| 1 | Effect of Nimesulide on proliferation and apoptosis of human hepatoma SMMC-7721 cells显示文摘AIM: Cyclooxygenase-2 (COX-2) has been suggested to beassociated with carcinogenesis. We sought to investigatethe effect of the selective COX-2 inhibitor, Nimesulide onproliferation and apoptosis of SMMC-7721 human hepatomacells.METHODS: This study was carried out on the culture ofhepatic carcinoma SMMC-7721 cell line. Variousconcentrations of Nimesulide (0、200 μmol/L、300 μmol/L、400μmol/L) were added and incubated. Cell proliferation wasdetected with MTT colorimetric assay, cell apoptosis byelectron microscopy, flow cytometry and TUNEL.RESULTS: Nimesulide could significantly inhibit SMMC-7721cells proliferation dose-dependent and in a dependentmanner compared with that of the control group. Theduration lowest inhibition rate produced by Nimesulide inSMMC-7721 cells was 19.06 %, the highest inhibition ratewas 58.49 %. After incubation with Nimesulide for 72 h, themost highest apoptosis rate and apoptosis index of SMMC-7721 cells comparing with those of the control were 21. 20 %+1.62% vs2.24% +0.26% and21.23+ 1.78 vs2.01+0.23(p<0.05).CONCLUSION: The selective COX-2 inhibitor, Nimesulide caninhibit the proliferation of SMMC-7721 cells and increaseapoptssis rate and apoptosis index of SMMC-7721 cells. | 2 | Galangin induces apoptosis of hepatocellular carcinoma cells via the mitochondrial pathway显示文摘AIM:To investigate the mechanism by which galangin,a polyphenolic compound derived from medicinal herbs,induces apoptosis of hepatocellular carcinoma(HCC) cells.METHODS:The 3-(4,5-Dimethyl-thiazol-2-yl)-2,5-diphenyl-tetrazolium bromide assay was used to measure cell viability.Apoptosis was evaluated by in situ uptake of propidium iodide and Hoechst 33258 and was then detected by fluorescence microscopy.Protein expressions were detected by Western blotting.To confirm the apoptotic pathway mediated by galangin,cells were transfected by bcl-2 gene to overexpress Bcl-2 or siRNA to down-regulate Bcl-2 expression.RESULTS:Galangin(46.25-370.0 μmol/L) exerted an anti-proliferative effect,induced apoptosis,and decreased mitochondrial membrane potential in a dose and time-dependent manner.Treatment with galangin induced apoptosis by translocating the pro-apoptotic protein Bax to the mitochondria,which released apoptosis-inducing factor and cytochrome c into the cytosol.Overexpression of Bcl-2 attenuated galangin-induced HepG2 cell apoptosis,while decreasing Bcl-2 expression enhanced galangin-induced cell apoptosis.CONCLUSION:Our data suggests that galangin mediates apoptosis through a mitochondrial pathway,and may be a potential chemotherapeutic drug for the treatment of HCC. | Hai-Tao Zhang Liu-Bo Lan Da-Hua Fan Kai-Dan Zhu Xiao-Yi Chen Min Wen Hui-Ming Liu Guangdong Province China Hui Luo | 2010 | World Journal of Gastroenterology2010,16,27: | 32 |
| 3 | A novel gene delivery system targeting cells expressing VEGF receptors显示文摘Two ligand oligopeptides GV1 and GV2 were designed according to the putative binding region of VEGF to its receptors. GV1, GV2 and endosome releasing oligopeptide HA20 were conjugated with poly-L-lysine or protamine and the resulting conjugates could interact with DNA in a noncovalent bond to form a complex. Using pSV2-β-galactosidase as a reporter gene, it has been demonstrated that exogenous gene was transferred into bovine aortic arch-derived endothelial cells (ABAE) andhuman malignant melanoma cell lines (A375) in vitro. In vivo experiments, exogenous gene was transferred into tumor vascular endothelial cells and tumor cells of subcutaneously transplanted human colon cancer LOVO, human malignant melanoma A375 and human hepatoma graft in nude mice. This system could also target gene to intrahepatically transplanted human hepatoma injected via portal vein in nude mice. | 4 | Grey GM(1,1) Model with Function-Transfer Method for Wear Trend Prediction and its Application显示文摘Trend forecasting is an important aspect in fault diagnosis and work state supervision. The principle, where Grey theory is applied in fault forecasting, is that the forecast system is considered as a Grey system; the existing known information is used to infer the unknown information’s character, state and development trend in a fault pattern, and to make possible forecasting and decisions for future development. It involves the whitenization of a Grey process. But the traditional equal time interval Grey GM (1,1) model requires equal interval data and needs to bring about accumulating addition generation and reversion calculations. Its calculation is very complex. However, the non equal interval Grey GM (1,1) model decreases the condition of the primitive data when establishing a model, but its requirement is still higher and the data were pre processed. The abrasion primitive data of plant could not always satisfy these modeling requirements. Therefore, it establishes a division method suited for general data modeling and estimating parameters of GM (1,1), the standard error coefficient that was applied to judge accuracy height of the model was put forward; further, the function transform to forecast plant abrasion trend and assess GM (1,1) parameter was established. These two models need not pre process the primitive data. It is not only suited for equal interval data modeling, but also for non equal interval data modeling. Its calculation is simple and convenient to use. The oil spectrum analysis acted as an example. The two GM (1,1) models put forward in this paper and the new information model and its comprehensive usage were investigated. | 6 | Endoscopic sphincterotomy associated cholangitis in patients receiving proximal biliary self-expanding metal stents显示文摘BACKGROUND: Endoscopic biliary stent placement during the procedure of endoscopic retrograde cholangiopancreatography (ERCP) is preferred to provide biliary drainage for unresectable malignant biliary obstruction. There is considerable controversy over the use of endoscopic sphincterotomy (ES) prior to stent placement. This study aimed to determine whether ES before intraductal self-expanding metal stent (SEMS) placement affects the clinical outcome and complications in patients with proximal malignant obstructive biliary diseases. METHODS: In a prospective randomized controlled trial, 82 patients with inoperable malignant biliary strictures were randomly assigned to biliary stenting groups with or without ES. Resolution of jaundice and the incidence of complications including acute cholangitis, pancreatitis and stent occlusion within 6 months were evaluated. RESULTS: SEMSs were successfully deployed in all patients, resulting in clinical and biochemical improvement of obstructive symptoms in both groups. The incidence of cholangitis was higher in the ES group than in the non-ES group (58.5% vs 31.7%, P=0.015). The interval between stent placement and the first acute cholangitis was much shorter in the ES group than in the non-ES group (P=0.024). The use of ES increased the incidence of cholangitis (P=0.004, risk ratio, 8.196). The rate of post-ERCP pancreatitis after stent placement was greater in the non-ES group than in the ES group (31.7% vs 9.8%, P=0.014). No significant differences were found in the rate of restenosis and the mortality rate between the two groups.CONCLUSIONS: ES prior to intraductal SEMS placement was associated with an increased incidence of acute cholangitis. | 7 | Spatial patterns in temperature sensitivity of soil respiration in China: Estimation with inverse modeling显示文摘Temperature sensitivity of soil respiration (Q10) is an important parameter in modeling the effects of global warming on ecosystem carbon release. Experimental studies of soil respiration have ubiquitously indicated that Q10 has high spatial heterogeneity. However, most biogeochemical models still use a constant Q10 in projecting future climate change and no spatial pattern of Q10 values at large scales has been derived. In this study, we conducted an inverse modeling analysis to retrieve the spatial pattern of Q10 in China at 8 km spatial resolution by assimilating data of soil organic carbon into a proc-ess-based terrestrial carbon model (CASA model). The results indicate that the optimized Q10 values are spatially heterogeneous and consistent to the values derived from soil respiration observations. The mean Q10 values of different soil types range from 1.09 to 2.38, with the highest value in volcanic soil, and the lowest value in cold brown calcic soil. The spatial pattern of Q10 is related to environmental factors, especially precipitation and top soil organic carbon content. | 9 | Blood flow activation in rat somatosensory cortex under sciatic nerve stimulation revealed by laser speckle imaging显示文摘In many functional neuroimaging research the change of local cerebral blood flow (CBF) induced by sensory stimulation is regarded as an indicator of the change in cortical neuronal activity although a precise and full spatio-temporal description of local CBF response coupled to neural activity has still not been laid out. Using the laser speckle imaging technique a relatively large exposed area in somatosensory cortex of rat was imaged for the observation of the variations of CBF during sciatic nerve stimulation. The results showed that cerebral blood flow activation was spatially localized and discretely distributed in the targeted microvasculature. Individual arteries, veins and capillaries in different diameters were activated with the time going. | 11 | Identification and functional analysis of a novel box C/D snoRNA from Schizosaccharomyces pombe显示文摘By constructing and screening the Schizosac-charomyces pombe nuclear cDNA library, a novel small nu-cleolar RNAs (snoRNA) was identified. The novel snoRNAdisplays structural features typical of C/D box snoRNA fam-ily and possesses a 10-nt-long rRNA antisense element whichis predicted to guide the 2’-O-methylation of the fission yeast 25S rRNA at G940. As expected, the rRNA ribose-methyla-tion site predicted by the novel snoRNA was preciselymapped by a deoxynucleoside triphosphate concentration-dependent primer extension assay. The comparison of func-tional element of guide snoRNAs among eukaryotes reveals that the novel snoRNA is a partial counterpart of the bud-ding yeast snR60 and was termed snR60-II. snR60-II genenested in the intron of a non-coding RNA gene with an un-known function, which is the first example of a yeastsnoRNA encoded in an intron of a non-coding RNA gene.Furthermore, a number of yeast snR60 homologues were also identified from other fungi and fly.
