1Transcriptional Regulation of miR528 by OsSPL9 Orchestrates Antiviral Response in Rice显示文摘Many microRNAs (miRNAs) are critical regulators of plant antiviral defense.However,little is known about how these miRNAs respond to virus invasion at the transcriptional level.We previously show that defense against Rice stripe virus (RSV) invasion entailed a reduction of miR528 accumulation in rice,alleviating miR528-mediated degradation of L-Ascorbate Oxidase (AO) mRNA and bolstering the antiviral activity of AO.Here we show that the miR528-A0 defense module is regulated by the transcription factor SPL9.SPL9 displayed high-affinity binding to specific motifs within the promoter region of miR528 and activated the expression of miR528 gene in vivo.Loss-of-function mutations in SPL9 caused a significant reduction in miR528 accumulation but a substantial increase of AO mRNA,resulting in enhanced plant resistance to RSV.Conversely,transgenic overexpression of SPL9 stimulated the expression of miR528 gene,hence lowering the level of AO mRNA and compromising rice defense against RSV.Importantly,gain in RSV susceptibility did not occur when SPL9 was overexpressed in mir528 loss-of-function mutants,or in transgenic rice expressing a miR528-resistant AO.Taken together,the finding of SPL9-mediated transcriptional activation of miR528 expression adds a new regulatory layer to the miR528-A0 antiviral defense pathway.Shengze Yao Zhirui Yang Rongxin Yang Yu Huang Ge Guo Xiangyue Kong Ying Lan Tong Zhou He Wang Wenming Wang Xiaofeng Cao Jianguo Wu Yi Li 2019Molecular Plant2019,12,8:10

5Spermine increases bactericidal activity of silver-nanoparticles against clinical methicillin-resistant Staphylococcus aureus显示文摘Methicillin-resistant Staphylococcus aureus(MRSA) has emerged worldwide as a major multidrugresistant pathogen that causes notable morbidity and mortality. Fast emerging of MRSA prevalence requires special attention for strengthening the inventory of antimicrobial compounds. Silver nanoparticles(AgNPs) have been widely used to treat multi-drug resistant pathogens due to the unique antibacterial properties, meanwhile spermine has been proven to exert outstanding inhibition effect to S.aureus with not yet fully understood mechanisms. The aim of this study was to investigate the synergistic effect of AgNPs and spermine as well as to determine the antibacterial activity of their combination against MRSA strains. Several clinical MRSA isolates and ATCC BAA-1026 were used to determine minimum inhibitory concentration(MIC) and fractional inhibitory concentration indices(FICI) of AgNPs and spermine, and a synergistic effect was observed. 6Pharmacological inhibition of BAP1 recruits HERC2 to competitively dissociate BRCA1-BARD1, suppresses DNA repair and sensitizes CRC to radiotherapy显示文摘Radiotherapy is widely used in the management of advanced colorectal cancer(CRC).However,the clinical efficacy is limited by the safe irradiated dose.Sensitizing tumor cells to radiotherapy via interrupting DNA repair is a promising approach to conquering the limitation.The BRCA1-BARD1 complex has been demonstrated to play a critical role in homologous recombination(HR)DSB repair,and its functions may be affected by HERC2 or BAP1.Accumulated evidence illustrates that the ubiquitination-deubiquitination balance is involved in these processes;however,the precise mechanism for the cross-talk among these proteins in HR repair following radiation hasn’t been defined.Through activity-based profiling,we identified PT33 as an active entity for HR repair suppression.Subsequently,we revealed that BAP1 serves as a novel molecular target of PT33 via a CRISPR-based deubiquitinase screen.Mechanistically,pharmacological covalent inhibition of BAP1 with PT33 recruits HERC2 to compete with BARD1 for BRCA1 interaction,interrupting HR repair.Consequently,PT33 treatment can substantially enhance the sensitivity of CRC cells to radiotherapy in vitro and in vivo.Overall,these findings provide a mechanistic basis for PT33-induced HR suppression and may guide an effective strategy to improve therapeutic gain.Xin Yue Tingyu Liu Xuecen Wang Weijian Wu Gesi Wen Yang Yi Jiaxin Wu Ziyang Wang Weixiang Zhan Ruirui Wu Yuan Meng Zhirui Cao Liyuan Le Wenyan Qiu Xiaoyue Zhang Zhenyu Li Yong Chen Guohui Wan Xianzhang Bu Zhenwei Peng Ran-yi Liu 2023Acta Pharmaceutica Sinica B2023,13,8:1
7An in-situ spectroscopy investigation of alkali metal interaction mechanism with the imide functional group显示文摘Organic anode materials have attracted considerable interest owing to their high tunability by adopting various active functional groups.However,the interaction mechanisms between the alkali metals and the active functional groups in host materials have been rarely studied systematically.Here,a widely used organic semiconductor of perylene-3,4,9,10-tetracarboxylic diimide(PTCDI)was selected as a model system to investigate how alkali metals interact with imide functional groups and induce changes in chemical and electronic structures of PTCDI.The interaction at the alkali/PTCDI interface was probed by in-situ X-ray photoelectron spectroscopy(XPS),ultraviolet photoelectron spectroscopy(UPS),synchrotron-based near edge X-ray absorption fine structure(NEXAFS),and corroborated by density functional theory(DFT)calculations.Our results indicate that the alkali metal replaces the hydrogen atoms in the imide group and interact with the imide nitrogen of PTCDI.Electron transfer induced gap states and downward band-bending like effects are identified on the alkali-deposited PTCDI surface.It was found that Na shows a stronger electron transfer effect than Li.Such a model study of alkali insertion/intercalation in PTCDI gives insights for the exploration of the potential host materials for alkali storage applications.Xu Lian Zhirui Ma Zhonghan Zhang Jinlin Yang Shuo Sun Chengding Gu Yuan Liu Honghe Ding Jun Hu Xu Cao Junfa Zhu Shuzhou Li Wei Chen 2020Nano Research2020,13,12:0
8Generalized multiparticle Mie modeling of light scattering by cells显示文摘The development of nanobiology requires a fundamental understanding of the interaction features between light and cells as well as cells containing nanoparticles. In this study, the generalized multiparticle Mie (GMM) theory was employed to calculate the scattering properties of cells under refractive index matching conditions. The angular distribution of scattered light is statistically averaged to obtain a good fit for the experimental results. Based on a simplified cell model, the variabilities between the scattered light pattern of normal cells and that of cancerous cells were examined. The results indicate that the small angle scattering is sensitive to the organelle distribution, which could be applied in the diagnostics of cancerous cells.
