1Hydrological and water cycle processes of inland river basins in the arid region of Northwest China显示文摘The increasing shortage in water resources is a key factor affecting sustainable socio-economic development in the arid region of Northwest China(ARNC). Water shortages also affect the stability of the region's oasis ecosystem. This paper summarizes the hydrological processes and water cycle of inland river basins in the ARNC, focusing on the following aspects: the spatial-temporal features of water resources(including air water vapor resources, runoff, and glacial meltwater) and their driving forces; the characteristics of streamflow composition in the inland river basins; the characteristics and main controlling factors of baseflow in the inland rivers; and anticipated future changes in hydrological processes and water resources. The results indicate that:(1) although the runoff in most inland rivers in the ARNC showed a significant increasing trend, both the glaciated area and glacial ice reserves have been reduced in the mountains;(2) snow melt and glacier melt are extremely important hydrological processes in the ARNC, especially in the Kunlun and Tianshan mountains;(3) baseflow in the inland rivers of the ARNC is the result of climate change and human activities, with the main driving factors being the reduction in forest area and the over-exploitation and utilization of groundwater in the river basins; and(4) the contradictions among water resources, ecology and economy will further increase in the future. 3RNA-binding protein LIN28B inhibits apoptosis through regulation of the AKT2/FOXO3A/BIM axis in ovarian cancer cells显示文摘LIN28B is an evolutionarily conserved RNA-binding protein that regulates mRNA translation and miRNA let-7 maturation in embryonic stem cells and developing tissues.Increasing evidence demonstrates that LIN28B is activated in cancer and serves as a critical oncogene.However,the underlying molecular mechanisms of LIN28B function in tumorigenesis are still largely unknown.Here we report that LIN28B was expressed in over half of the patients with epithelial ovarian cancer who were examined(n=584).Functional experiments demonstrated that LIN28B inhibited ovarian cancer cell apoptosis.Furthermore,we showed that the proapoptotic factor BIM played an essential role in the antiapoptotic function of LIN28B.RNA-IP microarray analysis suggested that LIN28B binds to mRNAs that are associated with the DNA damage pathway,such as AKT2,in ovarian cancer cells.By binding to AKT2 mRNA and enhancing its protein expression,LIN28B regulated FOXO3A protein phosphorylation and decreased the transcriptional level of BIM,which antagonized the antiapoptosis activity of LIN28B.Taken together,these results mechanistically linked LIN28B and the AKT2/FOXO3A/BIM axis to the apoptosis pathway.The findings may have important implications in the diagnosis and therapeutics of ovarian cancer.Xiaojuan Lin Jianfeng Shen Dan Peng Xinhong He Congjian Xu Xiaojun Chen Janos LTanyi Kathleen Montone Yi Fan Qihong Huang Lin Zhang Xiaomin Zhong 2018Signal Transduction and Targeted Therapy2018,3,1:8

13Non-viral targeted delivery system mediates transfection of thymidine kinase gene of herpes simplex virus into ovarian cancer cells:a comparison between one time and continuous mediation显示文摘Objective: To compare the transferring efficiency and killing effects of one time and continuous mediation with GE7,a non-viral targeted delivery system,in transfection of thymidine kinase gene of herpes simplex virus (HSV-tk) into ovarian cancer cells. Methods: GE7 was used to prepare recombinants with β-galactosidase (β-gal) and HSV1-tk; the re-combinants were then used to transfect human ovarian cancer line CaOV3 once and continuously. β-gal staining was used to compare the efficiencies of one time and continuous mediation with GE7 system. Ganciclovior (GCV) was introduced into HSV1-tk transfected ovarian cells. Through drawing the cell growth curve and flow cytometry,the killing effects of GCV on once and continuously GE7/HSV1-tk transfected cells were observed. Results: We found that the one time and continuous exogenous gene transfer efficiencies were about 80% and 85%,respectively. When 1μg/mL GCV was used to treat ovarian cell transfected with HSV1-tk gene,growth inhibiting rates of ovarian cells of one time and continuous transferring were 82% and 90%,respectively; their apoptosis indices were 15 and 30,respectively. Under same GCV concentration,continuous me-diation of GE7/pCMV-tk transfection into ovarian cancer cells had more significant inhibitory effect than one time mediation (P<0.05). Conclusion: Compared with one time mediation,continuous mediation of transfection with GE7 gene delivery system has higher efficiency. 14Alterations in immune cell heterogeneities in the brain of aged zebrafish using single-cell resolution显示文摘Immunocytes,including the microglia,are crucial in the neurodegenerative process in old people.However,the understanding regarding microglia heterogeneity and other involved immunocytes remains elusive.We analyzed 26,456 immunocytes from 12-and 26-month-old zebrafish brains at single-cell resolution.Microglia and T lymphocytes were detected in the brain at both time points.Two types of microglia were annotated,namely,ac+microglia and xr+microglia,which were clustered into subsets 1,2,3,4,5,and subsets 6,7,8,9,respectively.Diversified microglia predominated the adult brains and cooperated with T cells to perform the functions of immune response and neuronal nutrition.We validated the specific microglia markers.The novel transgenic lines,Tg(lgals3bpb:e GFP)and Tg(apoc1:e GFP),were created,which faithfully labeled ac+microglia and served as valuable labeling tools.However,the microglia population reduced while T cells of six subtypes intriguingly increased to serve as the primary immune cells in aged brains.Unlike in 12-month-old brains,T cells,together with microglia,exhibited a coordinated signature of inflammation in the 26-month-old brains.Our findings revealed the immunocytes atlas in aged zebrafish brains.It implied the involvement of microglia and T cells in the progression of neurodegeneration in aging.Jiangyong He Fangying Zhao Bingyue Chen Nianfei Cui Zhifan Li Jie Qin Lingfei Luo Congjian Zhao Li Li 2023Science China(Life Sciences)2023,66,6:0
