| 2 | Innate and adaptive immunity in inflammatory bowel disease显示文摘Inflammatory bowel diseases are the consequence of a dysregulated mucosal immune system. The mucosal immune system consists of two arms, innate and adaptive immunity, that have been studied separately for a long time. Functional studies from in vivo models of intestinal inflammation as well as results from genome-wide association studies strongly suggest a crossregulation of both arms. The present review will illustrate this interaction by selecting examples from innate immunity and adaptive immunity, and their direct impact on each other. | 7 | hsa-mir-183 is frequently methylated and related to poor survival in human hepatocellular carcinoma显示文摘AIM To screen clinically relevant micro RNAs (mi RNAs) silenced by DNA methylation in human hepatocellular carcinoma(HCC).METHODS Knockdown of DNA methyltransferases (DNMTs) using si RNAs and mi RNA profiling in HCC cell lines were performed to identify DNA hypermethylation-mediated mi RNA downregulation. Confirmation using individual quantitative real-time PCR (qR T-PCR) assays was thenperformed followed by DNA methylation quantification at the promoter of the mi RNA genes. Quantification of DNA methylation and mi RNA expression was then performed in primary HCC tumor samples and related with clinicopathological variables.RESULTS mi RNA profiling after DNMT knockdown in HCC cell lines revealed upregulation of mi R-23, mi R-25 and mi R-183. After q RT-PCR confirmation and Cp G island methylation quantification of these miR NAs in cell lines, further analysis in primary HCC specimens showed that hsa-mi R-183 is hypermethylated in 30% of HCC (n = 40). Expression of mature miR-183 showed an inverse correlation with DNA methylation levels. In HCC cells, DNMT knockdown and 5-aza-2'-deoxycytidine treatment reduced methylation and stimulated expression of mi R-183. In HCC patients, hypermethylation at hsami R-183 promoter significantly correlates with poor survival (log-rank test P = 0.03). However,in patients with previous coronary artery bypass graft operation(CABG) using internal mammary artery grafts,great vessel disease should also be considered. Herein we present two patients with history of CABG whose symptoms were suspicious for coronary ischemia. During cardiac catheterization reverse blood flow was observed from the left artery disease to the left internal mammary artery(LIMA) graft in both cases. | 18 | Loss of Aβ-nerve endings associated with the Merkel cell-neurite complex in the lesional oral mucosa epithelium of lichen planus and hyperkeratosis显示文摘The Merkel cell-neurite complex initiates the perception of touch and mediates Ab slowly adapting type I responses.Lichen planus is a chronic inflammatory autoimmune disease with T-cell-mediated inflammation,whereas hyperkeratosis is characterized with or without epithelial dysplasia in the oral mucosa.To determine the effects of lichen planus and hyperkeratosis on the Merkel cell-neurite complex,healthy oral mucosal epithelium and lesional oral mucosal epithelium of lichen planus and hyperkeratosis patients were stained by immunohistochemistry(the avidin-biotin-peroxidase complex and double immunofluorescence methods)using pan cytokeratin,20(K20,a Merkel cell marker),and neurofilament 200(NF200,a myelinated Ab-and Ad-nerve fibre marker)antibodies.NF200-immunoreactive(ir)nerve fibres in healthy tissues and in the lesional oral mucosa epithelium of lichen planus and hyperkeratosis were counted and statistically analysed.In the healthy oral mucosa,K20-positive Merkel cells with and without close association to the intraepithelial NF200-ir nerve fibres were detected.In the lesional oral mucosa of lichen planus and hyperkeratosis patients,extremely rare NF200-ir nerve fibres were detected only in the lamina propria.Compared with healthy tissues,lichen planus and hyperkeratosis tissues had significantly decreased numbers of NF200-ir nerve fibres in the oral mucosal epithelium.Lichen planus and hyperkeratosis were associated with the absence of Ab-nerve endings in the oral mucosal epithelium.Thus,we conclude that mechanosensation mediated by the Merkel cell-neurite complex in the oral mucosal epithelium is impaired in lichen planus and hyperkeratosis. | Daniela Calderon Carrion Yiiksel Korkmaz Britta Cho Marion Kopp Wilhelm Bloch Klaus Addicks Wilhelm Niedermeier | 2016 | International Journal of Oral Science2016,8,1: | 2 |
| 19 | Selenium Regulation of Selenium-dependent Glutathione Peroxidases in Animals and Transfected CHO Cells显示文摘Glutathione peroxidase (GPX1) was the first identified selenium-dependent enzyme, and this enzyme has been most useful as a biochemical indicator of selenium (Se) status and the parameter of choice for determining Se requirements. We have continued to study Se regulation of GPX1 to better understand the underlying mechanism and to gain insight into how cells themselves regulate nutrient status. In progressive Se deficiency in rats, GPX1 activity,protein and mRNA all decrease in a dramatic, coordinated and exponential fashion such that Se-deficient GPX1 mRNA levels are 6-15% of Sexadequate levels. mRNA levels for other Sedependent proteins are far less decreased in the same animals. The mRNA levels for a second Se-dependent peroxidase, phospholipid hydroperoxide glutathione peroxidase (GPX4 ), are little affected by Se deficiency, demonstrating that Se regulation of GPX1 is unique. Se regulation of GPX1 activity in growing male and female rats shows that the Se requirernent is 100 ng/g diet, based on liver GPX1 activity; use of GPX1 mRNA as the parameter indicates that the Se requirement is nearer to 50 ng Se/g diet in both male and female rats. This approach will readily detect an altered dietary Se requirement, as shown by the incremental increases in dietary Se requirement by 150, 100 or 50 ng Se/g diet in Seudeficient rat pups repleted with Se for 3, 7 or 14 d, respectively. Studies with CHO cells stably transfected with recombinant GPX1 also show that overexpression of GPX1 does not alter the minimum level of media Se necessary for Se-adequate levels of GPX1 activity or mRNA. | 20 | Multiphase-field modelling of concurrent grain growth and coarsening in complex multicomponent systems显示文摘Phase-field modelling of microstructural evolution in polycrystalline systems with phase-associated grains has largely been confined to continuum-field models.In this study,a multiphase-field approach,with a provision for introducing grain boundary and interphase diffusion,is extended to analyse concurrent grain growth and coarsening in multicomponent polycrystalline microstructures with chemically-distinct grains.The effect of the number of phases and components on the kinetics of evolution is investigated by considering binary and ternary systems of duplex and triplex microstructures,along with a single phase system.It is realised that the mere increase in the number of phases minimises the rate of concurrent grain growth and coarsening.However,the effect of components is substantially dependent on the respective kinetic coefficients.This work unravels that the disparity in the influence of phases and components is primarily due to the corresponding change introduced in the transformation mechanism.While the raise in number of phases convolutes the diffusion paths,the increase in number of component effects the rate of evolution through the interdiffusion,which introduces interdependency in the diffusing chemical-species.Additionally,the role of phase-fractions on the transformation rate of triplex microstructure is studied,and correspondingly,the interplay of interface-and diffusion-governed evolution is elucidated.A representative evolution of three-dimensional triplex microstructure with equal phase-fraction is comparatively analysed with the evolution of corresponding two-dimensional setup. | P.G.Kubendran Amos Ramanathan Perumal Michael Selzer Britta Nestler | 2020 | Journal of Materials Science & Technology2020,42,10: | 2 |
