| 1 | Typical atmospheric haze during crop harvest season in northeastern China:A case in the Changchun region显示文摘This study presents the mass concentrations of PM_(2.5),O_3,SO_2 and NOxat one urban,one suburban and two rural locations in the Changchun region from September 25 to October 272013. Major chemical components of PM_(2.5)at the four sites were daily sampled and analyzed. Most of daily concentrations of SO_2(7–82 μg/m^3),O_3(27–171 μg/m^3) and NOx(14–213 μg/m^3) were below the limits of the National Ambient Air Quality Standard(NAAQS)in China. However,PM_(2.5)concentrations(143–168 μg/m^3) were 2-fold higher than NAAQS.Higher PM_(2.5)concentrations(~ 150 μg/m^3) were measured during the pre-harvest and harvest at the urban site,while PM_(2.5)concentrations significantly increased from 250 to400 μg m^(-3) at suburban and rural sites with widespread biomass burning. At all sites,PM_(2.5)components were dominated by organic carbon(OC) and followed by soluble component sulfate(SO_4^(2-)),ammonium(NH_4^+) and nitrate(NO_3^-). Compared with rural sites,urban site had a higher mineral contribution and lower potassium(K^+and K) contribution to PM_(2.5).Severe atmospheric haze events that occurred from October 21 to 23 were attributed to strong source emissions(e.g.,biomass burning) and unfavorable air diffusion conditions.Furthermore,coal burning originating from winter heating supply beginning on October 18 increased the atmospheric pollutant emissions. | 2 | Temporal variability of atmospheric particulate matter and chemical composition during a growing season at an agricultural site in northeastern China显示文摘This study presents the observations of PM_(10) and PM_(2.5) concentrations at an agricultural site from April to October 2012 in Dehui city,China.Ambient air was sampled by filter-based samplers and online PM monitors.The filter samples were analyzed to determine the abundance of ionic/inorganic elements,organic carbon(OC) and elemental carbon(EC).The daily PM_(10) concentrations varied significantly over the monitoring period,with an average of168 ± 63(in the range of 52-277) μg/m^3 during the land preparation/planting period(26 April-15 June),85 ± 65(36-228) μg/m^3 during the growing season(16 June-25 September),and 207 ±88(103-310) μg/m^3 during the harvest period(26 September-31 October).PM_(2.5) accounted for44%,56%and 66%of atmospheric PM_(10) during these periods,respectively.The PM_(10) diurnal variation showed a distinct peak from 16:00 to 21:00(LST) during the growing and harvesting seasons,while a gradual increase throughout the daytime until 17:00 was observed during tilling season.Mineral dust elements(Al,Ca,Fe,and Mg) dominated the PM_(10) chemical composition during the tilling season;OC,NO_3^-,SO_4^(2-) and NH_4^+ during the growing season;and carbonaceous species(i.e.,OC and EC) during the harvesting season.Our results indicate that the soil particles emitted by farm tillage and organic matter released from straw burning are the two most significant sources of PM_(10) emissions contributing to the recurring high pollution events in this region.Therefore,development of agricultural PM inventories from soil tillage and straw burning is prioritized to support air quality modeling. | Weiwei Chen Daniel Tong Shichun Zhang Mo Dan Xuelei Zhang Hongmei Zhao | 2015 | Journal of Environmental Sciences2015,27,12: | 7 |

| 9 | Acquisition of taxane resistance by p53 inactivation in ovarian cancer cells显示文摘Ovarian cancer is one of the most common gynecologic malignancies in women and has a poor prognosis.Taxanes are a class of standard first-line chemotherapeutic agents for the treatment of ovarian cancer.However,tumor-intrinsic and acquired resistance to taxanes poses major challenges to improving clinical outcomes.Hence,there is an urgent clinical need to understand the mechanisms of resistance in order to discover potential biomarkers and therapeutic strategies to increase taxane sensitivity in ovarian cancer.Here,we report the identification of an association between the TP53 status and taxane sensitivity in ovarian cancer cells through complementary experimental and informatics approaches.We found that TP53 inactivation is associated with taxane resistance in ovarian cancer cells,supported by the evidence from(i)drug sensitivity profiling with bioinformatic analysis of large-scale cancer therapeutic response and genomic datasets and(ii)gene signature identification based on experimental isogenic cell line models.Further,our studies revealed TP53-dependent gene expression patterns,such as overexpression of ACSM3,as potential predictive biomarkers of taxane resistance in ovarian cancer.The TP53-dependent hyperactivation of the WNT/β-catenin pathway discovered herein revealed a potential vulnerability to exploit in developing combination therapeutic strategies.Identification of this genotype-phenotype relationship between the TP53 status and taxane sensitivity sheds light on TP53-directed patient stratification and therapeutic discoveries for ovarian cancer treatment. | Changfa Shu Xi Zheng Alafate Wuhafu Danielle Cicka Sean Doyle Qiankun Niu Dacheng Fan Kun Qian Andrey AIvanov Yuhong Du Xiulei Mo Haian Fu | 2022 | Acta Pharmacologica Sinica2022,43,9: | 1 |

| 16 | International study of the Complex Stress Reaction Syndrome:Implications for transdiagnostic clinical practice显示文摘BACKGROUND The debate regarding diagnostic classification systems in psychiatry(categorial vs dimensional systems)has essential implications for the diagnosis,prevention and treatment of stress reactions.We previously found a unique pattern of stress reaction in a study executed during the coronavirus disease 2019 pandemic using large representative samples in two countries,and termed it the Complex Stress Reaction Syndrome(CSRS).AIM To investigate CSRS,Type A(psychiatric symptoms,spanning anxiety,depression,stress symptoms,and posttraumatic stress disorder(PTSD)),with or without long-coronavirus disease(COVID)residuals(CSRS,Type B,neuropsychiatric symptoms spanning cognitive deficits and fatigue,excluding systemic symptoms).Our two-tailed hypothesis was that CSRS is a condition related to an unrecognized type of stress reaction in daily life in the general population(Type A)or that it is related to the severe acute respiratory syndrome coronavirus 2 infection and its long-COVID residuals(Type B).METHODS 977 individuals in four continents(North America,Europe,Australia and the Middle East)completed the online study questionnaire in six languages using the Qualtrics platform.The study was managed by six teams in six countries that promoted the study on social media.The questionnaire assessed anxiety,depression,stress symptoms and PTSD(CSRS,Type A),cognitive deficits and fatigue(CSRS,Type B).The data were analyzed using Proportion Analyses,Multivariate Analysis of Co-Variance(MANCOVA),linear regression analyses and validated clinical cutoff points.RESULTS The results of the Proportion Analyses showed that the prevalence of 4 symptoms spanning anxiety,depression,stress symptoms,and PTSD was significantly higher than the most prevalent combinations of fewer symptoms across 4 continents,age groups,and gender.This supports the transdiagnostic argument embedded in the CSRS(Type A).The same pattern of results was found in infected/recovered individuals.The prevalence of the 4 psychiatric symptoms combination was significantly greater than that of 5 and 6 symptoms,when adding cognitive deficits and fatigue,respectively.MANCOVA showed a significant three-way interaction(age×gender×continent).Further analyses showed that the sources of this three-way interaction were threefold relating to two sub-populations at-risk:(1)Individuals that self-identified as non-binary gender scored significantly higher on all 4 psychiatric symptoms of the CSRS,Type A at young age groups(<50 years old)in North America compared to(self-identified)women and men located in the 4 continents studied,and to other ages across the adult life span;and(2)This pattern of results(CSRS,Type A)was found also in women at young ages(<40 years old)in North America who scored higher compared to men and women in other continents and other ages.Linear regression analyses confirmed the MANCOVA results.CONCLUSION These results show a combined mental health risk factor related to stress reactivity,suggesting that the CSRS is sensitive to populations at risk and may be applied to future identification of other vulnerable sub-populations.It also supports the transdiagnostic approach for more accurate prevention and treatment.Time will tell if such transdiagnostic syndromes will be part of the discussions on the next revisions of the traditional classification systems or whether the crisis in psychiatry further evolves. | Sari Goldstein Ferber Aron Weller Adele M Hayes Tracy D Vannorsdall Yaroup Ajlouni Mo'nes Qudah Gil Zalsman Gal Shoval Tommaso Benedetto Jannini Racquel Fiedler Lily X Chen Danielle R Shayani Elin Kachuki Dory Dana Stolowicz-Melman Connor Evans Megan Trow Giorgio Di Lorenzo Rodolfo Rossi | 2023 | World Journal of Psychiatry2023,13,10: | 0 |
| 17 | 依据清创术后细菌培养结果确定开放骨折创面关闭时机显示文摘背景:目前,对开放骨折创面缝合关闭的时机尚没有客观的标准。许多学者对于开放骨折创面缝合关闭最佳时机提出了各种各样的建议,但均依据主观评判的参数。本研究的目的是应用前瞻性方法,在四肢开放骨折清创和冲洗木后,依据创面细菌培养结果来指导创面的缝合关闭时机。方法:本组对422例急诊开放骨折患者,进行了创面清创冲洗、骨折固定和开放创面处理。清创术后创面采样进行厌氧菌和有氧菌培养。清创术后48h,患者再次手术。如果初次细菌培养结果为阳性,则再次进行冲洗和清创术,之后再次留取菌培样本。创面不关闭直到细菌培养结果为阴性为止,否则重复进行冲洗和清创术。结果:在422例开放骨折中,有346例获得了长期随访。全组创面深部感染率为4|3%。GustiloⅡ型开放骨折创面深部感染率为4%,GustiloⅢ型开放骨折为5.7%。在GustiloⅢ型开放骨折中,依骨折类型不同,感染率出现变化,GustiloⅢA型开放骨折为1.8%,GustiloⅢB型开放骨折为10.6%,GustiloⅢC型开放骨折为20%。需要反复清创的骨折患者,伴有糖尿病患者以及体重指数增加的患者,感染率均出现上升。研究数据表明,开放骨折在菌培阳性条件下关闭创面(违反本研究原则)并未显著增加深部感染风险(p=0.0501)。结论:采用本文提出的标准处理方法,与历史对照方法相比,开放骨折深部感染率降到很低。但取得这样结果是以增加冲洗和清创术次数为代价。可信水平:治疗性研究Ⅳ级。关于证据等级的完整描述详见投稿须知。 | Christopher J. | 18 | Reducing children’s exposure to di(2-ethylhexyl)phthalate in homes and kindergartens in China:Impact on lifetime cancer risks and burden of disease显示文摘Exposure to di(2-ethylhexyl)phthalate(DEHP)in the indoor environment has been linked with significant health risks for Chinese children.Multi-phase DEHP concentrations in Chinese residences and kindergartens were estimated using a mass balance model based on the current baseline condition and control strategies(i.e.,increasing ventilation rate,reducing area of sources,using mechanical ventilation systems,and using portable air cleaners).The health benefits of each control strategy were quantified as the reduction in lifetime cancer risks(LCR)and burden of disease(BoD).In the current situation,the mean LCR and disability-adjusted life years(DALY)number attributable to indoor DEHP exposure for Chinese children were around 6.0×10^(−6) and 155 thousand,respectively.The mean LCR and DALY might be reduced by 25%-54%and 16%-40%,respectively,by increasing air exchange rates by 100%,reducing the use of source materials by two-thirds or deploying commercial air cleaners in naturally ventilated buildings.Meanwhile,avoidable DALYs could result in a reduction of mean economic losses of 2.2-5.3 billion RMB.Mechanical ventilation systems with filtration units may not be helpful for reducing children’s health risks.House-specific and tailor-made control measures are critical in lowering indoor exposure to DEHP to promote sustainable buildings and children’s health in China. | Dongsheng Tao Wen Sun Donghui Mo Yonghui Lin Wei Lv Daniel Mmereki Yousheng Xu Yanghui Ye Yuanjun Tang Chao Ye Cong Dong Zhongming Bu | 2024 | Building Simulation2024,17,3: | 0 |
| 19 | TMPRSS2 and SARS-CoV-2 SPIKE interaction assay for uHTS显示文摘SARS-CoV-2,the coronavirus that causes the disease COVID-19,has claimed millions of lives over the past 2 years.This demands rapid development of effective therapeutic agents that target various phases of the viral replication cycle.The interaction between host transmembrane serine protease 2(TMPRSS2)and viral SPIKE protein is an important initial step in SARS-CoV-2 infection,offering an opportunity for therapeutic development of viral entry inhibitors.Here,we report the development of a time-resolved fluorescence/Förster resonance energy transfer(TR-FRET)assay for monitoring the TMPRSS2–SPIKE interaction in lysate from cells co-expressing these proteins.The assay was configured in a 384-well-plate format for high-throughput screening with robust assay performance.To enable large-scale compound screening,we further miniaturized the assay into 1536-well ultrahigh-throughput screening(uHTS)format.A pilot screen demonstrated the utilization of the assay for uHTS.Our optimized TR-FRET uHTS assay provides an enabling platform for expanded screening campaigns to discover new classes of small-molecule inhibitors that target the SPIKE and TMPRSS2 protein–protein interaction. | Danielle Cicka Qiankun Niu Min Qui Kun Qian Eric Miller Dacheng Fan Xiulei Mo Andrey AIvanov Stefan GSarafianos Yuhong Du Haian Fu | 2023 | Journal of Molecular Cell Biology2023,15,3: | 0 |
