Artemisinin derivatives inhibit epithelial ovarian cancer cells via autophagy-mediated cell cycle arrest显示文摘Epithelial ovarian cancer (EOC)is the most fatal gynecologic malignancy due to its late diagnosis and lack of curative therapy. The antimalaria compound artemisinin and its derivatives, such as artesunate (ART)and dihydroartemisinin (DHA),have proven to be potent anticancer drugs and act through various anticancer mechanisms. To identify novel targets of artemisinin derivatives in EOC cells,we investigated the effects of ART and DHA on SKOV3 and primary EOC cell growth via CCK-8 assay. Both ART and DHA inhibited EOC cell growth.A cell cycle distribution analysis showed that ART and DHA caused G2/M cell cycle arrest. Moreover,ART and DHA induced autophagy in EOC cells, whereas autophagy inhibitors reversed the cell growth inhibition and cell cycle arrest induced by ART and DHA.Western blot analysis showed that ART and DHA also suppressed the cell cycle-related NF-κB-signaling pathway in EOC cells. These data suggest that artemisinin derivatives induce autophagy, block the cell cycle, and inhibit cell growth in EOC cells. Our research provides new targets for artemisinin derivatives for EOC treatment. | Boning Li Shixia Bu Junyan Sun Ying Guo Dongmei Lai | 2018 | Acta Biochimica et Biophysica Sinica2018,50,12: | 11
Consensus on clinical management of tumor-induced osteomalacia显示文摘Tumor-induced osteomalacia(TIO)is a rare paraneoplastic syndrome caused by excessive fibroblast growth factor 23(FGF23)production by a tumor, which often arises from a mesenchymal origin.[1-3]Most clinical symptoms of TIO are the consequences of prolonged FGF23-mediated hypophosphatemia as muscle weakness, bone pain, impaired mobility, and fractures.[4]Clinical diagnosis and management of TIO are challenging because knowledge about this condition is still restricted to a few specialized centers, leading to delay in diagnosis and appropriate treatment. The scope of the present consensus is to provide up-to-date guidance on the assessment and treatment of TIO. | Yan Jiang Xiang Li Li Huo Yong Liu Wei Lyu Lian Zhou Wei Yu Huan-Wen Wu Xiao-Ping Xing Mei Li Ou Wang Yue Chi Rui-Zhi Jiajue Yu Pei Jian-Min Liu Jian-Ming Ba Qiao Zhang Zhi-Feng Sheng Zhen-Lin Zhang Jia-Jun Zhao Salvatore Minisola Wei-Bo Xia on behalf of Chinese Society of Osteoporosis and Bone Mineral Research and Chinese Society of Endocrinology | 2021 | Chinese Medical Journal2021,,11: | 3
Improved genetic distance-based spatial deployment can effectively minimize inbreeding in seed orchard显示文摘Background:Inbreeding in seed orchards is expected to increase with the advancement of breeding cycles, which results in the delivery of crops with suboptimal genetic gain, reduced genetic diversity, and lower seed set. Here,a genetic distance-dependent method for clonal spatial deployment in seed orchards was developed and demonstrated, which reduced the inbreeding levels. The method’s main evaluation parameter of inbreeding is the genetic distance among individuals and the deployment method used an improved adaptive parallel genetic algorithm(IAPGA)based on Python language.Using inbreeding-prone Chinese Mongolian pine breeding population material originating from a single natural population, the proposed method was compared to a traditional orchard design and a distance-based design;namely, complete randomized block(RCB)and optimum neighborhood(ONA)designs, respectively. Results:With the advancement of selective breeding cycles, group separation among orchard related individuals is expected to increase.Based on the genetic distance among individuals, the IAPGA design was superior in significantly reducing the inbreeding level as compared to the two existing designs, confirming its suitability to advanced-generation orchards where relatedness among parents is common. In the 1st,2nd,and mixed generations clonal deployment schemes, the IAPGA design produced lower inbreeding with 87.22%,81.49%,and 87.23%of RCB,and 92.78%,91.30%,and 91.67%of ONA designs, respectively. Conclusions:The IAPGA clonal deployment proposed in this study has the obvious advantage of controlling inbreeding, and it is expected to be used in clonal deployment in seed orchards on a large-scale.Further studies are needed to focus on the actual states of pollen dispersal and mating in seed orchards, and more assumptions should be taken into account for the optimized deployment method. | Boning Yang Haihong Sun Jiandong Qi Shihui Niu Yousry AEl-Kassaby Wei Li | 2020 | Forest Ecosystems2020,7,1: | 1

Single-cell transcriptomic analysis uncovers intratumoral heterogeneity and drug-tolerant persister in ALK-rearranged lung adenocarcinoma显示文摘Dear Editor,Non-small cell lung carcinoma(NSCLC)accounts for more than 85%of all lung cancers[1].Lung adenocarcinomas(ADCs)arising from never-smokers have been frequently found to be associated with oncogenic driver mutations, including anaplastic lymphoma kinase(ALK)rearrangement.Echinoderm microtubule-associated protein-like 4(EML4)is the most common fusion partner of the ALK gene[2].Tyrosine kinase inhibitors(TKIs)are currently the first-line treatment for advanced EML4-ALKrearranged lung ADC. | Hoi-Hin Kwok Huiyu Li Jiashuang Yang Junyang Deng Nerissa Chui-Mei Lee Timmy Wing-Kuk Au Alva Ko-Yung Sit Michael Kuan-Yew Hsin Stephanie Kwai-Yee Ma LydiaWai-Ting Cheung Luc Girard Junya Fujimoto Ignacio Ivan Wistuba Boning Gao John Dorrance Minna David Chi-Leung Lam | 2023 | Cancer Communications2023,43,8: | 0
Machine Learning Modeling of Protein-intrinsic Features Predicts Tractability of Targeted Protein Degradation显示文摘Targeted protein degradation(TPD)has rapidly emerged as a therapeutic modality to eliminate previously undruggable proteins by repurposing the cell’s endogenous protein degradation machinery. However, the susceptibility of proteins for targeting by TPD approaches, termed“degradability”,is largely unknown. Here,we developed a machine learning model,model-free analysis of protein degradability(MAPD),to predict degradability from features intrinsic to protein targets.MAPD shows accurate performance in predicting kinases that are degradable by TPD compounds[with an area under the precision–recall curve(AUPRC)of 0.759 and an area under the receiver operating characteristic curve(AUROC)of 0.775]and is likely generalizable to independent non-kinase proteins. We found five features with statistical significance to achieve optimal prediction, with ubiquitination potential being the most predictive.By structural modeling,we found that E2-accessible ubiquitination sites, but not lysine residues in general, are particularly associated with kinase degradability. Finally,we extended MAPD predictions to the entire proteome to find964 disease-causing proteins(including proteins encoded by 278 cancer genes)that may be tractable to TPD drug development. | Wubing Zhang Shourya S.Roy Burman Jiaye Chen Katherine A.Donovan Yang Cao Chelsea Shu Boning Zhang Zexian Zeng Shengqing Gu Yi Zhang Dian Li Eric S.Fischer Collin Tokheim X.Shirley Liu | 2022 | Genomics, Proteomics & Bioinformatics2022,20,5: | 0
Efficient lignin biodegradation triggered by alkali-tolerant ligninolytic bacteria through improving lignin solubility in alkaline solution显示文摘Low lignin solubility in aqueous solution is one of the major bottlenecks for lignin biodegradation and bioconversion.Alkaline solution contributes to improving lignin solubility, whereas most microbes can not survive in alkaline conditions.Herein, lignin dissolution behaviors in different pH solutions were systematically investigated, which indicated that solution pH above 10.5 contributed to high solubility of alkali lignin. To match with alkaline lignin aqueous system, several alkali-tolerant ligninolytic bacteria were isolated, most of which are distinct to previously reported ones. Then, the ligninolytic capabilities of these isolates were assessed in different pH conditions by determining their assimilation on alkali lignin,lignin-derived monomers and dimers, their decolorization capabilities, and their lignin peroxidase activities.Thereafter, the underlying ligninolytic and alkali-tolerant mechanisms of Sutcliffiella sp.NC1,an alkalophilic bacterium, was analyzed on the basis of its genome information. The results not only provide valuable information for lignin biodegradation and lignin valorization, but also expand knowledge on alkali-tolerant bacteria. | Zhaoxian Xu Jie Li Pingping Li Chenggu Cai Sitong Chen Boning Ding Shuangmei Liu Mianshen Ge Mingjie Jin | 2023 | Journal of Bioresources and Bioproducts2023,8,4: | 0
Non-specific histological variant of dysembryoplastic neuroepithelial tumor:a diagnostic challenge显示文摘Objective The accurate diagnosis of the non-specific variant of dysembryoplastic neuroepithelial tumor(DNT) is very difficult because it is characterized by absence of the histological hallmark of the 'specific glioneuronal element' in lesions. We herein present two cases of the non-specific form of DNT to analyze the clinical, radiological, and histological features of this unusual subtype of DNT.Methods A 16-year-old and a 23-year-old patient had been treated for pharmacoresistant epilepsy for several years before undergoing referral to the hospital for further examination and treatment. Magnetic resonance imaging(MRI) revealed that both patients had a small, well-demarcated cystic lesion within the cortex of the brain without obvious contrast enhancement or peritumoral edema. The lesions were totally resected and routinely examined using histological and immunohistochemical analysis. Results Both lesions exhibited similar histological appearances with cyst formation and mural nodule architecture. The glial nodules were mainly composed of oligodendrocyte-like components, and partly of piloid cells resembling pilocytic astrocytoma. The cortex adjacent to the lesion in both cases was found to have the histological features of focal cortical dysplasia(FCD) Type I. Immunohistochemically, the oligodendrocyte-like components were diffusely positive for Syn and Olig-2, but staining for CD34, p53, and IDH1 R132 H was negative. The Ki-67(MIB-1) labeling index was low, approximately 1%. There was no 1p/19 q co-deletion in either lesion by fluorescence in situ hybridization(FISH) assay. Neither patient received postoperative adjuvant treatment, and both underwent regular follow-up for at least 24 months. No signs of recurrence or epileptic attacks were observed during the follow-up period. Conclusion The non-specific variant of DNT is a diagnostic challenge for pathologists in clinical practice, and differentiation from some low-grade gliomas needs to be considered. The careful inspection of radiologic and histopathologic findings, accompanied by analysis of patients' clinical manifestations, may be helpful in making an accurate diagnosis. | Yanyang Chen Bin Li Boning Luo Xiaoying Tian Zhi Li | 2016 | Oncology and Translational Medicine2016,2,2: | 0
