| 1 | Status and distribution changes of the endangered Green Peafowl (Pavo muticus) in China over the past three decades (1990s-2017)显示文摘Background: Understanding the changes in population dynamics, including demographics, distribution and threats, is essential for species status assessing. The endangered Green Peafowl(Pavo muticus) has experienced sharp population declines and distribution range diminishing both in China and Southeast Asia. Field population surveys have not been conducted in China since the 1990 s, which hindered conservation planning and decision-making.Methods: With interview and line transects methods, we figured out the population and distribution changes of Green Peafowl across its historical ranges over the past three decades in China during 2014-2017.Results: The Green Peafowl once habituated in 54 counties in China. Nearly 60% of the distribution counties were lost in the past three decades, with the left 22 counties distributed in central, southern and western Yunnan, SW China. Population decrease detected in all distribution areas except for Shuangbai and Xinping county where more than 60% of the total population is located. Only about 30% of the former bird population were recorded with the same interviewing method as 20 years ago. Three birds, 1 carcass, 6 calls and 12 footprints were detected along the 865 km line transects, indic-ating extremely low encounter rate of Green Peafowl in field. Sharp decreases in flock sizes were also detected, from 820 birds per flock in the 1990 s to 3-5 birds at present. Poaching and habitat conversion are two widespread and long-lasting threats, while poisoning caused mortality in the past and hydropower construction affect regional population's survival. Large flocks of 18–27 birds were discovered in the field, which increases our confidence of population recovery of this endangered pheasant in China.Conclusions: Only interviewed bird number and counts based on line transects were presented in this study, without further population estimation due to limitation of the data sets. | 3 | A mimotope of Pre-S_2 region of surface antigen of viral hepatitis B screened by phage display显示文摘To acquire the phage-displayed mimotopes which mimic the specificity of hepatitis B virus surface antigen (HBsAg), a random peptide library expressing linear peptide with 12 amino asids in length were used to screen with the serum from a hepatitis B virus infected patient in the recovery phase. After 3 rounds of biopanning, the positive phages were confirmed by competitive ELISA using HBsAg/P33. Two phagotopes were identified and one of them was confirmed as mimotope by competition experiment. Based on the mimotpe, a multiple antigenic peptide with four branches was synthesized by solid phase peptide synthesis. The antiginicity and specificity of the synthesized antigen was tested in BALB/c mice compared with the native epitope-based antigen. The results showed that the mimotope-based antigen could evoke higher titer of antibodies with the same specificity of the epitope-based antigen. | 4 | The double face of miR-320:cardiomyocytes-derived miR-320 deteriorated while fibroblasts-derived miR-320 protected against heart failure induced by transverse aortic constriction显示文摘MicroRNAs(miRNAs)are aberrantly expressed in the pathophysiologic process of heart failure(HF).However,the functions of a certain miRNA in different cardiac cell types during HF are scarcely reported,which might be covered by the globe effects of it on the heart.In the current study,Langendorff system was applied to isolate card io myocytes(CMs)and cardiac fibroblasts(CFs)from transverse aortic constriction(TAC)-induced mice.Slight increase of miR-320 expression was observed in the whole heart tissue of TAC mice.Interestingly,miR-320 was significantly elevated in CMs but decreased in CFs from TAC mice at different time points.Then,recombinant adeno-associated virus 9 with cell-type-specific promoters were used to manipulate miR-320 expressions in vivo.Both in vitro and in vivo experiments showed the miR-320 overexpression in CMs exacerbated cardiac dysfunction,whereas overexpression of miR-320 in CFs alleviated cardiac fibrosis and hypertrophy.Mechanically,downstream signaling pathway analyses revealed that miR-320 might induce various effects via targeting PLEKHM3 and IFITM1 in CMs and CFs,respectively.Moreover,miR-320 mediated effects could be abolished by PLEKHM3 re-expression in CMs or IFITM1 re-expression in CFs.Interestingly,miR-320 treated CFs were able to indirectly affect CMs function,but not vice versa.Meanwhile,upstream signaling pathway analyses showed that miR-320 expression and decay rate were rigorously manipulated by Ago2,which was regulated by a cluster of cell-type-specific TFs distinctively expressed in CMs and CFs,respectively.Together,we demonstrated that miR-320 functioned differently in various cell types of the heart during the progression of HF. | Xudong Zhang Shuai Yuan Huaping Li Jiabing Zhan Feng Wang Jiahui Fan Xiang Nie Yan Wang Zheng Wen Yanghui Chen Chen Chen Dao Wen Wang | 2021 | Signal Transduction and Targeted Therapy2021，6，3： | 4 |
| 5 | Long-term modifications of blood pressure in normotensive and spontane-ously hypertensive rats by gene delivery of rAAV-mediated cytochrome P450 arachidonic acid hydroxylase显示文摘Arachidonic acid cytochrome P-450 (CYP) hydroxylase 4A isoforms, including 4A1, 4A2, 4A3 and 4A8 in the rat kidney, catalyze arachidonic acid to produce 19/20-Hydroxyeicosatetraenoic acids (20-HETE), a biologically active metabolite, which plays an important role in the regulation of blood pressure. However, controversial results have been reported regarding the exact role of 20-HETE on blood pressure. In the present study, we used recombinant adeno- associated viral vector (rAAV) to deliver CYP 4A1 cDNA and antisense 4A1 cDNA into Sprague-Dawley (SD) rats and spontaneously hypertensive rats (SHR), respectively, to investigate the effects of long-term modifications of blood pressure and the potential for gene therapy of hypertension. The mean systolic pressure increased by 14.2±2.5 mm Hg in rAAV·4A1-treated SD rats and decreased by 13.7±2.2 mm Hg in rAAV·anti4A1-treated SHR rats 5 weeks after the injection compared with controls and these changes in blood pressure were maintained until the experiments ended at 24 weeks. In 4A1 treated animals CYP4A was overexpressed in various tissues, but preferentially in the kidney at both mRNA and protein levels. In anti-4A1-treated SHR, CYP4A mRNA in various tissues was probed, especially in kidneys, but 4A1 protein expression was almost completely inhibited. | 6 | A method for computing unsupported roof distance in roadway advancement and its in-situ application显示文摘A reasonable unsupported roof distance(URD) when advancing underground coal mine roadways can contribute greatly to safe and rapid roadway development.A mechanical model of the roof,using the relationship between the roof stress distribution and URD,obtained by the difference method,and roof stability according to the in-situ roof stress and rock mass strength was developed.We subsequently designed a proper range of URD,developed a testing method of URD with the function of mining protection,evaluated roof stability through analyzing the test data and then determined a reasonable URD.Considering the factors of the geological conditions,the immediate roof stability and the efficiency of the labor arrangement system,the URD of the advancing roadway of 9802 working face in Zhangshuanglou coal mine was determined to be 6 m using the proposed method.The results show that,when a 2 m length of roadway was reinforced by temporary support and high pre-stressed bolt support after the roadway advancement of 6 m per cycle,the speed and the security of the roadway development can be achieved and the advance rate can reach more than 400 m per month. | Chen Yong Bai Jianbiao Yan Shuai Hao Shengpeng Dao Viet Doan | 2016 | International Journal of Mining Science and Technology2016，26，4： | 2 |
| 7 | Effect of 1,25-Dihydroxyvitamin D_3 on Regulatory T Cells in Ovariectomized Mice显示文摘Objective To investigate the correlation between regulatory T(Treg) cells and postmenopausal osteoporosis and the antiosteoporotic effect of 1,25-dihydroxyvitamin D3 [1,25(OH)2D3] in relation to Treg cells. Methods Fifty female BALB/c mice were randomly divided into five groups: the basal control(BAS), Sham, ovariectomy(OVX), OVX+diethylstilbestrol(OVX+DES), and OVX+1,25(OH)2D3. Tibias were harvested and processed with decalcification for quantitative bone histomorphometry. Femurs were stained by immunohistochemistry to detect Foxp3 protein expression. Spleens were used to detect Treg and Foxp3 gene expression by flow cytometry and quantitative RT-PCR, respectively. Results In comparison with the Sham group, a significant decrease was found in the OVX group in such indices as trabecular bone volume/total tissue area(BV/TV), trabecular number(Tb.N) and trabecular thickness(Tb.Th). 1,25(OH)2D3 and DES partly prevented the decrease in BV/TV, Tb.N, Tb.Th in OVX mice. Treg cell number, Foxp3 mRNA expression in spleen and Foxp3 protein expression in femur significantly decreased in the OVX-treated group compared with those in the sham group. 1,25(OH)2D3 and DES significantly increased Treg cell number and Foxp3 expression. Treg cells and Foxp3 gene expression were related to bone histomorphometric parameters. Conclusion The decrease in Treg cell numbers is relevant to the postmenopausal osteoporosis.
