1Articular cartilage and osteochondral tissue engineering techniques:Recent advances and challenges显示文摘In spite of the considerable achievements in the field of regenerative medicine in the past several decades,osteochondral defect regeneration remains a challenging issue among diseases in the musculoskeletal system because of the spatial complexity of osteochondral units in composition,structure and functions.In order to repair the hierarchical tissue involving different layers of articular cartilage,cartilage-bone interface and subchondral bone,traditional clinical treatments including palliative and reparative methods have showed certain improvement in pain relief and defect filling.It is the development of tissue engineering that has provided more promising results in regenerating neo-tissues with comparable compositional,structural and functional characteristics to the native osteochondral tissues.Here in this review,some basic knowledge of the osteochondral units including the anatomical structure and composition,the defect classification and clinical treatments will be first introduced.Then we will highlight the recent progress in osteochondral tissue engineering from perspectives of scaffold design,cell encapsulation and signaling factor incorporation including bioreactor application.Clinical products for osteochondral defect repair will be analyzed and summarized later.Moreover,we will discuss the current obstacles and future directions to regenerate the damaged osteochondral tissues.Wenying Wei Honglian Dai 2021Bioactive Materials2021,6,12:14
2Effects of r-HuEPO on the biophysical characteristics of erythrocyte membrane in patients with anemia of chronic renal failure显示文摘Using electron spin resonance (ESR) spin labeling technique, we have studied the conformation of sulfhydryl groups(-SH) binding sites in membrane proteins and membrane fluidity of red blood cells (RBCs) from two groups of patients with anemia of chronic renal failure (ACRF). One of the groups is composed of patients who were untreated with recombinant human erythropoietin(r-HuEPO), and the other is composed of patients who were treated with r-HuEPO. The results indicated: 1) the conformation of SH group binding site in RBC membrane proteins from former group was different from those of health people. 2)the fluidity in the region near the surface of RBC membrane from former group was lower than those of healthy people. 3) However, the above biophysical properties of RBC membrane from later group were normal. 3Optical tweezers technique and its applications显示文摘Since their advent in the 1980s,optical tweezers have attracted more and more attention due to their unique non-contact and non-invasion characteristics and their wide applications in physics,biology,chemistry,medical science and nanoscience.In this paper,we introduce the basic principle,the history and typical applications of optical tweezers and review our recent experimental works on the development and application of optical tweezers technique.We will discuss in detail several technological issues,including high precision displacement and force measurement in single-trap and dual-trap optical tweezers,multi-trap optical tweezers with each trap independently and freely controlled by means of space light modulator,and incorporation of cylindrical vector optical beams to build diversified optical tweezers beyond the conventional Gaussian-beam optical tweezers.We will address the application of these optical tweezers techniques to study biophysical problems such as mechanical deformation of cell membrane and binding energy between plant microtubule and microtubule associated proteins.Finally we present application of the optical tweezers technique for trapping,transporting,and patterning of metallic nanoparticles,which can be harnessed to manipulate surface plasmon resonance properties of these nanoparticles.GUO HongLian LI ZhiYuan 2013Science China(Physics,Mechanics & Astronomy)2013,56,12:7
4Characterization, gene cloning and expression of new xylanase XYNB with high specific activity显示文摘Extracellular xylanase XYNB from Streptomy-ces olivaceoviridis A1 has been purified and characterized. The optimal pH value and temperature of XYNB for its ac-tivity are 5.2 and 60℃, respectively. The specific activity of XYNB is as high as 2869.78 U/mg. Metal cations, EDTA and SDS have no effects on enzyme activity of XYNB. The gene xynB coding mature protein of XYNB has been cloned by PCR. The forward oligonucleotide primer used in the PCR reaction was synthesized based on the N-terminal amino acid sequence of XYNB mature protein, and the reverse oligonu-cleotide primers are random oligonucleotide. The cloned gene xynB is 576 bp long and its G+C content is 64.3%. The xynB encodes 191 amino acid residues, and the putative mo-lecular weight of XYNB is 20.839 kD. 5Preparation and characterization of a degradable magnesium phosphate bone cement显示文摘A kind of degradable magnesium phosphate bone cement(MPBC)was fabricated by using the mixed powders of magnesium oxide(MgO),potassium dihydrogen phosphate(KH_(2)PO_(4))and calcium dihydrogen phosphate(Ca(H_(2)PO_(4))2.H_(2)O).As MgKPO_(4),the main product of MgO and KH_(2)PO_(4)was alkaline,the Ca(H_(2)PO_(4))2.H_(2)Owas added to neutralize the alkali of the system.And the effects of Ca(H_(2)PO_(4))2.H_(2)Oon the performance of MPBC were discussed.The results showed that the adding of Ca(H_(2)PO_(4))2.H_(2)Oextended the setting time,which was about 6 min to 18 min.The compressive strength increased first and then decreased,and maximum value reached 31.2MPa after setting for 24 h without any additional pressure.The MPBC was degradable in Tris–HCl solution,and the extracts of the cytotoxicity assay showed that the MPBC had good biocompatibility,indicating that the MPBC had good biodegradable and biocompatible properties.Ying Yu Chao Xu Honglian Dai 2016Regenerative Biomaterials2016,3,4:6
6Polymorphisms of chemokine receptors and its ligand alleles influencing genetic suscepti-bity to HIV-1 infection in eight ethnic groups in Chinese mainland显示文摘Limited genetic information is available concerning the polymorphisms of HIV-1 resistant genes in indigenous Chinese populations. The aim of this study is to identify the allelic frequencies of the chemokine and chemokine receptor genes in the Chinese mainland. Genomic DNA samples extracted from whole blood of 2318 subjects were analyzed by using PCR or PCR/restriction fragment length polymorphism (RFLP) assays, and further confirmed by direct DNA sequencing. Higher frequencies of mutant CCR2-64I (19.15%-28.79%) and SDF1-3’A (19.10% -29.86%) alleles were found in subjects of 8 ethnic groups in the Chinese mainland. In contrast, the △32 mutation in CCRS gene occurs at a very low frequency (0.0016, n=1287) in Han population. A relatively high frequency of CCR5-wt/△32 heterozygotes was observed in Uygurian and Mongolian populations. No A32 mutation allele was detected in Tibetan and other 4 ethnic groups in Yunnan Province. 7The Effects of Microwave Ablation and Surgical Resection on Hematogenous Dissemination of Cancer Cells in Treating Patients with Small Primary Hepatocellular Carcinoma显示文摘OBJECTIVE To conduct a comparative study of the effects of treatment using microwave ablation versus surgical resection on hematogenous dissemination of cancer cells, and on the level of immune cells of the peripheral blood in patients with small primary hepatocellular carcinoma (PHC,≤5 cm). METHODS Forty patients with small PHC (maximal diameter≤5 cm) were divided into a microwave group (19 cases) and a surgical operation group (21 cases). A real-time (RT) quantitative nested RT-PCR examination was performed for peripheral blood alpha-fetoprotein (AFP) mRNA. Studies were conducted to determine the level of CD3, CD4, CD8 and CD4/CD8 cells and for liver function at 30 min before, and 30 min,1 day and 3 days after the treatment. RESULTS Compared to the value before ablation, no obvious changes of CD3, CD4, CD8 and CD4/CD8 cells were found in patients of the microwave group within 7 days after ablation, but CD3, CD4 and CD4/CD8 cells in the operation group were lower compared to that before operation. The copy number of AFP mRNA in the peripheral blood samples of the patients of the 2 groups before operation was determined in 67.5% of the patients (27/40). There was an rise in the expression after treatment but no statistical difference was found in comparing the 2 groups. Follow-up of the patients was conducted for 1 to 16 months. For patients with continuous expression of peripheral blood AFP mRNA, the possibility of relapse and metastasis was increased. CONCLUSION Surgical resection or microwave ablation can cause more exfoliation of hepatoma carcinoma cells in the peripheral blood of patients with small PHC. The immune function of peripheral blood cells decreased in the patients after surgical resection, however, the immune function was better protected following microwave ablation. 9Ray-optics model for optical force and torque on a spherical metal-coated Janus microparticle显示文摘In this paper, we develop a theoretical method based on ray optics to calculate the optical force and torque on a metallo-dielectric Janus particle in an optical trap made from a tightly focused Gaussian beam. The Janus particle is a 2.8 μm diameter polystyrene sphere half-coated with gold thin film several nanometers in thickness. The calculation result shows that the focused beam will push the Janus particle away from the center of the trap,and the equilibrium position of the Janus particle, where the optical force and torque are both zero, is located in a circular orbit surrounding the laser beam axis. The theoretical results are in good agreement qualitatively and quantitatively with our experimental observation. As the ray-optics model is simple in principle, user friendly in formalism, and cost effective in terms of computation resources and time compared with other usual rigorous electromagnetics approaches, the developed theoretical method can become an invaluable tool for understanding and designing ways to control the mechanical motion of complicated microscopic particles in various optical tweezers.Jing Liu Chao Zhang Yiwu Zong Honglian Guo Zhi-Yuan Li 2015Photonics Research2015,3,5:4
10Laboratory-based Surveillance and Clinical Profile of Sporadic HEV Infection in Shanghai,China显示文摘The study aimed to describe the epidemiological,virological and clinical features of sporadic HEV infection in eastern China.A total of 6112 patient sera were tested for anti-HEV IgG or anti-HEV IgM during one consecutive year(between August 2018 and July 2019).HEV RNA presence was evaluated by RT-PCR and HEV sequences were phylogenetically analyzed.Clinical features of confirmed HEV-infected patients were delineated.The sero-positivity rate of anti-HEV IgG maintained stable around 40%,while an obvious winter spike of anti-HEV IgM prevalence was observed.A total of 111 patients were confirmed of HEV viremia by molecular diagnosis.Subtype 4d was predominant.Phylogenetic analyses suggest that certain strains circulate across species and around the country.Subjects with confirmed current HEV infection had a high median age(58 years)and males were predominant(62.2%).Most patients presented with jaundice(75.7%)and anorexia(68.0%).Significantly elevated levels of liver enzymes and bilirubin were observed.Remarkably,the baseline bilirubin level was positively correlated with illness severity.Pre-existing HBV carriage may deteriorate illness.The clinical burden caused by locally acquired HEV infection is increasing.Surveillance should be enforced especially during the transition period from winter to spring.Patients with higher level of bilirubin at disease onset had slower recovery from HEV infection.Jie Lu Qing Li Jiayuan Jiang Ziqiang Li Peiyun Wang Zike Sheng Rongtao Lai Huijuan Zhou Wei Cai Hui Wang Qing Guo Honglian Gui Qing Xie 2021Virologica Sinica2021,36,4:4
11Applying a new method for direct collection, volume quantification and determination of N_2 emission from water显示文摘The emission of N2 is important to remove excess N from lakes, ponds, and wetlands. To investigate the gas emission from water, Gao et al.(2013) developed a new method using a bubble trap device to collect gas samples from waters. However, the determination accuracy of sampling volume and gas component concentration was still debatable. In this study, the method was optimized for in situ sampling, accurate volume measurement and direct injection to a gas chromatograph for the analysis of N2 and other gases. By the optimized new method, the recovery rate for N2 was 100.28% on average; the mean coefficient of determination(R2) was 0.9997; the limit of detection was 0.02%. We further assessed the effects of the new method, bottle full of water, vs. vacuum bag and vacuum vial methods, on variations of N2 concentration as influenced by sample storage times of 1,2, 3, 5, and 7 days at constant temperature of 15°C, using indices of averaged relative peak area(%) in comparison with the averaged relative peak area of each method at 0 day.The indices of the bottle full of water method were the lowest(99.5%–108.5%) compared to the indices of vacuum bag and vacuum vial methods(119%–217%). Meanwhile, the gas chromatograph determination of other gas components(O2, CH4, and N2O) was also accurate. 13Preliminary experimental results by the prototype of Sanya Incoherent Scatter Radar显示文摘In the past decades,the Incoherent Scatter Radar(ISR)has been demonstrated to be one of the most powerful instruments for ionosphere monitoring.The Institute of Geology and Geophysics at the Chinese Academy of Sciences was founded to build a state-ofthe-art phased-array ISR at Sanya(18.3°N,109.6°E),a low-latitude station on Hainan Island,named the Sanya ISR(SYISR).As a first step,a prototype radar system consisting of eight subarrays(SYISR-8)was built to reduce the technical risk of producing the entire large array.In this work,we have summarized the preliminary experimental results based on the SYISR-8.The amplitude and phase among 256 channels were first calibrated through an embedded internal monitoring network.The mean oscillation of the amplitude and phase after calibration were about 1 dB and 5°,respectively,which met the basic requirements.The beam directivity was confirmed by crossing screen of the International Space Station.The SYISR-8 was further used to detect the tropospheric wind profile and meteors.The derived winds were evaluated by comparison with independent radiosonde and balloon-based GPS measurements.The SYISR-8 was able to observe several typical meteor echoes,such as the meteor head echo,range-spread trail echo,and specular trail echo.These results confirmed the validity and reliability of the SYISR-8 system,thereby reducing the technical risk of producing the entire large array of the SYISR to some extent.XinAn Yue WeiXing Wan Han Xiao LingQi Zeng ChangHai Ke BaiQi Ning Feng Ding BiQiang Zhao Lin Jin Chen Li MingYuan Li JunYi Wang HongLian Hao Ning Zhang 2020Earth and Planetary Physics2020,4,6:3
14Iron oxide nanoparticles with photothermal performance and enhanced nanozyme activity for bacteria-infected wound therapy显示文摘Metal-based nanomaterials usually have broad-spectrum antibacterial properties,low biological toxicity and no drug resistance due to their intrinsic enzyme-like catalytic properties and external field(magnetic,thermal,acoustic,optical and electrical)responsiveness.Herein,iron oxide(Fe_(3)O_(4))nanoparticles(IONPs)synthesized by us have good biosafety,excellent photothermal conversion ability and peroxidase-like catalytic activity,which can be used to construct a photothermal-enzymes combined antibacterial treatment platform.IONPs with peroxide-like catalytic activity can induce H_(2)O_(2)to catalyze the production of·OH in a slightly acidic environment,thus achieving certain bactericidal effects and increasing the sensitivity of bacteria to heat.When stimulated by near-infrared light,the photothermal effect could destroy bacterial cell membranes,resulting in cleavage and inactivation of bacterial protein,DNA or RNA.Meanwhile,it can also improve the catalytic activity of peroxidase-like and promote IONPs to catalyze the production of more·OH for killing bacteria.After IONPs synergistic treatment,the antibacterial rate of Escherichia coli and Staphylococcus aureus reached nearly 100%.It also has an obvious killing effect on bacteria in infected wounds of mice and can effectively promote the healing of S.aureusinfected wounds,which has great application potential in clinical anti-infection treatment.Jiaxin Guo Wenying Wei Yanan Zhao Honglian Dai 2022Regenerative Biomaterials2022,9,1:3

17Preparation of Nano-TiO_2 by Liquid Hydrolysis and Characterization of Its Antibacterial Activity显示文摘The nano-TiO2 particles were prepared by liquid hydrolysis method and characterized using XRD. Its antibacterial activity against two representative bacterial, Escherichia coli and Staphylococcus aureus, was also studied. The experimental results showed that the nano-TiO2 calcinated at 600-700 ℃ contained the obvious anatase phase and exerted excellent antibacterial activity. 18Reconfigurable nano-kirigami metasurfaces by pneumatic pressure显示文摘Tunable/reconfigurable metasurfaces that can actively control electromagnetic waves upon external stimuli are of great importance for practical applications of metasurfaces.Here,we demonstrate a reconfigurable nano-kirigami metasurface driven by pneumatic pressure operating in the near-infrared wavelength region.The metasurfaces consist of combined Archimedean spirals and are fabricated in a free-standing gold/silicon nitride nanofilm by employing focused ion beam(FIB)lithography.The deformable spirals are instantly transformed from two dimensional(2 D)to three-dimensional(3 D)by the FIB-based nano-kirigami process.The 2 D–to–3 D transformation induces a dramatic irreversible change of the plasmonic quadruple modes and results in significant modulation in reflection by 137%.The suspended porous nano-kirigami metasurface is further integrated with an optofluidics device,with which the optical resonance is reversibly modulated by the pneumatic pressure.This work provides a strategy for tunable/reconfigurable metasurfaces,which are useful to build a promising lab-ona-chip platform for microfluidics,biological diagnostics,chemical sensing,and pressure monitoring.Shanshan Chen Wei Wei Zhiguang Liu Xing Liu Shuai Feng Honglian Guo Jiafang Li 2020Photonics Research2020,8,7:2
19A New Type of Nanogel Carrier based on Mixed Pluronic Loaded with Low-Dose Antitumor Drugs显示文摘To design a new type of antitumor nanodrug carrier with good biocompatibility, a drug delivery system with a 2.19% drug-loading rate, measured by high-performance liquid chromatography(HPLC), was prepared by membrane hydration using a mixed polymer: Pluronic■ F-127, which binds folic acid(FA), Pluronic■ F-68 and triptolide(TPL)(FA-F-127/F-68-TPL). As a control, another drug delivery system based on a single polymer(FA-F-127-TPL) with a 1.90% drug-loading rate was prepared by substituting F-68 with F-127. The average particle sizes of FA-F-127/F-68-TPL and FA-F-127-TPL measured by a particle size analyzer were 30.7 nm and 31.6 nm, respectively. Their morphology was observed by atomic force microscopy(AFM). The results showed that FA-F-127-TPL self-assembled into nanomicelles, whereas FA-F-127/F-68-TPL self-assembled into nanogels. An MTT assay showed that a very low concentration of FA-F-127/F-68-TPL or FA-F-127-TPL could significantly inhibit the proliferation of multidrug-resistant(MDR) breast cancer cells(MCF-7/ADR cells) and induce cell death. The effects were significantly different from those of free TPL(P < 0.01). Using the fluorescent probe Nile red(Nr) as the drug model, FA-F-127/F-68-Nr nanogels and FAF-127-Nr nanomicelles were prepared and then incubated with human hepatocarcinoma(HepG2) and MCF-7/ADR cells, and the fluorescence intensity in the cells was measured by a multifunctional microplate reader. The results indicated that both FA-F-127/F-68-Nr and FA-F-127-Nr had sustained release in the cells, but HepG2 and MCF-7/ADR cells exhibited significantly higher endocytosis of FA-F-127/F-68-Nr than that of FA-F-127-Nr(P < 0.01). A nude mice transplanted tumor model was prepared to monitor FA-F-127/F-68-Nr in the tumor tissue and organs by whole-body fluorescent imaging. The results showed that FA-F-127/F-68-Nr targeted tumor tissues.
