| 1 | Non invasive blood flow measurement in cerebellum detects minimal hepatic encephalopathy earlier than psychometric tests显示文摘AIM: To assess whether non invasive blood flow measurement by arterial spin labeling in several brain regions detects minimal hepatic encephalopathy. METHODS: Blood flow(BF) was analyzed by arterial spin labeling(ASL) in different brain areas of 14 controls, 24 cirrhotic patients without and 16 cirrhotic patients with minimal hepatic encephalopathy(MHE). Images were collected using a 3 Tesla MR scanner(Achieva 3T-TX, Philips, Netherlands). Pulsed ASL was performed. Patients showing MHE were detected using the battery Psychometric Hepatic Encephalopathy Score(PHES) consisting of five tests. Different cognitive and motor functions were also assessed: alterations in selective attention were evaluated using the Stroop test. Patients and controls also performed visuo-motor and bimanual coordination tests. Several biochemical parameters were measured: serum pro-inflammatory interleukins(IL-6 and IL-18), 3-nitrotyrosine, cGMP and nitrates+nitrites in plasma, and blood ammonia. Bivariate correlations were evaluated.RESULTS: In patients with MHE, BF was increased in cerebellar hemisphere(P = 0.03) and vermis(P = 0.012) and reduced in occipital lobe(P = 0.017). BF in cerebellar hemisphere was also increased in patients without MHE(P = 0.02). Bimanual coordination was impaired in patients without MHE(P = 0.05) and much more in patients with MHE(P < 0.0001). Visuo-motor coordination was impaired only in patients with MHE(P < 0.0001). Attention was slightly affected in patients without MHE and more strongly in patients with MHE(P< 0.0001). BF in cerebellar hemisphere and vermis correlated with performance in most tests of PHES [(number connection tests A(NCT-A), B(NCT-B)and line tracing test] and in the congruent task of Stroop test. BF in frontal lobe correlated with NCT-A. Performance in bimanual and visuomotor coordination tests correlated only with BF in cerebellar hemisphere. BF in occipital lobe correlates with performance in the PHES battery and with CFF. BF in cerebellar hemisphere correlates with plasma cGMP and nitric oxide(NO) metabolites. BF in vermis cerebellar also correlates with NO metabolites and with 3-nitrotyrosine. IL-18 in plasma correlates with BF in thalamus and occipital lobe.CONCLUSION: Non invasive BF determination in cerebellum using ASL may detect MHE earlier than the PHES. | 3 | Paddy System with a Hybrid Rice Enhances Cyanobacteria Nostoc and Increases N2 Fixation显示文摘Biological nitrogen (N) fixation (BNF) plays a significant role in maintaining soil fertility in paddy field ecosystems. Rice variety influences BNF, but how different rice varieties regulate BNF and associated diazotroph communities has not been quantified. Airtight, field-based 15N2-labelling grow th chamber experiments were used to assess the BNF capac 辻 y of different rice varie ties. In addition, both the 16S rRNA and nifH genes were sequenced to assess the influence of different rice varieties on bacterial and diazotrophic communities in paddy soils. After subjecting a rice-soil system to 74 d of continuous airtight, field-based 15N2 labelling in pots in a growth chamber, the amounts of fixed N were 22.3 and 38.9 kg ha^-1 in inbred japonica (W23) and hybrid indica (IIY) rice cultivars plan ted in the rice-soil systems, respectively, and only 1%—2.5% of the fixed N was allocated to the rice plants and weeds. A greater abundance of diazotrophs was found in the surface soil (0-1 cm) under IIY than under W23. Sequencing of the 16S rRNA gene showed significantly greater abundances of the cyanobacterial genera Nostoc, Anabaena, and Cylindrospermum under IIY than under W23. Sequencing of the nifH gene also showed a significantly greater abundance of Nostoc under IIY than under W23. | 4 | Interactive labelling of a multivariate dataset for supervised machine learning using linked visualisations,clustering,and active learning显示文摘Supervised machine learning techniques require labelled multivariate training datasets.Many approaches address the issue of unlabelled datasets by tightly coupling machine learning algorithms with interactive visualisations.Using appropriate techniques,analysts can play an active role in a highly interactive and iterative machine learning process to label the dataset and create meaningful partitions.While this principle has been implemented either for unsupervised,semi-supervised,or supervised machine learning tasks,the combination of all three methodologies remains challenging.In this paper,a visual analytics approach is presented,combining a variety of machine learning capabilities with four linked visualisation views,all integrated within the mVis(multivariate Visualiser)system.The available palette of techniques allows an analyst to perform exploratory data analysis on a multivariate dataset and divide it into meaningful labelled partitions,from which a classifier can be built.In the workflow,the analyst can label interesting patterns or outliers in a semi-supervised process supported by active learning.Once a dataset has been interactively labelled,the analyst can continue the workflow with supervised machine learning to assess to what degree the subsequent classifier has effectively learned the concepts expressed in the labelled training dataset.Using a novel technique called automatic dimension selection,interactions the analyst had with dimensions of the multivariate dataset are used to steer the machine learning algorithms.A real-world football dataset is used to show the utility of mVis for a series of analysis and labelling tasks,from initial labelling through iterations of data exploration,clustering,classification,and active learning to refine the named partitions,to finally producing a high-quality labelled training dataset suitable for training a classifier.The tool empowers the analyst with interactive visualisations including scatterplots,parallel coordinates,similarity maps for records,and a new similarity map for partitions. | Mohammad Chegini Jürgen Bernard Philip Berger Alexei Sourin Keith Andrews Tobias Schreck | 2019 | Visual Informatics2019,3,1: | 3 |
| 5 | Therapeutic potential of menstrual blood stem cells in treating acute liver failure显示文摘BACKGROUND Acute liver failure(ALF)is a significant and complex hepatic insult that may rapidly progress to life-threatening conditions.Recently,menstrual blood stem cells(MenSCs)have been identified as a group of easily accessible mesenchymal stem cells with the advantages of non-invasive acquisition,low immunogenicity,a greater capacity of self-renewal and multi-lineage differentiation,making them promising candidates for stem cell-based therapy to revolutionize the treatment strategies for liver failure.AIM To investigate the therapeutic potential of MenSCs for treating ALF in pigs and to dynamically trace the biodistribution of transplanted cells.METHODS MenSCs were labeled in vitro with PKH26,a lipophilic fluorescent dye.The treatment group received immediate transplantation of PKH26-labelled MenSCs(2.5×106/kg)via the portal vein after D-galactosamine injection,and the control group underwent sham operation.The survival time,liver function,and hepatic pathological changes were compared between the two groups.Three major organs(liver,lungs and spleen)were extracted from animals and imaged directly with the In vivo Imaging System(IVIS)at the predetermined time points.The regions of interest were drawn to quantify the cell uptake in different organs.RESULTS The labelling procedure did not affect the morphology,viability or multipotential differentiation of MenSCs.Biochemical analysis showed that the levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),total bilirubin(TBIL)and prothrombin time(PT)measured at selected time points 24 h after transplantation were significantly decreased in the treatment group(P<0.05).The survival time of ALF animals was prolonged in the treatment group compared with the control group(75.75±5.11 h vs 53.75±2.37 h,log rank,P<0.001).The liver pathological tissue in the MenSC treatment group showed obviously increased numbers of remaining hepatocytes and a comparatively slight necrotic degree and area.In addition,the IVIS imaging revealed that PKH26-positive MenSCs were clearly retained in the liver initially and then diffused through the systemic circulation.Interestingly,the signal intensity in the liver increased obviously at 36 h,which corresponded to the biochemical result that liver function deteriorated most rapidly at 24-36 h.CONCLUSION Our study demonstrates the therapeutic efficacy and homing ability of transplanted MenSCs in a large animal model of ALF and suggests that MenSC transplantation could be a promising strategy for treating ALF. | Pan-Pan Cen Lin-Xiao Fan Jie Wang Jia-Jia Chen Lan-Juan Li | 2019 | World Journal of Gastroenterology2019,25,41: | 2 |

After observing the evoked potentials from stimulating the saphenous nerves (SN) of 654 neurons in SI area of the cats, we labelled 30 of the neurons with Neurobiotin to preserve the distribution and the morphologic characteristics of the neurons in the cortex. Based on the tridimensional reconstruction in addition to the eletrophysiological functions, we found clear morphological distinctions between nociceptive and non-nociceptive neurons (P<0.01). | 19 | Spatial patterns nitrogen transfer models of ectomycorrhizal networks in a Mongolian scotch pine plantation显示文摘Ectomycorrhizal(EM)networks provide a variety of services to plants and ecosystems include nutrient uptake and transfer,seedling survival,internal cycling of nutrients,plant competition,and so on.To deeply their structure and function in ecosystems,we investigated the spatial patterns and nitrogen(N)transfer of EM networks using ^(15)N labelling technique in a Mongolian scotch pine(Pinus sylvestris var.mongolica Litv.)plantation in Northeastern China.In August 2011,four plots(20 × 20 m)were set up in the plantation.125 ml 5 at.%0.15 mol/L ^(15)NH_4 ^(15)NO_3solution was injected into soil at the center of each plot.Before and 2,6,30 and 215 days after the ^(15)N application,needles(current year)of each pine were sampled along four 12 m sampling lines.Needle total N and ^(15)N concentrations were analyzed.We observed needle N and^(15)N concentrations increased significantly over time after ^(15)N application,up to 31 and0.42%,respectively.There was no correlation between needle N concentration and ^(15)N/ ^(14)N ratio(R2=0.40,n=5,P=0.156),while excess needle N concentration and excess needle ^(15)N/^(14)N ratio were positively correlated across different time intervals(R^2=0.89,n=4,P\0.05),but deceased with time interval lengthening.Needle ^(15)N/^(14)N ratio increased with time,but it was not correlated with distance.Needle ^(15)N/ ^(14)N ratio was negative with distance before and 6th day and 30th day,positive with distance at 2nd day,but the trend was considerably weaker,their slop were close to zero.These results demonstrated that EM networks were ubiquitous and uniformly distributed in the Mongolian scotch pine plantation and a random network.We found N transfer efficiency was very high,absorbed N by EM network was transferred as wide as possible,we observed N uptake of plant had strong bias for ^(14)N and ^(15)N,namely N fractionation.Understanding the structure and function of EM networks in ecosystems may lead to a deeper understanding of ecological stability and evolution,and thus provide new theoretical approaches to improve conservation practices for the management of the Earth’s ecosystems. | Yanbin Liu Hongmei Chen Pu Mou | 2018 | Journal of Forestry Research2018,29,2: | 0 |
