| 5 | MEF2C regulates osteoclastogenesis and pathologic bone resorption via c-FOS显示文摘Osteoporosis is a metabolic bone disease with dysregulated coupling between bone resorption and bone formation,which results in decreased bone mineral density.The MEF2C locus,which encodes the transcription factor MADS box transcription enhancer factor 2,polypeptide C(MEF2C),is strongly associated with adult osteoporosis and osteoporotic fractures.Although the role of MEF2C in bone and cartilage formation by osteoblasts,osteocytes,and chondrocytes has been studied,the role of MEF2C in osteoclasts,which mediate bone resorption,remains unclear.In this study,we identified MEF2C as a positive regulator of human and mouse osteoclast differentiation.While decreased MEF2C expression resulted in diminished osteoclastogenesis,ectopic expression of MEF2C enhanced osteoclast generation.Using transcriptomic and bioinformatic approaches,we found that MEF2C promotes the RANKL-mediated induction of the transcription factors c-FOS and NFATc1,which play a key role in osteoclastogenesis.Mechanistically,MEF2C binds to FOS regulatory regions to induce c-FOS expression,leading to the activation of NFATC1 and downstream osteoclastogenesis.Inducible deletion of Mef2c in mice resulted in increased bone mass under physiological conditions and protected mice from bone erosion by diminishing osteoclast formation in K/BxN serum induced arthritis,a murine model of inflammatory arthritis.Our findings reveal direct regulation of osteoclasts by MEF2C,thus adding osteoclasts as a cell type in which altered MEF2C expression or function can contribute to pathological bone remodeling. | Takayuki Fujii Koichi Murata Se-Hwan Mun Seyeon Bae Ye Ji Lee Tannia Pannellini Kyuho Kang David Oliver Kyung-Hyun Park-Min Lionel B.Ivashkiv | 2021 | Bone Research2021,9,1: | 4 |

| 7 | A microscale optical implant for continuous in vivo monitoring of intraocular pressure显示文摘Intraocular pressure(IOP)is a key clinical parameter in glaucoma management.However,despite the potential utility of daily measurements of IOP in the context of disease management,the necessary tools are currently lacking,and IOP is typically measured only a few times a year.Here we report on a microscale implantable sensor that could provide convenient,accurate,ondemand IOP monitoring in the home environment.When excited by broadband near-infrared(NIR)light from a tungsten bulb,the sensor’s optical cavity reflects a pressure-dependent resonance signature that can be converted to IOP.NIR light is minimally absorbed by tissue and is not perceived visually.The sensor’s nanodot-enhanced cavity allows for a 3–5 cm readout distance with an average accuracy of 0.29 mm Hg over the range of 0–40 mm Hg.Sensors were mounted onto intraocular lenses or silicone haptics and secured inside the anterior chamber in New Zealand white rabbits.Implanted sensors provided continuous in vivo tracking of short-term transient IOP elevations and provided continuous measurements of IOP for up to 4.5 months. | Jeong Oen Lee Haeri Park Juan Du Ashwin Balakrishna Oliver Chen David Sretavan Hyuck Choo | 2017 | Microsystems & Nanoengineering2017,3,1: | 4 |

TSI nano,TSI standard,Grimm,and MSP instruments were collocated at the Fresno Supersite and particle size distributions were measured continuously from August 18 through September 18,2005. For particles with diameters between 10 and 200 nm,differences among hourly-average ambient particle concentrations ranged from 0% between the TSI nano and Grimm in the 30-50 nm size range to 39% between the Grimm and MSP in the 10-30 nm size range. MSP concen-trations were 10-33% lower than those measured with the TSI standard for particles smaller than 200 nm. | 16 | Genetic and environmental control of fruit maturation,dry matter and firmness in apple(Malus×domestica Borkh.)显示文摘For any given genotype,the environment in which an apple is grown can influence the properties of the fruit considerably.While there has been extensive research on the mechanism of the genetic control of fruit quality traits,less effort has been made to investigate the way that these genetic mechanisms interact with the environment.To address this issue,we employed a large‘Royal Gala’בBraeburn’population of 572 seedlings replicated over sites in three climatically diverse apple-growing regions in New Zealand.Phenotyping for traits including fruit maturation timing,firmness and dry matter content was performed at each of these three sites for a single growing season(2011),and at two sites(Motueka and Hawke’s Bay)for two seasons(2009 and 2010).The phenotype data collected over 2 years at two sites enabled the detection of 190 quantitative trait loci(QTL)that controlled these traits regardless of year or growing location,as well as some chromosomal loci that influenced the traits in a single given environment or year.For those loci that were environmentally stable over three sites,there was an interdependency of fruit maturation date,dry matter content and storage potential within this population,with two regions on Linkage Groups(LGs)10 and 16 strongly contributing.If these loci were used in a marker-assisted selection programme to select for progeny bearing firmer fruit,this would have the unintentional consequence of selecting,high dry matter content,later maturing apples.In addition,a further 113 new QTLs with a smaller effect were identified,some of which were exhibited only in a single growing environment,demonstrating the underlying complexity of control of traits determining fruit quality,in addition to the need for being aware of environmental effects when developing new apple varieties. | David Chagne Daya Dayatilake Robert Diack Murray Oliver Hilary Ireland Amy Watson Susan E Gardiner Jason W Johnston Robert J Schaffer Stuart Tustin | 2014 | Horticulture Research2014,1,1: | 2 |
| 17 | Impacts of long-term land-use changes on herd size and mobility among pastoral households in Amboseli ecosystem,Kenya显示文摘Introduction:Long-term changes in rangeland ecosystems of the world have impacted on the livestock production,a key livelihood strategy in these areas.This paper presents perceptions of the pastoral community on the dynamics of pastoral livelihoods under three land-use types namely nomadic,semi-nomadic,and sedentary in Amboseli ecosystem located in southern Kenya.Structured questionnaire was used to collect data concerning household livestock herds,perceived livestock trends and their causes,importance of migration,sedentarization,and land subdivision,and were administered to respondents randomly selected from the three land-use types.Results:The study shows that the livestock herd size was higher in nomadic(117.1)and lower in sedentary(56.6)land-use site.The majority of the respondents in nomadic(79%),semi-nomadic(73%),and sedentary(64%)reported a declining trend in household herd size.The declines were mostly attributed to recurrent droughts and loss of grazing lands.Conclusions:Pastoralism remains an important livelihood strategy to majority of households in the study area;therefore,to achieve sustainable livelihood strategies,grazing lands should be maintained. | Kennedy Sakimba Kimiti David Western Judith Syombua Mbau Oliver Vivian Wasonga | 2018 | Ecological Processes2018,7,1: | 1 |
