2Structure of African swine fever virus p15 reveals its dual role for membrane-association and DNA binding显示文摘Dear Editor, African swine fever (ASF) is a highly contagious disease of domestic pigs caused by African swine fever virus (ASFV),with mortality rates approaching 90%-100%.The first ASF case was described in 1921 in Kenya.Over the past decades, ASF has spread from Sub-Saharan Africa to Europe and Asia, and pose a huge threat to global food safety.Since the first ASF case reported in August 2018, more than 10 million pigs had been culled in China at an enormous economic loss.The risk of ASFV mutating to infect humans is extremely low.However, ASF not only threatens food security but also severely strikes the world economy in many fields, such as the global supply of the blood thinner heparin,most of which is produced from pigs.Hence, ASF is a strictly monitored infectious animal disease by OIE (World Organization for Animal Health), unfortunately, there is no approved drug or vaccine against this lethal viral disease.Dan Fu Dongming Zhao Wei Zhang Guangshun Zhang Mingyu Li Zheng Zhang Yuhui Wang Dongdong Sun Peng Jiao Cheng Chen Yu Guo Zihe Rao 2020Protein & Cell2020,11,8:3
3A urinary proteomic landscape of COVID-19 progression identifies signaling pathways and therapeutic options显示文摘Signaling pathway alterations in COVID-19 of living humans as well as therapeutic targets of the host proteins are not clear.We analyzed 317 urine proteomes,including 86 COVID-19,55 pneumonia and 176 healthy controls,and identified specific RNA virus detector protein DDX58/RIG-I only in COVID-19 samples.Comparison of the COVID-19 urinary proteomes with controls revealed major pathway alterations in immunity,metabolism and protein localization.Biomarkers that may stratify severe symptoms from moderate ones suggested that macrophage induced inflammation and thrombolysis may play a critical role in worsening the disease.Hyper activation of the TCA cycle is evident and a macrophage enriched enzyme CLYBL is up regulated in COVID-19 patients.As CLYBL converts the immune modulatory TCA cycle metabolite itaconate through the citramalyl-CoA intermediate to acetyl-CoA,an increase in CLYBL may lead to the depletion of itaconate,limiting its anti-inflammatory function.These observations suggest that supplementation of itaconate and inhibition of CLYBL are possible therapeutic options for treating COVID-19,opening an avenue of modulating host defense as a means of combating SARS-CoV-2 viruses.Yuntao Liu Lan Song Nairen Zheng Jinwen Shi Hongxing Wu Xing Yang Nianci Xue Xing Chen Yimin Li Changqing Sun Cha Chen Lijuan Tang Xiaotian Ni Yi Wang Yaling Shi Jianwen Guo Guangshun Wang Zhongde Zhang Jun Qin 2022Science China(Life Sciences)2022,65,9:1

16Leucine-rich glioma-inactivated protein 1 antibody-mediated autoimmune encephalitis in a 4-year-old girl:a case report显示文摘Background:Leucine-rich glioma-inactivated protein 1(LGI1)antibody-mediated encephalitis is a rare subtype of autoimmune encephalopathy,which is associated with autoimmunity against the neuronal plasma membrane proteins.The characteristic symptoms of this disease are memory dysfunction,seizures,faciobrachial dystonic seizures,cognitive deficits,neuropsychiatric disturbances,and intractable hyponatremia.The diagnosis of this disease mainly depends on the presence of anti-LGI1 antibody in serum or cerebrospinal fluid of patients.LGI1 antibody encephalitis has been reported mostly in adults,with rare occurrences in children.Case presentation:In this report,we described a 4-year-old girl with typical seizures.Seizure types included focal seizures and generalized tonic-clonic seizures.The electroencephalogram findings showed focal discharges.Brain magnetic resonance imaging(MRI)showed normal.The cerebrospinal fluid(CSF)levels of cells,glucose,and chloride were within the normal range,and the culture did not reveal growth of any pathogen.Test of serum LGI1-Ab was positive,while the tests for autoimmune encephalitis antibody series in CSF were negative.The seizures of the patient were completely controlled after the therapy of immunoglobulin,methylprednisolone and antiepileptic drugs(AEDs),and the mental state almost returned to normal.Conclusion:To our knowledge,the patient described here may be the youngest case of LGI1 antibody encephalitis reported to date.Children with the LGI1 antibody-associated encephalitis may present only with single symptoms such as epileptic seizures and have good response to the therapy of immunoglobulin,methylprednisolone and antiepileptic drugs.Our case report will provide hints for pediatricians in the diagnosis and treatment of LGI1-antibody encephalitis.Junxia Luo Jianguo Shi Yehong Chen Wandong Hu Yujie Guo Guangshun Hou Zaifen Gao 2021Acta Epileptologica2021,3,1:1
