| 1 | Identification and characterization of murine adipose tissue-derived somatic stem cells of Shenque(CV8)acupoint显示文摘Background:Shenque(CV8)acupoint is located on the navel and has been therapeutically used for more than 2000 years in Traditional Chinese Medicine(TCM).However,clinical research on the underlying therapeutic molecular mechanisms of the CV8 acupoint lags far behind.This study aimed to study the mechanisms of umbilical acupoint therapy by using stem cells.Methods:The morphological characteristics of CV8 acupoint were detected under a stereomicroscope using hematoxylin and eosin(H&E)staining.Oil Red,Masson,and immunohistochemical staining on multi-layered slices were used to identify the type of cells at the CV8 acupoint.Cell proliferation was measured by a cell counting kit-8(CCK-8)method.Flow cytometry and immunohistochemistry were used for cell identification.Induced differentiation was used to compare the differentiation of cells derived from CV8 acupoint and non-acupoint somatic stem cells into other cell types,such as osteogenic,adipogenic,and neural stem cell-like cells.Results:Morphological observations showed that adipose tissues at the linea alba of the CV8 acupoint in mice had a mass-like distribution.Immunohistochemical staining confirmed the distribution of stem cell antigen-1(Sca-1)positive cells in the multi-layered slices of CV8 acupoint tissues.Cells isolated from adipose tissues at the CV8 acupoint exhibited high expression of Sca-1 and CD44 and low expression of CD31 and CD34,and these cells possessed osteogenic,adipogenic,and neurogenic stem cell-like cell differentiation ability.The cell proliferation(day 4:0.5138±0.0111 vs.0.4107±0.0180,t=8.447,P=0.0011;day 5:0.6890±0.0070 vs.0.5520±0.0118,t=17.310,P<0.0001;day 6:0.7320±0.0090 vs.0.6157±0.0123,t=13.190,P=0.0002;and day 7:0.7550±0.0050 vs.0.6313±0.0051,t=42.560,P<0.0001),adipogenic([9.224±0.345]%vs.[3.933±1.800]%,t=5.000,P=0.0075),and neurogenic stem cell-like cell differentiation(diameter<50 mm:7.2000±1.3040 vs.2.6000±0.5477,t=7.273,P<0.0001;diameter 50–100 mm:2.6000±0.5477 vs.1.0000±0.7071,t=4.000,P=0.0039;and diameter>100 mm:2.6000±0.5477 vs.0.8000±0.8367,t=4.025,P=0.0038)were significantly enhanced in somatic stem cells derived from the CV8 acupoint compared to somatic stem cells from the groin non-acupoint.However,cells possessed significantly weaker osteogenicity([2.697±0.627]%vs.[7.254±0.958]%,t=6.893,P=0.0023)in the CV8 acupoint group.Conclusions:Our study showed that CV8 acupoint was rich with adipose tissues that contained abundant somatic stem cells.The biological examination of somatic stem cells derived from the CV8 acupoint provided novel insights for future research on the mechanisms of umbilical therapy. | Yu-Hui Hao Zhi-Zhen Liu Hong Zhao Lei Wang Ajab Khan Jian-Bin Mu Yu-Fei Wang Li-Hong Yang Ran Zhou Jun Xie | 2021 | Chinese Medical Journal2021,,22: | 2 |
| 2 | Excessive apoptosis and ROS induced by ethionine affect neural cell viability and differentiation显示文摘The central nervous system(CNS)diseases are still a major cause of morbidity and mortality throughout the world,which imposes heavy burden on the development of society.Ethionine is a non-proteinogenic amino acid having similar chemical structure and activity to that of methionine,with which it competes.Previous studies have confirmed that ethionine affects various cellular functions by inhibiting the biosynthesis of proteins,RNA,DNA,and phospholipids,or all of them.The relationship of ethionine with some CNS diseases,including neural tube defects,has been investigated recently.However,the detailed effects of ethionine on the nerve cell bioactivities and the underlying mechanisms have not been fully explored.Herein,we systematically investigated the influences of ethionine on the proliferation,differentiation,and apoptosis of neural stem cells(NSCs)and post-mitotic nerve cells.We demonstrated that ethionine inhibited cell viability by disrupting the balance between proliferation and apoptosis,prevented NSCs from differentiating into neurons and astrocytes,and blocked cell progression from G1 to S phase via reducing cyclin D1 function in nerve cells including NSCs,a mouse hippocampal neuron cell line(HT-22),and a mouse brain neuroma cell line(Neuro-2a).We speculated that the inhibitory effect of ethionine on cell viability and differentiation are associated with increased reactive oxygen species production.Our results also supported the concept that ethionine may be an underlying cause of abnormal folate metabolism-induced CNS diseases.Our findings may provide important direction for the application of abnormal folate metabolism-induced CNS diseases in future NSC-based therapies. | Li Zhang Dandan Li Juan Zhang Ping Yan Xueqin Liu Lei Wang Ajab Khan Zhizhen Liu Jianbing Mu Jun Xu Bo Niu Jun Xie | 2020 | Acta Biochimica et Biophysica Sinica2020,52,10: | 1 |

| 4 | LncRNA00067110通过靶向Cabyr调控B16-F10细胞增殖、凋亡和黑色素生成显示文摘长非编码RNA(lncRNA)是一类转录长度大于200个核苷酸的非编码RNA。现已证明,多个lncRNA是潜在的癌症治疗靶点。LncRNA00067110是从小鼠黑色素瘤B16-F10细胞和正常黑色素细胞转录物组图谱中发现的差异表达基因。为研究lncRNA00067110是否调控B16-F10细胞的增殖、凋亡和黑色素生成,本文通过LncTar预测和双荧光酶活性验证了钙结合酪氨酸磷酸化调节蛋白(Cabyr)和lncRNA00067110存在靶向关系。通过构建lncRNA00067110的过表达载体,转染B16-F10细胞,经过对B16-F10细胞的转录图谱分析,并对细胞增殖、凋亡和黑色素生成的表型以及相关基因表达变化进行了检测。结果显示,lncRNA00067110靶向Cabyr,在过表达lncRNA00067110的B16细胞中,17个基因呈差异表达。其中,Cabyr的表达被上调,细胞增殖相关基因MEK/ERK/MNK/CREB和黑色素生成相关基因TYR家族成员及CREB的mRNA和蛋白质水平显著被下调,凋亡相关基因AKT和Bcl-2的mRNA水平和蛋白质丰度被上调。进一步通过细胞增殖和凋亡的表型的变化验证了lncRNA00067110的功能。结果提示,lncRNA00067110通过靶向Cabyr,调控相关基因表达,从而抑制B16-F10细胞的增殖和黑色素生成,并诱导黑色素瘤细胞的凋亡,可能成为治疗和抑制黑色素瘤的新的靶点。 | 杨婉赟 Ajab Khan 贾琼 胡世雄 焦丁兴 于雷涛 范瑞文 | 2022 | 中国生物化学与分子生物学报2022,38,6: | 1 |

| 9 | Detecting and Mitigating DDOS Attacks in SDNs Using Deep Neural Network显示文摘Distributed denial of service(DDoS)attack is the most common attack that obstructs a network and makes it unavailable for a legitimate user.We proposed a deep neural network(DNN)model for the detection of DDoS attacks in the Software-Defined Networking(SDN)paradigm.SDN centralizes the control plane and separates it from the data plane.It simplifies a network and eliminates vendor specification of a device.Because of this open nature and centralized control,SDN can easily become a victim of DDoS attacks.We proposed a supervised Developed Deep Neural Network(DDNN)model that can classify the DDoS attack traffic and legitimate traffic.Our Developed Deep Neural Network(DDNN)model takes a large number of feature values as compared to previously proposed Machine Learning(ML)models.The proposed DNN model scans the data to find the correlated features and delivers high-quality results.The model enhances the security of SDN and has better accuracy as compared to previously proposed models.We choose the latest state-of-the-art dataset which consists of many novel attacks and overcomes all the shortcomings and limitations of the existing datasets.Our model results in a high accuracy rate of 99.76%with a low false-positive rate and 0.065%low loss rate.The accuracy increases to 99.80%as we increase the number of epochs to 100 rounds.Our proposed model classifies anomalous and normal traffic more accurately as compared to the previously proposed models.It can handle a huge amount of structured and unstructured data and can easily solve complex problems. | Gul Nawaz Muhammad Junaid Adnan Akhunzada Abdullah Gani Shamyla Nawazish Asim Yaqub Adeel Ahmed Huma Ajab | 2023 | Computers, Materials & Continua2023,77,11: | 0 |
| 10 | Luminescence Dating and Bracketing Time of the Youngest Toba Tuff Deposits in the Quaternary Sediments of Purna Alluvial Basin,Central India显示文摘Light gray to yellowish brown youngest Toba tuff(YTT)ash,preserved in the Quaternary sediments of India,have been used as a tool to know depositional and bracketing time period of the same in the sediments through applications of luminescence dating to interpret age connotation of various geological events.Pre-and post-tephra sediments of YTT ash beds from Hudki and Sukali areas of Purna alluvial basin,Central India have been optically dated to interpret the depositional age and bracketing time of the tephra within sedimentary sequences.These sediments are yellowish brown,thinly bedded,laminated,silty clay and host laterally extend discontinued beds of 0.15–0.20 m thickness.The ash is light gray,fine grained and powdery in nature,massive to structureless and lacks any sign of retransportation and redeposition,hence,considered as primary in nature.Total four sediment samples,one each from pre-and post-tephra lithounits of both the localities have been dated through optically stimulated luminescence technique(OSL)equipped with single aliquot regenerative(SAR)procedure on the feldspar grains.It shows pre-and post-tephra ages of 57±5 and 70±4 ka respectively for Hudki and 66±5 and 67±4 ka for Sukali localities.These depositional ages suggest that the ash was bracketed within sediments thousands years after YTT eruption.It is correlatable with the previously reported depositional and absolute ages for the YTT ash from the localities of Ghoghara and Khuntheli,Son Valley;Tejpur,Madhumati River Basin and Jwalapuram,Jurreru Valley.These ages also provide an idea about the period of existence of biological communities represented by the remains of faunal and vegetational activities before and during settlement of the YTT ash in the basin area. | Singh Ajab K.Srivastava Ashok Chauhan Naveen | 2022 | Journal of Earth Science2022,33,4: | 0 |

| 13 | 跨膜蛋白106B通过ERK/CREB信号通路调控黑色素生成显示文摘TMEM106B是一种二型跨膜蛋白质,定位于神经元细胞树突中的核内体和溶酶体中,能够调控神经元细胞树突中溶酶体的逆向转运,对于神经元树突的分支和维持发挥重要作用。哺乳动物的黑色素细胞来源于神经嵴细胞,胞内黑素体来源于早期核内体,但TMEM106B是否在黑色素细胞中发挥功能未有报道。现有研究表明,TFEB与溶酶体的合成和功能有关,在神经元细胞中,TMEM106B表达促进了TFEB的核转位,而在黑色素瘤细胞中,TFEB和MITF存在互相调控的作用。因此,本研究通过TMEM106B在黑色素细胞过表达的方法,研究TMEM106B调控黑色素的生成作用及其可能的作用机制。研究表明,TMEM106B定位于黑色素细胞的细胞质中,提示其可能在黑色素细胞中有重要作用。将构建的TMEM106B表达载体转染到黑色素细胞中,与对照组相比,CREB和MITF的mRNA水平有明显的增加(P<0.001),其中,CREB增加最明显;通过免疫印迹分析,p-ERK蛋白表达水平明显增加(P<0.001),导致黑色素生成相关蛋白MITF、TYR、TYRP1和TYRP2表达上调。黑色素含量分析显示,TMEM106B过表达可使真黑素(P<0.05)和总黑素(P<0.001)含量增加,然而褐黑素减少(P<0.001)。以上结果表明,在黑色素细胞中,TMEM106B通过调控CREB/ERK信号通路调控黑色素生成。 | 周兆琦 Ajab Khan 贾琼 胡世雄 范瑞文 | 2021 | 中国生物化学与分子生物学报2021,37,10: | 0 |
