2Promotion of spinosad biosynthesis by chromosomal integration of the Vitreoscilla hemoglobin gene in Saccharopolyspora spinosa显示文摘To promote spinosad biosynthesis by improving the limited oxygen supply during high-density fermentation of Saccharopolyspora spinosa, the open reading frame of the Vitreoscilla hemoglobin gene was placed under the control of the promoter for the erythromycin resistance gene by splicing using overlapping extension PCR. This was cloned into the integrating vector pSET152, yielding the Vitreoscilla hemoglobin gene expression plasmid pSET152EVHB. This was then introduced into S. spinosa SP06081 by conjugal transfer, and integrated into the chromosome by site-specific recombination at the integration site ΦC31 on pSET152EVHB. The resultant conjugant, S. spinosa S078-1101, was genetically stable. The integration was further confirmed by PCR and Southern blotting analysis. A carbon monoxide differential spectrum assay showed that active Vitreoscilla hemoglobin was successfully expressed in S. spinosa S078-1101. Fermentation results revealed that expression of the Vitreoscilla hemoglobin gene significantly promoted spinosad biosynthesis under normal oxygen and moderately oxygen-limiting conditions (P<0.01). 4Enhancement of oxygen reduction activity and stability via introducing acid-resistant refractory Mo and regulating the near-surface Pt content显示文摘Although Pt Ni catalyst possesses good oxygen reduction activity, its poor stability is the main obstacle for the commercialization of proton exchange membrane fuel cells(PEMFCs). In this work, we introduce the acid-resistant refractory Mo to enhance the structure stability and modify the electronic structure of Pt in the prepared PtNi catalyst, improving the catalytic activity for oxygen reduction reaction(ORR). In addition, near-surface Pt content in the nanoparticle is also optimized to balance the ORR activity and stability. The electrochemical results show that the alloy formed by Mo and Pt Ni is obviously more stable than the PtNi alloy alone, because the acid-resistant Mo and its oxides effectively prevent the dissolution of Pt. Especially, the Pt3 Ni3 MoN/C exhibits the optimal ORR catalytic performance in O2-saturated 0.1 mol L^(-1) HClO4 aqueous solutions, with mass activity(MA) of 900 m A mg^(-1) Pt at 0.90 V vs. RHE, which is 3.75 times enhancement compared with the commercial Pt/C(240 mA mg^(-1) Pt). After 30 k accelerated durability tests, its MA(690 m A mg^(-1) Pt) is still 2.88 times higher than the pristine Pt/C. 163D printing CO_(2)-activated carbon nanotubes host to promote sulfur loading for high areal capacity lithium-sulfur batteries显示文摘Lithium-sulfur batteries(LSBs)have emerged as a promising high energy density system in miniaturized energy storage devices.However,serious issues rooted in large volume change(80%),poor intrinsic conductivity,“shuttle effect”of S cathode,and limited mass loading of traditional electrode still make it a big challenge to achieve high energy density LSBs in a limited footprint.Herein,an innovative carbon dioxide(CO_(2))assisted three-dimensional(3D)printing strategy is proposed to fabricate threedimensional lattice structured CO_(2)activated single-walled carbon nanotubes/S composite thick electrode(3DP S@CNTs-CO_(2))for high areal capacity LSBs.The 3D lattice structure formed by interwoven CNTs and printed regular macropores can not only act as fast electron transfer networks,ensuring good electronic conductivity of thick electrode,but is beneficial to electrolyte infiltration,effectively boosting ion diffusion kinetics even under a high-mass loading.In addition,the subsequent hightemperature CO_(2)in-situ etching can induce abundant nanopores on the wall of CNTs,which significantly promotes the sulfur loading as well as its full utilization as a result of shortened ion diffusion paths.Owing to these merits,the 3DP S@CNTs-CO_(2)electrode delivers an impressive mass loading of 10 mg·cm^(−2).More importantly,a desired attribute of linearly scale up in areal capacitance with increased layers is observed,up to an outstanding value of 5.74 mAh·cm^(−2),outperforming most reported LSBs that adopt strategies that physically inhibit polysulfides.This work provides a thrilling drive that stimulates the application of LSBs in new generation miniaturized electronic devices.Haiyan Chen Jingfeng Liu Wanqiu Cao Hanna He Xiaolong Li Chuhong Zhang 2023Nano Research2023,16,6:0
17SARS-CoV-2 infection characteristics among students and staffin a large high school COVID-19 outbreak and secondary transmission in households显示文摘Background:SARS-CoV-2 clinical presentation is associated with the patients’age group.Overall,young indi-viduals present higher proportions of asymptomatic or mild COVID-19 infection,compared to adults.Data on secondary COVID-19 transmission in households,according to the cases’age group,are accumulating.Methods:We performed a follow-up cohort study including all COVID-19 real-time polymerase chain reaction(RT-PCR)-confirmed cases(adolescent students and school staff)diagnosed in an epidemiological investigation of a large high school outbreak.We compared the adolescent and adult groups regarding clinical symptoms,time to negative COVID-19 RT-PCR tests,and infection transmission in households.Results:The study population included 817 persons.The confirmed COVID-19 RT-PCR outbreak cases(n=178)were followed(students aged 12-19 years,median age 14 years,n=153,school staffaged 24-67 years,median age 39 years,n=25)and the cases’household close contacts(n=639)were tested.The adolescents had lower symptomatic infection rates,shorter time to negative COVID-19 RT-PCR tests,and lower transmission rates to household members,compared to the adults.The general transmission rate among household contacts was 13.5%,(86/639)ranging from 8.6%in asymptomatic students’contacts to 27.3%in symptomatic staffcontacts.COVID-19 transmission rates were significantly higher in contacts of symptomatic cases compared to asymptomatic cases(odds ratio:2.06,95%CI 1.26-3.4)and higher in adults compared to adolescents(odds ratio:2.69,95%CI 1.43-4.89).Conclusions:Adolescents and adults diagnosed in an outbreak investigation differ as to COVID-19 clinical presen-tation and transmission.As adolescents may show mild or no symptoms,COVID-19 prevention in school settings is challenging.Implementing nonpharmaceutical measures and promoting vaccination programs in eligible staffand students should be considered.Chen Stein-Zamir Ora Sinai-Zaken Eti Zvulun Ghada Najajra Rinat Pinto Shahar Koren Hanna Shoob Nitza Abramson 2023Infectious Medicine2023,2,1:0

19Recombineering enables genome mining of novel siderophores in a non-model Burkholderiales strain显示文摘Iron is essential for bacterial survival,and most bacteria capture iron by producing siderophores.Burkholde-riales bacteria produce various types of bioactive secondary metabolites,such as ornibactin and malleobactin siderophores.In this study,the genome analysis of Burkholderiales genomes showed a putative novel siderophore gene cluster crb,which is highly similar to the ornibactin and malleobactin gene clusters but does not have pvdF,a gene encoding a formyltransferase for N-δ-hydroxy-ornithine formylation.Establishing the bacteriophage recom-binase Redγ-Redδβ7029 mediated genome editing system in a non-model Burkholderiales strain Paraburkholderia caribensis CICC 10960 allowed the rapid identification of the products of crb gene cluster,caribactins A-F(1-6).Caribactins contain a special amino acid residue N-δ-hydroxy-N-δ-acetylornithine(haOrn),which differs from the counterpart N-δ-hydroxy-N-δ-formylornithine(hOrn)in ornibactin and malleobactin,owing to the absence of pvdF.Gene inactivation showed that the acetylation of hOrn is catalyzed by CrbK,whose homologs proba-bly not be involved in the biosynthesis of ornibactin and malleobactin,showing possible evolutionary clues of these siderophore biosynthetic pathways from different genera.Caribactins promote biofilm production and en-hance swarming and swimming abilities,suggesting that they may play crucial roles in biofilm formation.This study also revealed that recombineering has the capability to mine novel secondary metabolites from non-model Burkholderiales species.Xingyan Wang Haibo Zhou Xiangmei Ren Hanna Chen Lin Zhong Xianping Bai Xiaoying Bian 2023Engineering Microbiology2023,3,3:0
