2Increase of TNFα-stimulated Osteoarthritic Chondrocytes Apoptosis and Decrease of Matrix Metalloproteinases 9 by NF-κB Inhibition显示文摘Objective To investigate the in vitro effect of caffeic acid phenethyl ester(CAPE),a NF-κB inhibitor,on the apoptosis of osteoarthritic(OA)chondrocytes and on the regulation of the gelatinases matrix metalloproteinase 2(MMP-2)and matrix metalloproteinase 9(MMP-9).Methods Annexin V-FITC/propidium iodide(PI)labeling and western blotting were used to observe and determine the apoptosis in TNFα-stimulated primary cultured osteoarthritic chondrocytes.Also,gelatin zymography was applied to examine MMP-2 and MMP-9 activities in supernatants.Results It was confirmed by both flow cytometry and western blotting that chondrocytes from OA patients have an apoptotic background.Use of CAPE in combination with 10 ng/mL of TNFα for 24 h facilitated the apoptosis.MMP-9 in the supernatant could be autoactivated(from proMMP-9 to active MMP-9),and the physiologic calcium concentration(2.5 mmol/L)could delay the autoactivation of MMP-9.The activities of MMP-2 and MMP-9 in the fresh supernatant increased significantly in response to stimulation by 10 ng/mL of TNFα for 24 h.The stimulatory effect of TNFα just on proMMP-9 was counteracted significantly by CAPE.Conclusion NF-κB could prevent chondrocytes apoptosis though its activation was attributed to the increase of proMMP-9 activity induced by TNFα(a pro-apoptotic factor).Therefore,therapeutic NF-κB inhibitor was a 'double-edged swords' to the apoptosis of chondrocytes and the secretion of MMP-9.WANG Yan LI De Ling ZHANG Xin Bo DUAN Yuan Hui WU Zhi Hong HAO Dong Sheng CHEN Bao Sheng QIU Gui Xing 2013Biomedical and Environmental Sciences2013,26,4:13

4Allergy-related Evidences in Relation to Serum IgE:Data from the China State Key Laboratory of Respiratory Disease,2008-2013显示文摘Objective To investigate the serum total IgE(tlgE) and specific IgE(slgE) to common allergens among allergic patients in Guangzhou,China.Methods 7 085 patients were examined for tlgE and slgE to 15 allergens,based on the protocols of reversed enzyme allergosorbent test and the sandwich enzyme-linked immunosorbent assay.Results 3 758(53.04%) patients tested positive for tlgE,and 4 640(65.49%) for slgE.Der pteronyssinus,Der farinae,eggs,and cow's milk were the most common allergens leading to higher positive rates of slgE responses.Several peaks of sensitization were:Der pteronyssinus,Der farinae,and Blomia tropicalis at age 10-12;cow's milk at age below 3;eggs at age 4-6.The mean level and positive rate of tlgE tended to increase in subjects sensitized to more allergens.Sensitization to Der pteronyssinus(OR,1.6;P<0.05),Der farinae(OR,1.5;P<0.05),Blomia tropicalis(OR,1.4;P<0.05),Blattella germanica(OR,1.5;P<0.05),cow's milk(OR,1.3;P<0.05),and soy beans(OR,2.0;P<0.05) were independently correlated with allergy-related conditions in preliminary diagnosis.Conclusion The major allergens in Guangzhou include Der pteronyssinus,Der farinae,cow's milk,and eggs.Sensitization to these allergens appears to be predictors of allergy-related disorder.SUN Bao Qing CHEN De Hui ZHENG Pei Yan HUANG Hui Min LUO Wen Ting ZENG Guang Qiao ZHANG Xiao Wen 2014Biomedical and Environmental Sciences2014,27,7:11

6Association of Overlapped and Un-overlapped Comorbidities with COVID-19 Severity and Treatment Outcomes: A Retrospective Cohort Study from Nine Provinces in China显示文摘Objective Several COVID-19 patients have overlapping comorbidities. The independent role of each component contributing to the risk of COVID-19 is unknown, and how some non-cardiometabolic comorbidities affect the risk of COVID-19 remains unclear.Methods A retrospective follow-up design was adopted. A total of 1,160 laboratory-confirmed patients were enrolled from nine provinces in China. Data on comorbidities were obtained from the patients’ medical records. Multivariable logistic regression models were used to estimate the odds ratio(OR) and 95% confidence interval(95% CI) of the associations between comorbidities(cardiometabolic or non-cardiometabolic diseases), clinical severity, and treatment outcomes of COVID-19.Results Overall, 158(13.6%) patients were diagnosed with severe illness and 32(2.7%) had unfavorable outcomes. Hypertension(2.87, 1.30–6.32), type 2 diabetes(T2 DM)(3.57, 2.32–5.49),cardiovascular disease(CVD)(3.78, 1.81–7.89), fatty liver disease(7.53, 1.96–28.96), hyperlipidemia(2.15, 1.26–3.67), other lung diseases(6.00, 3.01–11.96), and electrolyte imbalance(10.40, 3.00–26.10)were independently linked to increased odds of being severely ill. T2 DM(6.07, 2.89–12.75), CVD(8.47,6.03–11.89), and electrolyte imbalance(19.44, 11.47–32.96) were also strong predictors of unfavorable outcomes. Women with comorbidities were more likely to have severe disease on admission(5.46,3.25–9.19), while men with comorbidities were more likely to have unfavorable treatment outcomes(6.58, 1.46–29.64) within two weeks.Conclusion Besides hypertension, diabetes, and CVD, fatty liver disease, hyperlipidemia, other lung diseases, and electrolyte imbalance were independent risk factors for COVID-19 severity and poor treatment outcome. 7Effect of Simulated Microgravity and its Associated Mechanism on Pulmonary Circulation in Rats显示文摘Objective To study the effect of Simulated Microgravity and its Associated Mechanism on Pulmonary Circulation in Rats. Methods Rat tail-suspension model was used to simulate the physiological effects of microgravity and changes in pulmonary blood vessel morphology, pulmonary arterial and venous blood pressure, pulmonary vascular resistance, pulmonary vasomotoricity, as well as the regulation of pulmonary circulation by cytokines produced and released by the lung of rats were measured. Results The walls of pulmonary blood vessels of rats were thickened, and the pulmonary artery was reconstructed with increased pulmonary vascular resistance. The pulmonary blood vessels of rats became more prone to dilation as contractions increased. Rat epithelial Adrenomedulin gene transcription and protein expression were upregulated. The level of basic fibroblast growth Factor of rat was also elevated. 9Complement C3a activates osteoclasts by regulating the PI3K/PDK1/SGK3 pathway in patients with multiple myeloma显示文摘Objective:Myeloma bone disease(MBD)is the most common complication of multiple myeloma(MM).Our previous study showed that the serum levels of C3/C4 in MM patients were significantly positively correlated with the severity of bone disease.However,the mechanism of C3 a/C4 a in osteoclasts MM patients remains unclear.Methods:The formation and function of osteoclasts were analyzed after adding C3 a/C4 a in vitro.RNA-seq analysis was used to screen the potential pathways affecting osteoclasts,and the results were verified by Western blot,q RT-PCR,and pathway inhibitors.Results:The osteoclast area per view induced by 1μg/m L(mean±SD:50.828±12.984%)and 10μg/m L(53.663±12.685%)of C3 a was significantly increased compared to the control group(0μg/m L)(34.635±8.916%)(P<0.001 and P<0.001,respectively).The relative m RNA expressions of genes,OSCAR/TRAP/RANKL/cathepsin K,induced by 1μg/m L(median:5.041,3.726,1.638,and 4.752,respectively)and 10μg/m L(median:5.140,3.702,2.250,and 5.172,respectively)of C3 a was significantly increased compared to the control group(median:3.137,2.004,0.573,and 2.257,respectively)(1μg/m L P=0.001,P=0.003,P<0.001,and P=0.008,respectively;10μg/m L:P<0.001,P=0.019,P<0.001,and P=0.002,respectively).The absorption areas of the osteoclast resorption pits per view induced by 1μg/m L(mean±SD:51.464±11.983%)and 10μg/m L(50.219±12.067%)of C3 a was also significantly increased(33.845±8.331%)(P<0.001 and P<0.001,respectively)compared to the control.There was no difference between the C4 a and control groups.RNA-seq analysis showed that C3 a promoted the proliferation of osteoclasts using the phosphoinositide 3-kinase(PI3 K)signaling pathway.The relative expressions of PIK3 CA/phosphoinositide dependent kinase-1(PDK1)/serum and glucocorticoid inducible protein kinases(SGK3)genes and PI3 K/PDK1/p-SGK3 protein in the C3 a group were significantly higher than in the control group.The activation role of C3 a in osteoclasts of MM patients was reduced by the SGK inhibitor(EMD638683).Conclusions:C3 a activated osteoclasts by regulating the PI3 K/PDK1/SGK3 pathways in MM patients,which was reduced using a SGK inhibitor.Overall,our results identified potential therapeutic targets and strategies for MBD patients。Fengjuan Jiang Hui Liu Fengping Peng Zhaoyun Liu Kai Ding Jia Song Lijuan Li Jin Chen Qing Shao Siyang Yan Kim De Veirman Karin Vanderkerken Rong Fu 2021Cancer Biology & Medicine2021,18,3:3

12Study on Syntheses and Characterizations of Polymethylphenethylsilane, Polymethylcyclohexylsilane and their Copolymers显示文摘The effects of adding cosolvents of diglyme and 15 crown 5 to the reaction mixture of Wurtz type coupling of dichlorosilanes on the yield and relative molecular mass dispersity of polymethylphenethylsilane(PMPES) were discussed. The results show that addition of 10% (volume ratio of diglyme to toluene) diglyme as a cosolvent to the reaction mixture leads to the yield increase of PMPES with a monomodal distribution of relative molecular masses. Adding 10% diglyme to the reaction mixtures, the yields of polymethylcyclohexylsilane(PMCS) and copolymers (polymethylphenethylsilane co methylcyclohexylsilane), (the molar ratios of methylphenethyldichlorosilane to methylcyclohexyldichlorosilane were 2.0, 1.0 and 0.5, and the copolymers were abbreviated by Copolymers Ⅰ, Ⅱ, Ⅲ, respectively) are 47%, 52%, 54%, 53%, respectively. Their relative molecular masses ( w ) almost reach 10 5.
