| 1 | ADAMTS-7 promotes vascular smooth muscle cells proliferation in vitro and in vivo显示文摘Vascular smooth muscle cell(VSMC) proliferation and migration are pivotal for the pathogenesis of atherosclerosis and post-angioplasty restenosis. We have recently reported that a disintegrin and metalloproteinase with thrombospondin motifs-7(ADAMTS-7), a novel metalloproteinase, contributes directly to neointima formation by mediating VSMC migration. However, whether ADAMTS-7 affects VSMC proliferation remains unclear. In this study, we found that luminal adenoviral delivery of ADAMTS-7 aggravated intimal hyperplasia 7 d after injury, paralleled by an increased percentage of PCNA-positive cells in both intima and media. In contrast, perivascular administration of ADAMTS-7 si RNA, but not scrambled si RNA to injured arteries attenuated intimal thickening at day 7, paralleled with reduced intimal VSMC replication, without alteration of VSMC proliferation in the media. In accordance, [3H]-thymidine incorporation assay in primary cultured rat VSMCs revealed an enhanced replication rate(by 61%) upon ADAMTS-7 overexpression and retarded proliferation(by 23%) upon ADAMTS-7 si RNA administration. Our data demonstrates that ADAMTS-7 promotes VSMC proliferation both in vitro and in vivo. | 2 | Two-Year Observation of Fossil Fuel Carbon Dioxide Spatial Distribution in Xi’an City显示文摘The need for atmospheric carbon dioxide(CO2)reduction in the context of global warming is widely acknowledged by the global scientific community.Fossil fuel CO2(CO2ff)emissions occur mainly in cities,and can be monitored directly with radiocarbon(14 C).In this research,annual plants[Setaria viridis(L.)Beauv.]were collected from 26 sites in 2013 and2014 in the central urban district of Xi’an City.The△14C content of the samples were analyzed using a 3 MV Accelerator Mass Spectrometer,and CO2ff concentrations were calculated based on mass balance equations.The results showed that the CO2ff mixing ratio ranged from 15.9 to 25.0 ppm(part per million,equivalent toμmol mol-1),with an average of 20.5 ppm in 2013.The range of measured values became larger in 2014,from 13.9 ppm to 33.1 ppm,with an average of 23.5 ppm.The differences among the average CO2ff concentrations between the central area and outer urban areas were not statistically significant.Although the year-to-year variation of the CO2ff concentration was significant(P<0.01),there was a distinctly low CO2 ff value observed in the northeast corner of the city.CO2 ff emiissions from vehicle exhaust and residential sources appeared to be more significant than two thermal power plants,according to our observed CO2 ff spatial distribution.The variation of pollution source transport recorded in our observations was likely controlled by southwesterly winds.These results could assist in the optimal placement of regional CO2 monitoring stations,and benefit the local government in the implementation of efficient carbon emission reduction measures. | Xiaohu XIONG Weijian ZHOU Shugang WU Peng CHENG Hua DU Yaoyao HOU Zhenchuan NIU Peng WANG Xuefeng LU Yunchong FU | 2020 | Advances in Atmospheric Sciences2020,37,6: | 2 |
| 3 | Melatonin shapes bacterial clearance function of porcine macrophages during enterotoxigenic Escherichia coli infection显示文摘Due to the immature gastrointestinal immune system,weaning piglets are highly susceptible to pathogens,e.g.,enterotoxigenic Escherichia coli(ETEC).Generally,pathogens activate the immune cells(e.g.,macrophages)and shape intracellular metabolism(including amino acid metabolism);nevertheless,the metabolic cues of tryptophan(especially melatonin pathway)in directing porcine macrophage function during ETEC infection remain unclear.Therefore,this study aimed to investigate the changes in the serotonin pathway of porcine macrophages during ETEC infection and the effect of melatonin on porcine macrophage functions.Porcine macrophages(3D4/21 cells)were infected with ETEC,and the change of serotonin pathway was analysed by reverse transcription PCR and metabolomic analysis.The effect of melatonin on porcine macrophage function was also studied with proteomic analysis.In order to investigate the effect of melatonin on bacterial clearance function of porcine macrophages during ETEC infection,methods such as bacterial counting,reverse transcription PCR and western blotting were used to detect the corresponding indicators.The results showed that ETEC infection blocked melatonin production in porcine macrophages(P<0.05)which is largely associated with the heat-stable enterotoxin b(STb)of ETEC(P<0.05).Interestingly,melatonin altered porcine macrophage functions,including bacteriostatic and bactericidal activities based on proteomic analysis.In addition,melatonin pretreatment significantly reduced extracellular lactate dehydrogenase(LDH)activity(P<0.05),indicating that melatonin also attenuated ETEC-triggered macrophage death.Moreover,melatonin pretreatment resulted in the decrease of viable ETEC in 3D4/21 cells(P<0.05),suggesting that melatonin enhances bacterial clearance of porcine macrophages.These results suggest that melatonin is particularly important in shaping porcine macrophage function during ETEC infection. | Lingfei Du Bingnan Liu Ziyi Han Yaoyao Xia Miaomiao Wu Shaojuan Liu | 2022 | Animal Nutrition2022,,4: | 2 |

| 5 | Evaluating Tislelizumab, Lenvatinib, and FOLFOX4-HAIC as a Conversion Therapy for Unresectable Hepatocellular Carcinoma显示文摘Background and aims:Conversion therapy downstages tumors and renders patients with unresectable hepatocel-lular carcinoma(HCC)eligible for radical resection.This study aimed to evaluate the efficacy and safety of tislelizumab plus lenvatinib and hepatic artery infusion chemotherapy with oxaliplatin,fluorouracil,and leuco-vorin(FOLFOX4-HAIC)as a first-line conversion therapy.Methods:Clinical data from HCC patients who were treated with the triple therapy between April 2021 and April 2022 were retrospectively analyzed.The primary outcome included objective response rate(ORR),disease control rate(DCR),conversion resection rate(CRR),and treatment-related adverse events(TRAEs).Results:A total of 18 patients completed conversion therapy assessment,which ended on March 27,2023.The patients had a median age of 55.5(37–72)years,and 94.4%were male.According to mRECIST,tumor shrinkage was observed in all patients,with an ORR of 94.4%(17/18),a DCR of 94.4%(17/18),and a median time to response of 1.4(0.7–3.0)months.Successful conversion was observed in 61.1%(11/18)of patients(mRECIST).The CRR and pathological complete response were 38.9%(7/18)and 57.1%(4/7),respectively.The median progression-free survival(PFS)was 17.8 months,while median overall survival was not reached.The 6-and 9-month PFS rates were 83.3%and 66.7%,respectively.The most common TRAE(16/18 patients,88.9%)was an increase in aspartate aminotransferase levels.Conclusion:Tislelizumab combined with lenvatinib and FOLFOX4-HAIC achieved a high conversion rate and acceptable toxicity in patients with unresectable HCC,suggesting that this combination may represent a new conversion strategy for this population. | Kai Tan Xiaojun He Haoran Yuan Shoujie Zhao Chenyu Gong Yaoyao Zhao Haiyan Nan Li Zang Zhonghua Luo Xilin Du | 2023 | iLIVER2023,2,3: | 0 |
| 6 | Functional analysis of zona pellucida domain protein Dusky in Tribolium castaneum显示文摘The zona pellucida domain protein Dusky(Dy)plays a vital role in wing morphogenesis in insects,but little information on its function has been reported.In this study,we found that dy regulated wing cell size,larval and pupal duration,and the metabolism of amino acid and 20-hydroxyecdysone in Tribolium castaneum.Using RNA-seq,413 differentially expressed genes were identified between physiological buffer-injected and dy-double-stranded RNA-treated larvae,including 88 downregulated genes and 325 upregulated genes.Among these genes,dy knockdown increased CYP18A1 expression to elevate the 26-hydroxylation of 20-hydroxyecdysone,which ultimately led to growth defects in wing cells.Silencing of dy upregulated the transcription of genes encoding tyrosine aminotransferase,4-hydroxyphenylpyruvate dioxygenase,homogentisate 1,2-dioxygenase,and Pale to promote the catabolism of tyrosine and phenylalanine,which eventually reduced amino acid content.Furthermore,dy knockdown upregulated 4E-BP expression,and 4E-BP silencing partially phenocopied dy RNA interference-mediated wing morphogenesis.These results suggest that Dy controls 20-hydroxyecdysone and amino acid metabolism to regulate wing morphogenesis in the insect. | Chengjun Li Liu Yang Youwei Wang Huanyu Du Jiangyan Zhang Yaoyao Lu Bin Li Keping Chen | 2022 | Insect Science2022,29,2: | 0 |

| 10 | A Real-Time Small Target Vehicle Detection Algorithm with an Improved YOLOv5m Network Model显示文摘To address the challenges of high complexity,poor real-time performance,and low detection rates for small target vehicles in existing vehicle object detection algorithms,this paper proposes a real-time lightweight architecture based on You Only Look Once(YOLO)v5m.Firstly,a lightweight upsampling operator called Content-Aware Reassembly of Features(CARAFE)is introduced in the feature fusion layer of the network to maximize the extraction of deep-level features for small target vehicles,reducing the missed detection rate and false detection rate.Secondly,a new prediction layer for tiny targets is added,and the feature fusion network is redesigned to enhance the detection capability for small targets.Finally,this paper applies L1 regularization to train the improved network,followed by pruning and fine-tuning operations to remove redundant channels,reducing computational and parameter complexity and enhancing the detection efficiency of the network.Training is conducted on the VisDrone2019-DET dataset.The experimental results show that the proposed algorithmreduces parameters and computation by 63.8% and 65.8%,respectively.The average detection accuracy improves by 5.15%,and the detection speed reaches 47 images per second,satisfying real-time requirements.Compared with existing approaches,including YOLOv5m and classical vehicle detection algorithms,our method achieves higher accuracy and faster speed for real-time detection of small target vehicles in edge computing. | Yaoyao Du Xiangkui Jiang | 2024 | Computers, Materials & Continua2024,78,1: | 0 |
