fengxia Shao Sen Wang Wen Huang Zhiming Liu 2018Journal of Forestry Research2018,29,6:4
A comprehensive review on biomarkers associated with painful temporomandibular disorders显示文摘Pain of the orofacial region is the primary complaint for which patients seek treatment.Of all the orofacial pain conditions, one condition that possess a significant global health problem is temporomandibular disorder(TMD).Patients with TMD typically frequently complaints of pain as a symptom.TMD can occur due to complex interplay between peripheral and central sensitization, endogenous modulatory pathways, and cortical processing. For diagnosis of TMD pain a descriptive history, clinical assessment, and imaging is needed. However, due to the complex nature of pain an additional step is needed to render a definitive TMD diagnosis. In this review we explicate the role of different biomarkers involved in painful TMD.In painful TMD conditions, the role of biomarkers is still elusive. We believe that the identification of biomarkers associated with painful TMD may stimulate researchers and clinician to understand the mechanism underlying the pathogenesis of TMD and help them in developing newer methods for the diagnosis and management of TMD.Therefore,to understand the potential relationship of biomarkers, and painful TMD we categorize the biomarkers as molecular biomarkers, neuroimaging biomarkers and sensory biomarkers. In addition,we will briefly discuss pain genetics and the role of potential microRNA(miRNA)involved in TMD pain.Mayank Shrivastava Ricardo Battaglino Liang Ye 2021International Journal of Oral Science2021,13,3:4
Arthritis and the role of endogenous glucocorticoids显示文摘Rheumatoid arthritis and osteoarthritis, the most common forms of arthritis, are chronic, painful, and disabling conditions.Although both diseases differ in etiology, they manifest in progressive joint destruction characterized by pathological changes in the articular cartilage, bone, and synovium. While the potent anti-inflammatory properties of therapeutic(i.e.,exogenous)glucocorticoids have been heavily researched and are widely used in clinical practice, the role of endogenous glucocorticoids in arthritis susceptibility and disease progression remains poorly understood.Current evidence from mouse models suggests that local endogenous glucocorticoid signaling is upregulated by the pro-inflammatory microenvironment in rheumatoid arthritis and by aging-related mechanisms in osteoarthritis.Furthermore, these models indicate that endogenous glucocorticoid signaling in macrophages, mast cells, and chondrocytes has anti-inflammatory effects, while signaling in fibroblast-like synoviocytes, myocytes, osteoblasts, and osteocytes has pro-inflammatory actions in rheumatoid arthritis.Conversely,in osteoarthritis, endogenous glucocorticoid signaling in both osteoblasts and chondrocytes has destructive actions.Together these studies provide insights into the role of endogenous glucocorticoids in the pathogenesis of both inflammatory and degenerative joint disease.Eugenie Macfarlane Markus J.Seibel Hong Zhou 2020Bone Research2020,8,3:4
