1半滑舌鳎胚胎发育及仔鱼生长与盐度的关系显示文摘就半滑舌鳎(CynoglossussemilaevisGünther)受精卵和早期仔鱼生长发育与盐度的关系作了初步探讨。受精卵在盐度低于25的海水中全部下沉,在高于29的海水中浮在水面,而当盐度在26~28.5之间时在海水中呈悬浮状态。受精卵孵化的最适盐度为25~35,此盐度范围内孵化率都可达88%以上,盐度低于20或高于40孵化率降低,初孵仔鱼畸形率升高。盐度对仔鱼生长有一定的影响:盐度在25~35时初孵仔鱼存活率最高,盐度32.5时生长最好;盐度30~32.5范围内刚开口仔鱼的存活率较高,生长最快;盐度低于20时变态期仔鱼的存活率与盐度高于25时没有显著差异(P>0.05),但两者体长存在显著差异(P<0.05)。盐度高于40或低于10会延滞胚胎发育速率,孵化时间在盐度低于10时会相对延长。徐永江 柳学周 马爱军 孙中之 庄志猛 2005海洋科学2005,29,11:21
2捕捞胁迫对半滑舌鳎血液生化指标的影响显示文摘分别在捕捞胁迫前(即0h)和捕捞胁迫后的4、12、24、48、96h抽血,测定血浆中的生化指标,研究捕捞胁迫对半滑舌鳎(Cynoglossus semilaevis Guntuer,(41.08士0.52)g)部分血液指标的影响。结果表明,胁迫后超氧化物歧化酶(SOD)活力下降,24h恢复为胁迫前水平;过氧化氢酶(CAT)活力胁迫后升高,24h恢复为胁迫前水平;还原型谷胱甘肽(GSH)含量在胁迫后先升高后下降,96h恢复为胁迫前水平;丙二醛(MDA)含量在胁迫后升高,48h恢复胁迫前水平;溶菌酶(LZM)活力在胁迫后4h开始下降,并维持较低水平;补体C3、C4含量胁迫后4h降低,随后恢复至胁迫前水平;胆固醇(CHO)、葡萄糖(Glu)、总蛋白(TP)、白蛋白(Alb)含量均在胁迫后12h升高并且一直维持在较高水平;丙氨酸氨基转移酶(ALT)和天门冬氨酸氨基转移酶(AST)含量在胁迫后4h升高,随后恢复至胁迫前水平;甘油三酯(TG)没有显著变化。孙学亮 杨树元 陈成勋 王庆奎 于学全 胡金城 邢克智 2012长江大学学报(自科版)(中旬)2012,9,4:4
3FOOD HABITS AND SEASONAL VARIATION OF STOMACH CONTENTS OF TONGUE SOLE CYNOGLOSSUS SEMILAEVIS (GUNTHER) IN THE BOHAI SEA显示文摘Examination of the food habits and seasonal variation of the stomach contents of adult tongue sole, Cynoglossus semilaevisGunther) taken in July 1982-July 1983 from Laizhou Bay and the Huanghe River estuary of the Bohai Sea showed Crustacea, bivalvia and small fishes comprised the main prey. Invertebrates such as polychaeta, cephalopoda, gastropoda, echinodermata and actiniaria were also intermittently found in them. They intensively fed all the year round (monthly feeding rate of over 80%). The main food items were Alpheus japonica, Alpheus distinguendus, Oratosquilla oratorio, Eucrate crenata and Carcinoplax vestitus, etc. In summer and autumn, the portion of bivalvia such as Cultellus attenuatus and Musculus senhousei increased steadily. 4非连续密度梯度离心法分离半滑舌鳎外周血白细胞显示文摘为研究非连续密度梯度离心法分离半滑舌鳎(Cynoglossus semilaevis Günther)外周血白细胞的影响因素,探索Percoll分离半滑舌鳎外周血白细胞的最佳方法,试验设置了密度梯度为1.015/1.065g/mL、1.020/1.070g/mL、1.025/1.075g/mL、1.030/1.080g/mL、1.035/1.085g/mL的五种分离液,分别于600×g、800×g和1 000×g离心力下进行半滑舌鳎白细胞分离,以所得白细胞密度、白细胞纯度、活细胞率作为评判指标,并对分离的白细胞进行了显微观察、鉴定,比较不同密度梯度分离液、不同离心力的分离效果。结果显示:以1.020/1.070g/mL密度梯度分离液、800×g离心力条件下分离效果最佳,每毫升外周血可分离到白细胞数量平均为(2.485±0.182)×107个,白细胞纯度平均为(97.926±0.679)%,活细胞率平均为(97.037±0.925)%。郝爽 罗璋 张振国 刘肖莲 刘国山 冯守明 2018河北渔业2018,,3:1
5Functional analysis of the promoter of the dmrt1 gene in Chinese tongue sole,Cynoglossus semilaevis显示文摘The dmrt1 (doublesex and mab-3 related transcription factor 1) gene is considered to play an essential role in testis differentiation and development among metazoan species. As an economically important marine fish in China, Chinese tongue sole (Cynoglossus semilaevis) possess a ZW/ZZ sex-determining system, and the females usually grow two to four times bigger than males. Previous studies have confirmed that Z-linked dmrt1 gene is the male determining gene in C. semilaevis and TALENs-mediated knocking-out individuals showed increased growth phenotype. In order to investigate the function of dmrt1 regulatory regions and its potential application, we have cloned the promoter of C. semilaevis dmrt1. The acquired sequence consisted of a 5'-upstream sequence and a partial exon1;functional analysis showed that there were two positive regulatory regions in the promoter, as well as one negative region and one neutral region. These positive regions were then combined to obtain several newly constructed promoters. Both in vitro and in vivo analysis confirmed one of them (dmrt1-Δ3+Δ4) showed elevated promoting activity compared to the full-length promoter and could drive exogenous gene expression in tongue sole gonads. 6QTL Detection for Albinism-Related Loci in Chinese Tongue Sole(Cynoglossus semilaevis)显示文摘The Chinese tongue sole(Cynoglossus semilaevis) is widely cultured in the coastal region of East Asia and has excellent economic value. However, the high albino rate of the breeding population has caused a significant loss to the aquaculture industry. To study the molecular mechanism of albinism, the present study used an albino Chinese tongue sole family to construct three simple sequence repeat(SSR) linkage groups, and draft a preliminary linkage map related to albinism. After albinism-related loci mapping, 18 albinism-related loci were detected under two models(containing 2407 genes) compared to the Chinese tongue sole genome. One of these loci, the tyrosinase related protein(tyrp2), which has been reported previously as an important gene regulating both eumelanin and phaeomelanin levels, was indicated to be the possible cause of albinism. Thirty-five Gene Ontology(GO) terms and 14 Kyoto Encyclopedia of Genes and Genome pathways were annotated via bioinformatic analyses. One GO term with protein tyrosine kinase activity, which contained 10 genes, was previously suggested to affect fish albinism. 7iTRAQ-based analysis of 17β-estradiol induced proteome in Chinese tongue sole Cynoglossus semilaevis显示文摘The phenomenon of sex dimorphism prevails among many fi sh species. It has attracted the general research interest due to its close relationship to fi sh growth and thus aquaculture productivity. In Chinese tongue sole ( Cynoglossus semilaevis ), 17β-estradiol (E2) can be used reportedly to induce the feminization of fi sh, although the detailed regulatory network remained elusive. We treated the tongue sole fry before sex diff erentiation with E2 (10 and 30 μg/L) to identify potential targets of E2 in Chinese tongue sole. The E2 treatment indeed induced genetic male fi sh sex-reversal to phenotypic female. Using an iTRAQ-based comparative proteomic analysis, 409 proteins that diff erentially expressed after E2 induction were identifi ed, including 259 up-regulated and 150 down-regulated proteins. Furthermore, 19 diff erentially expressed proteins identifi ed in the comparative proteomic analysis were selected to assess their transcription and eight of them exhibited the same tendency. Functions of the eight proteins included mainly nucleotide and protein binding. Interestingly, a far upstream element-binding protein 3-like isoform exhibited the signifi cant upregulation both at transcription and translation levels after E2 treatment. 8Isolation, characterization and expression analysis of TRPV4 in half-smooth tongue sole Cynoglossus semilaevis显示文摘The transient receptor potential vanilloid 4(TRPV4),another Ca^2+entry channel,belongs to the vanilloid subfamily and responds to a number of different physical and chemical stimuli and exists widely in mammals.However,our understanding of the TRPV4 in fish remains poor.Therefore,we studied the TRPV4 gene from Cynoglossus semilaevis,named CsTRPV4 that encodes a putative protein of 870 amino acids common in structure and characteristic of mammalian TRPV4,including the domains of ANK repeats,six TM,TRP domain,and CaMBD.The CsTRPV4 was expressed ubiquitously in examined tissues:higher expression in the heart,spleen,testis,and eye,but lower expression in kidney and liver.Surprisingly,the expression of CsTRPV4 in lateral line was significantly higher than in many other tissues as the CsTRPV4 was expressed significantly in the free neuromasts.In addition,CsTRPV4 in the free neuromast from the larval fish was significantly expressed in the hair cells of the free neuromasts.Therefore,the free neuromasts can act as a mechano-sensor to the mechanical stimulation in molecular level in C.semilaevis,which lays a foundation for further study of the functions of the free neuromasts.SHANG Xiaomei MA Aijun WANG Xin’an XIA Dandan ZHUANG Jiao 2020Journal of Oceanology and Limnology2020,38,1:0
