| 14 | Population isolation shapes plant genetics, phenotype and germination in naturally patchy ecosystems显示文摘Aims Habitat connectivity is important in conservation since isolation can diminish the potential of a population for adaptation and increase its risk of extinction.However,conservation of naturally patchy ecosystems such as peatlands has mainly focused on preserving specific sites with exceptional characteristics,neglecting the poten-tial interconnectivity between patches.In order to better under-stand plant dynamics within a peatland network,we assessed the effect of population isolation on genetic distinctiveness,phenotypic variations and germination rates using the peatland-obligate white-fringed orchid(Platanthera blephariglottis).Methods Fifteen phenotypic traits were measured for 24 individuals per pop-ulation(20 distinct populations,Quebec,Canada)and germination rates of nearly 20000 seeds were assessed.Genetic distinctiveness was quantified for 26 populations using single nucleotide polymor-phism markers obtained via a pooled genotyping-by-sequencing approach.Geographic isolation was measured as the distance to the nearest population and as the number of populations occurring in concentric buffer zones(within a radius of 2,5 and 10 km)around the studied populations.Important Findings All phenotypic traits showed significant differences among popu-lations.Genetic results also indicated a pattern of isolation-by-distance,which suggests that seed and/or pollen exchange is restricted geographically.Finally,all phenotypic traits,as well as a reduced germination rate,were correlated with either geographic isolation or genetic distance.We conclude that geographic iso-lation likely restricts gene flow,which in turn may affect germi-nation.Consequently,it is imperative that conservation programs take into account the patchy nature of such ecosystems,rather than targeting a few specific sites with exceptional character for preservation. | Laurent De Vriendt Marc-AndréLemay Martine Jean Sébastien Renaut Stéphanie Pellerin Simon Joly François Belzile Monique Poulin | 2017 | Journal of Plant Ecology2017,10,4: | 0 |

Design: Prospective nonrandomized study conducted from November 1999 to November 2002. Setting: Eleven university or hospital dermatology departments in France. Patients: Sixty- four consecutive patients with newly diagnosed early- stage mycosis fungoides (stage IA, n=33; stage IB, n=26; stage IIA, n=5). Interventions: Patients were treated with twice- weekly applications of a 0.02% aqueous solution of mechlorethamine followed by an application of betamethasone cream during a 6- month period. Main OutcomeMeasures: The primary end point was the rate of complete response during the treatment. Secondary end points were mean delay to achieve complete response, rate of severe cutaneous reactions of intolerance, and rate of relapse after achieving complete response. Results: Thirty- seven patients (58% ) had a complete response after a mean± SD treatment duration of 3.6± 2.5 months: 20 (61% ) of 33 patients with stage IA disease, 15 (58% ) of 26 patients with stage IB disease, and 2 (40% ) of 5 patients with stage IIA disease. Eighteen patients (28% ) developed severe cutaneous reactions of intolerance that necessitated treatment discontinuation. Relapse was observed in 17 patients (46% )- after a mean± SD time of 7.7± 6.5 months. Conclusions: A regimen of twice- weekly applications of mechlorethamine and betamethasone cream is an effective treatment for early- stage mycosis fungoides. | 16 | Complete response in anaplastic lymphoma kinase–rearranged oncocytic thyroid cancer:A case report and review of literature显示文摘BACKGROUND Oncocytic carcinoma of the thyroid is a rare disease,characterized by a poor prognosis and low response rate to radioiodine therapy.Crizotinib is a specific anaplastic lymphoma kinase(ALK)inhibitor,which was initially developed in non-small cell lung cancer.Other solid tumors harboring a translocation in ALK have been described,such as renal carcinoma,thyroid,colorectal,ovarian cancers,and spitzoid melanoma.The research of ALK rearrangements in thyroid tumor is a promising therapeutic track,and treatments need to be explored.CASE SUMMARY We report the case of a 76-year-old woman with a history of multinodular goiter,who was hospitalized for impairment of her general condition.She was diagnosed with metastatic oncocytic thyroid cancer.Synchrone metastases were found:Multiple mediastinal lymphadenopathies,lytic bone lesions and bilateral mammary lumps.Fluorescence in situ hybridization analysis revealed an ALK rearrangement in 61%of cells.No other mutation was found.A tumor board discussion based on molecular characteristics of the tumor suggested initiating a daily treatment by crizotinib,a specific ALK inhibitor.A positron emission tomography scan performed 4 mo after the initiation of crizotinib showed a complete metabolic response.CONCLUSION This case highlights an unexpected efficacy of crizotinib in an ALK-rearranged thyroid tumor,and the need of further assessments. | Victoire de Salins Gokoulakrichenane Loganadane Charlotte Joly Mukedaisi Abulizi Maya Nourieh Hélène Boussion Yazid Belkacemi Christophe Tournigand Emmanuelle Kempf | 2020 | World Journal of Clinical Oncology2020,11,7: | 0 |
