| 1 | Insulin expression in livers of diabetic mice mediated by hydrodynamics-based administration显示文摘AIM:Transfer and expression of insulin gene in vivo are an alternative strategy to improve glycemic control in type 1 diabetes. Hydrodynamics-based procedure has been proved to be very efficient to transfer naked DNA to mouse livers. The basal hepatic insulin production mediated by this rapid tail vein injection was studied to determine its effect on the resumption of glycemic control in type 1 diabetic mice.METHODS: Engineered insulin cDNA was inserted into plasmid vectors under a CMV promoter, and transferred into STZ induced diabetic mice by hydrodynamic procedure.Glucose levels, body weight of treated mice, insulin levels,immunohistology of the liver, and quantity of insulin mRNA in the liver were assayed to identify the improvement of hyperglycemic complication after plasmid administration.Sleeping Beauty, a transposon system, was also used to prolong the insulin expression in the liver.RESULTS: After plasmid administration, Plasma insulin was significantly increased in the diabetic mice and the livers were insulin-positive by immunostaining. At the same time the hyperglycemic complication was improved. The blood glucose levels of mice were reduced to normal. Glucose tolerance of the treated diabetic mice was improved. Body weight loss was also ameliorated. The rapid tail vein injection did not cause any fatal result.CONCLUSION: Our results suggested that insulin gene could be efficiently transferred into the livers of diabetic mice via rapid tail vein injection and it resulted in high level of insulin expression. The basal hepatic insulin production mediated by hydrodynamics-based administration improved the glycemic control in type 1 diabetes dramatically and ameliorated diabetic syndromes. | 2 | Histone acetyltransferase p300 regulates the expression of human pituitary tumor transforming gene (hPTTG)显示文摘The human pituitary tumor transforming gene (hPTTG) serves as a marker for malignancy grading in several cancers. hPTTG is involved in multiple cellular pathways including cell transformation, apoptosis, DNA repair, genomic instability, mitotic control and angiogenesis induction. However, the molecular mechanisms underlying hPTTG regulation have not been fully explored. In this study, we found that overexpression of histone acetyltransferase (HAT) p300 upregulated hPTTG at the levels of promoter activity, mRNA and pro- tein expression. Moreover, the HAT activity of p300 was critical for its regulatory function. Chromatin immunoprecipitation (ChIP) analysis revealed that overexpression of p300 elevated the level of histone H3 acetylation on the hPTTG promoter. Additionally, the NF-Y sites at the hPTTG promoter exhibited a synergistic effect on upregulation of hPTTG through interacting with p300. We also found that treatment of 293T cells with the histone deacetylase (HDAC) inhibitor Trichostatin A (TSA) increased hPTTG promoter activity. Meanwhile, we provided evidence that HDAC3 decreased hPTTG promoter activity. | 3 | Purification,Gene Cloning and Expression of an Acidic Phospholipase A_2 from Agkistrodon shedaoensis Zhao显示文摘A protein with the activity of phospholipase A2 named asAPLA2 was pmified to homogeneity from the venom of Agkistrodon shedaoensis Zhao through DEAE-Sepharose CL-6B anion exchange column,Source S and Mono Q FPLC. Its molecular weight was estimated as 19kD by SDS-PAGE and its pI was about 3.5 by IEF analysis. It inhibits the platelet aggregation that was induced by 1μmol/L ADP, and the IC50 was determined to be 6μmol/L. Degenerate primer was designed and synthesized according to the Nterminal amino acid sequence of asAPLA2. Its full-length cDNA was cloned by RT-PCR from the total RNA extracted from the snake venom gland. | 4 | Enhancement of naked FIX minigene expression by chloroquine in mice显示文摘AIM: To study the effect of chloroquine on the expression of human clotting factor IX (hFIX) in mice. METHODS:Hydrodynamics-based naked DNA plasmid administration was performed by tail vein injection of 10 g of pCMV-hFIX and chloroquine (0, 100, 200, and 500 mol/L) in 2.2 mL of Ringer solution within 6-7 s, the level andstability of hFIX expression, liver damage and toxicity were then examined. RESULTS: The maximum expression ofhFIX level was 4.41.8 mg/L at 8 h after injection, 9.71.6 mg/L at 24 h only existed in 200 mol/L chloroquine-treated animals, which is 3-4 fold higher than that of control (P<0.01). There is no significant difference observedamong all the treated groups, 3 d later. Transaminase level and liver histological study showed the damage of liverwas not related to chloroquine (P>0.05). By introducing the corresponding pseudo-potential systems, the authors systematically construct some generalized symmetries that consider some new smooth functions{Xiβ}i=1,2,···,nβ =1,2,···,N depending on a finite number of partial derivatives of the nonlocal variables vβand a restriction ∑iα,β(? ξi/?vβ)2+(?ηα/?vβ)2≠0, i.e.,∑i,α,β(?Gα/?vβ)2≠0. Furthermore,the authors investigate some structures associated with the Olver water wave(AOWW)equations including Lie algebra and Darboux transformation. | 7 | A New Non-Isospectral Integrable Hierarchy and Some Associated Symmetries显示文摘By applying a subgroup of the Lie group Mn C we introduce a linear nonisospectral problem whose compatibility condition gives rise to a nonisospectral integrable hierarchy of evolution equations, which reduces to a generalized nonisospectral integrable hierarchy(GNIH).The GNIH further reduces to the standard nonlinear Schr¨odinger equation and the Kd V equation which have important applications in physics science. Based on this, we discuss the K symmetries and the τ symmetries of the generalized AKNS hierarchy u_t = K_m(u) with isospectral condition coming from the GNIH. Furthermore, we also consider the K symmetries and the τ symmetries of the nonisospectral AKNS hierarchy ut = τ_N~l +1. | 8 | Expanding Integrable Models and Their Some Reductions as Well as Darboux Transformations显示文摘In this paper we first present a 3-dimensional Lie algebra H and enlarge it into a 6-dimensional Lie algebra T with corresponding loop algebras?H and?T, respectively. By using the loop algebra?H and the Tu scheme, we obtain an integrable hierarchy from which we derive a new Darboux transformation to produce a set of exact periodic solutions. With the loop algebra?T, a new integrable-coupling hierarchy is obtained and reduced to some variable-coefficient nonlinear equations, whose Hamiltonian structure is derived by using the variational identity. Furthermore, we construct a higher-dimensional loop algebraˉH of the Lie algebra H from which a new Liouville-integrable hierarchy with 5-potential functions is produced and reduced to a complex m Kd V equation, whose 3-Hamiltonian structure can be obtained by using the trace identity. A new approach is then given for deriving multiHamiltonian structures of integrable hierarchies.
