| 3 | Construction and characterization of an HCV-derived multi-epitope peptide antigen containing B-cell HVR1 mimotopes and T-cell conserved epitopes显示文摘Hepatitis C(HCV) genome is highly variable,particularly in the hypervariable region 1 (HVR1) of its E2 envelope gene.The variability of HCV genome has been a major obstacle for de-veloping HCV vaccines.Due to B-cell HVR1 mimotopes mimicking the antigenicity of natural HVR1 epitopes and some T-cell epitopes from the consensus sequence of HCV genes conserving among the different HCV genotypes,we synthesized an minigene of HCV-derived multi-epitope peptide an-tigen(CMEP) ,which contains 9 B-cell HVR1 mimotopes in E2,2 conserved CTL epitopes in C,1 conserved CTL epitope in NS3 and 1 conserved Th epitope in NS3.This minigene was cloned into a GST expression vector to generate a fusion protein GST-CMEP.The immunogenic properties of CEMP were characterized by HCV infected patients' sera,and found that the reactivity frequency reached 75%.The cross reactivity of anti-CEMP antibody with different natural HVR1 variants was up to 90%.Meanwhile,we constructed an HCV DNA vaccine candidate,plasmid pVAX1.0-st-CMEP carrying the recombinant gene(st) of a secretion signal peptide and PADRE universal Th cell epitope sequence in front of the CMEP minigene.Immunization of rabbits with pVAX1.0-st-CMEP resulted in the production of antibody,which was of the same cross reactivity as the fusion protein GST-CMEP.Our findings indicate that the HCV-derived multi-epitope peptide antigen in some degree possessed the characteristics of neutralizing HCV epitopes,and would be of the value as a candidate for the development of HCV vaccines. | GAO Jun1,2,GONG Yuping1,ZHAO Ping1,ZHU Qing3,YANG Xiaoping1 & QI Zhongtian1 1.Department of Microbiology,Stake Key Laboratory of Medical Immunology,Second Military Medical University,Shanghai 200433,China 2.Department of Gastroenterology,Changhai Hospital,Second Military Medical University,Shanghai 200433,China 3.Department of Epidemiology,Naval Medical Research Institute,Shanghai 200433,China | 2006 | Science China(Life Sciences)2006,49,5: | 7 |

| 5 | Preliminary experimental results by the prototype of Sanya Incoherent Scatter Radar显示文摘In the past decades,the Incoherent Scatter Radar(ISR)has been demonstrated to be one of the most powerful instruments for ionosphere monitoring.The Institute of Geology and Geophysics at the Chinese Academy of Sciences was founded to build a state-ofthe-art phased-array ISR at Sanya(18.3°N,109.6°E),a low-latitude station on Hainan Island,named the Sanya ISR(SYISR).As a first step,a prototype radar system consisting of eight subarrays(SYISR-8)was built to reduce the technical risk of producing the entire large array.In this work,we have summarized the preliminary experimental results based on the SYISR-8.The amplitude and phase among 256 channels were first calibrated through an embedded internal monitoring network.The mean oscillation of the amplitude and phase after calibration were about 1 dB and 5°,respectively,which met the basic requirements.The beam directivity was confirmed by crossing screen of the International Space Station.The SYISR-8 was further used to detect the tropospheric wind profile and meteors.The derived winds were evaluated by comparison with independent radiosonde and balloon-based GPS measurements.The SYISR-8 was able to observe several typical meteor echoes,such as the meteor head echo,range-spread trail echo,and specular trail echo.These results confirmed the validity and reliability of the SYISR-8 system,thereby reducing the technical risk of producing the entire large array of the SYISR to some extent. | XinAn Yue WeiXing Wan Han Xiao LingQi Zeng ChangHai Ke BaiQi Ning Feng Ding BiQiang Zhao Lin Jin Chen Li MingYuan Li JunYi Wang HongLian Hao Ning Zhang | 2020 | Earth and Planetary Physics2020,4,6: | 3 |

| 8 | Serial deletion reveals structural basis and stability for the core enzyme activity of human glutaminase 1 isoforms:relevance to excitotoxic neurodegeneration显示文摘Background:Glutaminase 1 is a phosphate-activated metabolic enzyme that catalyzes the first step of glutaminolysis,which converts glutamine into glutamate.Glutamate is the major neurotransmitter of excitatory synapses,executing important physiological functions in the central nervous system.There are two isoforms of glutaminase 1,KGA and GAC,both of which are generated through alternative splicing from the same gene.KGA and GAC both transcribe 1–14 exons in the N-terminal,but each has its unique C-terminal in the coding sequence.We have previously identified that KGA and GAC are differentially regulated during inflammatory stimulation and HIV infection.Furthermore,glutaminase 1 has been linked to brain diseases such as amyotrophic lateral sclerosis,Alzheimer’s disease,and hepatic encephalopathy.Core enzyme structure of KGA and GAC has been published recently.However,how other coding sequences affect their functional enzyme activity remains unclear.Methods:We cloned and performed serial deletions of human full-length KGA and GAC from the N-terminal and the C-terminal at an interval of approximately 100 amino acids(AAs).Prokaryotic expressions of the mutant glutaminase 1 protein and a glutaminase enzyme activity assay were used to determine if KGA and GAC have similar efficiency and efficacy to convert glutamine into glutamate.Results:When 110 AAs or 218 AAs were deleted from the N-terminal or when the unique portions of KGA and GAC that are beyond the 550 AA were deleted from the C-terminal,KGA and GAC retained enzyme activity comparable to the full length proteins.In contrast,deletion of 310 AAs or more from N-terminal or deletion of 450 AAs or more from C-terminal resulted in complete loss of enzyme activity for KGA/GAC.Consistently,when both Nand C-terminal of the KGA and GAC were removed,creating a truncated protein that expressed the central 219 AA-550 AA,the protein retained enzyme activity.Furthermore,expression of the core 219 AA-550 AA coding sequence in cells increased extracellular glutamate concentrations to levels comparable to those of full-length KGA and GAC expressions,suggesting that the core enzyme activity of the protein lies within the central 219 AA-550 AA.Full-length KGA and GAC retained enzyme activities when kept at 4°C.In contrast,219 AA-550 AA truncated protein lost glutaminase activities more readily compared with full-length KGA and GAC,suggesting that the Nterminal and C-terminal coding regions are required for the stability KGA and GAC.Conclusions:Glutaminase isoforms KGA and GAC have similar efficacy to catalyze the conversion of glutamine to glutamate.The core enzyme activity of glutaminase 1 protein is within the central 219 AA-550 AA.The N-terminal and C-terminal coding regions of KGA and GAC help maintain the long-term activities of the enzymes. | Yuju Li Justin Peer Runze Zhao Yinghua Xu Beiqing Wu Yi Wang Changhai Tian Yunlong Huang Jialin Zheng | 2017 | Translational Neurodegeneration2017,6,1: | 1 |

| 10 | Effect of injection scheme on asymmetric phenomenon in rectangular and circular scramjets显示文摘The asymmetric separation has a crucial effect on the performance of the scramjet.In this study,the asymmetric separation and combustion in both rectangular and circular scramjets are investigated numerically,and the effect of injection scheme is analyzed.The characteristics of the flow field are analyzed based on sufficient code verification.In the rectangular scramjet,the separation tends to occur in the corner due to the corner boundary-layer effect.The separation is asym-metric and only two corners have serious separation.The fuel penetration depth in the separation zone increases and the combustion is intensified.When the injection scheme is uniform,both the combustion and separation become weak.In the circular scramjet,the separation and combustion are basically axisymmetric in the scramjet with one-row injection scheme.The asymmetric combustion becomes obvious in cases with multi-row injection scheme.When the injection orifices distribute intensively on the top and bottom sides,the strongest and weakest separations occur near these two sides respectively.When the distribution of orifices becomes uniform,the direction of separation cannot be predicted.For multi-row cases,the nonuniform injection scheme could result in violent combustion and asymmetric flow structures compared with the uniform injection scheme. | Guangwei MA Mingbo SUN Guoyan ZHAO Changhai LIANG Hongbo WANG Jiangfei YU | 2023 | Chinese Journal of Aeronautics2023,36,1: | 0 |
