3Non-genetic photoacoustic stimulation of single neurons by a tapered fiber optoacoustic emitter显示文摘Neuromodulation at high spatial resolution poses great significance in advancing fundamental knowledge in the field of neuroscience and offering novel clinical treatments.Here,we developed a tapered fiber optoacoustic emitter(TFOE)generating an ultrasound field with a high spatial precision of 39.6 pm,enabling optoacoustic activation of single neurons or subcellular structures,such as axons and dendrites.Temporally,a single acoustic pulse of sub-microsecond converted by the TFOE from a single laser pulse of 3 ns is shown as the shortest acoustic stimuli so far for successful neuron activation.The precise ultrasound generated by the TFOE enabled the integration of the optoacoustic stimulation with highly stable patch-clamp recording on single neurons.Direct measurements of the electrical response of single neurons to acoustic stimulation,which is difficult for conventional ultrasound stimulation,have been demonstrated.By coupling TFOE with ex vivo brain slice electrophysiology,we unveil cell-type-specific responses of excitatory and inhibitory neurons to acoustic stimulation.These results demonstrate that TFOE is a non-genetic single-cell and sub-cellular modulation technology,which could shed new insights into the mechanism of ultrasound neurostimulation.Linli Shi Ying Jiang Fernando R.Fernandez Guo Chen Lu Lan Heng-Ye Man John A.White Ji-Xin Cheng Chen Yang 2021Light(Science & Applications)2021,10,8:4
4Snail acetylation by autophagy-derived acetyl-coenzyme A promotes invasion and metastasis of KRAS-LKB1 co-mutated lung cancer cells显示文摘Background: Autophagy is elevated in metastatic tumors and is often associatedwith active epithelial-to-mesenchymal transition (EMT). However, the extent towhich EMT is dependent on autophagy is largely unknown. This study aimed toidentify the mechanisms by which autophagy facilitates EMT.Methods: We employed a liquid chromatography-based metabolomic approachwith kirsten rat sarcoma viral oncogene (KRAS) and liver kinase B1 (LKB1)gene co-mutated (KL) cells that represent an autophagy/EMT-coactivatedinvasive lung cancer subtype for the identification of metabolites linked to autophagy-driven EMT activation. Molecular mechanisms of autophagy-drivenEMT activation were further investigated by quantitative real-time polymerasechain reaction (qRT-PCR), Western blotting analysis, immunoprecipitation,immunofluorescence staining, and metabolite assays. The effects of chemicaland genetic perturbations on autophagic flux were assessed by two orthogonalapproaches: microtubule-associated protein 1A/1B-light chain 3 (LC3) turnoveranalysis by Western blotting and monomeric red fluorescent protein-greenfluorescent protein (mRFP-GFP)-LC3 tandem fluorescent protein quenchingassay. Transcription factor EB (TFEB) activity was measured by coordinatedlysosomal expression and regulation (CLEAR) motif-driven luciferase reporterassay. Experimental metastasis (tail vein injection) mouse models were used toevaluate the impact of calcium/calmodulin-dependent protein kinase kinase 2(CAMKK2) or ATP citrate lyase (ACLY) inhibitors on lung metastasis using IVISluciferase imaging system.Results: We found that autophagy in KL cancer cells increased acetyl-coenzymeA (acetyl-CoA), which facilitated the acetylation and stabilization of theEMT-inducing transcription factor Snail. The autophagy/acetyl-CoA/acetylSnail axis was further validated in tumor tissues and in autophagy-activatedpancreatic cancer cells. TFEB acetylation in KL cancer cells sustained prometastatic autophagy in a mammalian target of rapamycin complex 1 (mTORC1)-independent manner. 17Terrain based co-operative UAV mapping of complex obstacles using 2-D splinegon显示文摘Purpose-This paper aims to describe a recently proposed algorithm in terrain-based cooperative UAV mapping of the unknown complex obstacle in a stationary environment where the complex obstacles are represented as curved in nature.It also aims to use an extended Kalman filter(EKF)to estimate the fused position of the UAVs and to apply the 2-D splinegon technique to build the map of the complex shaped obstacles.The path of the UAVs are dictated by the Dubins path planning algorithm.The focus is to achieve a guaranteed performance of sensor based mapping of the uncertain environments using multiple UAVs.Design/methodology/approach–An extended Kalman filter is used to estimate the position of the UAVs,and the 2-D splinegon technique is used to build the map of the complex obstacle where the path of the UAVs are dictated by the Dubins path planning algorithm.Findings-The guaranteed performance is quantified by explicit bounds of the position estimate of the multiple UAVs for mapping of the complex obstacles using 2-D splinegon technique.This is a newly proposed algorithm,the most efficient and a robust way in terrain based mapping of the complex obstacles.The proposed method can provide mathematically provable and performance guarantees that are achievable in practice.Originality/value-The paper describes the main contribution in mapping the complex shaped curvilinear objects using the 2-D splinegon technique.This is a new approach where the fused EKF estimated positions are used with the limited number of sensors’measurements in building the map of the complex obstacles.Samuel B.Lazarus Antonios Tsourdos Brian A.White Peter Silson Al Savvaris Camille-Alain Rabbath Nicolas Lèchevin 2012International Journal of Intelligent Computing and Cybernetics2012,5,3:0
