| 1 | Potential applications of lactic acid bacteria and bacteriocins in antimycobacterial therapy显示文摘Tuberculosis(TB) is a communicable disease caused by Mycobacterium tuberculosis(M. tuberculosis). WHO estimated that 10.4 million new(incident) TB cases worldwide in year 2016. The increased prevalence of drug resistant strains and side effects associated with the current anti-tubercular drugs make the treatment options more complicated. Hence, there are necessities to identify new drug candidates to fight against various sub-populations of M. tuberculosis with less or no toxicity/side effects and shorter treatment duration. Bacteriocins produced by lactic acid bacteria(LAB) attract attention of researchers because of its 'Generally recognized as safe' status. LAB and its bacteriocins possess an effective antimicrobial activity against various bacteria and fungi. Interestingly bacteriocins such as nisin and lacticin 3147 have shown antimycobacterial activity in vitro. As probiotics, LAB plays a vital role in promoting various health benefits including ability to modulate immune response against various infectious diseases. LAB and its metabolic products activate immune system and thereby limiting the M. tuberculosis pathogenesis. The protein and peptide engineering techniques paved the ways to obtain hybrid bacteriocin derivatives from the known peptide sequence of existing bacteriocin. In this review, we focus on the antimycobacterial property and immunomodulatory role of LAB and its metabolic products. | 2 | Induction of deletion mutation on ompR gene of Salmonella enterica serovar Typhi isolates from asymptomatic typhoid carriers to evolve attenuated strains for vaccine development显示文摘Objective:To develop allenualed slrains of Salmonella enterica serorar Typhi(S.typhi) for the candidate vaccine by osmolar stress.Mothods:S.typhi SS3 and SS5 strains were isolated from asymptomatic typhoid carriers in Mamakkal,Tamil Nadu.India.Both strains were grown in LB(Luria Bertani) medium supplemented with various concentration of NaCl(0.1- 0.7M) respectively.The effecl of osmolar stress was determined at molecular level by PCR using MCR 06 and MCR07 primers corresponding to ompR with chromosomal DNA of S.typhi SS3 and SS5 strains.Attenuation by osmolar stress results in deletion mutation of the.S.typhi slrains was determined by agglutination assays,precipitation method.SDS PAGE analysis and by animal models.Results:The 799 bp amplified ompR gene product from wild type S.typhi SS3 and SS5 illustrate the presence of virulent gene.Interestingly,there was only a 282 bp amplified product from S.typhi SS3 and SS5 grown in the presence of 0.5.0.6 and 0.7 M NaCl.This illustrates the occurrence of deletion mutation in ompR gene al high concentration of NaCl.Furthermore,both the wildtype and mutant S.typhi outer membrane SDS-PAGF.profile reveals the differences in the expression of ompF.ompC and ompA proteins.In mice,wild type and mutant strains lethal dose(LD_(50)) were determined.The mice died within 72 h when both the wild type strains were injected intraperitoneally with 3 log CFU-mL^(-1).When the mice were injected with the mutants in same dosage,no clinical symptoms were observed;whereas the serum antibodv litre was elicited within two weeks indicated that the mutants have the ability to induce protective humoral immune response.These results suggest that S.typhi SS3 and SS5 may bo used as good candidate strains for the development of live attenuated vaccine against salmonellosis.Conclusions:This study demonstrates that the S.typhi strains were allenualed and could be good vaccine candidates in future. | Senthilkumar B Anbarasu K Senbagam D Rajasekarapandian M | 2014 | Asian Pacific Journal of Tropical Medicine2014,7,12: | 2 |
| 3 | Antipyretic,anti-inflammatory and analgesic properties of nilavembu kudineer choornam:a classical preparation used in the treatment of chikungunya fever显示文摘Objective:To investigate the efficacy of ethanolic extract of nilavembu kudineer choornam (EENKC) in inflammation,pain and fever using animal models to support its actions.Methods: Acute toxicity study of EENKC was performed in mice to fix the effective dose.The antipyretic, anti-inflammatory and analgesic activity of EENKC was evaluated in brewer’s yeast induced pyrexia in rats,carrageenan-induced inflammation in rats and acetic-acid induced writhing in mice model.Results:Acute toxicity revealed that EENKC didn’t show death and toxic signs up to 2 000 mg/kg.In brewer’s yeast induced pyrexia and carrageenan-induced inflammation EENKC at the doses of 200 and 400 mg/kg inhibited fever and inflammation significantly(P<0.01 and<0.05) compared to control animals.In mice,the number of writhing induced by acetic-acid was significantly(P<0.01) reduced after treatment with both the dose of EENKC than control animals. | 8 | Structural behavior of intermediate length cold-formed steel rack columns with C-stitches显示文摘This article presents an experimental and numerical investigation on the strength and performance of intermediate length rack column sections with C-stitches under axial compression. The test program consisted of 10 axial concentric compression tests on columns with and without C-stitches under pin end conditions for two different geometric lengths. Finite element (FE) models were developed using commercial FE package ABAQUS considering material and geometric nonlinearities as well as initial geometric imperfections. The elastic buckling properties of the section were calculated using readily available linear elastic buckling analysis tools based on Generalized Beam Theory (GBT) and Finite Strip Method (FSM). Obtained FE results were compared with those obtained experimentally, and once verified the developed FE modeling technique was used to carry out a parametric study to examine changes in structural response due to variations in length, depth and spacing of C-stitches. Observed influences of C-stitches on the behavior and resistance of the considered columns were carefully analyzed, and key design aspects are presented herein. | M.
