1Verteporfin-mediated on/off photoswitching functions synergistically to treat choroidal vascular diseases显示文摘Choroidal vascular diseases,such as age-related macular degeneration,are the leading cause of vision impairment and are characterized by pathological angiogenesis.Verteporfin-mediated photodynamic therapy is a current strategy that selectively occludes choroidal neovasculature.However,the clinically used large-dose systemic administration increases the risk of systemic adverse events,such as phototoxicity to superficial tissues.In this study,we developed an in situ verteporfin delivery system with a photoswitching synergistic function that disassembles in response to intraocular inflammatory enzymes.Under light-on conditions,verteporfin-mediated photodynamic therapy effectively occurs and this leads to vascular occlusion.Under light-off conditions,non-photoactive verteporfin negatively regulates vascular endothelial growth factor-induced angiogenesis as a yes-associated protein inhibitor.Taken together,our system serves as an intraocular verteporfin reservoir to improve the bioavailability of verteporfin by innovatively exploiting its photochemical and biological functions.This work provides a promising strategy with synergistic antiangiogenic effects for the treatment of choroidal vascular diseases.Yahan Ju Xiaochan Dai Zhimin Tang Zunzhen Ming Ni Ni Dongqing Zhu Jing Zhang Bo Ma Jiajing Wang Rui Huang Siyu Zhao Yan Pang Ping Gu 2022Bioactive Materials2022,7,8:0
2Oriented Generation of Novel Thermo-Sensitive Genic Male Sterile Lines with Improved Grain Shape and Outcrossing Rate in Early-Season Rice显示文摘Two-line hybrid rice with excellent quality is preferred in the Chinese market.However,there is a trade-off between reducing costs for hybrid seed production and lowering the outcrossing rate of the sterile line,which is largely determined by the stigma exsertion rate(SER).In this study,we constructed mutants of male sterility lines with improved grain length(GL)and SER in three elite early-season indica rice varieties through targeted manipulation of the TMS5 and GS3 genes using CRISPR/Cas9-mediated multiplex systems.We obtained a series of marker-free gs3 single mutants and gs3tms5 double mutants with significantly higher SER,longer grains,and increased 1000-grain weight compared with the wild type(WT).Importantly,the typically thermo-sensitive genic male sterile(TGMS)trait with a higher SER was observed in gs3tms5 mutants,and their F1 hybrids exhibited remarkable improvements in grain shape and yield-related traits.Our findings provided an efficient method to generate new valuable TGMS germplasm with improved SER through the mutagenesis of GS3 and TMS5 synergistically,and demonstrated that GS3 had pleiotropic effects on grain size,SER,and grain quality in early-season indica rice.ZHANG Huali CHEN Junyu LI Ruiqing WANG Huimei DAI Dongqing LIANG Minmin WU Mingyue MA Liangyong 2024Rice science2024,31,2:0
3A backbone parent contributes core genomic architecture to pedigree breeding of early-season indica rice显示文摘Due to the capacity to deliver favorable target traits to offspring via breeder selection,backbone parents carrying accumulated favorable agronomic traits have been used widely in breeding programs in crop species,such as rice,wheat,cotton,and maize(Zhou et al.,2016;Fradgley et al.,2019;Li et al.,2019;Ma et al.,2019;Han et al.,2020).It is estimated that 3656(~70%)of the major Chinese rice varieties released from the year 1950e2008 were found to be derived from as few as 35 backbone parents(Tang et al.,2012).Large-scale genome sequencing of diverse rice accessions has been emerged as a promising technology for the identification of key genomic regions or loci under selection during rice domestication(Huang et al.,2012;Wang et al.,2018)and genetic improvement(Xie et al.,2015).On the other hand,sequence information derived from breeding pedigrees could also help to unravel how favorable genomic regions were transmitted from parents to their offspring,as demonstrated by the researches on the pedigrees of the wellknown rice varieties Minghui 63 and Huanghuazhan(Zhou et al.,2016;Chen et al.,2017;Huang et al.,2018).These researches enhanced the understanding of the cumulative effects of beneficial alleles at limited loci in selected offspring varieties.A few examples include IPA1,Gn1a,GW5,GS3,and GS5 for high grain yield,Waxy for good grain eating quality,and Xa21 for high adversity adaptability.Junyu Chen Huali Zhang Shuhan Deng Huilong Du Zhuo Chen Yuhui Zhao Dongqing Dai Chengzhen Liang Ximing Li Chengzhi Liang Rui Zhang Liangyong Ma 2021Journal of Genetics and Genomics2021,48,11:0
