1A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 8Potassium channel in peripheral blood lymphocytes of turbot (Scophthalmus maximus)显示文摘In order to provide pertinent evidence of ion channel with immune response in the fish,whole cell patch-clamp technique was employed for potassium ion channel study in turbot(Scophthalmus maximus) . Lymphocytes were isolated by Percoll density gradient centrifugation from peripheral blood samples,and electrophysiological characters of the channel were analyzed. In the recorded cells,activated voltage of the channels was-42.5 ± 3.7 mV and the average peak current was 313.12 ± 28.2 pA. 9Phylogenetic analysis and in vitro culture of mosses fromthe Antarctic Fildes Peninsula显示文摘Molecular genetic techniques have proven very useful for initial analysis of the extent of genetic variation and dispersal in several Antarctic moss species.In the present study,the small subunit ribosomal RNA(SSU rDNA)and internal transcribed spacers of the nuclear ribosomal DNA(ITS rDNA)were sequenced in nine individuals of different mosses from the Fildes Peninsula of Antarctica.Sequence alignment showed that the extreme environment tended to increase the genetic diversity of Antarctic mosses.In addition,in our phylogenetic analysis,one previously unidentified Antarctic moss species was characterized by comparison with SSU and ITS rDNA sequences of known moss species.Moreover,the optimal culture medium and conditions for surface explant sterilization and protonemata induction in tissue culture of Pohlia nutans were investigated.The successful establishment of a tissue culture protocol together with the phylogenetic analysis of Antarctic mosses will provide technological support to establish an effective resource regeneration method for discovering new functional genes and gaining novel insights into the mechanisms of stress acclimation.LIU Shenghao ZHANG Zhaohui WANG Nengfei CONG Bailin ZHANG Pengying LIN Xuezheng HUANG Xiaohang 2014Advances in Polar Science2014,25,2:0
