| 1 | Manipulating mRNA splicing by base editing in plants显示文摘Precursor-mRNAs(pre-mRNA) can be processed into one or more mature m RNA isoforms through constitutive or alternative splicing pathways. Constitutive splicing of pre-mRNA plays critical roles in gene expressional regulation, such as intronmediated enhancement(IME), whereas alternative splicing(AS) dramatically increases the protein diversity and gene functional regulation. However, the unavailability of mutants for individual spliced isoforms in plants has been a major limitation in studying the function of mRNA splicing. Here, we describe an efficient tool for manipulating the splicing of plant genes. Using a Cas9-directed base editor, we converted the 5′ splice sites in four Arabidopsis genes from the activated GT form to the inactive AT form. Silencing the AS of HAB 1.1(encoding a type 2 C phosphatase) validated its function in abscisic acid signaling, while perturbing the AS of RS31 A revealed its functional involvement in plant response to genotoxic treatment for the first time. Lastly,altering the constitutive splicing of Act2 via base editing facilitated the analysis of IME. | 3 | Rational design of novel ultra-small amorphous Fe_(2)O_(3)nanodots/graphene heterostructures for all-solid-state asymmetric supercapacitors显示文摘Constructing graphene-based heterostructures with large interfacial area is an efficient approach to enhance the electrochemical performance of supercapacitors but remains great challenges in their synthesis.Herein,a novel ultra-small amorphous Fe_(2)O_(3)nanodots/graphene heterostructure(a-Fe_(2)O_(3)NDs/RGO)aerogel was facilely synthesized via excessive metal-ion-induced self-assembly and subsequent calcination route using Prussian blue/graphene oxide(PB/GO)composite aerogel as precursors.The deliberately designed a-Fe_(2)O_(3)NDs/RGO heterostructure offers a highly interconnected porous conductive network,large heterostructure interfacial area,and plenty of accessible active sites,greatly facilitating the electron transfer,electrolyte diffusion,and pseudocapacitive reactions.The obtained a-Fe_(2)O_(3)NDs/RGO aerogel could be used as flexible free-standing electrodes after mechanical compression,which exhibited a significantly enhanced specific capacitance of 347.4 F·g^(-1)at 1 A·g^(-1),extraordinary rate capability of 184 F·g^(-1)at 10 A·g^(-1),and decent cycling stability.With the as-prepared a-Fe_(2)O_(3)NDs/RGO as negative electrodes and the Co_(3)O_(4)NDs/RGO as positive electrodes,an all-solid-state asymmetric supercapacitor(a-Fe_(2)O_(3)NDs/RGO//Co_(3)O_(4)NDs/RGO asymmetric supercapacitor(ASC))was assembled,which delivered a high specific capacitance of 69.1 F·g^(-1)at 1 A·g^(-1)and an impressive energy density of 21.6 W·h·k·g^(-1)at 750 W·k·g^(-1),as well as good cycling stability with a capacity retention of 94.3%after 5,000 cycles.This work provides a promising avenue to design high-performance graphene-based composite electrodes and profound inspiration for developing advanced flexible energy-storage devices. | Chenxiao Wu Zhifang Zhang Zhonghui Chen Zuimin Jiang Huiyu Li Haijing Cao Yongsheng Liu Yanyan Zhu Zebo Fang Xiangrong Yu | 2021 | Nano Research2021,14,4: | 6 |

| 5 | Transient expression of a TaGRF4-TaGIF1 complex stimulates wheat regeneration and improves genome editing显示文摘Genome editing is an unprecedented technological breakthrough but low plant regeneration frequencies and genotype dependence hinder its implementation for crop improvement. Here, we found that transient expression of a complex of the growth regulators TaGRF4 and TaGIF1(TaGRF4-TaGIF1) increased regeneration and genome editing frequency in wheat. When we introduced synonymous mutation in the miR396 target site of TaGRF4, the resulting complex(mTaGRF4-TaGIF1) performed better than original TaGRF4-TaGIF1. Use of m TaGRF4-TaGIF1 together with a cytosine base editor targeting TaALS resulted in 2-9-fold increases in regeneration and transgene-free genome editing in 11 elite common wheat cultivars. | 6 | Genome editing in plants with MAD7 nuclease显示文摘MAD7 is an engineered nuclease of the Class 2 type V-A CRISPR-Cas(Cas12 a/Cpf1)family with a low level of homology to canonical Cas12 a nucleases.It has been publicly released as a royalty-free nuclease for both academic and commercial use.Here,we demonstrate that the CRISPR-MAD7 system can be used for genome editing and recognizes T-rich PAM sequences(YTTN)in plants.Its editing efficiency in rice and wheat is comparable to that of the widely used CRISPR-Lb Cas12 a system.We develop two variants,MAD7-RR and MAD7-RVR that increase the target range of MAD7,as well as an M-AFID(a MAD7-APOBEC fusion-induced deletion)system that creates predictable deletions from 50-deaminated Cs to the MAD7-cleavage site.Moreover,we show that MAD7 can be used for multiplex gene editing and that it is effective in generating indels when combined with other CRISPR RNA orthologs.Using the CRISPR-MAD7 system,we have obtained regenerated mutant rice and wheat plants with up to 65.6%efficiency. | Qiupeng Lin Zixu Zhu Guanwen Liu Chao Sun Dexing Lin Chenxiao Xue Shengnan Li Dandan Zhang Caixia Gao Yanpeng Wang Jin-Long Qiu | 2021 | Journal of Genetics and Genomics2021,48,6: | 4 |

| 8 | PD-based trajectory tracking control in automatic cell injection system显示文摘Trajectory tracking technology has been the focus of industrial manipulatory applications for many years, and its research has been found in micromanipulation in bioengineering recently. In this paper, a hybrid vision and force control method is applied to the automatic cell injection. The three-dimensional cell injection process involves the trajectory tracking in free space and the force control in contact space. A PD plus feedforward compensation control method is applied to the trajectory tracking in 3D space. Further, a PD-based robust controller is introduced into trajectory tracking while the systemic uncertainty of the cell injection is additionally considered. Both of the two control methods are theoretically proved to be exponential convergent. | 10 | Multistage-batch bipolar membrane electrodialysis for base production from high-salinity wastewater显示文摘Bipolar membrane electrodialysis(BMED)is considered a state-of-the-art technology for the conversion of salts into acids and bases.However,the low concentration of base generated from a traditional BMED process may limit the viability of this technology for a large-scale application.Herein,we report an especially designed multistage-batch(two/three-stage-batch)BMED process to increase the base concentration by adjusting different volume ratios in the acid(Vacid),base(Vbase),and salt compartments(Vsalt).The findings indicated that performance of the two-stage-batch with a volume ratio of Vacid:Vbase:Vsalt=1:1:5 was superior in comparison to the threestage-batch with a volume ratio of Vacid:Vbase:Vsalt=1:1:2.Besides,the base concentration could be further increased by exchanging the acid produced in the acid compartment with fresh water in the second stage-batch process.With the two-stage-batch BMED,the maximum concentration of the base can be obtained up to 3.40 mol∙L^(-1),which was higher than the most reported base production by BMED.The low energy consumption and high current efficiency further authenticate that the designed process is reliable,cost-effective,and more productive to convert saline water into valuable industrial commodities. | Arif Hussain Haiyang Yan Noor Ul Afsar Chenxiao Jiang Yaoming Wang Tongwen Xu | 2022 | Frontiers of Chemical Science and Engineering2022,16,5: | 2 |

| 14 | Gelation of Hole Transport Layer to Improve the Stability of Perovskite Solar Cells显示文摘To achieve high power conversion efficiency(PCE) and long-term stability of perovskite solar cells(PSCs), a hole transport layer(HTL) with persistently high conductivity, good moisture/oxygen barrier ability, and adequate passivation capability is important. To achieve enough conductivity and effective hole extraction, spiro-OMe TAD, one of the most frequently used HTL in optoelectronic devices, often needs chemical doping with a lithium compound(LiTFSI). However, the lithium salt dopant induces crystallization and has a negative impact on the performance and lifetime of the device due to its hygroscopic nature. Here, we provide an easy method for creating a gel by mixing a natural small molecule additive(thioctic acid, TA) with spiro-OMe TAD. We discover that gelation effectively improves the compactness of resultant HTL and prevents moisture and oxygen infiltration. Moreover, the gelation of HTL improves not only the conductivity of spiro-OMe TAD, but also the operational robustness of the devices in the atmospheric environment. In addition, TA passivates the perovskite defects and facilitates the charge transfer from the perovskite layer to HTL. | 15 | Multi-year assessment of paralytic shellfish toxins in hard clam species along the coastline of Jiangsu Province, China显示文摘Paralytic shellfish toxins(PSTs) are notorious neurotoxins that threaten public health and food safety worldwide.Although PST monitoring programs have recently been established throughout China, the profiles and variation of PSTs in important commercial clams(e.g., Mactra veneriformis, Ruditapes philippinarum, and Meretrix meretrix) along the Jiangsu Province coastline remain largely unexplored. In this study, a validated hydrophilic interaction liquid chromatography–tandem mass spectrometry(HILIC-MS/MS) method was used to examine PST profiles and levels in 540 clam samples from natural production areas along Jiangsu Province coastline during2014–2016. Although the PST levels(≤6.38 μg saxitotoxin equivalents(eq)/kg) were consistently below European Union regulatory limits(≤800 μg saxitotoxin eq/kg) during this time period, saxitotoxin, decarbamoylsaxitotoxin,and gonyautoxins 1 and 4 were detected, and nearly 40% of the samples were saxitotoxin-positive. The PST levels also varied significantly by seasons, with peak values observed in May during 2014–2016. Compared with the existing results, the newly obtained bounds are less conservative and more practical, which means that the condition for the existence of the upper bounds derived here is much weaker.
