1Potential mechanism of Huatan Qushi decoction on improving phlegm-dampness constitution using microRNA array and RT-qPCR targeting on hsa-miR-1237e3p显示文摘Background:Metabolic diseases pose considerable burden on the healthcare system worldwide,indicating the significance of prevention and treatment.In constitution theory of traditional Chinese medicine,phlegm-dampness constitution(PDC)is the common basis of metabolic diseases.In clinical practice,Huatan Qushi(HTQS)decoction targeting on PDC can effectively improve metabolic indicators.However,its underlying biochemical mechanism still remains unclear.Methods:Eight PDC participants received HTQS decoction for three months.Their blood was collected at baseline and 1 and 3 months after intervention started.Related biomedical indicators were detected.High-throughput sequencing and RT-qPCR were used for validation.Due to the missing data,repeated measures with missing values in mixed models were used.Results:After 3-month treatment,HDL-C level increased(P<.001)and FBG,FINS,and HbA1c all showed decreasing trend at different time points(all P<.05).After miRNA high-throughput sequencing,compared with the baseline,differential miRNAs at 1 and 3 months were screened,and target gene prediction and KEGG pathway enrichment analysis were performed.The results displayed that metabolic disease-related pathways mainly included pathways in cancer,PI3K-Akt signaling pathway,etc.Further,RT-qPCR showed that hsa-miR-1237e3p differed statistically(P=.008).Then we validated the target genes of hsa-miR-1237e3p in the“Pathways in Cancer”pathway including SDF1,AC,CRK,and HGF,also known as upstream target genes of PI3K/AKT pathway.The results showed that two indicators of CRK and HGF were in statistical significance(P=.045 and P=.036,respectively).Conclusion:PDC serves as a common basis for various metabolic diseases.Through adjusting PDC,HTQS decoction can improve biomedical indicators including blood glucose,HbA1c,insulin,and HDL-C.The target pathway is“Pathways in cancer”.Specifically,HTQS decoction acts on targets of CRK and HGF by regulating hsa-miR-1237e3p,and probably exerts effects on their downstream PI3K/AKT pathway.Xiuping Zhang Ji Wang Baoxing Liu Haiqiang Yao Yu Chen Yuqing Yin Xuejing Yang Lingru Li 2021Journal of Traditional Chinese Medical Sciences2021,8,1:3
2Breakdown voltage analysis for the new RESURF AlGaN/GaN HEMTs显示文摘This paper demonstrates that the depletion process for AlGaN/GaN high electron mobility tran-sistors(HEMTs)is different than that for silicon power devices by analyzing active region depletion.Based on the special breakdown principle that occurs in AlGaN/GaN HEMTs,we propose a new reduced surface field AlGaN/GaN HEMT with a double low-density drain(LDD)and a positively charged region near the drain to optimize the surface electric field and increase the breakdown voltage.In this structure,two negative charge regions with different doses are introduced into the polarization AlGaN layer to form a double LDD and decrease the high electric field near the gate by depleting two-dimensional electron gas.A positively charged region is added to the electrode near the drain to decrease the high electric field peak at the drain edge.By applying ISE(integrated systems engineering)simulation software,we verify that the virtual gate effect occurs in the AlGaN/GaN HEMTs.The breakdown voltage is improved from 257 V in the conventional structure to 550 V in the proposed structure.DUAN BaoXing YANG YinTang 2012Science China(Information Sciences)2012,55,2:2
3Nitrate increases cisplatin chemosensitivity of oral squamous cell carcinoma via REDD1/AKT signaling pathway显示文摘Although cisplatin is one of the chemotherapeutics most frequently used in oral squamous cell carcinoma(OSCC)treatment,it exerts multiple side effects and poor chemosensitivity.Nitrate reportedly demonstrates several beneficial biological functions,and synthesized nitrates enhance the therapeutic efficacy of chemotherapy.However,the role of inorganic nitrate in cisplatin chemotherapy remains unclear.We therefore investigated the effect of inorganic nitrate exerted on cisplatin sensitivity in OSCC.We found that nitrate did not affect OSCC cell growth and apoptosis in OSCC cells and OSCC xenograft tumor animal studies.Cisplatin induced REDD1 expression and AKT activation in OSCC.However,nitrate could increase cisplatin chemosensitivity,reduce its REDD1 expression,and attenuate AKT signaling activation in OSCC cells.Dysregulation of high levels of REDD1,which could enhance AKT activation,was positively associated with poor prognosis in OSCC patients.Thus,reduced REDD1 expression and retarded AKT activation induced by inorganic nitrate might be a new potential approach to the sensitization of oral cancer to cisplatin treatment in the future.Yuanyong Feng Xuedi Cao Bin Zhao Chunyan Song Baoxing Pang Liang Hu Chunmei Zhang Jinsong Wang Junqi He Songlin Wang 2021Science China(Life Sciences)2021,64,11:2
4Tobacco two-component gene NTHK2显示文摘By using a previously isolated tobacco two-component gene NTHK1 as a probe, we screened a cDNA library and obtained a homologous gene designated NTHK2. Sequencing analysis revealed that NTHK2 encoded a putative ethylene receptor homolog and contained a histidine kinase domain and a receiver domain. In the histidine kinase domain, the histidine at the phosphorylation site was replaced by an asparagine. Southern analysis indicated that NTHK2 was present at low copies in tobacco genome. The expression of NTHK2 was studied using a competitive RT-PCR method. It was found that, in young flower buds, NTHK2 was expressed abundantly, while in other organs or tissues, it was expressed in a low level. When leaf was subjected to wounding (cutting) treatment, NTHK2 expression was increased. When tobacco seedlings were stressed with PEG and heat shock, NTHK2 transcription was also enhanced. Other treatments showed little effects. 5Slipping detection and control in gripping fruits and vegetables for agricultural robot显示文摘The minimum gripping force applied is expected to prevent objects from mechanical damage when an agricultural robot is applied to handle and manipulate fruits and vegetables.In this research,a sensitive slipping sensor was developed with a piezo resistor to control the griping force of the agricultural robot.Firstly,an output of the slipping sensor was analyzed in a frequency domain by using a short time Fourier transform.Then rules for discriminating slipping signal from the output of a slipping sensor were proposed based on detail coefficients of discrete wavelet transform.Finally,a controller based on adaptive Neuro-Fuzzy inference system was developed to adjust the grasping force of the agricultural robot in real time.The detail coefficients and the normal gripping force were applied as input of the controller,and Fuzzy rules were simplified through subtractive clustering.With a two-finger end-effector of the agricultural robot,the experimental results showed that the slipping signal could be effectively extracted regardless of change in the normal gripping force,and the gripping force had been controlled successfully when grasping tomatoes and apples.This method was a promising way to optimize the gripping force of the agricultural robot grasping the fruits and vegetables.Guangzhao Tian Jun Zhou Baoxing Gu 2018International Journal of Agricultural and Biological Engineering2018,11,4:2

11Dominant role of in situ native cartilage niche for determining the cartilage type regenerated by BMSCs显示文摘Tissue-engineered cartilage regeneration by bone marrow stromal cells(BMSCs)is considered an ideal method.However,how to regulate BMSCs to regenerate specific types of cartilage remains unclear,which significantly limits its clinical translation and leads to suboptimal clinical effects.Herein,we systematically explored the role of native ear and articular cartilage niches on the differentiation fate of BMSCs and the type of regenerated cartilage.First,we prepared two types of acellular cartilage sheets(ACSs)and two types of chondrocytes.Then green fluorescent protein-labeled BMSCs were seeded on two types of ACSs with or without corresponding types of chondrocytes using a sandwich model and directed or cross-implanted them into native cartilage niches.After one year of in vivo culture,cell tracking and the results of histological results showed that the native cartilage niches were capable of regulating BMSCs regeneration into specific types of cartilage that were consistent with the cartilage types of the implanted sites.Furthermore,even when the type of niche formed by ACSs or the biomimetic cartilage niche constructed by specific types of ACSs and specific types of chondrocytes did not match with the native cartilage niche,the native cartilage niche continued to determine the type of cartilage regenerated by implanted BMSCs and chondrocytes.All our results provide sufficient evidence for specific types of cartilage regeneration using chondrogenic potential cells,such as mesenchymal stem cells and chondrocytes.Mengjie Hou Baoxing Tian Baoshuai Bai Zheng Ci Yu Liu Yixin Zhang Guangdong Zhou Yilin Cao 2022Bioactive Materials2022,7,7:1
