2Antigenicity of tissues and organs from GGTA1/CMAH/β4GalNT2 triple gene knockout pigs显示文摘Clinical xenotransplantations have been hampered by human preformed antibody-mediated damage of the xenografts.To overcome biological incompatibility between pigs and humans,one strategy is to remove the major antigens[Gal,Neu5 Gc,and Sd(a)]present on pig cells and tissues.Triple gene(GGTAI,CMAH,and β4 GalNT2)knockout(TKO)pigs were produced in our laboratory by CRISPR-Cas9 targeting.To investigate the antigenicity reduction in the TKO pigs,the expression levels of these three xenoantigens in the cornea,heart,liver,spleen,lung,kidney,and pancreas tissues were examined.The level of human IgG/IgM binding to those tissues was also investigated,with wildtype pig tissues as control.The results showed that aGal,Neu5 Gc,and Sd(a)were markedly positive in all the examined tissues in wildtype pigs but barely detected in TKO pigs.Compared to wildtype pigs,the liver,spleen,and pancreas of TKO pigs showed comparable levels of human IgG and IgM binding,whereas corneas,heart,lung,and kidney of TKO pigs exhibited significantly reduced human IgG and IgM binding.These results indicate that the antigenicity of TKO pig is significantly reduced and the remaining xenoantigens on porcine tissues can be eliminated via a gene targeting approach.Ronggen Wang Miaomiao Ruan Runjie Zhang Lei Chen Xiaoxue Li Bin Fang Chu Li Xueyang Ren Jiying Liu Qiang Xiong Lining Zhang Yong Jin Lin Li Rongfeng Li Ying Wang Haiyuan Yang Yifan Dai 2019The Journal of Biomedical Research2019,33,4:3
3Acetytation of ACAP4 regutates CCL18-elicited breast cancer cell migration and invasion显示文摘Tumor metastasis represents the main causes of cancer-related death.Our recent study showed that chemokine CCL18 secreted from tumor-associated macrophages regulates breast tumor metastasis,but the underlying mechanisms remain less clear.Here, we show that ARF6 GTPase-activating protein ACAP4 regulates CCL18-elicited breast cancer cell migration via the acetyltransferase PCAF-mediated acetylation.CCL18 stimulation elicited breast cancer cell migration and invasion via PCAF-dependent acetylation.ACAP4 physically interacts with PCAF and is a cognate substrate of PCAF during CCL18 stimulation.The acetylation site of ACAP4 by PCAF was mapped to Lys311 by mass spectrometric analyses.Importantly,dynamic acetylation of ACAP4 is essential for CCL18-induced breast cancer cell migration and invasion,as overexpression of the persistent acetylation-mimicking or nonacetylatable ACAP4 mutant blocked CCL18-elicited cell migration and invasion.Mechanistically,the acetylation of ACAP4 at Lys311 reduced the lipid-binding activity of ACAP4 to ensure a robust and dynamic cycling of ARF6-ACAP4 complex with plasma membrane in response to CCL18 stimulation.Thus,these results present a previously undefined mechanism by which CCL18-elicited acetylation of the PH domain controls dynamic interaction between ACAP4 and plasma membrane during breast cancer cell migration and invasion.Xiaoyu Song Wei Liu Xiao Yuan Jiying Jiang Wanjuan Wang McKay Mullen Xuannv Zhao Yin Zhang Fusheng Liu Shihao Du Adeel Rehman Ruijun Tian Jian Li Andra Frost Zhenwei Song Hadiyah-Nicole Green Calmour Henry Xing Liu Xia Ding Dongmei Wang Xuebiao Yao 2018Journal of Molecular Cell Biology2018,10,6:3

6Comparison of pH Value, TBA Value, Glycogen and Lactic Acid Content in Muscle of Landrace and Yorkshire under Different Storage Time and Conditions显示文摘In order to assess the meat quality under different storage time and conditions,the pH value,TBA value,glycogen and lactic acid content in muscle of Landrace and Yorkshire were measured. The results showed that within 10 h post slaughtering,post slaughtering time had extremely significant influence on pH value and lactic acid content of Landrace,but had no significant influence on glycogen content and TBA value. On the other hand,post slaughtering time had extremely significant influence on pH value of Yorkshire,but had no significant influence on glycogen,lactic acid content and TBA value. The pH values of Yorkshire at 3,5 and 10 h post slaughtering were 5. 21%( P < 0. 01),5. 64%( P < 0. 05) and 5. 59%( P < 0. 05) higher than those of Landrace,respectively; the lactic acid content in muscle of Landrace at 10 h post slaughtering was 56. 04%( P < 0. 05) higher than that of Yorkshire; the TBA value in muscle of Yorkshire at 10 h post slaughtering was 89. 19%( P < 0. 01) higher than that of Landrace; there was no significant difference in glycogen content between Yorkshire and Landrace. 7A Bama miniature pig model of monoallelic TSC1 mutation for human tuberous sclerosis complex显示文摘Tuberous sclerosis complex(TSC)is a dominant genetic neurocutaneous syndrome characterized by multiple organ hamartomas.Although rodent models bearing a germline mutation in either TSC1 or TSC2 gene have been generated,they do not develop pathogenic lesions matching those seen in patients with TSC because of the significant differences between mice and humans,highlighting the need for an improved large animal model of TSC.Here,we successfully generate monoallelic TSC1-modified Bama miniature pigs using the CRISPR/Cas9 system along with somatic cell nuclear transfer(SCNT)technology.The expression of phosphorylated target ribosomal protein S6 is significantly enhanced in the piglets,indicating that disruption of a TSC1 allele activate the mechanistic target of rapamycin(mTOR)signaling pathway.Notably,differing from the mouse TSC models reported previously,the TSC1^(+/−)Bama miniature pig developed cardiac rhabdomyoma and subependymal nodules,resembling the major clinical features that occur in patients with TSC.These TSC1^(+/−)Bama miniature pigs could serve as valuable large animal models for further elucidation of the pathogenesis of TSC and the development of therapeutic strategies for TSC disease.Xiaoxue Li Tingdong Hu Jiying Liu Bin Fang Xue Geng Qiang Xiong Lining Zhang Yong Jin Xiaorui Liu Lin Li Ying Wang Rongfeng Li Xiaochun Bai Haiyuan Yang Yifan Dai 2020Journal of Genetics and Genomics2020,47,12:2
8All-fiber spatiotemporal mode-locking lasers with large modal dispersion显示文摘It is a challenging problem to balance the modal walk-off(modal dispersion) between multiple transverse modes and chromatic dispersion in long step-index multimode fibers(MMFs). By properly designing the oscillator, we have overcome the difficulty and successfully obtained an all-fiber spatiotemporal mode-locked laser based on step-index MMFs with large modal dispersion for the first time, to our knowledge. Various proofs of spatiotemporal mode-locking(STML) such as spatial, spectral, and temporal properties, are measured and characterized.This laser works at a fundamental frequency of 28.7 MHz, and achieves a pulse laser with single pulse energy of 8 nJ, pulse width of 20.1 ps, and signal-to-noise ratio of-70 dB. 11Mathematical Model of Decarburization of Ultra Low Carbon Steel during RH Treatment显示文摘According to the balance of carbon and oxygen, a decarburization model for the RH treatment has been developed. in which the influence of the mass transfer of carbon and oxygen in the liquid steel and the stirring energy (ε) in the vacuum vessel on decarburization rate has been considered. 12Influence of long-period pesticide force on genetic polymorphism of wolf spider Pardosa pseudoannulata (Lycosidae: Araneae)显示文摘Wolf spiders are predators in large quantities in the fields. How wolf spiders keep their dominant species status under long-period pesticide force? To answer this question, eight geographical populations of Pardosa pseudoannulata were used as materials to test the influence of geographical habitats on their genomic DNA polymorphism. The RAPD pattern showed polymorphic variations among and within different populations. Total 84 bands amplified by 10 random primers, of which 62 (73.81%) are polymorphic, were generated from 55 individuals of eight geographical populations. Meanwhile, Shannon’s index (Ho= 0.5177) showed a rich genetic diversity of P. pseudoannulata, and most of the genetic variation (64.24%) was found within populations. 16Combined Treatment with Electrocauterization,Carbon Dioxide Laser,and Microneedle Fractional Radiofrequency for Facial Angiofibromas in Tuberous Sclerosis Complex:A Case Report and Literature Review显示文摘Tuberous sclerosis complex is a type of genetic multisystem disease that causes hamartomas in various organs.Facial angiofibromas commonly occur in 80%of patients and are prominently distributed over the cheek,chin,and nasolabial folds with severe disfigurement and emotional distress.Recently,photoelectric devices have been identified for the treatment of angiofibromas with great efficacy and fewer side effects.We report a case of a 42-year-old man with facial angiofibromas,who was treated with a combination of high-frequency electrocauterization,Ultrapulse CO_(2) laser,and microneedle fractional radiofrequency with 7 sessions and a 6-month follow-up.The patient showed great improvement in relation to the elevated lesions and nodules.A low recurrence rate was observed.This is the first study to investigate the efficacy of high-frequency electrocauterization and microneedle fractional radiofrequency in angiofibromas.It may provide an optimal approach for clinicians wherein a combined treatment of various lasers and electric devices is effective for complicated,protuberant,and firm angiofibromas of specific patients.Jiying DONG Shen WANG Min YAN Jing MI Ying ZENG Huyan LIN Yiqiu ZHANG Min YAO 2021Chinese Journal Of Plastic and Reconstructive Surgery2021,3,1:1
