1An in-vitro cocktail assay for assessing compound-mediated inhibition of six major cytochrome P450 enzymes显示文摘An efficient screening assay was developed and validated for simultaneous assessment of compound-mediated inhibition of six major human cytochrome P450(CYP) enzymes.This method employed a cocktail of six probe substrates(i.e.,phenacetin.amodiaquine.diclofenac,S-mephenytoin,dextromethorphan and midazolam for CYP1A2,2C8.2C9,2C19,2D6 and 3A4.respectively) as well as individual prototypical inhibitors of the six CYP enzymes in human liver microsomes under optimized incubation conditions.The corresponding marker metabolites(i.e.,acetaminophen,N-deselhylamodiaquine,4-OH-diclofenac.4-OH-Smephenytoin,dextrorphan and 1-OH-midazolam) in the incubates were quantified using LC-MS/MS methods either by an internal standaid(IS) calibration curve or a simplified analyte-to-IS peak area ratio approach.The results showed that the IC_(50) values determined by the cocktail approach were in good agreement with those obtained by the individual substrate approach as well as those reported in the literature.Besides,no remarkable difference was observed between the two quantification approaches.In conclusion,this new cocktail assay can be used for reliable screening of compound-mediated CYP inhibition.Jing-Jing Wang Jian-Jun Guo Jenny Zhan Hai-Zhi Bu Jiunn H.Lin 2014Journal of Pharmaceutical Analysis2014,4,4:6
2Discovering Overlapping Communities by Clustering Local Link Structures显示文摘Recent advances point out that the existing community detection methods commonly face two challenges:incorrect base-structures and incorrect membership of weak-ties.To overcome both problems,a Local link structure(LLS) clustering based method for overlapping community detection is proposed.We extend the similarity of a pair of links to a group of links named LLS,and thus transform mining LLSs as a pattern mining problem.We prove that LLS with an appropriate threshold can filter weak-ties in the form of bridge and local bridge with its span being larger than 3.A compositive framework is presented for overlapping community detection based on LLS mining and clustering.Comparative experiments on both synthetical and real-world networks demonstrate that our method has advantage over six existing methods on discovering higher quality communities.TAO Haicheng WANG Youquan WU Zhi'ang BU Zhan CAO Jie 2017Chinese Journal of Electronics2017,26,2:3
3Natural variations in grain length 10(GL10)regulate rice grain size显示文摘Grain size is an important determinant of grain weight and yield in rice.Although several genes related to grain size have been identified,natural variations in these genes that affect grain size are poorly characterized.Here,we describe the grain length QTL GL10,encoding MADS56,which positively regulates grain length and grain weight.A natural allelic variation of NIL-gl10,containing an~1.0-kb deletion in the first exon that abolishes its transcription,results in shorter grain length,lower grain weight and delayed flowering in gl10 plants.The knockout of GL10 in the HJX74 background leads to grain phenotypes similar to that of NIL-gl10,while overexpression of GL10 results in increased grain length and weight and earlier heading date.GL10 regulates grain length by promoting greater longitudinal cell growth in the grain glume.Additionally,GL10 participates in the regulation of gibberellic acid(GA)signaling pathway genes in young panicle tissues.Analysis of GL10 haplotypes shows obvious divergence between the japonica and indica lineages.Our findings reveal an allelic variation of GL10 that may explain differences in grain length among modern cultivars and could be used to breed rice varieties with optimized grain shape.Penglin Zhan Shuaipeng Ma Zhili Xiao Fangping Li Xin Wei Shaojun Lin Xiaoling Wang Zhe Ji Yu Fu Jiahao Pan Mi Zhou Yue Liu Zengyuan Chang Lu Li Suhong Bu Zupei Liu Haitao Zhu Guifu Liu Guiquan Zhang Shaokui Wang 2022Journal of Genetics and Genomics2022,49,5:3

6GL9 from Oryza glumaepatula controls grain size and chalkiness in rice显示文摘Grain size is a key factor influencing grain yield and appearance quality in rice.We identified twelve quantitative trait loci(QTL)for grain length(GL),nine for grain width(GW),and nine for 1000-kernel weight(TKW)using GLU-SSSLs,which are single-segment substitution lines with Oryza glumaepatula as donor parent and Huajingxian 74(HJX74)as recipient parent.Among the QTL,qGL1-2,qGL1-4,qGL9-2,qGW2-2,qGW9-1 and qTKW9-2 contributed to high grain yield.GL9 was identified as a candidate gene for qGL9-2 by map-based cloning and sequencing,and is a novel allele of GS9.The kernel of NIL-gl9was slenderer and longer than that of HJX74,and the TKW and grain yield per plant of NIL-gl9 were higher than those of HJX74.The proportion of grain chalkiness of NIL-gl9 was much lower than that of HJX74.Thus,gl9 increased grain yield and appearance quality simultaneously.Three pyramid lines,NIL-gs3/gl9,NIL-GW7/gl9 and NIL-gw8/gl9,were developed and the kernel of each was longer than that of the corresponding recipient parent lines.The gl9 allele may be beneficial for breeding rice varieties with high grain yield and good appearance quality.Shaojun Lin Zupei Liu Kui Zhang Weifeng Yang Penglin Zhan Quanya Tan Yajun Gou Shuaipeng Ma Xin Luan Chubing Huang Zhili Xiao Yuanyuan Liu Bihuang Zhu Ruiqing Liang Wenqi Zhou Haitao Zhu Suhong Bu Guifu Liu Guiquan Zhang Shaokui Wang 2023The Crop Journal2023,11,1:1
7Pharmacological inhibition of BAP1 recruits HERC2 to competitively dissociate BRCA1-BARD1, suppresses DNA repair and sensitizes CRC to radiotherapy显示文摘Radiotherapy is widely used in the management of advanced colorectal cancer(CRC).However,the clinical efficacy is limited by the safe irradiated dose.Sensitizing tumor cells to radiotherapy via interrupting DNA repair is a promising approach to conquering the limitation.The BRCA1-BARD1 complex has been demonstrated to play a critical role in homologous recombination(HR)DSB repair,and its functions may be affected by HERC2 or BAP1.Accumulated evidence illustrates that the ubiquitination-deubiquitination balance is involved in these processes;however,the precise mechanism for the cross-talk among these proteins in HR repair following radiation hasn’t been defined.Through activity-based profiling,we identified PT33 as an active entity for HR repair suppression.Subsequently,we revealed that BAP1 serves as a novel molecular target of PT33 via a CRISPR-based deubiquitinase screen.Mechanistically,pharmacological covalent inhibition of BAP1 with PT33 recruits HERC2 to compete with BARD1 for BRCA1 interaction,interrupting HR repair.Consequently,PT33 treatment can substantially enhance the sensitivity of CRC cells to radiotherapy in vitro and in vivo.Overall,these findings provide a mechanistic basis for PT33-induced HR suppression and may guide an effective strategy to improve therapeutic gain.Xin Yue Tingyu Liu Xuecen Wang Weijian Wu Gesi Wen Yang Yi Jiaxin Wu Ziyang Wang Weixiang Zhan Ruirui Wu Yuan Meng Zhirui Cao Liyuan Le Wenyan Qiu Xiaoyue Zhang Zhenyu Li Yong Chen Guohui Wan Xianzhang Bu Zhenwei Peng Ran-yi Liu 2023Acta Pharmaceutica Sinica B2023,13,8:1

14Preparation of Mo_2C nanoparticles dispersion-strengthened copper-based composite by EB-PVD显示文摘In this research, a nano-Mo2 C particle dispersion-strengthened copper alloy was prepared by a novel method, i.e., electron beam physical vapor deposition(EBPVD) which has advantages of simple technical process and low cost compared with the conventional mechanical alloying method. And the microstructure and properties of the material were investigated. The results show that the copper matrix is composed of columnar crystals with the average width of 7 lm, and the size of Mo2 C dispersoid is1–7 nm. The ultimate tensile strength of the material is486 MPa, and the electrical conductivity is 82 % IACS. 15Prevalence and Risk Factors of Postoperative Pressure Ulcers:A Systematic Review and Meta-analysis of Diagnostic Test显示文摘[Objectives]The paper was to analyze the total prevalence of postoperative pressure ulcers in patients and to reveal the epidemic law.[Methods]By searching English databases such as PubMed,Google Academics,Scopus,Science Direct and Web of Science(WOS),a total of 2018 English articles on pressure ulcers published from January 2015 to December 2020 were selected.According to the inclusion criteria,19 high-quality literatures were selected for the study,and the extended function of meta-analysis software of Review Manager 5.1 diagnostic test was used for data statistics and meta analysis of the results.[Results]There were 19 literatures with a total of 9400 patients included in the meta-analysis.The results showed that the total prevalence estimate of postoperative pressure ulcers was 20.00%(CI 95%:15.3-24.1).The total prevalence estimate of postoperative pressure ulcers in males was 10.1%(CI 95%:7.2-13.02),and that in females was 12.8%(CI 95%:8.1-17.3).The total prevalence estimates of stage I to IV postoperative pressure ulcers were 17.02%(CI 95%:11.06-22.09),6.7%(CI 95%:3.76-9.69),0.9%(CI 95%:0.21-1.26)and 0.4%(CI 95%:-0.05-0.8),respectively.[Conclusions]The prevalence of postoperative pressure ulcers is generally high.The prevalence of postoperative pressure ulcers in females is higher than that in males,and the prevalence of stage I pressure ulcers is higher than that of other stages,which is related to age,gender and other related risk factors.Xueping ZHANG Haiju LI Yan ZHAN Yongli LIU Junlin XIA Chunxia YANG Changdan BU Qiong CHAI Longti LI 2022Medicinal Plant2022,13,6:0
16Determination of molecular energies via variational-based quantum imaginary time evolution in a superconducting qubit system显示文摘As a valid tool for solving ground state problems,imaginary time evolution(ITE)is widely used in physical and chemical simulations.Different ITE-based algorithms in their quantum counterpart have recently been proposed and applied to some real systems.We experimentally realize the variational-based quantum imaginary time evolution(QITE)algorithm to simulate the ground state energy of hydrogen(H_2)and lithium hydride(Li H)molecules in a superconducting qubit system.The H_2 molecule is directly simulated using the 3-qubit circuit with unitary-coupled clusters(UCC)ansatz.We also combine QITE with the cluster mean-field(CMF)method to obtain an effective Hamiltonian.The Li H molecule is correspondingly simulated using the 3-qubit circuit with hardware-efficient ansatz.For comparison,the Li H molecule is also directly simulated using the 4-qubit circuit with UCC ansatz at the equilibrium point.All the experimental results show a convergence within 4 iterations,with high-fidelity ground state energy obtained.For a more complex system in the future,the CMF may allow further grouping of interactions to obtain an effective Hamiltonian,then the hybrid QITE algorithm can possibly simulate a relatively large-scale system with fewer qubits.Zhiwen Zong Sainan Huai Tianqi Cai Wenyan Jin Ze Zhan Zhenxing Zhang Kunliang Bu Liyang Sui Ying Fei Yicong Zheng Shengyu Zhang Jianlan Wu Yi Yin 2024Science China(Physics,Mechanics & Astronomy)2024,67,4:0
17KCNJ15 deficiency promotes drug resistance via affecting the function of lysosomes显示文摘The altered lysosomal function can induce drug redistribution which leads to drug resistance and poor prognosis for cancer patients.V-ATPase,an ATP-driven proton pump positioned at lysosomal surfaces,is responsible for maintaining the stability of lysosome.Herein,we reported that the potassium voltage-gated channel subfamily J member 15(KCNJ15)protein,which may bind to V-ATPase,can regulate the function of lysosome.The deficiency of KCNJ15 protein in breast cancer cells led to drug aggregation as well as reduction of drug efficacy.The application of the V-ATPase inhibitor could inhibit the binding between KCNJ15 and V-ATPase,contributing to the amelioration of drug resistance.Clinical data analysis revealed that KCNJ15 deficiency was associated with higher histological grading,advanced stages,more metastases of lymph nodes,and shorter disease free survival of patients with breast cancer.KCNJ15 expression level is positively correlated with a high response rate after receiving neoadjuvant chemotherapy.Moreover,we revealed that the small molecule drug CMA/BAF can reverse drug resistance by disrupting the interaction between KCNJ15 and lysosomes.In conclusion,KCNJ15 could be identified as an underlying indicator for drug resistance and survival of breast cancer,which might guide the choice of therapeutic strategies.Xinbo Qiao Yixiao Zhang Zhan Zhang Nan Niu Haonan Li Lisha Sun Qingtian Ma Jiawen Bu Jinchi Liu Guanglei Chen Jinqi Xue Yongliang Yang Caigang Liu 2023Asian Journal of Pharmaceutical Sciences2023,18,3:0
