| 2 | 5-Hydroxymethylome in Circulating Cell-free DNA as A Potential Biomarker for Non-small-cell Lung Cancer显示文摘Non-small-cell lung cancer(NSCLC), the most common type of lung cancer accounting for 85% of the cases, is often diagnosed at advanced stages owing to the lack of efficient early diagnostic tools. 5-Hydroxymethylcytosine(5 hmC) signatures in circulating cell-free DNA(cfDNA) that carries the cancer-specific epigenetic patterns may represent the valuable biomarkers for discriminating tumor and healthy individuals, and thus could be potentially useful for NSCLC diagnosis. Here,we employed a sensitive and reliable method to map genome-wide 5 hmC in the cfDNA of Chinese NSCLC patients and detected a significant 5 hmC gain in both the gene bodies and promoter regions in the blood samples from tumor patients compared with healthy controls. Specifically, we identified six potential biomarkers from 66 patients and 67 healthy controls(mean decrease accuracy>3.2, P < 3.68E-19) using machine-learning-based tumor classifiers with high accuracy. | 3 | Effects of Rice-Fish Co-culture on Oxygen Consumption in Intensive Aquaculture Pond显示文摘Rice-fish co-culture has gained increasing attention to remediate the negative environmental impacts induced by intensive aquaculture. However, the effect of rice-fish co-culture on oxygen depletion has rarely been investigated. We constructed a rice-fish co-culture system in yellow catfish(Pelteobagrus fulvidraco) and freshwater shrimp(Macrobrachium nipponense) ponds using a new high-stalk rice variety, and conducted a field experiment to investigate the effect of rice-fish co-culture on water parameters and oxygen consumption. The results showed that rice-fish co-culture reduced the nutrients(total nitrogen, ammonia-N, total phosphorous and potassium) and the dissolved oxygen content in fish and shrimp ponds. However, they showed similar seasonal change of dissolved oxygen in the water of fish and shrimp ponds. Rice-fish co-culture reduced the total amount of oxygen consumption and optimized the oxygen consumption structure in pond. The respiration rates in water and sediment were significantly reduced by 66.1% and 31.7% in the catfish pond, and 64.4% and 38.7% in the shrimp pond, respectively, by additional rice cultivation. Rice-fish co-culture decreased the proportions of respiration in sediment and water, and increased the proportion of fish respiration. | 4 | AEROELASTIC PROPERTIES OF SANDWICH BEAM WITH PYRAMIDAL LATTICE CORE CONSIDERING GEOMETRIC NONLINEARITY IN THE SUPERSONIC AIRFLOW显示文摘The equation of motion of sandwich beam with pyramidal lattice core in the supersonic flow considering geometric nonlinearity is formulated using Hamilton's principle. The piston theory is used to evaluate aerodynamic pressure. The structural aeroelastic properties are analyzed using frequency- and time-domain methods, and some interesting phenomena are observed. It is noted that the flutter of sandwich beam occurs under the coupling effect of low order modes. The critical flutter aerodynamic pressure of the sandwich beam is higher than that of the isotropic beam with the same weight, length and width. | 6 | Electrochemical corrosion behavior of the cobalt modified aluminide coating in 3.5 wt% NaCl solutions显示文摘In this paper, we achieved the Co modified aluminide coating on nickel base superalloys by pack cementation process. The corrosion currents(i_(corr)) of coated and uncoated substrate after 1 h of immersion tested by potentiodynamic polarization in 3.5 wt% NaCl solutions are 9.73 b × 10^(-7) A/cm^2 and 1.31 × 10^(-6) A/cm^2, respectively. With the immersion time extension, the icorrof the coating decreases and the values of |Z| by EIS increase, indicating that the electrochemical reaction rate of the coating becomes slower. | 7 | USP51/PD-L1/ITGB1-deployed juxtacrine interaction plays a cell-intrinsic role in promoting chemoresistant phenotypes in non-small cell lung cancer显示文摘Background:Programmed death ligand 1(PD-L1)has been demonstrated to facilitate tumor progression and therapeutic resistance in an immuneindependent manner.Nevertheless,the function and underlying signaling network(s)of cancer cell-intrinsic PD-L1 action remain largely unknown.Herein,we sought to better understand how ubiquitin-specific peptidase 51(USP51)/PD-L1/integrin beta-1(ITGB1)signaling performs a cell-intrinsic role in mediating chemotherapeutic resistance in non-small cell lung cancer(NSCLC).Methods:Western blotting and flow cytometry were employed for PD-L1 detection in NSCLC cell lines.Coimmunoprecipitation and pulldown analyses,protein deubiquitination assay,tissue microarray,bioinformatic analysis and molecular biology methods were then used to determine the significance of PD-L1 in NSCLC chemoresistance and associated signaling pathways in several different cell lines,mouse models and patient tissue samples.Ubiquitin-7-amido-4-methylcoumarin(Ub-AMC)-based deubiquitinase activity,cellular thermal shift and surface plasmon resonance(SPR)analyses were performed to investigate the activity of USP51 inhibitors.Results:We provided evidence that cancer cell-intrinsic PD-L1 conferred the development of chemoresistance by directly binding to its membrane-bound receptor ITGB1 in NSCLC.At the molecular level,PD-L1/ITGB1 interaction subsequently activated the nuclear factor-kappa B(NF-κB)axis to elicit poor response to chemotherapy.We further determined USP51 as a bona fide deubiquitinase that targeted the deubiquitination and stabilization of the PD-L1 protein in chemoresistant NSCLC cells.Clinically,we found a significant direct relationship between the USP51,PD-L1 and ITGB1 contents in NSCLC patients with chemoresistant potency.The elevated USP51,PD-L1 and ITGB1 levels were strongly associated with worse patient prognosis.Of note,we identified that a flavonoid compound dihydromyricetin(DHM)acted as a potential USP51 inhibitor and rendered NSCLC cells more sensitive to chemotherapy by targeting USP51-dependent PD-L1 ubiquitination and degradation in vitro and in vivo.Conclusions:Together,our results demonstrated that the USP51/PD-L1/ITGB1 network potentially contributes to the malignant progression and therapeutic resistance in NSCLC.This knowledge is beneficial to the future design of advanced cancer therapy. | Jianjun Li Xuechun Xiao Yang Ou Lixia Cao Min Guo Chunchun Qi Zhaoyang Wang Yuxin Liu Qiuying Shuai Hang Wang Peiqing Sun Yi Shi Guang Yang Shuang Yang | 2023 | Cancer Communications2023,43,7: | 0 |
| 8 | Differential Transcriptomic Landscapes of SARS-CoV-2 Variants in Multiple Organs from Infected Rhesus Macaques显示文摘Severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)caused the persistent coronavirus disease 2019(COVID-19)pandemic,which has resulted in millions of deaths worldwide and brought an enormous public health and global economic burden.The recurring global wave of infections has been exacerbated by growing variants of SARS-CoV-2.In this study,the virological characteristics of the original SARS-CoV-2 strain and its variants of concern(VOCs;including Alpha,Beta,and Delta)in vitro,as well as differential transcriptomic landscapes in multiple organs(lung,right ventricle,blood,cerebral cortex,and cerebellum)from the infected rhesus macaques,were elucidated.The original strain of SARS-CoV-2 caused a stronger innate immune response in host cells,and its VOCs markedly increased the levels of subgenomic RNAs,such as N,Orf9b,Orf6,and Orf7ab,which are known as the innate immune antagonists and the inhibitors of antiviral factors.Intriguingly,the original SARS-CoV-2 strain and Alpha variant induced larger alteration of RNA abundance in tissues of rhesus monkeys than Beta and Delta variants did.Moreover,a hyperinflammatory state and active immune response were shown in the right ventricles of rhesus monkeys by the up-regulation of inflammation-and immune-related RNAs.Furthermore,peripheral blood may mediate signaling transmission among tissues to coordinate the molecular changes in the infected individuals.Collectively,these data provide insights into the pathogenesis of COVID-19 at the early stage of infection by the original SARS-CoV-2 strain and its VOCs. | Tingfu Du Chunchun Gao Shuaiyao Lu Qianlan Liu Yun Yang Wenhai Yu Wenjie Li Yong Qiao Sun Cong Tang Junbin Wang Jiahong Gao Yong Zhang Fangyu Luo Ying Yang Yun-Gui Yang Xiaozhong Peng | 2023 | Genomics, Proteomics & Bioinformatics2023,21,5: | 0 |
