| 1 | Development of SNP markers using RNA-seq technology and tetraprimer ARMS-PCR in sweetpotato显示文摘The information of single nucleotide polymorphisms(SNPs) is quite unknown in sweetpotato. In this study,two sweetpotato varieties(Xushu 18 and Xu 781) were sequenced by Illumina technology,as well as de novo transcriptome assembly,functional annotation,and in silico discovery of potential SNP molecular markers. Tetra-primer Amplification Refractory Mutation System PCR(ARMS-PCR) is a simple and sufficient method for detecting different alleles in SNP locus. Total 153 sets of ARMS-PCR primers were designed to validate the putative SNPs from sequences. PCR products from 103 sets of primers were different between Xu 781 and Xushu 18 via agarose gel electrophoresis,and the detection rate was 67.32%. We obtained the expected results from 32 sets of primers between the two genotypes. Furthermore,we ascertained the optimal annealing temperature of 32 sets of primers. These SNPs might be used in genotyping,QTL mapping,or marker-assisted trait selection further in sweetpotato. To our knowledge,this work was the first study to develop SNP markers in sweetpotato by using tetra-primer ARMS-PCR technique. | 2 | Parallel selection of distinct Tof5 alleles drove the adaptation of cultivated and wild soybean to high latitudes显示文摘Photoperiod responsiveness is a key factor limiting the geographic distribution of cultivated soybean and its wild ancestor.In particular,the genetic basis of the adaptation in wild soybean remains poorly understood.In this study,by combining whole-genome resequencing and genome-wide association studies we identified a novel locus,Time of Flowering 5(Tof5),which promotes flowering and enhances adaptation to high latitudes in both wild and cultivated soybean.By genomic,genetic and transgenic analyses we showed that Tof5 en-codes a homolog of Arabidopsis thaliana FRUITFULL(FUL).Importantly,further analyses suggested that different alleles of Tof5 have undergone parallel selection.The Tof5H1 allele was strongly selected by humans after the early domestication of cultivated soybean,while Tof5H2 allele was naturally selected in wild soybean,and in each case facilitating adaptation to high latitudes.Moreover,we found that the key flowering repressor E1 suppresses the transcription of Tof5 by binding to its promoter.In turn,Tof5 physically associates with the promoters of two important FLOWERING LOCUS T(FT),FT2a and FT5a,to upregulate their transcription and promote flowering under long photoperiods.Collectively,ourfindings provide insights into how wild soybean adapted to high latitudes through natural selection and indicate that cultivated soybean underwent changes in the same gene but evolved a distinct allele that was artificially selected after domestication. | Lidong Dong Qun Cheng Chao Fang Lingping Kong Hui Yang Zhihong Hou Yongli Li Haiyang Nan Yuhang Zhang Qingshan Chen Chunbao Zhang Kun Kou Tong Su Lingshuang Wang Shichen Li Haiyang Li Xiaoya Lin Yang Tang Xiaohui Zhao Sijia Lu Baohui Liu Fanjiang Kong | 2022 | Molecular Plant2022,15,2: | 5 |
| 3 | Synthesis, DNA-binding and photocleavage studies of ruthenium(Ⅱ) complexes [Ru(btz)_3]^(2+) and [Ru(btz)(dppz)_2]^(2+)显示文摘Two new ruthenium(Ⅱ) complexes, [Ru(btz)3](ClO4)2 (1) and [Ru(btz)(dppz)2](ClO4)2 (2) (btz = 4,4′-bithi-azole, dppz = dipyrido[3,2-a:2′,3′-c]phenazine), have been synthesized and characterized by elemental analysis, 1H NMR, ES-MS and X-ray crystallography. The DNA binding behaviors of two complexes have been studied by spectroscopic and viscosity measurements. The results suggest that complex 1 binds to CT-DNA via an electrostatic mode, while complex 2 via an intercalative mode. Under irradiation at 365 nm, both complexes were found to promote the cleavage of plasmid pBR 322 DNA from supercoiled form Ⅰ to nicked form Ⅱ. | 4 | Ipomoea batatas HKT1 transporter homolog mediates K^+ and Na^+ uptake in Saccharomyces cerevisiae显示文摘Soil salinity causes the negative effects on the growth and yield of crops. In this study,two sweet potato(Ipomoea batatas L.) cultivars,Xushu 28(X-28) and Okinawa 100(O-100),were examined under 50 and 100 mmol L^(–1) Na Cl stress. X-28 cultivar is relatively high salt tolerant than O-100 cultivar. Interestingly,real-time quantitative polymerase chain reaction(RT-q PCR) results indicated that sweet potato high-affinity K^+ transporter 1(IbHKT1) gene expression was highly induced by 50 and 100 mmol L^(–1) Na Cl stress in the stems of X-28 cultivar than in those of O-100 cultivar,but only slightly induced by these stresses in the leaves and fibrous roots in both cultivars. To characterize the function of IbHKT1 transporter,we performed ion-flux analysis in tobacco transient system and yeast complementation. Tobacco transient assay showed that IbHKT1 could uptake sodium(Na^+). Yeast complementation assay showed that IbHKT1 could take up K^+ in 50 mmol L^(–1) K^+ medium without the presence of Na Cl. Moreover,Na^+ uptake significantly increased in yeast overexpressing IbHKT1. These results showed that IbHKT1 transporter could have K^+-Na^+ symport function in yeast. Therefore,the modes of action of IbHKT1 in transgenic yeast could differ from the mode of action of the other HKT1 transporters in class I. | 6 | Transarterial chemoembolization with hepatic arterial infusion chemotherapy plus S-1 for hepatocellular carcinoma显示文摘BACKGROUND Transarterial chemoembolization(TACE)and hepatic arterial infusion chemotherapy(HAIC)have shown promising local benefits for advanced hepatocellular carcinoma(HCC).S-1,a composite preparation of a 5-fluorouracil prodrug,has proven to be a convenient oral chemotherapeutic agent with definite efficacy against advanced HCC.AIM To evaluate the efficacy and safety of TACE followed by HAIC with or without oral S-1 for treating advanced HCC.METHODS In this single-center,open-label,prospective,randomized controlled trial,117 participants with advanced HCC were randomized to receive TACE followed by oxaliplatin-based HAIC either with(TACE/HAIC+S-1,n=56)or without(TACE/HAIC,n=61)oral S-1 between December 2013 and September 2017.Two participants were excluded from final analysis for withdrawing consent.The primary endpoint was progression-free survival(PFS)and secondary endpoints included overall survival(OS),objective response rate,disease control rate and safety.RESULTS In total,115 participants(100 males and 15 females;mean age,57.7 years±11.9)were analyzed.The median PFS and OS were 5.0 mo(0.4–58.6 mo)(95%confidence interval(CI):3.82 to 6.18)vs 4.4 mo(1.1–54.4 mo)(95%CI:2.54 to 6.26;P=0.585)and 8.4 mo(0.4–58.6 mo)(95%CI:6.88 to 9.92)vs 8.3 mo(1.4–54.4 m)(95%CI:5.71 to 10.96;P=0.985)in the TACE/HAIC+S-1 and TACE/HAIC groups,respectively.The objective response rate and disease control rate were 30.9%vs 18.4%and 72.7%vs 56.7%in the TACE/HAIC+S-1 and TACE/HAIC groups,respectively.Grade 3/4 adverse events had a similar frequency in both treatment groups.CONCLUSION No improvements in tumor response rates,PFS or OS were observed with the addition of S-1 to TACE/HAIC in advanced HCC.Both treatment regimens had a similar safety profile. | Jian-Hai Guo Shao-Xing Liu Song Gao Fu-Xin Kou Xin Zhang Di Wu Xiao-Ting Li Hui Chen Xiao-Dong Wang Peng Liu Peng-Jun Zhang Hai-Feng Xu Guang Cao Lin-Zhong Zhu Ren-Jie Yang Xu Zhu | 2020 | World Journal of Gastroenterology2020,26,27: | 3 |

| 12 | Association between interleukin-21 gene rs907715 polymorphism and gastric precancerous lesions in a Chinese population显示文摘BACKGROUND The single nucleotide polymorphisms of interleukin-21(IL-21)gene were confirmed to be related to various diseases,but no studies have examined the possible role of IL-21 single nucleotide polymorphisms(SNPs)(rs907715,rs2221903,and rs12508721)in gastric precancerous lesions.AIM To explore the associations between SNPs of IL-21 gene(rs907715,rs2221903,and rs12508721)and gastric precancerous lesions in a Chinese population.METHODS Three SNPs of IL-21 were genotyped using polymerase chain reaction–ligase detection reaction in 588 cases and 290 healthy controls from May 2013 to December 2016 in northwestern China.Gastric precancerous lesions were confirmed by endoscopic examination and categorized as non-atrophic gastritis,atrophic gastritis,and intestinal metaplasia.Descriptive statistic and logistic regression were used for data analyses.RESULTS IL-21 rs907715 genotype CC and C frequencies were higher in in patients with gastric precancerous lesions than in the controls(OR=1.59,95%CI:1.06-2.38,P=0.013;OR=1.28,95%CI:1.01-2.22,P=0.044,respectively)after adjusting for confounding factors.For SNP rs907715 in intestinal metaplasia patients,significant differences between cases and controls were observed in the frequencies of genotype CC and C(OR=1.92,95%CI:1.24-2.98,P=0.004;OR=1.53,95%CI:1.04-2.24,P=0.028,respectively);for non-atrophic gastritis and atrophic gastritis patients,the CC and C genotypes showed no significant association with risk in all models.No association between either rs2221903 or rs12508721 and gastric precancerous lesions was found in the present study.In the haplotype analysis,the TC haplotype(rs907715 and rs12508721)and TT haplotype(rs2221903 and rs907715)were more frequent in the case group than control group(P<0.05).CONCLUSION Our findings indicate that SNP rs907715 of IL-21 gene is associated with gastric precancerous lesions.The TC haplotype(rs907715 and rs12508721)and TT haplotype(rs2221903 and rs907715)increased the risk of gastric precancerous lesions.If confirmed,these findings will shed light on the etiology of precancerous lesions. | Xiao-Qin Wang Yang Li Paul D Terry Wen-Jing Kou Yue Zhang Zhao-Zhao Hui Xiao-Han Ren Ming-Xu Wang | 2020 | World Journal of Gastrointestinal Oncology2020,12,3: | 2 |
