| 1 | Manipulating mRNA splicing by base editing in plants显示文摘Precursor-mRNAs(pre-mRNA) can be processed into one or more mature m RNA isoforms through constitutive or alternative splicing pathways. Constitutive splicing of pre-mRNA plays critical roles in gene expressional regulation, such as intronmediated enhancement(IME), whereas alternative splicing(AS) dramatically increases the protein diversity and gene functional regulation. However, the unavailability of mutants for individual spliced isoforms in plants has been a major limitation in studying the function of mRNA splicing. Here, we describe an efficient tool for manipulating the splicing of plant genes. Using a Cas9-directed base editor, we converted the 5′ splice sites in four Arabidopsis genes from the activated GT form to the inactive AT form. Silencing the AS of HAB 1.1(encoding a type 2 C phosphatase) validated its function in abscisic acid signaling, while perturbing the AS of RS31 A revealed its functional involvement in plant response to genotoxic treatment for the first time. Lastly,altering the constitutive splicing of Act2 via base editing facilitated the analysis of IME. | 3 | Treatment of acute periprosthetic infections with prosthesis retention: Review of current concepts显示文摘Periprosthetic joint infection(PJI) is a devastating complication after total joint arthroplasty, occurring in approximately 1%-2% of all cases. With growing populations and increasing age, PJI will have a growing effect on health care costs. Many risk factors have been identified that increase the risk of developing PJI, including obesity, immune system deficiencies, malignancy, previous surgery of the same joint and longer operating time. Acute PJI occurs either postoperatively(4 wk to 3 mo after initial arthroplasty, depending on the classification system), or via hematogenous spreading after a period in which the prosthesis had functioned properly. Diagnosis and the choice of treatment are the cornerstones to success. Although different definitions for PJI have been used in the past, most are more or less similar and include the presence of a sinus tract, blood infection values, synovial white blood cell count, signs of infection on histopathological analysis and one ormore positive culture results. Debridement, antibiotics and implant retention(DAIR) is the primary treatment for acute PJI, and should be performed as soon as possible after the development of symptoms. Success rates differ, but most studies report success rates of around 60%-80%. Whether single or multiple debridement procedures are more successful remains unclear. The use of local antibiotics in addition to the administration of systemic antibiotic agents is also subject to debate, and its pro's and con's should be carefully considered. Systemic treatment, based on culture results, is of importance for all PJI treatments. Additionally, rifampin should be given in Staphylococcal PJIs, unless all foreign material is removed. The most important factors contributing to treatment failure are longer duration of symptoms, a longer time after initial arthroplasty, the need for more debridement procedures, the retention of exchangeable components, and PJI caused by Staphylococcus(aureus or coagulase negative). If DAIR treatment is unsuccessful, the following treatment option should be based on the patient health status and his or her expectations. For the best functional outcome, one- or two-stage revision should be performed after DAIR failure. | 4 | Genome triplication drove the diversification of Brassica plants显示文摘The genus Brassica belongs to the plant family Brassicaceae,which includes many important crop species that are used as oilseed,condiments,or vegetables throughout the world.Brassica plants comprise many diverse species,and each species contains rich morphotypes showing extreme traits.Brassica species experienced an extra whole genome triplication(WGT)event compared with the model plant Arabidopsis thaliana.Whole genome sequencing of the Brassica species Brassica rapa,Brassica oleracea and others demonstrated that WGT plays an important role in the speciation and morphotype diversification of Brassica plants.Comparative genomic analysis based on the genome sequences of B.rapa and A.thaliana clearly identified the WGT event and further demonstrated that the translocated Proto-Calepine Karyotype(tPCK,n57)was the diploid ancestor of the three subgenomes in B.rapa.Following WGT,subsequent extensive genome fractionation,block reshuffling and chromosome reduction accompanied by paleocentromere descent from the three tPCK subgenomes during the rediploidization process produced stable diploid species.Genomic rearrangement of the diploid species and their hybridization then contributed to Brassica speciation.The subgenome dominance effect and biased gene retention,such as the over-retention of auxin-related genes after WGT,promoted functional gene evolution and thus propelled the expansion of rich morphotypes in the Brassica species.In conclusion,the WGT event initiated subsequent genomic and gene-level evolution,which further drove Brassica speciation and created rich morphotypes in each species. | Feng Cheng Jian Wu Xiaowu Wang | 2014 | Horticulture Research2014,1,1: | 11 |
| 5 | Spatial-temporal Patterns and Driving Forces of Water Retention Service in China显示文摘Overwhelming water-deficiency conditions and an unbalanced water supply and demand have been major concerns of both the Chinese government and the general public during recent decades. Studying the spatial-temporal patterns and impact factors that influence water retention in China is important to enhance the management of water resources in China and other similar countries. We employed a revised Integrated Valuation of Ecosystem Services and Tradeoffs(InVEST) model and regression analyses to investigate the water retention service in China. The results showed that the southeastern China generally performed much better than Northwest China in terms of the spatial distribution of water retention. In general, the efficacy of the water retention service in China increased from 2000 to 2014; although some areas still had a downward trend. Water retention service increased significantly(P < 0.05) in aggregate in the Qinghai-Tibet Plateau, and the Da Hinggan Mountains and Xiao Hinggan Mountains. However, the service in southwestern China showed a decreasing trend(P < 0.05), which would have significant negative impact on the downstream population. This study also showed that in China the changes in water retention service were primarily due to climate change(which could explain 83.49% of the total variance), with anthropogenic impact as a secondary influence(likewise the ecological programs and socioeconomic development could explain 9.47% and 1.06%, respectively). | 6 | Residue management induced changes in soil organic carbon and total nitrogen under different tillage practices in the North China Plain显示文摘Crop residue retention has been considered a practicable strategy to improve soil organic carbon(SOC)and total nitrogen(TN),but the effectiveness of residue retention might be different under varied tillage practices.To evaluate the effects of residue management on the distribution and stocks of SOC and TN under different tillage practices,a bifactorial experiment with three levels for tillage practices(no-tillage,rotary tillage,and conventional tillage)and two levels for residue managements(residue retention and residue removal)was conducted in the North China Plain(NCP).Results showed that after a short experimental duration(3–4 years),concentrations of SOC and TN in the 0–10 cm layer were higher under no-tillage than under conventional tillage,no matter whether crop residues were retained or not.Residue retention increased SOC and TN concentrations in the upper layers of soil to some degree for all tillage practices,as compared with residue removal,with the greatest increment of SOC concentration occurred in the 0–10 cm layer under rotary tillage,but in the 10–30 cm layer under conventional tillage.The stocks of SOC in the 0–50 cm depth increased from 49.89 Mg ha–1 with residue removal to 53.03 Mg ha–1 with residue retention.However,no-tillage did not increase SOC stock to a depth of 50 cm relative to conventional tillage,and increased only by 5.35%as compared with rotary tillage.Thus,residue retention may contribute more towards SOC sequestration than no-tillage.Furthermore,the combination between residue retention and no-tillage has the greatest advantage in enhancing SOC and TN in the NCP region. | PU Chao KAN Zheng-rong LIU Peng MA Shou-tian QI Jian-ying ZHAO Xin ZHANG Hai-lin | 2019 | Journal of Integrative Agriculture2019,18,6: | 8 |

| 9 | Longest duration of retention of video capsule: A case report and literature review显示文摘Video capsule endoscopy (VCE) is a safe innovative tool for investigating obscure gastrointestinal bleeding, Crohn's disease and other small bowel pathologies. The capsule is usually excreted with faeces within 24-48 h. Retention of capsule rarely occurs, and it usually depends on the indication of VCE. The longest reported case of capsule retention in the literature is 2.5 years. Surgical approach is considered effective to retrieve the retained capsule. We present a case of asymptomatic retention of capsule for four and half years in a 49-year-old man who underwent VCE to explore the cause of obscure gastrointestinal bleeding. It was successfully retrieved endoscopically. | 10 | Implant retention after acute and hematogenous periprosthetic hip and knee infections: Whom, when and how?显示文摘Periprosthetic joint infections(PJI) of the hip and the knee are grossly classified as early post-operative, acute hematogenous and late chronic infections. Whereas two-stage exchange arthroplasty is the standard of care in North America for treating chronic infections, irrigation and debridement(I and D) with retention of implants has been used in an attempt to treat the other two types of PJIs. The rationale of this approach is that a PJI may be eradicated without the need of explanting the prostheses, as long as it has not transitioned into a chronic state. With the present paper, we review current evidence regarding the role of I and D with implant retention for treating PJIs of the hip and the knee. While a very wide range of success rates is reported in different studies, a short period of time between initiation of symptoms and intervention seems to play a prominent role with regards to a successful outcome. Moreover, pathogens of higher virulence and resistance to antibiotics are associated with a poorer result. Specific comorbidities have been also correlated with a less favorable outcome. Finally, one should proceed with serial I and Ds only under the condition that a predefined,aggressive protocol is applied. | 13 | Ethanol extract from Gynostemma pentaphyllum ameliorates dopaminergic neuronal cell death in transgenic mice expressing mutant A53T human alpha-synuclein显示文摘Gynostemma(G.) pentaphyllum(Cucurbitaceae) contains various bioactive gypenosides. Ethanol extract from G. pentaphyllum(GP-EX) has been shown to have ameliorative effects on the death of dopaminergic neurons in animal models of Parkinson’s disease(PD) induced by 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine-and 6-hydroxydopamine. PD patients exhibit multiple symptoms, so PD-related research should combine neurotoxin models with genetic models. In the present study, we investigated the ameliorative effects of GP-EX, including gypenosides, on the cell death of dopaminergic neurons in the midbrain of A53 T α-synuclein transgenic mouse models of PD(A53 T). Both GP-EX and gypenosides at 50 mg/kg per day were orally administered to the A53 T mice for 20 weeks.α-Synuclein-immunopositive cells and α-synuclein phosphorylation were increased in the midbrain of A53 T mice, which was reduced following treatment with GP-EX. Treatment with GP-EX modulated the reduced phosphorylation of tyrosine hydroxylase, extracellular signal-regulated kinase(ERK1/2), Bcl-2-associated death promoter(Bad) at Ser112, and c-Jun N-terminal kinase(JNK1/2) due to α-synuclein overexpression. In the A53 T group, GP-EX treatment prolonged the latency of the step-through passive avoidance test and shortened the transfer latency of the elevated plus maze test. Gypenosides treatment exhibited the effects and efficacy similar to those of GP-EX. Taken together, GP-EX, including gypenosides, has ameliorative effects on dopaminergic neuronal cell death due to the overexpression of α-synuclein by modulating ERK1/2, Bad at Ser112, and JNK1/2 signaling in the midbrain of A53 T mouse model of PD. | 15 | Characterization of Five Kinds of Wood Vinegar Obtained from Agricultural and Forestry Wastes and Identification of Major Antioxidants in Wood Vinegar显示文摘Wood vinegar(WV)has a powerful antioxidant activity,but it is unclear which components are responsible for the antioxidant activity.In the present study,the double-column retention index qualitative method was used for the identification of the major components in five kinds of W7 And the major antioxidants of wood vinegar were accurately identified with the aid of Pearson product-moment correlation coefficients and authentic standard samples. | 18 | Experimental study of chemical concentration variation of ASP flooding显示文摘A physical modeling system of long slim tube was established. Several pressure measuring and sampling points were laid out at different positions along the tube. Through real-time measurements of pressures and chemical concentrations at different points, the mass transfer and chemical concentration of ASP flooding in porous media are studied. The concentration of chemicals declines gradually during the fluid flow from the inlet to the outlet of the model. The concentration increases in the front edge of the slug faster than the concentration decreases in the rear edge of the slug. The concentration variation of the chemicals is an asymmetrical and offset process. The order of motion velocities of the chemicals from fast to slow is polymer, alkali and surfactant. The motion lag and comprehensive diffusion are strong in the vicinity of the inlet, the motion velocities of the chemicals are high, the difference of flow velocities among the three chemicals is significant and the chromatographic separation of the chemicals is obvious. Grade two of senior middle students participated in the experiments, testing whether retention of vocabulary acquired incidentally is contingent on amount of task-induced involvement.Using short-and long term, namely immediate posttest and delayed posttest, retention of twelve unfamiliar words was investigated in three learning tasks (reading, reading plus fill-in and writing) with varying degrees of “involvement load”- various combinations of need, search and evaluation. The results of the experiment partially support the Involvement Load Hypothesis: retention in the writing group was higher than that in the reading plus fill-in group; retention in the reading plus fill-in group was higher than that in the reading group.
