| 1 | Identification and Fine Mapping of rhm1 Locus for Resistance to Southern Corn Leaf Blight in Maize显示文摘rhm1 is a major recessive disease resistance locus for Southern corn leaf blight (SCLB).To further narrow down its genetic position,F 2 population and BC 1 F 1 population derived from the cross between resistant (H95 rhm) and susceptible parents (H95) of maize (Zea mays) were constructed.Using newly developed markers,rhm1 was initially delimited within an interval of 2.5 Mb,and then finally mapped to a 8.56 kb interval between InDel marker IDP961-503 and simple sequence repeat (SSR) marker A194149-1.Three polymorphic markers IDP961-504,IDP B2-3 and A194149-2 were shown to be co-segregated with the rhm1 locus.Sequence analysis of the 8.56 kb DNA fragment revealed that it contained only one putative gene with a predicted amino acid sequence identical to lysine histidine transporter 1 (LHT1).Comparative sequence analysis indicated that the LHT1 in H95 rhm harbors a 354 bp insertion in its third exon as compared with that of susceptible alleles in B73,H95 and Mo17.The 354 bp insertion resulted in a truncation of the predicted protein of candidate resistance allele (LHT1-H95 rhm).Our results strongly suggest LHT1 as the candidate gene for rhm1 against SCLB.The tightly linked molecular markers developed in this study can be directly used for molecular breeding of resistance to Southern corn leaf blight in maize. | Yuanzeng Zhao Xiaomin Lu Chaoxian Liu Haiying Guan Mei Zhang Zhongfeng Li Hongwei Cai Jinsheng Lai | 2012 | Journal of Integrative Plant Biology2012,54,5: | 4 |
| 2 | Fosmid library construction and screening for the maize mutant gene Vestigial glume 1显示文摘The maize mutant gene Vestigial glume 1(Vg1) has been fine-mapped to a narrow region by map-based cloning and the candidate gene for Vg1 spanned 19.5 kb. Here we report Vg1 genomic fosmid library construction and screening. The fosmid library of Vg1 consisted of574,000 clones with an average insert size of 36.4 kb, representing 7.9-fold coverage of the maize genome. Fosmid stability assays indicated that clones were stable during propagation in the fosmid system. Using Vg1 candidate gene-specific primers, a positive clone was successfully identified. | 3 | Characterization of the imprinting and expression patterns of ZAG2 in maize endosperm and embryo显示文摘ZAG2 has been identified as a maternally expressed imprinted gene in maize endosperm.Our study revealed that paternally inherited ZAG2 alleles were imprinted in maize endosperm and embryo at 14 days after pollination(DAP), and consistently imprinted in endosperm at 10, 12, 16, 18, 20, 22, 24, 26, and 28 DAP in reciprocal crosses between B73 and Mo17. ZAG2 alleles were also imprinted in reciprocal crosses between Zheng 58 and Chang7-2 and between Huang C and 178. ZAG2 alleles exhibited differential imprinting in hybrids of 178 × Huang C and B73 × Mo17, while in other hybrids ZAG2 alleles exhibited binary imprinting. | 5 | Scientific literature addressing detection of monosialoganglioside A 10-year bibliometric analysis显示文摘OBJECTIVE: The study was undertaken to explore a bibliometric approach to quantitatively assess the research on detection of monosialoganglioside from 2002 to 2011. DATA RETRIEVAL: A bibliometric analysis based on the publications on Web of Science was performed using key words such as 'monosialoganglioside', 'colloidal gold', 'high performance liquid chromatography' and 'detection'. SELECTION CRITERIA: (1) Research articles on the detection of monosialoganglioside; (2) researches on human and animal fundamentals, clinical trials and case reports; (3) article types: article, review, proceedings paper, note, letter, editorial material, discussion, book chapter; (4) Publication year: 2002-2011. Exclusion criteria: (1) unrelated articles; (2) type of articles: correction; (3) articles from following databases: all databases related to social science and arts & humanities in Web of Science were excluded. MAIN OUTCOME MEASURES: (1) distribution of subject areas; (2) number of publications annually; (3) document type and language of publications; (4) distribution of institutions; (5) distribution of output in journals; (6) the number of countries in which the article is published; (7) top cited paper. RESULTS: Overall population stands at 1 880 research articles addressing detection of monosialoganglioside in Web of Science during the study period. Articles (1 599) were the most frequently used document type comprising 85.05%, followed by meeting abstracts, reviews and proceedings papers. The distribution of subject categories showed that monosialoganglioside research covered both clinical and basic science research. The USA, Japan, and Italy were the three most productive countries, and the publication numbers in the USA were highest with 559 papers. The University of Milan, Nagoya University, and Kinki University are the most productive institutions regarding detection of monosialoganglioside. In 559 articles published by Americans, Medical College of Georgia ranked the first with 30 articles, followed by University of Medicine and Dentistry of New Jersey (28 articles), Cornell University (24 articles) and Johns Hopkins University (24 articles). In 442 articles published by Japanese, Nagoya University ranked the first with 40 articles, followed by Kinki University (36 articles), and Dokkyo University (31 articles). Though the total number of publications by Japanese is smaller than Americans, the top three institutions published more publications than American institutions. There is a markedly increase in the number of publications on the subject detection of monosialoganglioside in 2004, which the peak in the past 10 years. The valley bottom of the subject appeared in 2005. In total, the research is increased with time prolonged. Journal of Neurochemistry, Journal of Biological Chemistry and Journal of Neuroimmunology were core subject journals in monosialoganglioside studies. | 7 | Molecular mapping and candidate gene analysis of the semi-dominant gene Vestigial glume1 in maize显示文摘The glume is an organ of the maize spikelet and plays important roles in anther and kernel development.Vestigial glume1(Vg1)is a classic mutant associated with ligule and glume development.Here we report the phenotypic characterization,fine mapping,and candidate gene analysis of the Vg1 mutant.Vg1 is a semi-dominant and pleiotropic gene,and also affects plant height,ear height,and tassel length.Vg1 ligule degeneration begins at the first leaf,and the Vg1 tassel and ear can be distinguished from those of wild-type plants when their lengths reach respectively 55 mm and 51 mm.Using a BC3 mapping population of 11,445 plants,we delimited the Vg1 functional site to an interval of 7.4 kb,flanked by the markers InDelLM and CRM6.A putative cyclopropane fatty-acid synthase gene(ZmCPA-FAS1)was hypothesized to underlie the mutant phenotype.We detected a Helitron insertion in the sixth intron of ZmCPAFAS1.Its presence caused abnormal alternative splicing of ZmCPA-FAS1 that conferred new characteristics on the Vg1 mutant.These findings are a basis for further discovery of the molecularmechanism underlying glume development and a potential guide formaize breeding of small-glume varieties,especially sweet corn breeding. | Chaoxian Liu Yuanzeng Zhao Yang Bai Xiaomin Lu Weibin Song Liping Qin Yilin Cai | 2019 | The Crop Journal2019,7,5: | 1 |

| 12 | Identification and Characterization of ZF-HD Genes in Response to Abscisic Acid and Abiotic Stresses in Maize显示文摘The zinc finger homeodomain(ZF-HD)genes belong to the homeobox gene family,playing critical roles in flower development and stress response.Despite their importance,however,to date there has been no genome-wide identification and characterization of the ZF-HD genes that are probably involved in stress responses in maize.In this study,24 ZF-HD genes were identified,and their chromosomal locations,protein properties,duplication patterns,structures,conserved motifs and expression patterns were investigated.The results revealed that the ZF-HD genes are unevenly distributed on nine chromosomes and that most of these genes lack introns.Six and two ZF-HD genes have undergone segmental and tandem duplication,respectively,during genome expansion.These 24 ZF-HD transcription factors were classified into six major groups on the basis of protein molecular evolutionary relationship.The expression profiles of these genes in different tissues were evaluated,resulting in producing two distinct clusters.ZF-HD genes are preferentially expressed in reproductive tissues.Furthermore,expression profiles of the 24 ZF-HD genes in response to different kinds of stresses revealed that ten genes were simultaneously up-regulated under ABA,salt and PEG treatments;meanwhile four genes were simultaneously down-regulated.These findings will pave the way for deciphering the function and mechanism of ZF-HD genes on how to implicate in abiotic stress. | Xiaojie Jing Chunyan Li Chengjuan Luo Chaonan Yao Jiahao Zhang Tingting Zhu Jiuguang Wang Chaoxian Liu | 2023 | Phyton-International Journal of Experimental Botany2023,92,3: | 0 |
| 13 | Imprinting, methylation, and expression characterization of the maize ETHYLENE-INSENSITIVE 2-like gene显示文摘The endosperm plays essential roles in embryogenesis and seed germination and provides abundant resources for human food and industrial products. Identification of genes regulating the development of the endosperm and elucidation of their functions is of great importance for maize genetics and breeding. This study showed that the genespecific imprinted gene, ETHYLENE-INSENSITIVE 2-like(EIN2-like), is maternally expressed in both endosperm and embryo. The maternally expressed pattern was maintained throughout later seed developmental stages. Bisulfite sequencing using DNA obtained from hybrid endosperm tissues showed that the upstream regions of the alleles of EIN2-like were highly methylated at symmetrical sites(CG and CHG). A differentially methylated region in the upstream part of the maternal allele of EIN2-like was identified and found to be hypomethylated. Expression analysis showed that EIN2-like was highly expressed in the maize endosperm as well as at different stages of cell differentiation(8–12 days after pollination) in the hybrid endosperm.
