| 2 | Role of apoptosis-inducing factor in perinatal hypoxic-ischemic brain injury显示文摘Perinatal complications,such as asphyxia,can cause brain injuries that are often associated with subsequent neurological deficits,such as cerebral palsy or mental retardation.The mechanisms of perinatal brain injury are not fully understood,but mitochondria play a prominent role not only due to their central function in metabolism but also because many proteins with apoptosis-related functions are located in the mitochondrion.Among these proteins,apoptosis-inducing factor has already been shown to be an important factor involved in neuronal cell death upon hypoxia-ischemia,but a better understanding of the mechanisms behind these processes is required for the development of more effective treatments during the early stages of perinatal brain injury.In this review,we focus on the molecular mechanisms of hypoxic-ischemic encephalopathy,specifically on the importance of apoptosis-inducing factor.The relevance of apoptosis-inducing factor is based not only because it participates in the caspase-independent apoptotic pathway but also because it plays a crucial role in mitochondrial energetic functionality,especially with regard to the maintenance of electron transport during oxidative phosphorylation and in oxidative stress,acting as a free radical scavenger.We also discuss all the different apoptosis-inducing factor isoforms discovered,focusing especially on apoptosis-inducing factor 2,which is only expressed in the brain and the functions of which are starting now to be clarified.Finally,we summarized the interaction of apoptosis-inducing factor with several proteins that are crucial for both apoptosis-inducing factor functions(prosurvival and pro-apoptotic)and that are highly important in order to develop promising therapeutic targets for improving outcomes after perinatal brain injury. | Juan Rodriguez Tao Li Yiran Xu Yanyan Sun Changlian Zhu | 2021 | Neural Regeneration Research2021,16,2: | 11 |
| 3 | Effects of Buyang Huanwu decoction on cell proliferation and differentiation in the hippocampal dentate gyrus of aged rats following cerebral ischemia/reperfusion显示文摘BACKGROUND:The mobilization of endogenous stem cells is an effective way to promote repair following ischemic brain damage.Buyang Huanwu decoction(BHD) can effectively improve cerebral blood flow and protect against cerebral ischemia/reperfusion damage. OBJECTIVE:To study the effects of BHD on cell proliferation and differentiation in the hippocampal dentate gyrus of rats following cerebral infarction,to investigate the protective effects of BHD against cerebral infarction,and to analyze the dose-effect relationship. DESIGN,TIME AND SETTING:This randomized,controlled,animal study was performed at the Laboratory of Department of Physiology,Henan College of Traditional Chinese Medicine,China from June 2007 to February 2008. MATERIALS:A total of 36 male,Sprague Dawley rats,aged 20-21 months,were equally and randomly assigned to the following groups:sham operation,model control,and nimodipine,as well as high-dose,moderate-dose,and low-dose BHD.BHD was composed of milkvetch root,Chinese angelica,red peony root,earthworm,peach seed,safflower,and Szechwan Iovage rhizome,which were provided by the Outpatient Department,Henan College of Traditional Chinese Medicine, China. METHODS:The Chinese medicinal ingredients described above were decocted.The external carotid artery was ligated in rats from the sham operation group.Rat models of focal cerebral infarction were established by middle cerebral artery occlusion in the model control and nimodipine groups,as well as the high-dose,moderate-dose,and low-dose BHD groups.The drugs were administered by gavage 5 days,as well as 2 hours,prior to model induction.Rats in the nimodipine group were daily administered a 6 mg/kg nimodipine suspension by gavage.Rats in the high-dose, moderate-dose,and low-dose BHD groups were administered daily 26,13,and 6.5 g/kg BHD, respectively.Rats in the sham operation and model control groups were treated with an equal volume of saline. MAIN OUTCOME MEASURES:The effects of BHD on neurological dysfunction score,brain water content,cell proliferation and differentiation in the hippocampal dentate gyrus,and pathological changes in the ischemic brain hemisphere were measured in cerebral infarction rats. RESULTS:Compared with the sham operation group,the neurological dysfunction score,brain water content,number of BrdU-positive cells,BrdU/NeuN-positive cells,and BrdU/GFAP-positive cells in the hippocampal dentate gyrus significantly increased in the model control group(P<0.01). Compared with the model control group,neurological dysfunction score and brain water content were significantly decreased(P<0.01 or 0.05),as were the number of BrdU-positive and BrdU/NeuN-positive cells(P<0.01 or 0.05).The number of BrdU/GFAP-positive cells was significantly reduced(P<0.05) in the nimodipine group,high-dose,moderate-dose,and low-dose BHD groups.Compared with the nimodipine group,the neurological dysfunction score was significantly reduced in the moderate-dose BHD group(P<0.05).However,the number of BrdU-positive cells was significantly increased in the rat hippocampal dentate gyrus in the high-dose and moderate-close BHD groups(P<0.01 or 0.05).The following was determined by microscopy: slightly disarranged neural cells,mild vascular dilatation,inflammatory cell infiltration,and light tissue edema were observed in the nimodipine group;inflammatory cell infiltration was reduced in the low-dose BHD group;cerebral edema and inflammatory cell infiltration were significantly reduced in the high-dose and in the moderate-dose BHD group.Electron microscopy revealed lipofuscin, slightly swollen mitochondria,and normal rough endoplasmic reticulum in the high-dose and moderate-dose BHD groups.Improvement was best in the moderate-dose BHD group. | 4 | Reduced grain chalkiness and its possible physiological mechanism in transgenic rice overexpressing L-GalLDH显示文摘Chalkiness is one of the key factors determining rice quality and price. Ascorbic acid(Asc) is a major plant antioxidant that performs many functions in plants. L-Galactono-1,4-lactone dehydrogenase(L-Gal LDH, EC1.3.2.3) is an enzyme that catalyzes the final step of Asc biosynthesis in plants. Here we show that the L-Gal LDH-overexpressing transgenic rice, GO-2,which has constitutively higher leaf Asc content than wild-type(WT) plants, exhibits significantly reduced grain chalkiness. Higher foliar ascorbate/dehydroascorbate(Asc/DHA)ratios at 40, 60, 80, and 100 days of plant age were observed in GO-2. Further investigation showed that the enhanced level of Asc resulted in a significantly higher ribulose-1,5-bisphosphate(Ru BP) carboxylase/oxygenase(Rubisco) protein level in GO-2 at 80 days. In addition, levels of abscisic acid(ABA) and jasmonic acid(JA) were lower in GO-2 at 60, 80, and100 days. | 5 | Right anterior cingulate gyrus in encephalic region associated with integrating and processing Chinese words information in working memory: A functional magnetic resonance imaging study显示文摘BACKGROUND: Many experimental results have been found in previous studies on whether laterality of brain cingulate gyrus exists in processing Chinese words information. OBJECTIVE: To observe the function of right anterior cingulate gyrus and activation showed by MRI in processing Chinese words in a visual working memory judgment tasks, and evaluate the laterality of activated brain regions. DESIGN: Observation experiment. SETTING: The Medical Psychological Research Center, Second Xiangya Hospital of Central South University. PARTICIPANTS: Thirteen healthy volunteers (7 males and 6 females), aged (29±7)years ranging from 23 to 36 years old, participated in this fMRI study during August 2003 to Febuary 2004. They gave informed consent in accordance with guidelines set by Second Xiangya Hospital of Central South University. All subjects were native Chinese speakers and strongly right handed. METHODS: In this study, Chinese numeral words were used as test materials and test mode of Sternberg was used in cognitive task, it was aimed to investigate the encephalic region involved in the storage and processing of Chinese numerical information. ①The head of subject was fixed with dense foam pad. Visual working task was performed according to the requests. Verbal working task was that the subject saw target items for four numerals written in Chinese, presenting for 1.5 s and then, after a short time by blank for 3.0 s. The subject saw one numeral presenting for 1.5 s and had to press one button if it was one of the shown numbers and another button if it did not belong to the presented numbers. There were two visual working memory stimuli in each stimulation block. The experiment was conducted in a single run, which consisted of seven blocks of visual working memory tasks. Different Chinese numerals were displayed in each block to avoid any practice effect (The response rate of the whole working memory task was 50%). ②The activation of brain was scanned with a 1.5T MRI scanner (GE Signa Twinspeed) (slice thickness 5 mm, slice gap 1.5 mm, slice parallel to line between pars geniculate and splenium in corpus callosum from corona capitis to superior part of cerebellum, totally 16 to 18 layers). ③The obtained images were pre-processed and statistically analysed with SPM 99 software. The procedure included timeslice adjusted, realigned, nomalized and smoothed. According to experimental task, data from each subject were analysed to obtain t value of each voxel. Brain activation image was got by Student’s t test and statistic was presented by pseudo-color. Statistical parameter image was formed by overlapping brain activation image on three-dimensional structure image, and the threshold value was set at P < 0.05 with ten or more continous voxels (T ≥ 4.64, K ≥ 10 voxels). The brain activation images of all the subjects were calculated and overlapped into mean brain activation images. The regions with different activation density were found out. The activation voxels in regions-of-interest were checked to calculate a laterality index for each condition. The negative value indicated right hemisphere dominance. MAIN OUTCOME MEASURES: fMRI activation and laterality coefficient of Chinese numerial working task. RESULTS: Thirteen healthy subjects participated in the result analysis. ①fMRI activation of Chinese numerial working task: The results showed that the working memory task with Chinese words not only activated left cerebral cortex including left superior frontal gyrus (BA6/10), left middle frontal gyrus (BA9), left inferior frontal gyrus (BA45/9/47), but also activated right cerebral cortex including right middle frontal gyrus (BA10/46/8), right inferior frontal lobe (BA47). Specially, peak activation was located in right anterior cingulate gyrus (BA32) with an activation volume of 879 (voxels). It indicated that superior, middle and inferior frontal gyrus, bilateral Broca regions and anterior cingutate involved in the working memory of Chinese words. ②Asymmertry index : The asymmetry index was -0.47 for frontal lobe, +0.42 for temporal lobe, +0.14 for parietal lobe and -1.00 for occipital lobe. It indicated that the right frontal lobe, left temporal lobe, left parietal lobe and right occipital lobe were more involved in the Chinese numerial working task. CONCLUSION: Brain activation in processing Chinese words in the visual working memory task broadly involves both the left and right hemisphere cortex. | 6 | Expression of multi-domain type III antifreeze proteins from the Antarctic eelpout (Lycodichths dearborni) in transgenic tobacco plants improves cold resistance显示文摘Type III antifreeze proteins(AFPIIIs)are a group of small globular proteins found in some polar fishes to protect them against freezing damage.Transgenic expression of AFPs has been shown to confer cold tolerance to commercially important plants and animals.We have previously isolated multiple AFPIII genes in the Antarctic eelpout(Lycodichthys dearborni)that encode larger AFPIII isoforms with up to 12 of the conventional domains.Here we have introduced the fish AFPIII genes that encode for the monomer(ld1),dimer(ld2),trimer(ld3)and tetramer(ld4)AFPIII isoforms in tobacco plants.Pot-grown 4-week-old transgenic tobacco plants were exposed to cold stress at 4◦C for 30 days and the results show that ld1,ld2,ld3 and ld4 transgenic plants present relatively lower electrolyte leakage and lower content of malondialdehyde(MDA),but accumulated higher content of proline when compared to control plants.This indicates considerable improved membrane integrity under low temperature stress and improvement of the plant cold resistance.The plants transformed with the AFPIII tetramer-and trimer-domains demonstrated a higher cold-tolerant levels when compared with plants transformed with the dimer-and monomer AFPIII domains.Our study further supports that fish AFPIIIs,especially the multidomain proteins,protect cells from non-freezing hypothermic stresses,apart from there well-known function as ice inhibitors molecules at freezing temperature. | Qiao Huang Ruiqin Hu Hui zhu Changlian Peng Liangbiao Chen | 2021 | Aquaculture and Fisheries2021,6,2: | 2 |

| 11 | Brain activation regions in schizophrenia patients performing the game piece memory task显示文摘BACKGROUND:Go,a traditional Chinese chess-like game,requires many unknown functions of the brain including attention,imaging,problem solving and processing of spatial working memory. To date,it remains uncertain whether the intellectual activities required to play Go are related to the frontal lobe. OBJECTIVE:To investigate various patterns of brain region activity while schizophrenic patients and normal subjects engaged in memorizing piece placement in the Chinese game of Go.Spatial working memory was measured in order to validate whether the prefrontal lobe participates in this memory process. DESIGN,TIME AND SETTING:Non-randomized,concurrent control trial was performed at Second Xiangya Hospital of Central South University,between May and December 2004. PARTICIPANTS:A total of nine Chinese schizophrenic patients with no brain or bodily diseases and not undergoing electroshock treatment,who were in accordance with the DSM-Ⅳcriteria for schizophrenia,as well as thirteen healthy staffs and students with matched age,sex,and education were included.Patients and control subjects had no neurological disorders or mental retardation.In addition,all participants were right-handed. METHODS:The cognitive task for functional magnetic resonance imaging was a block design experiment.Both groups were asked to remember the placement of pieces in the Chinese game of Go on a computer screen.A brain activation map was analyzed in SPM99. MAIN OUTCOME MEASURES:Brain responses were compared with regard to activation region size,volume,and asymmetry indices. RESULTS:Compared with the control group,the reaction time was significantly delayed in schizophrenics performing the working memory task(P<0.05).When performing the tasks, normal subjects showed significant activation of the bilateral dorsolateral prefrontal lobe with left dominance;the asymmetry indices were:frontal lobe,+0.32;temporal lobe,0.58;parietal lobe, 0.41;and occipital lobe,-0.34.On the other hand,schizophrenics showed right dominance and had a broader activation region of the prefrontal lobe(asymmetry indices:frontal lobe,-0.10; temporal lobe,+0.38;parietal lobe,+0.24;and occipital lobe,0.00).When comparing the normal group subtracted with the schizophrenic group,no significant lateralization was found in the frontal lobes but significant activation was found in the left anterior central gyrus,left middle frontal gyrus and in both sides of the cingulate gyrus.Comparing the schizophrenic group subtracted with the normal group,there was significant right lateralization of the frontal lobe and abnormally activated regions on both sides of the anterior central gyrus,middle frontal gyrus,left inferior frontal gyrus, right medial frontal gyrus and the right insular lobe.
