2Variable frequency of pulse hydraulic fracturing for improving permeability in coal seam显示文摘Variable frequency,a new pattern of pulse hydraulic fracturing,is presented for improving permeability in coal seam.A variable frequency pulse hydraulic fracturing testing system was built,the mould with triaxial loading was developed.Based on the monitor methods of pressure sensor and acoustic emission,the trials of two patterns of pulse hydraulic fracturing of single frequency and variable frequency were carried out,and at last fracturing mechanism was analyzed.The results show that the effect of variable frequency on fracture extension is better than that of single frequency based on the analysis of macroscopic figures and AE.And the shortage of single frequency is somewhat remedied when the frequency is variable.Under variable frequency,the pressure process can be divided into three stages:low frequency band,pressure stability band and high frequency band,and rupture pressure of the sample is smaller than that of the condition of single frequency.Based on the Miner fatigue theory,the effect of different loading sequences on sample rupture is discussed and the results show that it is better to select the sequence of low frequency at first and then high frequency.Our achievements can give a basis for the improvement and optimization of the pulse hydraulic fracturing technology.Li Quangui Lin Baiquan Zhai Cheng Ni Guanhua Peng Shen Sun Chen Cheng Yanying 2013International Journal of Mining Science and Technology2013,23,6:12
3Sinomenine reduces iNOS expressionvia inhibiting the T-bet IFN-γ pathway in experimental autoimmune encephalomyelitis in rats显示文摘Sinomenine is a bioactive alkaloid isolated from the Chinese medicinal plant Sinomenium acutum.It is widely used as an immunosuppressive drug for treating rheumatic and arthritic diseases.In our previous studies,we found that sinomenine reduced cellular infiltration within the spinal cord and alleviated experimental autoimmune encephalomyelitis(EAE) in rats.In this study,we further investigated the mechanisms of sinomenine treatment in EAE rats.In EAE rats,treatment with sinomenine exerted an anti-inducible NO synthase(anti-iNOS) effect,which is related to the reductions of Th1 cytokine interferon-γ(IFN-γ) and its transcription factor,T-bet,in spinal cords.Moreover,sinomenine treatment of splenocytes stimulated with anti-CD3 antibody and recombinant rat interleukin 12 reduced the expression of T-bet and IFN-γ in vitro and also reduced the capability of supernatants of splenocyte culture to induce iNOS expression by primary astrocytes.However,sinomenine had no direct inhibitory effect on iNOS produced by astrocytes cultured with IFN-γ and tumor necrosis factor α in vitro.In conclusion,the anti-iNOS effect of sinomenine on EAE is mediated via the suppression of T-bet /IFN-γ pathway.Bingjie Gu Yanying Zeng Cheng Yin Huijiuan Wang Xiaofan Yang Song Wang Xiaohui Ji 2012The Journal of Biomedical Research2012,26,6:11
4Establishment and application of drilling sealing model in the spherical grouting mode based on the loosing-circle theory显示文摘There are quite a few studies that have been done on borehole sealing theory both domestically and internationally.The existing researches usually consider drilling of the surroundings as a dense homogeneous elastic body which does not meet the characteristics of real drilling of the fractured body.Based on the loosing-circle theory and analyses of the surrounding rock stress field,cracks and seepage fields,combined with Newtonian fluid spherical grouting model,we deduced the dynamic relationship between the seepage coefficient and rock or grouting parameters of the drilling sealing fluid mode of spherical fissure grouting.In this experiment,mucus was injected in the simulated coal seam and the permeability coefficient of the sealing body was calculated by using the model.To verify the validity of the model,the calculated sealing body number was compared with the extreme negative pressure that the sealing body could withstand.The theoretical model revealed the drilling sealing fluid mechanism,provided a method for the quantitative calculation of the drilling sealing fluid effect by grouting mode and a reference for the subsequent research of sealing mechanism.Hao Zhiyong Lin Baiquan Gao Yabin Cheng Yanying 2012International Journal of Mining Science and Technology2012,22,6:6
5Measurement of electric properties of the single supersaturated aerosol droplet显示文摘A system for measuring the electric properties of single aerosol droplet is designed and applied to the NaClO4 aerosol droplet in different relative humidity (RH). The conductance and capacitance are ob-tained within the whole RH range, especially in the supersaturated state which cannot be acquired from the bulk solution. 6Rac1 Modulates Excitatory Synaptic Transmission in Mouse Retinal Ganglion Cells显示文摘Ras-related C3 botulinum toxin substrate 1(Racl),a member of the Rho GTPase family which plays important roles in dendritic spine morphology and plasticity,is a key regulator of cytoskeletal reorganization in dendrites and spines.Here,we investigated whether and how Racl modulates synaptic transmission in mouse retinal ganglion cells(RGCs)using selective conditional knockout of Racl(Racl-cKO).Racl-cKO significantly reduced the frequency of AMPA receptor-mediated miniature excitatory postsynaptic currents,while glycine/GABA_A receptor-mediated miniature inhibitory postsynaptic currents were not affected.Although the total GluA1 protein level was increased in Racl-cKO mice,its expression in the membrane component was unchanged.RaclcKO did not affect spine-like branch density in single dendrites,but significantly reduced the dendritic complexity,which resulted in a decrease in the total number of dendritic spine-like branches.These results suggest that Racl selectively affects excitatory synaptic transmission in RGCs by modulating dendritic complexity.Ling-Zhu Li Ning Yin Xue-Yan Li Yanying Miao Shuo Cheng Fang Li Guo-Li Zhao Shu-Min Zhong Xin Wang Xiong-Li Yang Zhongfeng Wang 2019Neuroscience Bulletin2019,35,4:2
7Dopamine D2 Receptor-Mediated Modulation of Rat Retinal Ganglion Cell Excitability显示文摘Ganglion cells(RGCs) are the sole output neurons of the retinal circuity. Here, we investigated whether and how dopamine D2 receptors modulate the excitability of dissociated rat RGCs. Application of the selective D2 receptor agonist quinpirole inhibited outward K^+ currents, which were mainly mediated by glybenclamide-and 4-aminopyridine-sensitive channels, but not the tetraethylammonium-sensitive channel. In addition,quinpirole selectively enhanced Nav1.6 voltage-gated Na^+ currents. The intracellular c AMP/protein kinase A,Ca^2+/calmodulin-dependent protein kinase Ⅱ, and mitogen-activated protein kinase/extracellular signal-regulated kinase signaling pathways were responsible for the effects of quinpirole on K^+ and Na^+ currents, while phospholipase C/protein kinase C signaling was not involved. Under current-clamp conditions, the number of action potentials evoked by positive current injection was increased by quinpirole. 8Association of ALOX5AP and PDE4D with the risk of lacunar infarct in people from Jiangsu Province,China显示文摘The genes for 5-lipoxygenase activating protein (ALOX5AP) and phosphodiesterase 4D (PDE4D) have been demonstrated as susceptibility genes for lacunar in the Icelandic and Pakistani populations,but little is known about the role of these genes in Chinese populations.The present study utilized polymerase chain reaction and ligase detection reaction to detect single nucleotide polymorphisms (SNPs) in 280 consecutive stroke patients and 258 unrelated population-based controls from Nanjing,Jiangsu Province,China.The allele frequency,genotypes,and haplotypes of the two SNPs (rs456009 and rs966221) in PDE4D were similar between the two groups.However,A allele frequency of rs4073259 (A/G) and rs4769055 (A/C) in the ALOX5AP gene exhibited differences in two groups,and especially the haplotype of the SNP was significantly different between the two groups.Results suggested that the ALOX5AP gene might be involved in lacunar infarct,while PDE4D gene was not a risk factor for lacunar infarct in individuals from Jiangsu Province,China.Hong Cheng Qingwen Jin Lixin Li Xinsheng Ding Xinjian Song Yanying Zeng 2011Neural Regeneration Research2011,6,12:2

10Study on conductance of supersaturated chloride microdroplets显示文摘By using the measuring system previously designed by the authors, the conductance of KCl, NaCl and NH4Cl microdroplets is obtained in the whole measuring RH range, especially in the supersaturation region, which cannot be acquired from the bulk solutions and fills the gap of lack of experimental data of conductance under the supersaturated state. The ERH and DRH of these three kinds of microdroplets observed from a microscope are 80.5% and 95.4% (KCl), 75.7% and 93.3% (NaCl), and 69.9% and 96.6% (NH4Cl), respectively. 14The existence of a nonclassical TCA cycle in the nucleus that wires the metabolic-epigenetic circuitry显示文摘The scope and variety of the metabolic intermediates from the mitochondrial tricarboxylic acid(TCA)cycle that are engaged in epigenetic regulation of the chromatin function in the nucleus raise an outstanding question about how timely and precise supply/consumption of these metabolites is achieved in the nucleus.We report here the identification of a nonclassical TCA cycle in the nucleus(nTCA cycle).We found that all the TCA cycle-associated enzymes including citrate synthase(CS),aconitase 2(ACO2),isocitrate dehydrogenase 3(IDH3),oxoglutarate dehydrogenase(OGDH),succinyl-CoA synthetase(SCS),fumarate hydratase(FH),and malate dehydrogenase 2(MDH2),except for succinate dehydrogenase(SDH),a component of electron transport chain for generating ATP,exist in the nucleus.We showed that these nuclear enzymes catalyze an incomplete TCA cycle similar to that found in cyanobacteria.We propose that the nTCA cycle is implemented mainly to generate/consume metabolic intermediates,not for energy production.We demonstrated that the nTCA cycle is intrinsically linked to chromatin dynamics and transcription regulation.Together,our study uncovers the existence of a nonclassical TCA cycle in the nucleus that links the metabolic pathway to epigenetic regulation.Xujun Liu Wenzhe Si Lin He Jianguo Yang Yani Peng Jie Ren Xiaoping Liu Tong Jin Huajing Yu Zihan Zhang Xiao Cheng Wenting Zhang Lu Xia Yunchao Huang Yue Wang Shumeng Liu Lin Shan Yu Zhang Xiaohan Yang Haixia Li Jing Liang Luyang Sun Yongfeng Shang 2021Signal Transduction and Targeted Therapy2021,6,12:0
15Preventive and therapeutic benefits of nelfinavir in rhesus macaques and human beings infected with SARS-CoV-2显示文摘Effective drugs with broad spectrum safety profile to all people are highly expected to combat COVID-19 caused by SARS-CoV-2.Here we report that nelfinavir,an FDA approved drug for the treatment of HIV infection,is effective against SARS-CoV-2 and COVID-19.Preincubation of nelfinavir could inhibit the activity of the main protease of the SARS-CoV-2(IC50=8.26μM),while its antiviral activity in Vero E6 cells against a clinical isolate of SARS-CoV-2 was determined to be 2.93μM(EC50).In comparison with vehicle-treated animals,rhesus macaque prophylactically treated with nelfinavir had significantly lower temperature and significantly reduced virus loads in the nasal and anal swabs of the animals.At necropsy,nelfinavir-treated animals had a significant reduction of the viral replication in the lungs by nearly three orders of magnitude.A prospective clinic study with 37 enrolled treatment-naive patients at Shanghai Public Health Clinical Center,which were randomized(1:1)to nelfinavir and control groups,showed that the nelfinavir treatment could shorten the duration of viral shedding by 5.5 days(9.0 vs.14.5 days,P=0.055)and the duration of fever time by 3.8 days(2.8 vs.6.6 days,P=0.014)in mild/moderate COVID-19 patients.The antiviral efficiency and clinical benefits in rhesus macaque model and in COVID-19 patients,together with its well-established good safety profile in almost all ages and during pregnancy,indicated that nelfinavir is a highly promising medication with the potential of preventative effect for the treatment of COVID-19.Zhijian Xu Danrong Shi Jian-Bao Han Yun Ling Xiangrui Jiang Xiangyun Lu Chuan Li Likun Gong Guangbo Ge Yani Zhang Yi Zang Tian-Zhang Song Xiao-Li Feng Ren-Rong Tian Jia Ji Miaojin Zhu Nanping Wu Chunhui Wu Zhen Wang Yechun Xu Cheng Peng Min Zheng Junling Yang Feifei Du Junliang Wu Peipei Wang Jingshan Shen Jianliang Zhang Yong-Tang Zheng Hangping Yao Weiliang Zhu 2023Signal Transduction and Targeted Therapy2023,8,5:0
16Effects of Different Afforestation Techniques on Soil Nutrients in Populus alba Woodland显示文摘[Objective] The research aimed to study the influence of different afforestation technology measures on soil nutrients of P. alba woodland in arid valley region of Rikaze,Tibet. It aimed to improve afforestation technology measure in arid valley region of Rikaze,Tibet,and provide theoretic basis for restoring the degraded ecosystem at upper reaches of the Yarlung Zangbo River. [Method] In arid valley region of Duojiao Village,Nanmulin County,Rikaze City,the planted land using rooting powder,egg shell,leaves,mud and water retention agent was taken as research object,while the land without any afforestation measures was taken as the control,to determine soil nutrients under different afforestation measures.[Result]Under different afforestation measures,the contents of various soil nutrients were all obviously higher than control,in which available potassium,available phosphorus,alkali hydrolysable nitrogen and organic matter in soil obviously increased than control,but there was no similar rule among different measures. 18Review of constraints and critical success factors of developing urban underground space显示文摘The development of urban underground space(UUS)is of great significance for sustainable urban development because it has potential to solve many urban problems.However,the development of UUS is limited because there are still a lot of constraints and challenges.To better understand the constraining factors of developing UUS and improve the development and utilization of UUS,this study conducts a comprehensive survey of existing literature retrieved from the Web of Science Core Collection database.Five main constraining factors,including the limited geological conditions,high cost,lack of natural light and ventilation,high incidence,and difficulty in collecting data and information on UUS,have been discussed to better understand how they affect the development of UUS.Based on the survey,the critical success factors of developing UUS,such as the effective governance structure,planning system for integrating UUS into urban development,coordination of the design of ground and underground,delineation of land property rights of UUS,and application of information techniques in the use of UUS,have been identified and discussed.In addition,knowledge gaps in the studies on development of UUS have been identified.Finally,the research trends and future directions have been proposed for better understanding and practice of UUS development.Yani Lai Yuling Wang Jing Cheng Xiangsheng Chen Quan Liu 2023Underground Space2023,,5:0
