1Diagnosis of gluten related disorders: Celiac disease, wheat allergy and non-celiac gluten sensitivity显示文摘Cereal crops and cereal consumption have had a vital role in Mankind's history. In the recent years gluten ingestion has been linked with a range of clinical disorders. Gluten-related disorders have gradually emerged as an epidemiologically relevant phenomenon with an estimated global prevalence around 5%. Celiac disease, wheat allergy and non-celiac gluten sensitivity represent different gluten-related disorders. Similar clinical manifestations can be observed in these disorders, yet there are peculiar pathogenetic pathways involved in their development. Celiac disease and wheat allergy have been extensively studied, while non-celiac gluten sensitivity is a relatively novel clinical entity, believed to be closely related to other gastrointestinal functional syndromes. The diagnosis of celiac disease and wheat allergy is based on a combination of findings from the patient's clinical history and specific tests, including serology and duodenal biopsies in case of celiac disease, or laboratory and functional assays for wheat allergy. On the other hand, non-celiac gluten sensitivity is still mainly a diagnosis of exclusion, in the absence of clear-cut diagnostic criteria. A multimodal pragmatic approach combining findings from the clinical history, symptoms, serological and histological tests is required in order to reach an accurate diagnosis. 3Constipation,deficit in colon contractions and alpha-synuclein inclusions within the colon precede motor abnormalities and neurodegeneration in the central nervous system in a mouse model of alphasynucleinopathy显示文摘Background:Gastrointestinal dysfunction can affect Parkinson’s disease(PD)patients long before the onset of motor symptoms.However,little is known about the relationship between gastrointestinal abnormalities and the development of PD.Contrary to other animal models,the human A53T alpha-synuclein(αS)transgenic mice,Line G2–3,developsαS-driven neurological and motor impairments after 9 months of age,displaying a long presymptomatic phase free of central nervous system(CNS)dysfunction.Methods:To determine whether this line can be suitable to study constipation as it occurs in prodromal PD,gastrointestinal functionality was assessed in young mice through a multidisciplinary approach,based on behavioral and biochemical analysis combined with electrophysiological recordings of mouse intestinal preparations.Results:We found that the A53TαS mice display remarkable signs of gastrointestinal dysfunction that precede motor abnormalities andαS pathology in the CNS by at least 6 months.YoungαS mice show a drastic delay in food transit along the gastrointestinal tract,of almost 2 h in 3 months old mice that increased to more than 3 h at 6 months.Such impairment was associated with abnormal formation of stools that resulted in less abundant but longer pellets excreted,suggesting a deficit in the intestinal peristalsis.In agreement with this,electrically evoked contractions of the colon,but not of the ileum,showed a reduced motor response in both longitudinal and circular muscle layers inαS mice already at 3 months of age,that was mainly due to an impaired cholinergic transmission of the underlying enteric nervous system.Interestingly,the presence of insoluble and aggregatedαS was found in enteric neurons in both myenteric and submucosal plexi only in the colon of 3 months oldαS mice,but not in the small intestine,and exacerbated with age,mimicking the increase in transit delay and the contraction deficit showed by behavioral and electrical recordings data.Conclusions:Gastrointestinal dysfunction in A53TαS mice represents an early sign ofαS-driven pathology without concomitant CNS involvement.We believe that this model can be very useful to study disease-modifying strategies that could extend the prodromal phase of PD and haltαS pathology from reaching the brain.Lucia Rota Carolina Pellegrini Laura Benvenuti Luca Antonioli Matteo Fornai Corrado Blandizzi Antonino Cattaneo Emanuela Colla 2019Translational Neurodegeneration2019,8,1:5

9Multi-objective forest harvesting under sustainable and economic principles显示文摘Selective logging is well-recognized as an effective practice in sustainable forest management.However,the ecological efficiency or resilience of the residual stand is often in doubt.Recovery time depends on operational variables,diversity,and forest structure.Selective logging is excellent but is open to changes.This may be resolved by mathematical programming and this study integrates the economic-ecological aspects in multi-objective function by applying two evolutionary algorithms.The function maximizes remaining stand diversity,merchantable logs,and the inverse of distance between trees for harvesting and log landings points.The Brazilian rainforest database(566 trees)was used to simulate our 216-ha model.The log landing design has a maximum volume limit of 500 m3.The nondominated sorting genetic algorithm was applied to solve the main optimization problem.In parallel,a sub-problem(p-facility allocation)was solved for landing allocation by a genetic algorithm.Pareto frontier analysis was applied to distinguish the gradientsα-economic,β-ecological,andγ-equilibrium.As expected,the solutions have high diameter changes in the residual stand(average removal of approximately 16 m^(3) ha^(-1)).All solutions showed a grouping of trees selected for harvesting,although there was no formation of large clearings(percentage of canopy removal<7%,with an average of 2.5 ind ha^(-1)).There were no differences in floristic composition by preferentially selecting species with greater frequency in the initial stand for harvesting.This implies a lower impact on the demographic rates of the remaining stand.The methodology should support projects of reduced impact logging by using spatial-diversity information to guide better practices in tropical forests.Talles Hudson Souza Lacerda Luciano Cavalcante de Jesus Franca Isáira Leite e Lopes Sammilly Lorrayne Souza Lacerda Evandro OrfanóFigueiredo Bruno Henrique Groenner Barbosa Carolina Souza Jarochinski e Silva Lucas Rezende Gomide 2023Journal of Forestry Research2023,34,5:0
10Appropriate search techniques to estimate Weibull function parameters in a Pinus spp.plantation显示文摘The Weibull function,a continuous probability distribution,is widely used for diameter distribution modelling,in which parameter estimation performance is affected by stand attributes and fitting methods.The Weibull cumulative distribution function is nonlinear,and classical fitting methods may provide a not optimal solution.Invoking the use of artificial intelligence by metaheuristics is reasonable for this optimisation task.Therefore,aimed and compared(1)the metaheuristics genetic algorithm and simulated annealing performance over the moment and percentile methods;(2)the hybrid strategy merging the metaheuristics tested and the percentile method and,(3)the metaheuristics fitness functions under basal area and density errors.A long-term experiment in a Pinus taeda stand subjected to crown thinning was used.According to our findings,all methods have a similar performance,independent of the thinning regimes and age.The pattern of the estimated parameters tends to be acceptable,as b increases over time and c increases after thinning.Overall,our findings suggest that methods based on metaheuristics have a higher precision than classical methods for estimating Weibull parameters.According to the classification test,the methods that involved simulated annealing stood out.The hybrid method involving this metaheuristic also stood out,with accurate estimates.Classical methods showed the poorest performance,and we therefore suggest the use of simulated annealing due to its faster processing time and high-quality solution.Lais Almeida Araujo Rafael Menali Oliveira Mario Dobner Jr Carolina Souza Jarochinskie Silva Lucas Rezende Gomide 2021Journal of Forestry Research2021,32,6:0
