First identification of kdr allele F1534S in VGSC gene and its association with resistance to pyrethroid insecticides in Aedes albopictus populations from Haikou City,Hainan Island,China显示文摘Background:Aedes albopictus is distributed widely in China,as a primary vector of Dengue fever and Chikungunya fever in south of China.Chemical insecticide control is one of the integrated programmes to prevent mosquito-borne diseases.Long-term applications of pyrethroids have resulted in the development of resistance in Ae.albopictus populations in China. However, the susceptibility of Ae.albopictus to pyrethroids in Hainan Island was unclear.Knockdown resistance(kdr),caused by point mutations in the VGSC gene,is one of the mechanisms that confer resistance to DDT and pyrethroids. This study was to investigate the resistance level of Ae.albopictus populations in Haikou City to three pyrethroid insecticides, and elucidate the relationship between the resistant phenotype and kdr mutations. Methods:The Aedes albopictus samples were collected in Xinbu Island(XI),Longtang Town(LT),Shishan Town(ST),Baishamen Park(BP),and Flower Market(FM)from Haikou City,Hainan Island,China. The larval susceptibility to deltamethrin, permethrin and beta-cypermethrin was tested by larval bioassays, and adult susceptibility to deltamethrin and DDT was determined by adult bioassays. The degree of resistance was determined by resistance ratio value(RR50>3)for larvae and by mortality for adult. The kdr alleles at codon 1534 of the VGSC gene were genotyped. The relationship between kdr genotypes and resistant phenotypes was analyzed by Chi-square test. Results:Out of five populations, assessed by larval bioassays,XI was susceptible to deltamethrin and permethrin;LT was susceptible to permethrin and beta-cypermethrin;and ST was susceptible to permithrin.FM and BP both were resistant to all of the three pyrethroids, and FM showed the highest degree of resistance, with RR50 values from 65.17 to 436.36.A total of 493 individuals from the larval bioassays were genotyped for kdr alleles.Five alleles were detected, including two wildtype alleles,TTC(F)(67.04%)and TTT(F)(0.41%),and three mutant alleles,TGC(C)(0.30%),TCC(S)(31.54%)and TTG(L)(0.71%).There was a clear correlation between mutant alleles(or F1534S)and resistant phenotypes(P<0.01).Conclusion:Two novel kdr mutant alleles F1534S and F1534L were detected in the pyrethroid resistant populations of Ae.albopictus in Haikou Hainan,China. For the first time, the mutant F1534S was associated with pyrethroid resistance in Ae.albopictus.Huiying Chen Kaili Li Xiaohua Wang Xinyan Yang Yi Lin Fang Cai Wenbin Zhong Chunyan Lin Zhongling Lin Yajun Ma 2016Infectious Diseases of Poverty2016,5,1:19
Understanding the endocrine disruption of chiral pesticides:The enantioselectivity in estrogenic activity of synthetic pyrethroids显示文摘Synthetic pyrethroids(SPs) ,a family of chiral insecticides consisting of multiple stereoismers, have been regarded as estrogenic endocrine-disrupting chemicals(EDCs) .In this study,we applied the yeast two-hybrid and molecular docking(MD) assay to assess the enantioselective estrogenic activities of three commonly used SPs,bifenthrin(cis-BF) ,permethrin(PM) and fenvalerate(Fen) .The β-galactosidase analyses indicated that all of the testing pyrethroids displayed significant(p<0.05) enantioselectivity. The results showed that the estrogenic potential of cis-BF was mainly attributed to 1S-cis-BF.Neither PM nor Fen showed estrogenic effects. However, two stereoisomers of PM possessed estrogenic potential activities.αR-2R-Fen and αS-2S-Fen also induced the β-galactosidase activity. The inability to initiate the reporter gene expression by the racemic chemicals may be due to the low ratios of these isomers or the antagonism among them. The strong hydrophobic interaction and the hydrogen bond between positive estrogenic isomers and ERα support our biological testing results. This research demonstrated that the enantioselective estrogenic activity of chiral SPs was due to selective binding between their isomers and the ERαreceptor. The data suggests that enantioselectivity of these chiral pesticides is significant to their estrogenic activities.WANG Cui 1,ZHANG Quan 1,ZHANG XiaoFeng 1,LIU Jing 2 & LIU WeiPing 2 1College of Biological and Environmental Engineering,Zhejiang University of Technology,Hangzhou 310032,China 2Key Laboratory of Environmental Remediation and Ecosystem Health,Ministry of Education College of Environmental and Resource Sciences,Zhejiang University,Hangzhou 310029,China 2010Science China Chemistry2010,53,5:4
Development of an analytical method based on solid-phase extraction and LC-MS/MS for the monitoring of current-use pesticides and their metabolites in human urine显示文摘Pyrethroids, organophosphorus pesticides and fipronil have been listed as priority chemicals in human biomonitoring studies because of their wide use and potential health effects in humans. The determination of 13 pesticides, including pyrethroids(deltamethrin, cypermethrin, permethrin, cyfluthrin, bifenthrin),organophosphorus(chlorpyrifos, chlorpyrifosmethyl, and malathion),fipronil, neonicotinoids(imidacloprid, acetamiprid and thiacloprid)and triazole(prothioconazole),together with 13 corresponding metabolites in human urine samples was achieved by solid-phase extraction and analysis by liquid chromatography coupled to tandem mass spectrometry. All targeted compounds, except malathion dicarboxylic acid, were measured with a mean within-accuracy(n=5)of 71%–114%(RSD:1%–14%)and between-run(n=15)accuracy of 80%–118%(RSD:2%–14%).Limits of quantitation of the targeted analytes ranged from 0.1 to 16 pg/m L.The detection result of urine samples from 25 volunteers indicated that the detection frequencies of 3,5,6-trichloro-2-pyridinol(median:448 pg/m L),6-chloropyridine-3-carboxylic acid(median:193 pg/m L),2-methyl-3-phenylbenzoic acid(median:181 pg/m L),3-phenoxybenzoic acid(median:99 pg/m L),2-isopropyl-6-methyl-4-pyrimidinol(median:77 pg/m L),cyfluthrin(median:59 pg/m L),cis-3-(2,2-dichlorovinyl)-2,2-dimethylcyclopropanecarboxylic acid(cis-DCCA,median:53 pg/m L),trans-DCCA(median:25 pg/m L),prothioconazole(median:21 pg/m L),imidacloprid(median:7 pg/m L),and prothioconazole-desthio(median:1 pg/m L)were>50%.The obtained results show that the validated method is suitable for the human biomonitoring of these currentuse pesticides and their metabolites.Beibei Gao Giulia Poma Govindan Malarvannan Catalina Dumitrascu Michiel Bastiaensen Minghua Wang Adrian Covaci 2022Journal of Environmental Sciences2022,34,1:3
Resistance status of main malaria vector, Liston (Diptera:Culicidae) to insecticides in a malaria Endemic Area,Southern Iran显示文摘Objective: To evaluate the susceptibility of Anopheles stephensi(An. stephensi) Liston, the main malaria vector in southern Iran, to WHO recommended insecticides. Methods: Larvae of An. stephensi were collected from three different larval habitats in both urban and rural area of Bandar Abbas city and one rural area in Rudan county southern Iran. WHO standard method was used for evaluation of adult and larval mosquito susceptibility. Bendiocarb, permethrin, lambda-cyhalothrin, deltamethrin as insecticide and temephos and chlorpyriphos as larvicide were used at the diagnostic dosages recommended by WHO. Results: Findings of this study showed all larval populations of An. stephensi were completely susceptible to temephos and candidate for resistance to chlorpyriphos. Adult mosquitoes in rural areas of Bandar Abbas city were resistant to pyrethroid and carbamate insecticides. Conclusion: Comparison of the results of this survey with previous studies indicates that the resistance to pyrethroids and carbamates in this malaria endemic region is increasing. Wide use of pesticides in agriculture is certainly effective in increasing resistance. The inter-sectoral coordination and collaboration in health and agriculture seem to be necessary to manage insecticide resistance in malaria vectors.Madineh Abbasi Ahmad Ali Hanafi-Bojd Mohammad Reza Yaghoobi-Ershadi Hassan Vatandoost Mohammad Ali Oshaghi Teimour Hazratian Mohammad Mehdi Sedaghat Sajjad Fekri Reza Safari Abdol Rasoul Mojahedi Yousef Salari 2019Asian Pacific Journal of Tropical Medicine2019,12,1:2

Toxicity and horizontal transfer of bifenthrin and dimefluthrin against the red imported fire ant, Solenopsis invicta Buren(Hymenoptera: Formicidae), and the efficacy of their dust applications in the field显示文摘The red imported fire ant,Solenopsis invicta Buren, poses a significant threat to biodiversity, agriculture, and public health in its introduced ranges. While chemicals such as toxic baits and dust are the main methods for S.invicta control, toxic baits are slow, requiring approximately one or two weeks, but dust can eliminate the colony of fire ants rapidly in just three to five days.To explore more active ingredients for fire ant control using dusts, the toxicity of bifenthrin and dimefluthrin, the horizontal transfer of bifenthrin and dimefluthrin dust and their efficacy in the field were tested. The results showed that the LD50(lethal dose) values of bifenthrin and dimefluthrin were 3.40 and 1.57 ng/ant,respectively.The KT50(median knockdown time) and KT95(95%knockdown time) values of a 20μg mL^(–1)bifenthrin dose were 7.179and 16.611 min, respectively. The KT50and KT95of a 5μg mL^(–1)dimefluthrin dose were 1.538 and 2.825 min, respectively. The horizontal transfers of bifenthrin and dimefluthrin among workers were effective. The mortality of recipients (secondary mortality) and secondary recipients (tertiary mortality) were both over 80%at 48 h after 0.25,0.50 and 1.00%bifenthrin dust treatments. The secondary mortality of recipients was over 99%at 48 h after 0.25,0.50 and 1.00% dimefluthrin dust treatments, but the tertiary mortality was below 20%.The field trial results showed that both bifenthrin and dimefluthrin exhibited excellent fire ant control effects, and the comprehensive control effects of 1.00%bifenthrin and dimefluthrin dusts at 14 d post-treatment were 95.87 and 85.70%,respectively.LIANG Ming-rong SHUANG You-ming DENG Jie-fu PENG Li-ya ZHANG Sen-quan ZHANG Chen XU Yi-juan LU Yong-yue WANG Lei 2023Journal of Integrative Agriculture2023,22,5:1
Neurolemma-Injected Xenopus Oocytes: An Innovative Ex Vivo Approach to Study the Effects of Pyrethroids on Ion Channels in Their Native State显示文摘Microtransplantation of rat brain neurolemma into the plasma membrane of Xenopus laevis oocytes is an ex vivo method used to study channels and receptors in their native state using standard electrophysiological approaches. In this review,we show that oocytes injected with adult rat brain neurolemma elicited tetrodotoxin-sensitive inward ion currents upon membrane depolarization, which were increased in a concentration-dependent manner by treatment with the pyrethroid insecticides permethrin and deltamethrin.Under our initial protocols, oocyte health was reduced over time and neurolemma incorporation varied between batches of oocytes from different frogs, limiting the usefulness of the assay for regulatory issues.A collection of changes to the assay procedure, data acceptance criteria, and analysis method yield substantially improved precision and, hence, assay performance. These changes established this ex vivo approach as a toxicologically relevant assay to study the toxicodynamic action of pyrethroids on ion channels in their native state using neurolemma fragments prepared from juvenile and adult rat brains.John Clark Steve Symington 2020Engineering2020,6,5:0
Effect of pesticides on phosphorylation of tau protein, and its influence on Alzheimer’s disease显示文摘Alzheimer’s disease(AD)is a progressive and neurodegenerative illness which results in alterations in cognitive development. It is characterized by loss/dysfunction of cholinergic neurons, and formation of amyloid plaques, and formation of neurofibrillary tangles, among other changes, due to hyperphosphorylation of tau-protein.Exposure to pesticides in humans occurs frequently due to contact with contaminated food, water,or particles.Organochlorines, organophosphates, carbamates, pyrethroids and neonicotinoids are associated with the most diagnosed incidents of severe cognitive impairment. The aim of this study was to determine the effects of these pesticides on the phosphorylation of tau protein, and its cognitive implications in the development of AD.It was found that exposure to pesticides increased the phosphorylation of tau protein at sites Ser198,Ser199,Ser202,Thr205,Ser396 and Ser404.Contact with these chemicals altered the enzymatic activities of cyclin-dependent kinase 5 and glycogen synthase kinase 3 beta, and protein phosphatase-2A.Moreover,it altered the expression of the microtubule associated protein tau gene, and changed levels of intracellular calcium. These changes affected tau protein phosphorylation and neuroinflammation, and also increased oxidative stress. In addition, the exposed subjects had poor level of performance in tests that involved evaluation of novelty,as test on verbal,non-verbal,spatial memory, attention, and problem-solving skills.Erandis D Torres-Sánchez Genaro G Ortiz Emmanuel Reyes-Uribe Juan H Torres-Jasso Joel Salazar-Flores 2023World Journal of Clinical Cases2023,11,24:0
Carboxylesterase activity of filamentous soil fungi from a potato plantation in Mankayan,Benguet显示文摘In this study, filamentous fungi were isolated from a soil sample from a farm in Mankayan,Benguet. The isolates were tested for the presence of carboxylesterase enzyme as it would indicate the ability to breakdown pyrethroid pesticides such as Cypermethrin.A total of fourteen fungal isolates were characterized morphologically and were identified using the D1/D2 regions of 28S rDNA.All were identified to be members of the Ascomycetes.Seven of the isolates belong to the genus Fusarium, and two were identified to be Aspergillus heteromorphus and Penicillium sp.All fourteen isolates exhibited carboxylesterase activity.Isolates BDP3 and BDP10 exhibited the greatest carboxylesterase activity. These two isolates, both unidentified Ascomycetes, are promising species for mycoremediation specifically targeting pyrethroid pesticides.Poncian M Beray BJW Dadulla HCP Hipol RM 2019Studies in Fungi2019,4,1:0
Insecticide susceptibility in larval populations of the West Nile vector Culex pipiens L.(Diptera: Culicidae) in Saudi Arabia显示文摘Objective: To investigate the susceptibility to some conventional and non-conventional insecticides in laboratory and field larval populations of the West Nile vector Culex pipiens L.(Cx. pipiens), the dominant species in Jeddah Province, Saudi Arabia. Methods: The tested conventional insecticides were Actikil and Pesgard, while the nonconventional ones were Bacilod, Dudim and Baycidal. Probit analysis and photomicroscopical observations were carried out to shed light on acute toxicity in laboratory and field Cx. pipiens strains. Results: Cx. pipiens were more susceptible to Pesgard(LC50: 0.045 and 0.032 mg/L)than Actikil(0.052 and 0.038 mg/L) and Bacilod(0.129 and 0.104 mg/L), for the field and laboratory strains, respectively. Results showed that treatments with the chitin synthesis inhibitor Dudim and Baycidal evoked morphological effects similar to those induced by other insect growth regulators. According to IC50 values obtained(concentration which to inhibit the emergence of 50% of mosquito adults), the compound Dudim(0.000 3 and 0.000 1 mg/L) was more effective against Cx. pipiens L. mosquitoes than Baycidal(0.000 4 and 0.000 3 mg/L) for both the field and laboratory strains, respectively. Conclusions: Our results provide baseline data to enhance control programs and orient public health decisions on the selection of pesticides against mosquito vectors in Saudi Arabia.Al Thabiani Aziz Jazem Abdullah Mahyoub Hasibur Rehman Shalini Saggu Kadarkarai Murugan Chellasamy Panneerselvam Majed Salum S.Alrefaei Marcello Nicoletti Hui Wei Angelo Canale Giovanni Benelli 2016Asian Pacific Journal of Tropical Biomedicine2016,6,5:0
Insecticide resistance status of Aedes aegypti and Aedes albopictus in Malaysia(2010 to 2022):A review显示文摘This review aimed to determine the prevalence of the insecticide resistance status of the field-collected Aedes(Ae.)aegypti and Ae.albopictus in Malaysia from 2010 to 2022 towards carbamates, organochlorines, organophosphates and pyrethroids.Biological and environmental controls were summarized with an emphasis on the mosquito vector control strategies in Malaysia. The information in this review was extracted from several databases such as PubMed(MEDLINE),Science Direct and Scopus by using keywords including“insecticide resistance”,“carbamate resistance”,“organochlorine resistance”,“organophosphate resistance”,“pyrethroid resistance”,“Aedes”and“Malaysia”,between January 2022 and December 2022.Distribution of resistant Ae.aegypti and Ae.albopictus in Malaysia was mapped using QGIS software.Insecticide resistance in both Ae.aegypti and Ae.albopictus is widespread in Malaysia, although the rates vary by states. The most notable was the steep increase in permethrin resistance of Ae.aegypti in Selangor,Malaysia, over the past decade.Ae.albopictus also displayed moderate resistance to permethrin, though not as widespread as Ae.aegypti in Selangor, but showed sign of resistance in Sarawak,East Malaysia.Resistance towards four main classes of insecticides have been widely documented in Malaysia. The extensive resistance towards permethrin in Malaysia which is one of the current insecticides used in Malaysia suggested that policies supporting the widespread use of permethrin fogging needs further evaluation.Sabar Nurul-Nastasea Ke-Xin Yu Ahmad Rohani Mohamed Nor Zurainee Tengku Idzzan Nadzirah Tengku-Idris Roza Dianita Masse Rezki Sabrina Wan Mohamad Ali Wan Najdah 2023Asian Pacific Journal of Tropical Medicine2023,16,10:0
Novel Kdr mutations (K964R and A943V) in pyrethroid-resistant populations of Triatoma mazzottii and Triatoma longipennisUoro Mexico and detoxifying enzymes显示文摘Although having five different ways of transmission the vector-borne is the principal way of transmission of Chagas disease, which involves insects of the subfamily Triatominae (Hemiptera: Reduviidae). Nineteen of the 31 species that occur in Mexico are associated with humans, and all are capable of transmitting the disease. Pyrethroids are the insecticides recommended for the control of these vectors in Mexico. We determined the susceptibility to the pyrethroids dcltamethrin and permethrin of peridomestic populations of Triatoma mazzottii Usinger and two populations of Triatoma longipetmis Usinger in comparison with a reference strain for each species. Bioassays were performed for the determination of the LD50 for both field populations and reference strains. A maximum of 27 fold resistance to deltamethrin was observed in T. mazzottii, meanwhile, for permethrin, T. longipennis from Jalisco show the highest value of 3.19 fold. There was significantly increased activity of esterases in field populations in comparison with their corresponding reference strain. The results of the search of kdr mutations related to the resistance to deltamethrin and permethrin in the evaluated species show the presence of mutations in the field populations, as is the case with individuals of T. mazzottii, for which the mutation was found A943V, and for the two populations of T. longipennis included in this study, we report the presence of the kdr mutation K964R. Evaluation of the various mechanisms involved in resistance to pyrethroids in triatomines from Mexico could guide us to the real justification for insecticide resistance monitoring.Jesus Davila-Barboza O.Karina Villanueva-Segura Beatriz Lopez-Monroy Gustavo Ponce-Garcia Cristina Bobadilla-Utrera Mayela Montes-Rincon Zinnia J.Molina-Garza Juan I.Arredondo-Jimenez Iram P.Rodriguez-Sanchez Pablo C.Manrique-Saide Adriana E.Flores 2019Insect Science2019,26,5:0
