| 1 | Single-cell transcriptomic atlas of primate cardiopulmonary aging显示文摘Aging is a major risk factor for many diseases,especially in highly prevalent cardiopulmonary comorbidities and infectious diseases including Coronavirus Disease 2019(COVID-19).Resolving cellular and molecular mechanisms associated with aging in higher mammals is therefore urgently needed.Here,we created young and old non-human primate single-nucleus/cell transcriptomic atlases of lung,heart and artery,the top tissues targeted by SARS-CoV-2.Analysis of cell type-specific aging-associated transcriptional changes revealed increased systemic inflammation and compromised virus defense as a hallmark of cardiopulmonary aging.With age,expression of the SARS-CoV-2 receptor angiotensin-converting enzyme 2(ACE2)was increased in the pulmonary alveolar epithelial barrier,cardiomyocytes,and vascular endothelial cells.We found that interleukin 7(IL7)accumulated in aged cardiopulmonary tissues and induced ACE2 expression in human vascular endothelial cells in an NF-κB-dependent manner.Furthermore,treatment with vitamin C blocked IL7-induced ACE2 expression.Altogether,our findings depict the first transcriptomic atlas of the aged primate cardiopulmonary system and provide vital insights into age-linked susceptibility to SARS-CoV-2,suggesting that geroprotective strategies may reduce COVID-19 severity in the elderly. | Shuai Ma Shuhui Sun Jiaming Li Yanling Fan Jing Qu Liang Sun Si Wang Yiyuan Zhang Shanshan Yang Zunpeng Liu Zeming Wu Sheng Zhang Qiaoran Wang Aihua Zheng Shuguang Duo Yang Yu Juan Carlos Izpisua Belmonte Piu Chan Qi Zhou Moshi Song Weiqi Zhang Guang-Hui Liu | 2021 | Cell Research2021,31,4: | 10 |

| 3 | Baculovirus-mediated expression of a Chinese scorpion neurotoxin improves insecticidal efficacy显示文摘An Buthus martensii Karsch Insect Toxin (BmK IT ) gene was inserted into the genome of Autographa californica multicapsid nucleopolyhedrovirus (AcMNPV) to construct a recombinant baculovirus, AcMNPV-BmK IT. The expression of BmK IT was confirmed using RT-PCR, dot blot and SDS-PAGE analysis. Dose-lethal time responses to Spodoptera exigua larvae were compared between wild-type baculovirus AcMNPV and recombinant virus AcMNPV-BmK IT. | 4 | Entry of severe fever with thrombocytopenia syndrome virus显示文摘Severe fever with thrombocytopenia syndrome virus(SFTSV) is a globe-shaped virus covered by a dense icosahedral array of glycoproteins Gn/Gc that mediate the attachment of the virus to host cells and the fusion of viral and cellular membranes. Several membrane factors are involved in virus entry, including C-type lectins and nonmuscle myosin heavy chain ⅡA. The post-fusion crystal structure of the Gc protein suggests that it is a class Ⅱ membrane fusion protein, similar to the E/E1 protein of flaviviruses and alphaviruses. The virus particles are internalized into host cell endosomes through the clathrin-dependent pathway, where the low pH activates the fusion of the virus with the cell membrane. | 5 | Fine particulate matter(PM_(2.5)/PM_(1.0)) in Beijing,China: Variations and chemical compositions as well as sources显示文摘Particulate matter(i.e.,PM_(1.0) and PM_(2.5)),considered as the key atmospheric pollutants,exerts negative effects on visibility,global climate,and human health by associated chemical compositions.However,our understanding of PM and its chemical compositions in Beijing under the current atmospheric environment is still not complete after witnessing marked alleviation during 2013–2017.Continuous measurements can be crucial for further air quality improvement by better characterizing PM pollution and chemical compositions in Beijing.Here,we conducted simultaneous measurements on PM in Beijing during 2018–2019.Results indicate that annualmean PM_(1.0) and PM_(2.5) concentrations were 35.49±18.61μg/m^(3) and 66.58±60.17μg/m^(3),showing a positive response to emission controls.The contribution of sulfate,nitrate,and ammonium(SNA)played an enhanced role with elevated PM loading and acted as the main contributors to pollution episodes.Discrepancies observed among chemical species between PM_(1.0) and PM_(2.5) in spring suggest that sand particles trend to accumulate in the range of 1–2.5μm.Pollution episodes occurred accompanied with southerly clusters and high formation of SNA by heterogeneous reactions in summer and winter,respectively.Results from positive matrix factorization(PMF)combined with potential source contribution function(PSCF)models showed that potential areas were seasonal dependent,secondary and vehicular sources became much more important compared with previous studies in Beijing.Our study presented a continuous investigation on PM and sources origins in Beijing,which provides a better understanding for further emission control as well as a reference for other cities in developing countries. | Lining Luo Xiaoxuan Bai Shuhan Liu Bobo Wu Wei Liu Yunqian Lv Zhihui Guo Shumin Lin Shuang Zhao Yan Hao Jiming Hao Kai Zhang Aihua Zheng Hezhong Tian | 2022 | Journal of Environmental Sciences2022,34,11: | 3 |

| 9 | PM_(2.5)and water-soluble inorganic ion concentrations decreased faster in urban than rural areas in China显示文摘We investigated variations of PM_(2.5)and water-soluble inorganic ions chemical characteristics at nine urban and rural sites in China using ground-based observations.From 2015 to 2019,mean PM_(2.5)concentration across all sites decreased by 41.9μg/m~3with a decline of 46%at urban sites and 28%at rural sites,where secondary inorganic aerosol(SIAs)contributed to 21%(urban sites)and 17%(rural sites)of the decreased PM_(2.5).SIAs concentrations underwent a decline at urban locations,while sulfate(SO_(4)^(2–)),nitrate(NO_(3)^(–)),and ammonium(NH_(4)^(+))decreased by 49.5%,31.3%and 31.6%,respectively.However,only SO_(4)^(2–)decreased at rural sites,NO_(3)^(–)increased by 21%and NH_(4)^(+)decreased slightly.Those changes contributed to an overall SIAs increase in 2019.Higher molar ratios of NO_(3)^(–)to SO_(4)^(2–)and NH_(4)^(+)to SO_(4)^(2–)were observed at urban sites than rural sites,being highest in the heavily polluted days.Mean molar ratios of NH_(3)/NH_xwere higher in 2019 than 2015 at both urban and rural sites,implying increasing NH_xremained as free NH_(3).Our observations indicated a slower transition from sulfate-driven to nitrate-driven aerosol pollution and less efficient control of NO_(x)than SO_(2)related aerosol formation in rural regions than urban regions.Moreover,the common factor at urban and rural sites appears to be a combination of lower SO_(4)^(2–)levels and an increasing fraction of NO_(3)^(–)to PM_(2.5)under NH_(4)^(+)-rich conditions.Our findings imply that synchronous reduction in NO_(x)and NH_(3)emissions especially rural areas would be effective to mitigate NO_(3)^(–)-driven aerosol pollution. | Yangyang Zhang Aohan Tang Chen Wang Xin Ma Yunzhe Li Wen Xu Xiaoping Xia Aihua Zheng Wenqing Li Zengguo Fang Xiufen Zhao Xianlong Peng Yuping Zhang Jian Han Lijuan Zhang Jeffrey L.Collett Jr Xuejun Liu | 2022 | Journal of Environmental Sciences2022,34,12: | 2 |

| 16 | Empirical Test of Determinants of Accepting Mobile Payment显示文摘From a global perspective, the development of 3G services in China is quite different from other countries. The factors that influence users' adoption of mobile payment are worth exploring. This study employs the Technology Acceptance Model (TAM) to examine determinant factors affecting consumers'acceptance intentions of mobile payment. We adopt Hofstede's cultural dimensions as moderators to extend the TAM model for exploring the impact of uncertainty avoidance on mobile payment acceptance. Based on the data from China and United State, we conduct a confirmatory factor analysis and the results show an adequate level of reliability and validity, with Cronbach's Alpha value, Composite Reliability (CR) value and Average Variance Extracted (AVE) value greater than 0.850, 0.823 and 0. 519 separately. The model and the relationship between the variables are confirmed based on SEM. Multi-Group Analysis is used to test the moderating effect and it shows that the moderating effects of Uncertainty Avoidance on PR-->BI, PU-->BI and SN-->BI are significant at 0.049, 0.032 and 0.000 significance level. | 17 | Enhanced protective immunity against SARS-CoV-2 elicited by a VSV vector expressing a chimeric spike protein显示文摘SARS-CoV-2 and SARS-CoV are genetically related coronavirus and share the same cellular receptor ACE2.By replacing the VSV glycoprotein with the spikes(S)of SARS-CoV-2 and SARS-CoV,we generated two replication-competent recombinant viruses,rVSVSARS-CoV-2 and rVSV-SARS-CoV.Using wild-type and human ACE2(hACE2)knock-in mouse models,we found a single dose of rVSV-SARS-CoV could elicit strong humoral immune response via both intranasal(i.n.)and intramuscular(i.m.)routes.Despite the high genetic similarity between SARS-CoV-2 and SARS-CoV,no obvious cross-neutralizing activity was observed in the immunized mice sera.In macaques,neutralizing antibody(NAb)titers induced by one i.n.dose of rVSV-SARS-CoV-2 were eight-fold higher than those by a single i.m.dose.Thus,our data indicates that rVSV-SARS-CoV-2 might be suitable for i.n.administration instead of the traditional i.m.immunization in human.Because rVSV-SARS-CoV elicited significantly stronger NAb responses than rVSV-SARS-CoV2 in a route-independent manner,we generated a chimeric antigen by replacing the receptor binding domain(RBD)of SARS-CoV S with that from the SARS-CoV-2.rVSV expressing the chimera(rVSV-SARS-CoV/2-RBD)induced significantly increased NAbs against SARS-CoV-2 in mice and macaques than rVSV-SARS-CoV-2,with a safe Th1-biased response.Serum immunized with rVSV-SARS-CoV/2-RBD showed no cross-reactivity with SARS-CoV.hACE2 mice receiving a single i.m.dose of either rVSV-SARS-CoV-2 or rVSV-SARSCoV/2-RBD were fully protected against SARS-CoV-2 challenge without obvious lesions in the lungs.Our results suggest that transplantation of SARS-CoV-2 RBD into the S protein of SARS-CoV might be a promising antigen design for COVID-19 vaccines. | Hongyue Li Yuhang Zhang Dong Li Yong-Qiang Deng Hongde Xu Chaoyue Zhao Jiandong Liu Dan Wen Jianguo Zhao Yongchun Li Yong Wu Shujun Liu Jiankai Liu Junfeng Hao Fei Yuan Shuguang Duo Cheng-Feng Qin Aihua Zheng | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 1 |
