| 2 | Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. Then a virtual simulation-based approach to implement the evaluations at three hierarchy levels is mainly analyzed with a detailed example, which validates the feasibility and effectiveness of evaluation architecture. | 4 | De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. | 5 | Adsorption of phenol from aqueous solution by a hierarchical micro-nano porous carbon material显示文摘A hierarchical micro-nano porous carbon material (MNC) was prepared using expanded graphite (EG), sucrose, and phosphoric acid as raw materials, followed by sucrose-phosphoric acid solution impregnation, solidification, carbonization and activation. Nitrogen adsorption and mercury porosimetry show that mixed nanopores and micropores coexist in MNC with a high specific surface area of 1978 m2·g-1 and a total pore volume of 0.99 cm3·g-1. In addition, the MNC is found to consist of EG and activated carbon with the latter deposited on the interior and the exterior surfaces of the EG pores. The thickness of the activated carbon layer is calculated to be about one hundred nanometers and is further confirmed by scanning electron microscope (SEM) and transmission election microscope (TEM). A maximum static phenol adsorption of 241.2 mg·g-1 was obtained by using MNC, slightly higher than that of 220.4 mg·g-1 by using commercial activated carbon (CAC). The phenol adsorption kinetics were investigated and the data fitted well to a pseudo-second-order model. Also, an intra-particle diffusion mechanism was proposed. Furthermore, it is found that the dynamic adsorption capacity of MNC is nearly three times that of CAC. | 6 | Dab2 attenuates brain injury in APP/PS1 mice via targeting transforming growth factor-beta/SMAD signaling显示文摘Transforming growth factor-beta(TGF-β)type II receptor(TβRII)levels are extremely low in the brain tissue of patients with Alzheimer’s disease.This receptor inhibits TGF-β1/SMAD signaling and thereby aggravates amyolid-beta deposition and neuronal injury.Dab2,a specific adapter protein,protects TβRII from degradation and ensures the effective conduction of TGF-β1/SMAD signaling.In this study,we used an adenoviral vector to overexpress the Dab2 gene in the mouse hippocampus and investigated the regulatory effect of Dab2 protein on TGF-β1/SMAD signaling in a mouse model of Alzheimer’s disease,and the potential neuroprotective effect.The results showed that the TβRII level was lower in APP/PS1 mouse hippocampus than in normal mouse hippocampus.After Dab2 expression,hippocampal TβRII and p-SMAD2/3 levels were significantly increased,while amyloid-beta deposition,microglia activation,tumor necrosis factor-βand interleulin-6 levels and neuronal loss were significantly attenuated in APP/PS1 mouse brain tissue.These results suggest that Dab2 can exhibit neuroprotective effects in Alzheimer’s disease by regulating TGF-β1/SMAD signaling. | Lei Song Yue Gu Jing Jie Xiaoxue Bai Ying Yang Chaoying Liu Qun Liu | 2014 | Neural Regeneration Research2014,9,1: | 4 |

| 8 | Genome assembly of the maize inbred line A188 provides a new reference genome for functional genomics显示文摘The current assembled maize genomes cannot represent the broad genetic diversity of maize germplasms.Acquiring more genome sequences is critical for constructing a pan-genome and elucidating the linkage between genotype and phenotype in maize.Here we describe the genome sequence and annotation of A188,a maize inbred line with high phenotypic variation relative to other lines,acquired by single-molecule sequencing and optical genome mapping.We assembled a 2210-Mb genome with a scaffold N50 size of 11.61 million bases(Mb),compared to 9.73 Mb for B73 and 10.2 Mb for Mo17.Based on the B73_Ref Gen_V4 genome,295 scaffolds(2084.35 Mb,94.30%of the final genome assembly)were anchored and oriented on ten chromosomes.Comparative analysis revealed that~30%of the predicted A188 genes showed large structural divergence from B73,Mo17,and W22 genomes,which causes high protein divergence and may lead to phenotypic variation among the four inbred lines.As a line with high embryonic callus(EC)induction capacity,A188 provides a convenient tool for elucidating the molecular mechanism underlying the formation of EC in maize.Combining our new A188 genome with previously reported QTL and RNA sequencing data revealed eight genes with large structural variation and two differentially expressed genes playing potential roles in maize EC induction. | Fei Ge Jingtao Qu Peng Liu Lang Pan Chaoying Zou Guangsheng Yuan Cong Yang Guangtang Pan Jianwei Huang Langlang Ma Yaou Shen | 2022 | The Crop Journal2022,10,1: | 2 |

| 10 | Acupoint electrogymnastics therapy in stroke hemiplegia显示文摘BACKGROUND: Electrogymnastics can offer a range of proprioceptive, motor, and cutaneous sensation impulses to the central nerve system. The center receives perception of the paralyzed muscle through the aid of these impulses. During this process, functional reorganization of connecting network between segment and intersegment takes place. OBJECTIVE: To observe the therapeutic efficacy of acupoint electrogymnastics and traditional electroacupuncture on stroke hemiplegia. DESIGN, TIME AND SETTING: A multicenter, randomized, controlled, blinded, clinical study was performed at the College of Acu-moxibustion and Massage in Shanghai University of Traditional Chinese Medicine from May 2004 to September 2006. PARTICIPANTS: A total of 153 patients suffering from stroke hemiplegia, comprising 83 males and 70 females, aged 63-70 years, were admitted to outpatient and inpatient at LongHua Hospital Affiliated Shanghai University of Traditional Chinese medicine, Putuo District Traditional Chinese Medicine Hospital and Changqiao Street Community Health Service Center of Shanghai. METHODS: The patients were randomly divided into treatment (n = 77) and control (n = 76) groups. They were treated with acupoint electrogymnastics and traditional electroacupuncture, respectively. In the treatment group, two pairs of positive and negative JD-2008 type electrodes from a hemiplegia treatment apparatus were directly pasted on the Shousanli (LI 10) and Waiguan (SJ 5) acupoints of the upper limb, as well as the Zusanli (S 36) and Yanglingquan (GB 34) acupoints of the lower limb, respectively. In the control group, needles were consecutively inserted into the above acupoints. Using the method of lifting-inserting and twisting-rotating, the needle was manipulated with small amplitude of 5-7 mm and a fast frequency of 80-120 times/min when the needle was inserted to a suitable depth. When the sensation of needling was attained, the two pairs of positive and negative electrodes of type G6805-II electro-acupuncture apparatus were connected to the Shousanli (LI 10) and Waiguan (SJ 5) acupoints of the upper limb, as well as the Zusanli (S 36) and Yanglingquan(GB 34) acupoints of the lower limb, respectively. MAIN OUTCOME MEASURES: The Fugl-Meyer evaluation method was used to assess upper limb movement, lower limb movement, sensory stimulation of all limbs, balance, and joint range of motion. RESULTS: Upper-lower limb motor function, limb sensory function, balance, and joint range of motion were significantly ameliorated following treatment, compared with pre-treatment (P < 0.01). There was no significant difference in various indexes between the two groups after treatment (P > 0.05). | 20 | Multi-dimensional and long-term time series monitoring and early warning of landslide hazard with improved cross-platform SAR offset tracking method显示文摘Multi-dimensional, long-term time series displacement monitoring is crucial for generating early warnings for active landslides and for mitigating geohazards. The synthetic aperture radar(SAR) interferometry method has been widely applied to achieve small-gradient landslide displacement monitoring;however, measuring the landslide displacement with a steep gradient has posed certain challenges. In comparison, the SAR offset tracking method is a powerful tool for mapping large-gradient landslide displacement in both the slant-range and azimuth directions. Nevertheless, there are some limitations in the existing SAR offset tracking approaches:(i) the measurement accuracy is greatly reduced by the extreme topography relief in high mountain areas,(ii) a fixed matching window from expert experience is usually adopted in the calculation of cross-correlation,(iii) estimating the long-term displacements between the SAR images from cross-platforms and with longer spatiotemporal baselines is a challenging task, and(iv) it is difficult to calculate the three-dimensional(3D) landslide displacements using a single SAR dataset.Additionally, only a few studies have focused on how to realize early warning of landslide deformation using SAR measurements.To address these issues, this paper presents an improved cross-platform SAR offset tracking method, which can not only estimate high-precision landslide displacements in two and three dimensions but also calculate long-term time series displacements over a decade using cross-platform SAR offset tracking measurements. Initially, we optimize the traditional SAR offset tracking workflow to estimate high-precision ground displacements, in which three improvements are highlighted:(i) an“ortho-rectification” operation is applied to restrain the effect of topography relief,(ii) an “adaptive matching window” is adopted in the cross-correlation computation, and(iii) a new strategy is proposed to combine all the possible offset pairs and optimally design the displacement inversion network based on the “optimization theory” of geodetic inversion. Next, robust mathematical equations are built to estimate the two-dimensional(2D) and 3D long-term time series landslide displacements using cross-platform SAR observations. The M-estimator is introduced into the 2D displacement inversion equation to restrain the outliers, and the total least squares criterion is adopted to estimate the 3D displacements considering the random errors in both the design matrix and observations. We take the Laojingbian landslide, Wudongde Reservoir Area(China), as an example to demonstrate the proposed method using ALOS/PALSAR-1 and ALOS/PALSAR-2 images. The results reveal that the proposed method significantly outperforms traditional methods. We also retrieve the movement direction of each pixel of the Laojingbian landslide using the proposed method, thus allowing us to understand the fine-scale landslide kinematics.
