| 1 | Microstructural Evolution and Mechanical Properties of Inconel 625 Alloy during Pulsed Plasma Arc Deposition Process显示文摘Pulsed plasma arc deposition(PPAD),which combines pulsed plasma cladding with rapid prototyping,is a promising technology for manufacturing near net shape components due to its superiority in cost and convenience of processing.In the present research,PPAD was successfully used to fabricate the Ni-based superalloy Inconel 625 components.The microstructures and mechanical properties of deposits were investigated by scanning electron microscopy(SEM),optical microscopy(OM),transmission electron microscopy(TEM) with energy dispersive spectrometer(EDS),microhardness and tensile testers.It was found that the as-deposited structure exhibited homogenous columnar dendrite structure,which grew epitaxially along the deposition direction.Moreover,some intermetallic phases such as Laves phase,minor MC(NbC, TiC) carbides and needle-likeδ-Ni_3Nb were observed in y-Ni matrix.Precipitation mechanism and distribution characteristics of these intermetallic phases in the as-deposited 625 alloy sample were analyzed.In order to evaluate the mechanical properties of the deposits,microhardness was measured at various location (including transverse plane and longitudinal plane).The results revealed hardness was in the range of 260—285 HV_(0.2).In particular,microhardness at the interface region between two adjacent deposited layers was slightly higher than that at other regions due to highly refined structure and the disperse distribution of Laves particles.Finally,the influence of precipitation phases and fabrication strategies on the tensile properties of the as-deposited samples was investigated.The failure modes of the tensile specimens were analyzed with fractography. | Fujia Xu Yaohui Lv Yuxin Liu Fengyuan Shu Peng He Binshi Xu | 2013 | Journal of Materials Science & Technology2013,29,5: | 33 |
| 2 | Human ESC-derived expandable hepatic organoids enable therapeutic liver repopulation and pathophysiological modeling of alcoholic liver injury显示文摘We report the generation of human ESC-derived,expandable hepatic organoids(hEHOs)using our newly established method with wholly defined(serum-free,feeder free)media.The hEHOs stably maintain phenotypic features of bipotential liver stem/progenitor cells that can differentiate into functional hepatocytes or cholangiocytes.The hEHOs can expand for 20 passages enabling large scale expansion to cell numbers requisite for industry or clinical programs.The cells from hEHOs display remarkable repopulation capacity in injured livers of FRG mice following transplantation,and they differentiate in vivo into mature hepatocytes.If implanted into the epididymal fat pads of immune-deficient mice,they do not generate non-hepatic lineages and have no tendency to form teratomas.We further develop a derivative model by incorporating human fetal liver mesenchymal cells(hFLMCs)into the hEHOs,referred to as hFLMC/hEHO,which can model alcoholic liver disease-associated pathophysiologic changes,including oxidative stress generation,steatosis,inflammatory mediators release and fibrosis,under ethanol treatment.Our work demonstrates that the hEHOs have considerable potential to be a novel,ex vivo pathophysiological model for studying alcoholic liver disease as well as a promising cellular source for treating human liver diseases. | Shuyong Wang Xuan Wang Zuolong Tan Yuxin Su Juan Liu Mingyang Chang Fang Yan Jie Chen Tao Chen Chuanjiang Li Jie Hu Yunfang Wang | 2019 | Cell Research2019,29,12: | 20 |

| 4 | Melatonin alleviates intervertebral disc degeneration by disrupting the IL-1β/NF-κB-NLRP3 inflammasome positive feedback loop显示文摘The inflammatory response is induced by the overexpression of inflammatory cytokines, mainly interleukin(IL)-1β, and is one of the main causes of intervertebral disc degeneration(IVDD). NLR pyrin domain containing 3(NLRP3) inflammasome activation is an important source of IL-1β. As an anti-inflammatory neuroendocrine hormone, melatonin plays various roles in different pathophysiological conditions. However, its roles in IVDD are still not well understood and require more examination. First, we demonstrated that melatonin delayed the progression of IVDD and relieved IVDD-related low back pain in a rat needle puncture IVDD model;moreover, NLRP3 inflammasome activation(NLRP3, p20, and IL-1β levels) was significantly upregulated in severely degenerated human discs and a rat IVDD model. Subsequently, an IL-1β/NF-κB-NLRP3 inflammasome activation positive feedback loop was found in nucleus pulposus(NP) cells that were treated with IL-1β. In these cells, expression of NLRP3 and p20 was significantly increased, NF-κB signaling was involved in this regulation, and mitochondrial reactive oxygen species(mt ROS)production increased. Furthermore, we found that melatonin disrupted the IL-1β/NF-κB-NLRP3 inflammasome activation positive feedback loop in vitro and in vivo. Melatonin treatment decreased NLRP3, p20, and IL-1β levels by inhibiting NF-κB signaling and downregulating mt ROS production. Finally, we showed that melatonin mediated the disruption of the positive feedback loop of IL-1β in vivo. In this study, we showed for the first time that IL-1β promotes its own expression by upregulating NLRP3 inflammasome activation. | 6 | Variation characteristics of heavy metals and nutrients in the core sediments of Taihu Lake and their pollution history显示文摘The sedimentary environment change, trophic evolution and heavy metals pollution history of the northern Taihu Lake in the last 100 years are studied according to the sedimentary geochemical proxies of the core sediments, such as grain size, nutrients, heavy metals, diatom, etc. The nutrients in the sediments depended mainly on the lake internal circulation and the heavy metals were from natural geogenic sources before the 1920s, which were not influenced by human activities generally, and grain size was one of the key factors influencing heavy metals content in the sediments.The alternation of manner and strength of human activities in Taihu Lake catchment before and after the 1920s made the lake sediments coarser, and hence heavy metals and TP content decreased contrasted with that before the 1920s. TP content in sediments and water increased from the 1950s to late 1970s due to anthropogenic pollutants discharge, and the lake belonging to mesotrophic state.TN and TOC content and C/N ratio increased due to the increasing external pollutants into Taihu Lake by human activities, TP content also increased obviously in water and sediments, and the diatom association was dominated by eutrophic species since the late 1970s, indicating the eutrophication state of Taihu Lake in this period. Meanwhile the increasing in heavy metals content, such as Cu, Mn,Ni, Pb and Zn, and their proportion of valid fractions in the sediments indicates that they all result from human pollutants since the late 1970s. | 8 | Holocene aeolian stratigraphic sequences in the eastern portion of the desert belt(sand seas and sandy lands) in northern China and their palaeoenvironmental implications显示文摘This paper presents the environmental history and its responses to palaeoclimatic changes since the start of the Holocene in the eastern portion of the desert belt(sand seas and sandy lands) in northern China by comparing the aeolian sandpalaeosol sequences and their palaeoclimatic proxies. The optically stimulated luminescence(OSL) ages of the aeolian sandpalaeosol sedimentary sequences and a series of palaeoenvironmental proxies show that:(1) The large-scale dune landscape currently in the Kubuqi Sand Sea was formed during the Holocene in general;and the palaeosol was generally developed during the period of 4–2 ka, indicating conditions favorable for vegetation growth, soil development, and organic carbon accumulation due to increased precipitation or effective moisture and weakened aeolian activities;the large-scale expansion of dunes in the recent 2 ka is closely linked to human activities. The variable discharge of the Yellow River with diversions for irrigation may have resulted in a more consistent supply of aeolian particles for dune field expansion.(2) The dune landscape of the Hunshandake Sandy Land was likely formed around 12 ka, and before this, the western part of the Hunshandake Sandy Land would have been covered by a single large lake;it was obviously wetter than today in the sandy land during the period of 9.6–3 ka and the palaeosols were developed at the same time. But the aeolian activities have not been completely dormant in this long-lasting wetter epoch;because the Holocene wetter period was likely time-transgressive across the region.(3) The palaeosol of the Hulunbuir Sandy Land began to develop as early as 14.5 ka, probably continuing until the last 2 ka. The palaeosol development of various dune fields in the eastern portion of the desert belt(sand seas and sandy lands) in northern China is spatially heterogeneous, and even the palaeosol development time in different locations within each sandy land is inconsistent. During the middle Holocene(especially the 7.5–3.5 ka), all the sandy lands were stabilized in general and the intensity of aeolian activities was significantly weakened. The number of palaeoenvironmental records in the eastern portion of the desert belt(sand seas and sandy lands) in northern China has increased rapidly in the past decade, but the amount of published data still does not match the vast extent of the dune fields. | 9 | miR-21 promotes the differentiation of hair folliclederived neural crest stem cells into Schwann cells显示文摘Hair follicle-derived neural crest stem cells can be induced to differentiate into Schwann cells in vivo and in vitro. However, the underlying regulatory mechanism during cell differentiation remains poorly understood. This study isolated neural crest stem cells from human hair follicles and induced them to differentiate into Schwann cells. Quantitative RT-PCR showed that microRNA(miR)-21 expression was gradually increased during the differentiation of neural crest stem cells into Schwann cells. After transfection with the miR-21 agonist(agomir-21), the differentiation capacity of neural crest stem cells was enhanced. By contrast, after transfection with the miR-21 antagonist(antagomir-21), the differentiation capacity was attenuated. Further study results showed that SOX-2 was an effective target of miR-21. Without compromising SOX2 mRNA expression, miR-21 can down-regulate SOX protein expression by binding to the 3′-UTR of miR-21 mRNA. Knocking out the SOX2 gene from the neural crest stem cells significantly reversed the antagomir-21 inhibition of neural crest stem cells differentiating into Schwann cells. | 10 | The enrichment characteristics of mercury in the sediments of Dongjiu and Xijiu, Taihu Lake catchment, in the past century显示文摘The concentration and the enrichment factors of mercury (Hg) in the sediment cores of Dongjiu and Xijiu, Taihu Lake catchment, were studied. The accumulation fluxes, anthropogenic input concentration and anthropogenic accumulation fluxes of Hg in recent 100 years were also analyzed based on the 210Pb dating. The results indicate that the increasing concentrations of Hg in the sediments are influ-enced by natural factors and anthropogenic input simultaneously. Generally, about 2/3 of the Hg in the sediment was from anthropogenic sources. In the early 20th century, the anthropogenic input was owing to the urban development and fossil fuel consumptions surrounding the Taihu Lake and the worldwide atmospheric deposition of Hg since the industrial revolution. The concentration and an-thropogenic fluxes of Hg increased with the industrial development in the catchment since the 1930s. | 12 | T cells promote the regeneration of neural precursor cells in the hippocampus of Alzheimer's disease mice显示文摘Alzheimer's disease is closely associated with disorders of neurogenesis in the brain, and growing evidence supports the involvement of immunological mechanisms in the development of the disease. However, at present, the role of T cells in neuronal regeneration in the brain is unknown. We injected amyloid-beta 1–42 peptide into the hippocampus of six BALB/c wild-type mice and six BALB/c-nude mice with T-cell immunodeficiency to establish an animal model of Alzheimer's disease. A further six mice of each genotype were injected with same volume of normal saline. Immunohistochemistry revealed that the number of regenerated neural progenitor cells in the hippocampus of BALB/c wild-type mice was significantly higher than that in BALB/c-nude mice. Quantitative fluorescence PCR assay showed that the expression levels of peripheral T cell-associated cytokines(interleukin-2, interferon-γ) and hippocampal microglia-related cytokines(interleukin-1β, tumor necrosis factor-α) correlated with the number of regenerated neural progenitor cells in the hippocampus. These results indicate that T cells promote hippocampal neurogenesis in Alzheimer's disease and T-cell immunodeficiency restricts neuronal regeneration in the hippocampus. The mechanism underlying the promotion of neuronal regeneration by T cells is mediated by an increased expression of peripheral T cells and central microglial cytokines in Alzheimer's disease mice. | 13 | Elevated Neurosteroids in the Lateral Thalamus Relieve Neuropathic Pain in Rats with Spared Nerve Injury显示文摘Neurosteroids are synthesized in the nervous system from cholesterol or steroidal precursors imported from peripheral sources. These compounds are important allosteric modulators of c-aminobutyric acid A receptors(GABA_ARs), which play a vital role in pain modulation in the lateral thalamus, a main gate where somatosensory information enters the cerebral cortex. Using high-performance liquid chromatography/tandem mass spectrometry,we found increased levels of neurosteroids(pregnenolone,progesterone, deoxycorticosterone, allopregnanolone, and tetrahydrodeoxycorticosterone) in the chronic stage of neuropathic pain(28 days after spared nerve injury) in rats.The expression of the translocator protein TSPO, the upstream steroidogenesis rate-limiting enzyme, increased at the same time. In vivo stereotaxic microinjection of neurosteroids or the TSPO activator AC-5216 into the lateral thalamus(AP-3.0 mm, ML ±3.0 mm, DV 6.0 mm)alleviated the mechanical allodynia in neuropathic pain,while the TSPO inhibitor PK 11195 exacerbated it. The analgesic effects of AC-5216 and neurosteroids were significantly attenuated by the GABA_AR antagonist bicuculline. The crystal structure of(R)-N-(2-methyl-1-(methylthio)propan-2-yl)-2-(4-nitrophenyl)-4,5-dihydrothiazole-4-carboxamide(13 d) was determined to show R configuration. The bioasssy results indicated that most title compounds displayed good and broad spectrum antifungal activities against several phytopathogenic fungi. The structure activity relationships were discussed. Based on the antifungal activity of title compounds against Phytophthora capsici, a CoMSIA calculation was performed to establish a 3 D-QSAR model, which revealed that electrostatic and hydrophobic fields were the two most significant factors for antifungal activity. Performance of dispersion simulations using the computational fluid dynamics(CFD)-based tool FLACS can get an uneven and irregular cloud. For the performance of gas explosion study with FLACS, the equivalent stoichiometric fuel-air cloud concept is widely applied to get a representative distribution of explosion loads. The Q9 cloud model that is employed in FLACS is an equivalent fuel-air cloud representation, in which the laminar burning velocity with first order SL and volume expansion ratio are taken into consideration. However,during an explosion in congested areas, the main part of the combustion involves turbulent flame propagation. Hence, to give a more reasonable equivalent fuel-air size, the turbulent burning velocity must be taken into consideration. | 17 | Selectivity regulation of CO2 electroreduction through contact interface engineering on superwetting Cu nanoarray electrodes显示文摘Electrocatalytic CO2 reduction is a promising way to mitigate the urgent energy and environmental issues, but how to increase the selectivity for desired product among multiple competi ng reaction pathways remains a bottle neck. Here, we demonstrate that engineering the gas-liquid-solid contact interface on the electrode surface could tailor the selectivity of CO2 reducti on and meanwhile suppress H2 production through regulated reacti on kin etics. Specifically, polytetrafl uoroethyle ne (PTFE) was utilized to modify a Cu nano array electrode as an example, which is able to cha nge the electrode surface from aerophobic to aerophilic state. The en riched nano-tunnels of the Cu nano array electrode can facilitate CO2 transportation and pin gaseous products on the electrode surface. The latter is believed to be the reason that boosts the Faradaic efficie ncy of liquid products by 67% and limits the H2 producti on to less than half of before. This in terface engin eeri ng strategy also lowered H2O (proton) affinity, therefore promoting CO and HCOOH production. | 18 | A novel selective autophagy receptor,CCDC50,delivers K63 polyubiquitination-activated RIG-I/MDA5 for degradation during viral infection显示文摘Autophagy is a con served process that delivers cytosolic substa nces to the lysosome for degradatio n,but its direct role in the regulation of antiviral inn ate immu nity remains poorly un derstood.Here,through high-throughput screeni ng,we discovered that CCDC50 functions as a previously unknown autophagy receptor that negatively regulates the type I interferon(IFN)signaling pathway initiated by RIG-I-like receptors(RLRs),the sensors for RNA viruses.The expression of CCDC50 is enhanced by viral infection,and CCDC50 specifically recognizes K63-polyubiquitinated RLRs,thus delivering the activated RIG-I/MDA5 for autophagic degradation.The associatio n of CCDC50 with phagophore membrane protei n LC3 is con firmed by crystal structure analysis.In contrast to other known autophagic cargo receptors that associate with either the LIR-docking site(LDS)or the UIMdocking site(UDS)of LC3,CCDC50 can bind to both LDS and UDS,representing a new type of cargo receptor.In mouse models with RNA virus infection,CCDC50 deficiency reduces the autophagic degradation of RIG-I/MDA5 and promotes type I IFN responses,resulting in enhaneed viral resistance and improved survival rates.These results reveal a new link between autophagy and antiviral innate immune resp on ses and provide additional in sights into the regulatory mecha nisms of RLR-mediated antiviral signaling. | Panpan Hou Kongxiang Yang Penghui Jia Lan Liu Yuxin Lin Zibo Li Jun Li Shuliang Chen Shuting Guo Ji'An Pan Junyu Wu Hong Peng Weijie Zeng Chunmei Li Yingfang Liu Deyin Guo | 2021 | Cell Research2021,31,1: | 5 |
