1Establishment and characterization of a cholangiocarcinoma cell line (RMCCA-1) from a Thai patient显示文摘AIM: To establish and characterize a new cell line derived from peripheral cholangiocarcinoma of a Thai patient.METHODS: The peripheral cholangiocarcinoma specimen surgically obtained from the patient was aseptically processed by washing and mincing before culturing in Ham’s F12 medium containing 10% fetal bovine serum. After 3 mo, when the cell line has become homogeneous and stabilized, several features were investigated, including growth characteristics, immunofluorescence staining for cytokeratins, expression of tumor markers, chromosomal analysis by G-banding and multicolour fluorescence in situ hybridization (mFISH), in vitro migration and invasion characteristics. RESULTS: The RMCCA-1 cell line has been established. These cells proliferated as a monolayer with a population doubling time of 48 h. Immunofluorescence staining showed positive staining for human cytokeratin 7 and 19 verifying the biliary epithelial origin. RMCCA-1 secreted carbohydrate antigen 19-9 (CA19-9), but insignificant levels of carcinoembryonic antigen (CEA) and α-fetoprotein (AFP). Chromosome analysis identified aneuploidy karyotypes with a modal chromosome number of 59. RMCCA-1 exhibited a low level of in vitro invasiveness, but a high degree of motility. The cell line exhibited a significant number of chromosomal aberrations as shown by mFISH and G-banding methods.CONCLUSION: A new cell line derived from peripheral cholangiocarcinoma of a Thai patient has been established. This cell line shows a low level of in vitro invasiveness, but a high degree of motility. 2Hepatitis E virus infection: Epidemiology and treatment implications显示文摘Hepatitis E virus(HEV) infection is now established as an emerging enteric viral hepatitis. Standard treatments in acute and chronic hepatitis E remain to be established. This study undertakes a review of the epidemiology, treatment implication and vaccine prevention from published literature. HEV infection is a worldwide public health problem and can cause acute and chronic hepatitis E. HEV genotypes 1 and 2 are primarily found in developing countries due to waterborne transmission, while the zoonotic potential of genotypes 3 and 4 affects mostly industrialized countries. An awareness of HEV transmission through blood donation, especially in the immunocompromised and solid organ transplant patients, merits an effective anti-viral therapy. There are currently no clear indications for the treatment of acute hepatitis E. Despite concerns for side effects, ribavirin monotherapy or in combination with pegylatedinterferon alpha for at least 3 mo appeared to show significant efficacy in the treatment of chronic hepatitis E. However, there are no available treatment options for specific patient population groups, such as women who are pregnant. Vaccination and screening of HEV in blood donors are currently a global priority in managing infection. 3Advancement and new perspectives of using formulated reactive amine blends for post-combustion carbon dioxide (CO_(2)) capture technologies显示文摘Chemical absorption using amine-based solvents have proven to be the most studied,as well as the most reliable and efficient technology for capturing carbon dioxide(CO_(2))from exhaust gas streams and synthesis gas in all combustion and industrial processes.The application of single amine-based solvents especially the very reactive monoethanolamine(MEA)is associated with a parasitic energy demand for solvent regeneration.Since regeneration energy accounts for up to threeequarters of the plant operating cost,efforts in its reduction have prompted the idea of using blended amine solvents.This review paper highlights the success achieved in blending amine solvents and the recent and future technologies aimed at increasing the overall volumetric mass transfer coefficient,absorption rate,cyclic capacity and greatly minimizing both degradation and the energy for solvent regeneration.The importance of amine biodegradability(BOD)and low ecotoxicity as well as low amine volatility is also highlighted.Costs and energy penalty indices that influences the capital and operating costs of CO_(2) capture process was also highlighted.A new experimental method for simultaneously estimating amine cost,degradation rate,regeneration energy and reclaiming energy is also proposed in this review paper.Chikezie Nwaoha Teeradet Supap Raphael Idem Chintana Saiwan Paitoon Tontiwachwuthikul Mohammed JAL-Marri Abdelbaki Benamor 2017Petroleum2017,3,1:4
4Effects of hexahydrocurcumin in combination with 5-fluorouracil on dimethylhydrazine-induced colon cancer in rats显示文摘AIM:To investigate the effects of hexahydrocurcumin(HHC),and its combination with 5-fluorouracil(5-FU) on dimethylhydrazine(DMH)-induced colon cancer in rats.METHODS:Male Wistar rats weighing 100-120 g were used as subject models.Aberrant crypt foci(ACF),early preneoplastic lesions of colon cancer,were induced by subcutaneous injection of DHM(40 mg/kg) twice a week for two weeks.After the first DMH injection,rats were treated daily with vehicle(n = 12),curcumin(CUR)(50 mg/kg)(n = 12),HHC(50 mg/kg) orally(n = 12),and treated weekly with an intraperitoneal injection of 5-FU(50 mg/kg)(n = 12),or a combination of 5-FU plus CUR(n = 12) and HHC(n = 12) at the same dosage(s) for 16 wk.The total number of ACF and large ACF were assessed.Cyclooxygenase(COX)-1 and COX-2 expression were detected by immunohistochemistry in colon tissues.The quantitative data of both COX-1 and COX-2 expression were presented as the percentage of number of positive-stained cells to the total number of cells counted.Apoptotic cells in colon tissues were also visualized using the dUTP-biotin nick end labeling method.Apoptotic index(AI) was determined as the percentage of labeled nuclei with respect to the total number of nuclei counted.RESULTS:The total number of ACF was highest in the DMH-vehicle group(1558.20 ± 17.37),however,the number of ACF was significantly reduced by all treatments,5-FU(1231.20 ± 25.62 vs 1558.20 ± 17.37,P < 0.001),CUR(1284.20 ± 25.47 vs 1558.20 ± 17.37,P < 0.001),HHC(1086.80 ± 53.47 vs 1558.20 ± 17.37,P < 0.001),DMH-5-FU + CUR(880.20 ± 13.67 vs 1558.20 ± 17.37,P < 0.001) and DMH-5-FU + HHC(665.80 ± 16.64 vs 1558.20 ± 17.37,P < 0.001).Interestingly,the total number of ACF in the combined treatment groups,the DMH-5-FU + CUR group(880.20 ± 13.67 vs 1231.20 ± 25.62,P < 0.001;880.20 ± 13.67 vs 1284.20 ± 25.47,P < 0.001) and the DMH-5-FU + HHC group(665.80 ± 16.64 vs 1231.20 ± 25.62,P < 0.001;665.80 ± 16.64 vs 1086.80 ± 53.47,P < 0.001) were significantly reduced as compared to 5-FU or each treatment alone.Large ACF were also significantly reduced in all treatment groups,5-FU(111.00 ± 7.88 vs 262.20 ± 10.18,P < 0.001),CUR(178.00 ± 7.33 vs 262.20 ± 10.18,P < 0.001),HHC(186.60 ± 21.51 vs 262.20 ± 10.18,P < 0.001),DMH-5-FU + CUR(122.00 ± 5.94 vs 262.20 ± 10.18,P < 0.001) and DMH-5-FU + HHC(119.00 ± 17.92 vs 262.20 ± 10.18,P < 0.001) when compared to the vehicle group.Furthermore,in the DMH-5-FU + CUR and DMH-5-FU + HHC groups the formation of large ACF was significantly reduced when compared to CUR(122.00 ± 5.94 vs 178.00 ± 7.33,P < 0.005) or HHC treatment alone(119.00 ± 17.92 vs 186.60 ± 21.51,P < 0.001),however,this reduction was not statistically different to 5-FU monotherapy(122.00 ± 5.94 vs 111.00 ± 7.88,P = 0.217;119.00 ± 17.92 vs 111.00 ± 7.88,P = 0.619,respectively).The levels of COX-1 protein after all treatments were not different from normal rats.A marked increase in the expression of COX-2 protein was observed in the DMH-vehicle group.Over-expression of COX-2 was not significantly decreased by 5-FU treatment alone(95.79 ± 1.60 vs 100 ± 0.00,P = 0.198).However,over-expression of COX-2 was significantly suppressed by CUR(77.52 ± 1.68 vs 100 ± 0.00,P < 0.001),HHC(71.33 ± 3.01 vs 100 ± 0.00,P < 0.001),5-FU + CUR(76.25 ± 3.32 vs 100 ± 0.00,P < 0.001) and 5-FU + HHC(68.48 ± 2.24 vs 100 ± 0.00,P < 0.001) in the treated groups compared to the vehicle group.Moreover,CUR(77.52 ± 1.68 vs 95.79 ± 1.60,P < 0.001),HHC(71.33 ± 3.01 vs 95.79 ± 1.60,P < 0.001),5-FU + CUR treatments(76.25 ± 3.32 vs 95.79 ± 1.60,P < 0.001) and 5-FU + HHC(68.48 ± 2.24 vs 95.79 ± 1.60,P < 0.001) markedly decreased COX-2 protein expression more than 5-FU alone.Furthermore,the AI in all treated groups,5-FU(38.86 ± 4.73 vs 23.56 ± 2.12,P = 0.038),CUR(41.78 ± 6.92 vs 23.56 ± 2.12,P < 0.001),HHC(41.06 ± 4.81 vs 23.56 ± 2.12,P < 0.001),5-FU + CUR(49.05 ± 6.75 vs 23.56 ± 2.12,P < 0.001) and 5-FU + HHC(53.69 ± 8.59 vs 23.56 ± 2.12,P < 0.001) significantly increased when compared to the DMH-vehicle group.However,the AI in the combination treatments,5-FU + CUR(49.05 ± 6.75 vs 41.78 ± 6.92,P = 0.192;49.05 ± 6.75 vs 38.86 ± 4.73,P = 0.771) and 5-FU + HHC(53.69 ± 8.59 vs 41.06 ± 4.81,P = 0.379;53.69 ± 8.59 vs 38.86 ± 4.73,P = 0.245) did not reach significant levels as compared with each treatment alone and 5-FU monotherapy,respectively.CONCLUSION:The combined effects of HHC with 5-FU exhibit a synergistic inhibition by decreasing ACF formation mediated by down-regulation of COX-2 expression.Khanitta Srimuangwong Chainarong Tocharus Jiraporn Tocharus Apichart Suksamrarn Pornphorm Yoysungnoen Chintana 2012World Journal of Gastroenterology2012,18,47:2

15Carbon dioxide adsorption/desorption performance of single-and blended-amines-impregnated MCM-41 mesoporous silica in post-combustion carbon capture显示文摘High-surface-area,hexagonal-structured mesoporous silica,MCM-41,was synthesized and wet impregnated with three different amines of 2-(ethylamino)ethanol(EAE),ethylenediamine(EDA),and tetraethylenepentamine(TEPA)for use as solid adsorbents in post-combustion CO_(2) capture application.The CO_(2) adsorption test was performed at 25℃and atmospheric pressure using 15/85 vol%of CO_(2)/N2 at a 20-mL/minute flow rate.Desorption was carried out at 100℃under 20 mL/minute of N2 flow.The results show that the capacity and rate of CO_(2) adsorption obtained from all the amine-modified adsorbents were significantly increased with increasing amine loading due to carbamate formation.Desorption efficiency and heat duty for regeneration were also affected by the amount of amine loading.The more stable the carbamate produced,the higher the energy was required.They exhibited the highest adsorption-desorption performance at 60 wt%amines used for impregnation.Blended EAE/TEPA at different weight ratios at a total concentration at 60 wt%amines was impregnated on MCM-41 adsorbent.Sorbent impregnated with 50%/10%of EAE/TEPA showed the best performance of 4.25 mmolCO_(2)/g at a high adsorption rate,a low heat duty of 12 kJ/mmolCO_(2) and with 9.4%reduction of regeneration efficiency after five repeated adsorption-desorption cycles.Pailin Muchan Chintana Saiwan Manit Nithitanakul 2022Clean Energy2022,6,3:1

17Hexahydrocurcumin enhances inhibitory effect of 5-fluorouracil on HT-29 human colon cancer cells显示文摘AIM:To investigate the ability of hexahydrocurcumin(HHC) to enhance 5-fluorouracil(5-FU) in inhibiting the growth of HT-29 cells by focusing on cyclooxygenase(COX)-2 expression.METHODS:Antiproliferative effects of HHC and 5-FU,alone and in combination,on growth of HT-29 human colon cancer cells were assessed using 5-diphenyltetrazolium bromide(MTT) reduction assay.In combination treatment,low doses of 5-FU were used combined with various concentrations of HHC to minimize the toxicity and side effects of 5-FU.The therapeutic effects of these drugs on down-regulation of COX-2 mRNA and protein expression were examined using semi-quantitative reverse transcription-polymerase chain reaction(RT-PCR) and Western blotting analysis.RESULTS:MTT reduction assay indicated that HHC alone markedly decreased the viability of HT-29 human colon cancer cells compared to control.Semi-quantitative RT-PCR analysis indicated that HHC is a selective COX-2 inhibitor.This finding was supported by the observation that HHC significantly down-regulates COX-2 mRNA expression compared to the control(control:100.05% ± 0.03% vs HHC:61.01% ± 0.35%,P < 0.05) but does not alter COX-1 mRNA.In combined treatment,addition of HHC to a low dose of 5-FU exerts a synergistic effect against the growth of HT-29 cells by markedly reducing cell viability to a greater degree than monotherapy.Semi-quantitative RT-PCR indicated that 5-FU at the concentration of 5 μmol/L in combination with HHC at the concentration of 25 μmol/L significantly down-regulates COX-2 mRNA expression when compared with values in cells treated with 5-FU or HHC alone(HHC + 5-FU:31.93% ± 5.69%,5-FU:100.66% ± 4.52% vs HHC:61.01% ± 0.35%,P < 0.05).CONCLUSION:HHC together with 5-FU exerts a synergistic effect and may prove chemotherapeutically useful in treating human colon cancer.Khanitta Srimuangwong Chainarong Tocharus Pornphrom Yoysungnoen Chintana Apichart Suksamrarn Jiraporn Tocharus 2012World Journal of Gastroenterology2012,18,19:1

19Cytokine levels in patients with chikungunya virus infection显示文摘Objective: To investigate cytokine profile in patients with chikungunya virus (CHIKV) infection. Methods: Twenty eight pairs of serum samples collected from CHIKV infected patients during the outbreak of chikungunya fever in South Thailand in 2008 were obtained. A multiple cytokine assay for detection of 17 cytokines was performed. Results: In the acute stage of CHIKV infection, the patients had significantly higher levels of interleukin-6, granulocyte colony-stimulating factor, granulocyte-macrophage colony-stimulating factor, monocyte chemotactic protein 1 and tumor necrosis factor alpha than the control ( P<0.001, P=0.023, P=0.015, P <0.001 and P=0.024, respectively). When the disease developed to the recovery stage, the patients had significantly lower levels of interleukin-6, granulocyte-macrophage colony-stimulating factor, monocyte chemotactic protein 1 and macrophage inflammatory protein beta than in the acute stage ( P<0.001). 20Investigation of adsorption/desorption performance by aminopropyltriethoxysilane grafted onto different mesoporous silica for post-combustion CO_(2) capture显示文摘Mesoporous silicas with hexagonal structure(MCM-41 and SBA-15)and cubical interconnected pore structure(KIT-6)were synthesized and modified with aminopropyltriethoxysilane(APTES)for using as adsorbents in carbon-dioxide(CO_(2))-adsorption application.The CO_(2)-adsorption experiment was carried out at room temperature and atmospheric pressure using 15%CO_(2) with a flow rate of 20 mL/min and the desorption experiment was carried out at 100℃ under N_(2) balance with a flow rate of 20 mL/min.The adsorption capacity and adsorption rate of all modified mesoporous silicas were enhanced due to the presence of primary amine in the structure,which was able to form a fast chemical reaction with CO_(2).All adsorbents showed good adsorption performance stability after using over five adsorption/desorption cycles.Due to the effect of the adsorbents’porous structure on the adsorption/desorption process,an adsorbent with sufficient pore-size diameter and pore volume together with interconnected pore,KIT-6,represents a promising adsorbent that gave the optimum adsorption/desorption performance among others.It showed reasonable adsorption capacity with a high rate of adsorption.In addition,it could also be regenerated with 99.72% efficiency using 12.07 kJ/mmol CO_(2) of heat duty for regeneration.Pailin Muchan Chintana Saiwan Manit Nithitanakul 2020Clean Energy2020,4,2:0
